2HDAC6 inhibition regulates substrate stiffness-mediated inflammation signaling in chondrocytes显示文摘Osteoarthritis(OA)is a chronic disease and is difficult to cure.Chondrocytes are highly mechanosensitive.Therefore,mechanical therapies have received attention as a therapeutic direction for OA.The stiffness,as a critical cue of the extracellular matrix(ECM),affects cell growth,development,and death.In this study,we use polydimethylsiloxane(PDMS)to create substrates with varying stiffness for chondrocyte growth,interleukin-1β(IL-1β)treatment to mimic the inflammatory environment,and Tubastatin A(Tub A)to inhibit histone deacetylase 6(HDAC6).Our results show that stiff substrates can be anti-inflammatory and provide a better matrix environment than soft substrates.Inhibition of HDAC6 improves the inflammatory environment caused by IL-1βand coordinates with inflammation to spread the chondrocyte area and primary cilia elongation.Without IL-1βand Tub A treatments,the length of the primary cilia rather than frequency is stiffness-dependent,and their length on stiff substrates are greater than that on soft substrates.In conclusion,we demonstrate that stiff substrates,inflammation,and inhibition of HDAC6 enhance the mechanosensitivity of primary cilia and mediate substrate stiffness to suppress inflammation and protect the matrix.Yang Zhang Godfred K Tawiah Yanjun Zhang Xiaohu Wang Xiaochun Wei Weiyi Chen Xiaohong Qiao Quanyou Zhang 2023Acta Biochimica et Biophysica Sinica2023,55,12:1

4Fracture Analysis of Cast Iron Materials with Cracks Based on Elastoplastic Extended Finite Element Method显示文摘Based on the ABAQUS platform for finite element analysis, the extended finite element method (XFEM) considering elastoplastic constitutive relationship is developed, by which the displacement discontinuity across the crack surface and the strong nonlinearity near the crack tip can be described more accurately. The strip specimens with unilateral cracks and central cracks under uniaxial tension are simulated using the XFEM and the FEM, respectively. The J-integral across the crack on each incremen t is calcula ted using the equivale nt domain integral met hod (EDIM), the interaction integral met hod and the FEM, respectively. The effec ts of mesh size and mesh shape near the crack tip, element type and different calculation methods on the accuracy of J-integral are analyzed. The simulation results show that the XFEM is more accurate than the FEM with the same element size and type. The fracture tests of cast iron specimens with unilateral cracks under uniaxial tension are performed, and the J-integral criterion is valid to predict the fracture initiation in numerical simulation. The critical value of J-integral is calculated using the EDIM of the XFEM. 6Enhanced optical performance of multifocal metalens with conic shapes显示文摘A multifocal metalens,which focuses incident light at multiple foci,has many applications in imaging systems and optical communications.However,the traditional design strategy of a multifocal metalens combines several lenses that have different focal points into a planar integrated unit,resulting in low imaging quality because of the high background noise.Here we show that the defects of the traditional method can be overcome by designing a metalens with conic shapes(the ellipse and the hyperbola);this approach could improve the imaging performance and substantially decrease the background noise of multifocal metalenses.These benefits arise from the intrinsic properties of the two conic curves,which can focus incident light constructively at all of the foci of the metalens.We further demonstrate that the proposed conicshaped metalens can function well within a broadband operation wavelength that ranges from 600 to 900 nm with the dual polarity actively controlled by the incident circular polarized light.The great agreement between the experimental and simulation results demonstrates that our proposed metalens has significant potential for use in future integrated nanophotonic devices.Yanjun Bao Qiao Jiang Yimin Kang Xing Zhu Zheyu Fang 2017Light(Science & Applications)2017,6,1:1
7Complex genetic architecture underlying the plasticity of maize agronomic traits显示文摘Phenotypic plasticity is the ability of a given genotype to produce multiple phenotypes in response to changing environmental conditions.Understanding the genetic basis of phenotypic plasticity and establishing a predictive model is highly relevant to future agriculture under a changing climate.Here we report findings on the genetic basis of phenotypic plasticity for 23 complex traits using a diverse maize population planted at five sites with distinct environmental conditions.We found that latituderelated environmental factors were the main drivers of across-site variation in flowering time traits but not in plant architecture or yield traits.For the 23 traits,we detected 109 quantitative trait loci(QTLs),29 for mean values,66 for plasticity,and 14 for both parameters,and 80%of the QTLs interacted with latitude.The effects of several QTLs changed in magnitude or sign,driving variation in phenotypic plasticity.We experimentally validated one plastic gene,ZmTPS14.1,whose effect was likely mediated by the compensation effect of ZmSPL6 from a downstream pathway.By integrating genetic diversity,environmental variation,and their interaction into a joint model,we could provide site-specific predictions with increased accuracy by as much as 9.9%,2.2%,and 2.6%for days to tassel,plant height,and ear weight,respectively.This study revealed a complex genetic architecture involving multiple alleles,pleiotropy,and genotype-byenvironment interaction that underlies variation in the mean and plasticity of maize complex traits.It provides novel insights into the dynamic genetic architecture of agronomic traits in response to changing environments,paving a practical way toward precision agriculture.Minliang Jin Haijun Liu Xiangguo Liu Tingting Guo Jia Guo Yuejia Yin Yan Ji Zhenxian Li Jinhong Zhang Xiaqing Wang Feng Qiao Yingjie Xiao Yanjun Zan Jianbing Yan 2023Plant Communications2023,4,3:1
8NGDcrm:a numeric graph dependency-based conflict resolution method for knowledge graph显示文摘Knowledge graph(KG)conflict resolution is to solve knowledge conflicts problem in the construction of KG.Aiming at the problem of KG conflict resolution,a KG conflict resolution algorithm NGDcrm is proposed,which is a numeric graph dependency-based conflict resolution method.NGDcrm utilizes the dependency graph to perform arithmetic calculation and predicate comparison of numerical entity knowledge in the KG.NGDcrm first uses a parallel segmentation method to segment the KG;then,it extracts the features of the KG according to KG embedding;finally,it uses numerical graph dependencies to detect and correct the wrong facts in the KG based on the extracted features.The experimental results on real data show that NGDcrm is better than the state-of-the-art knowledge conflict resolution method.Among them,the AUC value of NGDcrm on the DBpedia dataset is 15.4%higher than the state-of-the-art method.马江涛 Wang Yanjun Chen Xueting Qiao Yaqiong 2021High Technology Letters2021,27,2:1
9Monitoring of Avian Influenza Virus and Newcastle Disease Virus in Suichuan Region of Jiangxi Province显示文摘[Objective] The research aimed to investigate the epidemiological characteristics of Avian influenza virus( AIV) and Newcastle disease virus( NDV)of wild Ardeid birds in Suichuan region of Jiangxi Province. [Method] A total of 110 nasopharyngeal swabs and 110 cloacal swabs of Ardeid birds were collected from Suichuan region of Jiangxi Province in October of 2014. The swabs were conducted the virus isolation using SPF chicken embryos,suspected positive samples were screened by hemagglutination( HA) test and identified by Reverse Transcription-Polymerase Chain Reaction( RT-PCR). [Results] HA titre of all 110 test samples was 0 except four samples. Detection on AIV and NDV of the test samples by RT-PCR showed that all the samples were negative,which indicated that the infection risk of avian influenza( AI) and newcastle disease( ND) of Ardeid birds in Suichuan region in Jiangxi Province in the summer of 2014 was low. 10Study on Tissue Culture and Rapid Propagation of Platycodon grandiflorus显示文摘With seeds as experimental materials,MS was used as the basic medium to combine different species and concentrations of auxin and mitogen for proliferation,elongation and rooting culture. The best combination of medium and the most suitable medium were selected. The results showed that the best formula for the medium was MS + 0. 5 mg/L BA + 0. 5 mg/L IAA + 30 g/L sucrose in the proliferation culture,MS + 0. 25 mg/L BA + 0. 5 mg/L IAA + 30 g/L sucrose in the elongation culture,and MS + 0. 5 mg/L IAA + 20 g/L sucrose in the rooting culture respectively.
