| 1 | A method for the production and expedient screening of CRISPR/Cas9-mediated non-transgenic mutant plants显示文摘Developing CRISPR/Cas9-mediated non-transgenic mutants in asexually propagated perennial crop plants is challenging but highly desirable.Here,we report a highly useful method using an Agrobacterium-mediated transient CRISPR/Cas9 gene expression system to create non-transgenic mutant plants without the need for sexual segregation.We have also developed a rapid,cost-effective,and high-throughput mutant screening protocol based on Illumina sequencing followed by high-resolution melting(HRM)analysis.Using tetraploid tobacco as a model species and the phytoene desaturase(PDS)gene as a target,we successfully created and expediently identified mutant plants,which were verified as tetra-allelic mutants.We produced pds mutant shoots at a rate of 47.5%from tobacco leaf explants,without the use of antibiotic selection.Among these pds plants,17.2%were confirmed to be non-transgenic,for an overall non-transgenic mutation rate of 8.2%.Our method is reliable and effective in creating non-transgenic mutant plants without the need to segregate out transgenes through sexual reproduction.This method should be applicable to many economically important,heterozygous,perennial crop species that are more difficult to regenerate. | Longzheng Chen Wei Li Lorenzo Katin-Grazzini Jing Ding Xianbin Gu Yanjun Li Tingting Gu Ren Wang Xinchun Lin Ziniu Deng Richard J.McAvoy Frederick G.Gmitter Jr. Zhanao Deng Yunde Zhao Yi Li | 2018 | Horticulture Research2018,5,1: | 12 |

| 5 | Establishment of an Objective Standard for the Definition of Binary Tropical Cyclones in the Western North Pacific显示文摘To develop an objective standard for defining binary tropical cyclones(BTCs)in the western North Pacific(WNP),two best-track datasets,from the China Meteorological Administration and the Joint Typhoon Warning Center,were adopted for statistical analyses on two important characteristics of BTCs-two TCs approaching each other,and counterclockwise spinning.Based on the high consistency between the two datasets,we established an objective standard,which includes a main standard for defining BTCs and a secondary standard for identifying typical/atypical BTCs.The main standard includes two requirements:two coexisting TCs are a pair of BTCs if(i)the separation distance is≤1800 km,and(ii)this separation maintains for at least 12 h.Meanwhile,the secondary standard defines a typical BTC as one for which there is at least one observation when the two TCs approach each other and spin counterclockwise simultaneously.Under the standard,the ratio of typical BTCs increases as the BTC duration increases or the minimum distance between the two TCs decreases.Then,using the JTWC dataset,it was found that there are 505 pairs of BTCs during the period 1951−2014,including 328 typical BTCs and 177 atypical BTCs,accounting for 65.0%and 35.0%of the total,respectively.In addition,a study of two extreme phenomena-the maximum approaching speed and the maximum counterclockwise angular velocity in typical BTCs-shows that the configuration of the circulation conditions and the distribution of the BTCs favor the formation of these extreme phenomena. | Fumin REN Yanjun XIE Biwen YIN Mingyang WANG Guoping LI | 2020 | Advances in Atmospheric Sciences2020,37,11: | 3 |
| 6 | Identification of genes for drought resistance and prediction of gene candidates in soybean seedlings based on linkage and association mapping显示文摘Drought is one of the primary abiotic stress factors affecting the yield,growth,and development of soybeans.In extreme cases,drought can reduce yield by more than 50%.The seedling stage is an important determinant of soybean growth:the number and vigor of seedlings will affect growth and yield at harvest.Therefore,it is important to study the drought resistance of soybean seedlings.In this study,a recombinant inbred line(RIL)population comprising 234 F_(6:10)lines(derived from Zhonghuang35×Jindou 21)and a panel of 259 soybean accessions was subjected to drought conditions to identify the effects on phenotypic traits under these conditions.Using a genetic map constructed by single nucleotide polymorphism(SNPs)markers,18 quantitative trait loci(QTL)on 7 soybean chromosomes were identified in two environments.This included 9 QTL clusters identified in the RIL population.Fifty-three QTL were identified in 19 soybean chromosomes by genome-wide association analysis(GWAS)in the panel of accessions,including 69 significant SNPs(-log_(10)(P)≥3.97).A combination of the two populations revealed that two SNPs(-log_(10)(P)≥3.0)fell within two of the QTL(qPH7-4 and qPH7-6)confidence intervals.We not only re-located several previously reported drought-resistance genes in soybean and other crops but also identified several non-synonymous stress-related mutation site differences between the two parents,involving Glyma.07 g093000,Glyma.07 g093200,Glyma.07 g094100 and Glyma.07 g094200.One previously unreported new gene related to drought stress,Glyma.07 g094200,was found by regional association analysis.The significant SNP CHR7-17619(G/T)was within an exon of the Glyma.07 g094200 gene.In the RIL population,the DSP value of the'T'allele of CHR7-17619 was significantly(P<0.05)larger than the'G'allele in different environments.The results of our study lay the groundwork for cloning and molecular marker-assisted selection of droughtresistance genes in soybeans at the seedling stage. | Yanjun Zhang Zhangxiong Liu Xingrong Wang Yue Li Yongsheng Li Zuowang Gou Xingzhen Zhao Huilong Hong Honglei Ren Xusheng Qi Lijuan Qiu | 2022 | The Crop Journal2022,10,3: | 3 |

| 9 | Smoke suppressant in flame retarded thermoplastic polyurethane composites: Synergistic effect and mechanism study显示文摘Considerable smoke and toxic volatiles generation has compromised the application of thermoplastic polyurethane (TPU) and caused a great threat to human life. Here, nano-MgFe layered double hydroxide (MgFe-LDH) with uniform particle size was synthesized to reduce smoke density and toxic gases of TPU composites using ammonium polyphosphate (APP) as a flame retardant agent. The results show that the combination of 16 wt.% APP and 4 wt.% MgFe-LDH greatly decreased the smoke density (D20min and Ds, max), smoke production rate (SPR) and heat release rate (HRR) of TPU composites. Furthermore, the MgFe-LDH synergist demonstrated high efficiency in decreasing total volatiled products and toxic volatiles evolved, such as the CO, HCN and isocyanates. The reason was mainly attributed to the chemical reaction between MgFe-LDH and APP, which can promote the compactness of char layers with fine microstructure formed in the decomposition process of MgFe-LDH/APP/TPU composites. | 10 | Design of Full-Ocean-Depth Self-Floating Sampler and Analysis of Factors Affecting Core Penetration Depth显示文摘The hadal zone(ocean depths of 6 – 11 km) is one of the least-understood habitats on Earth because of its extreme conditions such as high pressure, darkness, and low temperature. With the development of deep-sea vehicles such as China's 7000 m manned submersible Jiaolong, abyssal science has received greater attention. For decades, gravity-piston corers have been widely used to collect loose subsea-sediment long-core samples. However, the weight and length of the gravity sampler cables and the operating environment limit sampling capacity at full ocean depths. Therefore, a new self-floating sediment sampler with a spring-loaded auto-trigger release and that incorporates characteristics from traditional gravity-driven samplers is designed. This study analyzes the process by which a gravity-piston corer penetrates the sediment and the factors that affect it. A formula for obtaining the penetration depth is deduced. A method of optimizing the sampling depth is then developed based on structure design and parametric factor modeling. The parameters considered in the modeling include the sampling depth, balance weight, ultimate stress friction coefficient, dimensions of the sampler, and material properties. Thus, a new deep-sea floating parametric sampler designed based on virtual prototyping is proposed. Accurate values for all the design factors are derived from calculations based on the conservation of energy with penetration depth, analyses of the factors affecting the penetration depth, and analyses of the pressure bar stability. | 15 | The impact of demographic dynamics on food consumption and its environmental outcomes:Evidence from China显示文摘With increasing population and changing demographics,food consumption has experienced a significant transition in quantity and quality.However,a dearth of knowledge remains regarding its environmental impacts and how it responds to demographic dynamics,particularly in emerging economies like China.Using the two-stage Quadratic Almost Demand System(QUAIDS)model,this study empirically examines the impact of demographic dynamics on food consumption and its environmental outcomes based on the provincial data from 2000 to 2020 in China.Under various scenarios,according to changes in demographics,we extend our analysis to project the long-term trend of food consumption and its environmental impacts,including greenhouse gas(GHG)emissions,water footprint(WF),and land appropriation(LA).The results reveal that an increase in the proportion of senior people significantly decreases the consumption of grain and livestock meat and increases the consumption of poultry,egg,and aquatic products,particularly for urban residents.Moreover,an increase in the proportion of males in the population leads to higher consumption of poultry and aquatic products.Correspondingly,in the current scenario of an increased aging population and sex ratio,it is anticipated that GHG emissions,WF,and LA are likely to decrease by 1.37,2.52,and 3.56%,respectively.More importantly,in the scenario adhering to the standards of nutritional intake according to the Dietary Guidelines for Chinese Residents in 2022,GHG emissions,WF,and LA in urban areas would increase by 12.78,20.94,and 18.32%,respectively.Our findings suggest that changing demographics should be considered when designing policies to mitigate the diet-environment-health trilemma and achieve sustainable food consumption. | Shaoting Li Xuan Chen Yanjun Ren Thomas Glauben | 2024 | Journal of Integrative Agriculture2024,23,2: | 1 |

| 18 | Do credit constraints affect households'economic vulnerability?Empirical evidence from rural China显示文摘Poverty alleviation is still one of the major challenges in developing countries,especially in transitional economy like China.From the perspective of anti-poverty,this paper examines the impact of formal credit constraints(FCCs)and informal credit constraints(IFCCs)on economic vulnerability(EV)using the data from the China Household Income Project(CHIP)survey for 2013(CHIPs 2013)of rural households.The potential endogeneity problem of credit constraints(CCs)is addressed by applying the control function approach within an ordered probit model.The results show that both FCCs and IFCCs have a robust positive and significant impact on the EV of rural households and that the impact of FCCs is greater than that of IFCCs.To identify the potential mechanisms through which CCs affect EV,the seemingly unrelated regressions are used and the potential intercorrelation among these mechanisms is examined.We find that the impact of CCs on EV is partly mediated by health,trust,per capita financial assets and per capita income,whereby health and per capita income contribute to most of the total indirect effect.Thus,policies focus on supply-side and demand-side to improve credit accessibility could reduce rural households'EV,especially through its positive effect on health and per capita income. | PENG Yan-ling Yanjun REN LI Hou-jian | 2021 | Journal of Integrative Agriculture2021,20,9: | 0 |
| 19 | Spatiotemporal characteristics of cultivated land use eco-efficiency and its influencing factors in China from 2000 to 2020显示文摘Improving cultivated land use eco-efficiency(CLUE)can effectively promote agricultural sustainability,particularly in developing countries where CLUE is generally low.This study used provincial-level data from China to evaluate the spatiotemporal evolution of CLUE from 2000 to 2020 and identified the influencing factors of CLUE by using a panel Tobit model.In addition,given the undesirable outputs of agricultural production,we incorporated carbon emissions and nonpoint source pollution into the global benchmark-undesirable output-super efficiency-slacks-based measure(GB-US-SBM)model,which combines global benchmark technology,undesirable output,super efficiency,and slacks-based measure.The results indicated that there was an upward trend in CLUE in China from 2000 to 2020,with an increase rate of 2.62%.The temporal evolution of CLUE in China could be classified into three distinct stages:a period of fluctuating decrease(2000-2007),a phase of gradual increase(2008-2014),and a period of rapid growth(2015-2020).The major grain-producing areas(MPAs)had a lower CLUE than their counterparts,namely,non-major grain-production areas(non-MPAs).The spatial agglomeration effect followed a northeast-southwest strip distribution;and the movement path of barycentre revealed a'P'shape,with Luoyang City,Henan Province,as the centre.In terms of influencing factors of CLUE,investment in science and technology played the most vital role in improving CLUE,while irrigation index had the most negative effect.It should be noted that these two influencing factors had different impacts on MPAs and non-MPAs.Therefore,relevant departments should formulate policies to enhance the level of science and technology,improve irrigation condition,and promote sustainable utilization of cultivated land. | LI Shaoting MU Na REN Yanjun Thomas GLAUBEN | 2024 | Journal of Arid Land2024,16,3: | 0 |
| 20 | Enrichment of health-promoting lutein and zeaxanthin in tomato fruit through metabolic engineering显示文摘Carotenoids constitute a large group of natural pigments widely distributed in nature.These compounds not only provide fruits and flowers with distinctive colors,but also have significant health benefits for humans.Lutein and zeaxanthin,both oxygen-containing carotenoids,are considered to play vital roles in promoting ocular development and maintaining eye health.However,humans and mammals cannot synthesize these carotenoid derivatives,which can only be taken from certain fruits or vegetables.Here,by introducing four endogenous synthetic genes,SlLCYE,SlLCYB,SlHYDB,and SlHYDE under fruit-specific promoters,we report the metabolic engineering of lutein/zeaxanthin biosynthesis in tomato fruit.Transgenic lines overexpression of one(SlLCYE),two(SlLCYE and SlLCYB;SlLCYB and SlHYDB),and all these four synthetic genes re-established the lutein/zeaxanthin biosynthetic pathways in the ripe tomato fruit and thus resulted in various types of carotenoid riched lines.Metabolic analyses of these engineered tomato fruits showed the strategy involved expression of SlLCYE tends to produceα-carotene and lutein,as well as a higher content of β-carotene and zeaxanthin was detected in lines overexpressing SlLCYB.In addition,the different combinations of engineered tomatoes with riched carotenoids showed higher antioxidant capacity and were associated with a significantly extended shelf life during postharvest storage.This work provides a successful example of accurate metabolic engineering in tomato fruit,suggesting the potential utility for synthetic biology to improve agronomic traits in crops.These biofortified tomato fruits could be also exploited as new research subjects for studying the health benefits of carotenoid derivatives. | Yanjun Wu Yong Yuan Wenqian Jiang Xin Zhang Siyan Ren Hsihua Wang Xiaolan Zhang Yang Zhang | 2022 | Synthetic and Systems Biotechnology2022,7,4: | 0 |
