| 2 | Diurnal and Seasonal Dynamics of Soil Respiration at Temperate Leymus Chinensis Meadow Steppes in Western Songnen Plain, China显示文摘To evaluate the diurnal and seasonal variations in soil respiration(Rs) and understand the controlling factors, we measured carbon dioxide(CO2) fluxes and their environmental variables using a LI-6400 soil CO2 flux system at a temperate Leymus chinensis meadow steppe in the western Songnen Plain of China in the growing season(May–October) in 2011 and 2012. The diurnal patterns of soil respiration could be expressed as single peak curves, reaching to the maximum at 11:00–15:00 and falling to the minimum at 21:00–23:00(or before dawn). The time-window between 7:00 and 9:00 could be used as the optimal measuring time to represent the daily mean soil CO2 efflux. In the growing season, the daily value of soil CO2 efflux was moderate in late spring(1.06–2.51 μmol/(m2·s) in May), increased sharply and presented a peak in summer(2.95–3.94 μmol/(m2·s) in July), and then decreased in autumn(0.74–0.97 μmol/(m2·s) in October). Soil temperature(Ts) exerted dominant control on the diurnal and seasonal variations of soil respiration. The temperature sensitivity of soil respiration(Q10) exhibited a large seasonal variation, ranging from 1.35 to 3.32, and decreased with an increasing soil temperature. Rs gradually increased with increasing soil water content(Ws) and tended to decrease when Ws exceeded the optimum water content(27%) of Rs. | 7 | Micro RNA profiles and potential regulatory pattern during the early stage of spermatogenesis in mice显示文摘Spermatogenesis is a complicated and poorly understood process that relies on the precise regulation of the self-renewal and differentiation of spermatogonia. In many organisms, micro RNAs(mi RNAs) are involved in multiple developmental processes as critical regulators of transcriptional and post-transcriptional gene silencing. This study investigated the expression pattern of mi RNAs in type B spermatogonia cells(BSc) and primary spermatocytes(PSc) of mice, using a high-throughput small RNA sequencing system. The results revealed that the expression levels of Let-7 family mi RNAs were remarkably high in both cell types. Furthermore, the expression levels of mi R-21, mi R-140-3p, mi R-103, mi R-30 a, mi R-101 b and mi R-99 b were decreased during the transformation from BSc to PSc. These mi RNAs target vital genes that participate in apoptosis, cell proliferation and differentiation, junction assembly and cell cycle regulation. | 9 | maturity of associating liver partition and portal vein ligation for staged hepatectomy-derived liver regeneration in a rat model显示文摘AIM To establish a rat model for evaluating the maturity of liver regeneration derived from associating liver partition and portal vein ligation for staged hepatectomy(ALPPS).METHODS In the present study, ALPPS, partial hepatecotmy(PHx), and sham rat models were established initially, which were validated by significant increase of proliferative markers including Ki-67, proliferating cell nuclear antigen, and cyclin D1. In the setting of accelerated proliferation in volume at the second and fifth day after ALPPS, the characteristics of newborn hepatocytes, as well as specific markers of progenitor hepatic cell, were identified. Afterwards, the detection of liver function followed by cluster analysis of functional gene expression were performed to evaluate the maturity.RESULTS Compared with PHx and sham groups, the proliferation of f LR was significantly higher in ALPPS group(P = 0.023 and 0.001 at second day, P = 0.034 and P < 0.001 at fifth day after stage I). Meanwhile, the increased expression of proliferative markers including Ki-67, proliferating cell nuclear antigen, and cyclin D1 verified the accelerated liver regeneration derived from ALPPS procedure. However, ALPPS-induced Sox9 positive hepatocytes significantly increased beyond the portal triad, which indicated the progenitor hepatic cell was potentially involved. And the characteristics of ALPPSinduced hepatocytes indicated the lower expression of hepatocyte nuclear factor 4 and anti-tryptase in early proliferative stage. Both suggested the immaturity of ALPPS-derived liver regeneration. Additionally, the detection of liver function and functional genes expression confirmed the immaturity of renascent hepatocytes derived in early stage of ALPPS-derived liver regeneration.CONCLUSION Our study revealed the immaturity of ALPPS-derived proliferation in early regenerative response, which indicated that the volumetric assessment overestimated the functional proliferation. | 14 | Impacts of land use and salinization on soil inorganic and organic carbon in the middle-lower Yellow River Delta显示文摘Soil inorganic carbon(SIC)is an important reservoir of carbon(C)in arid,semi-arid,and semi-humid regions.However,knowledge is incomplete on the dynamics of SIC and its relationship with soil organic C(SOC)under different land use types in the semi-humid region,particularly in coastal zones impacted by soil salinization.We collected 170 soil samples from 34 profiles across various land use types(maize-wheat,cotton,paddy,and reed)in the middle-lower Yellow River Delta(YRD),China.We measured soil pH,electrical conductivity(EC),water-soluble salts,and SOC and SIC contents.Our results showed significant differences in both SOC and SIC among land use types.The dry cropland(maize-wheat and cotton)soils had significantly higher SOC and SIC densities(4.71 and 15.46 kg C m^(-2),respectively)than the paddy soils(3.28 and 14.09 kg C m^(-2),respectively)in the 0–100 cm layer.Compared with paddy soils,reed soils contained significantly higher SOC(4.68 kg C m^(-2))and similar SIC(15.02 kg C m^(-2))densities.There was a significant positive correlation between SOC and SIC densities over a 0–100 cm soil depth in dry cropland soils,but a negative relationship in the paddy soils.On average,SOC and SIC densities under maize-wheat cropping were 15%and 4%lower,respectively,in the salt-affected soils in the middle-lower YRD than the upper YRD.This study indicated that land use types had great influences on both SOC and SIC and their relationship,and salinization had adverse effect on soil C storage in the YRD. | Yang GUO Xiujun WANG Xianglan LI Minggang XU Yuan LI Haonan ZHENG Yongming LUO Pete SMITH | 2021 | Pedosphere2021,31,6: | 4 |

| 17 | Diurnal Variation of Soil CO2 Efflux and Its Optimal Measuring Time-window of Temperate Meadow Steppes in Western Songnen Plain, China显示文摘In order to study the diurnal variation of soil CO_2 efflux from temperate meadow steppes in Northeast China, and determine the best time for observation, a field experiment was conducted with a LI-6400 soil CO_2 flux system under five typical plant communities(Suaeda glauca(Sg), Chloris virgata(Cv), Puccinellia distans(Pd), Leymus chinensis(Lc) and Phragmites australis(Pa)) and an alkali-spot land(As) at the meadow steppe of western Songnen Plain. The results showed that the diurnal variation of soil CO_2 efflux exhibited a single peak curve in the growing season. Diurnal maximum soil respiration(Rs) often appeared between 11:00 and 13:00, while the minimum occurred at 21:00–23:00 or before dawn. Air temperature near the soil surface(Ta) and soil temperature at 10 cm depth(T10) exerted dominant control on the diurnal variations of soil respiration. The time-windows 7:00–9:00 could be used as the optimal measuring time to represent the daily mean soil CO_2 efflux at the Cv, Pd, Lc and Pa sites. The daily mean soil CO_2 efflux was close to the soil CO_2 efflux from 15:00 to 17:00 and the mean of 2 individual soil CO_2 efflux from 15:00 to 19:00 at the As and Sg sites, respectively. During nocturnal hours, negative soil CO_2 fluxes(CO_2 downwards into the soil) were frequently observed at the As and Sg sites, the magnitude of the negative CO_2 fluxes were 0.10–1.55 μmol/(m^2·s) and 0.10–0.69 μmol/(m^2·s)at the two sites. | 19 | HSP70 alleviates sepsis-induced cardiomyopathy by attenuating mitochondrial dysfunction-initiated NLRP3 inflammasome-mediated pyroptosis in cardiomyocytes显示文摘Background:Sepsis-induced cardiomyopathy(SIC)is an identified serious complication of sepsis that is associated with adverse outcomes and high mortality.Heat shock proteins(HSPs)have been implicated in suppressing septic inflammation.The aim of this study was to investigate whether HSP70 can attenuate cellular mitochondrial dysfunction,exuberated inflammation and inflammasome-mediated pyroptosis for SIC intervention.Methods:Mice with cecal ligation plus perforation(CLP)and lipopolysaccharide(LPS)-treated H9C2 cardiomyocytes were used as models of SIC.The mouse survival rate,gross profile,cardiac function,pathological changes and mitochondrial function were observed by photography,echocardiography,hematoxylin-eosin staining and transmission electron microscopy.In addition,cell proliferation and the levels of cardiac troponin I(cTnI),interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were determined by Cell Counting Kit-8,crystal violet staining and enzyme-linked immunosorbent assay.Moreover,mitochondrial membrane potential was assessed by immunofluorescence staining,and dynamin-related protein 1 and pyroptosis-related molecules[nucleotide-binding domain,leucine-rich-repeat containing family pyrin domain-containing 3(NLRP3),caspase-1,gasdermin-D(GSDMD),gasdermin-D N-terminal(GSDMD-N)]were measured by western blotting,immunoprecipitation and immunoblotting.Finally,hsp70.1 knockout mice with CLP were used to verify the effects of HSP70 on SIC and the underlying mechanism.Results:Models of SIC were successfully established,as reduced consciousness and activity with liparotrichia in CLP mice were observed,and the survival rate and cardiac ejection fraction(EF)were decreased;conversely,the levels of cTnI,TNF-αand IL-1βand myocardial tissue damage were increased in CLP mice.In addition,LPS stimulation resulted in a reduction in cell viability,mitochondrial destabilization and activation of NLRP3-mediated pyroptosis molecules in vitro.HSP70 treatment improved myocardial tissue damage,survival rate and cardiac dysfunction caused by CLP.Additionally,HSP70 intervention reversed LPS-induced mitochondrial destabiliza-tion,inhibited activation of the NLRP3 inflammasome,caspase-1,GSDMD and GSDMD-N,and decreased pyroptosis.Finally,knockout of hsp70.1 mice with CLP aggravated cardiac dysfunction and upregulated NLRP3 inflammasome activity,and exogenous HSP70 significantly rescued these changes.It was further confirmed that HSP70 plays a protective role in SIC by attenuating mitochondrial dysfunction and inactivating pyroptotic molecules.Conclusions:Our study demonstrated that mitochondrial destabilization and NLRP3 inflammasome activation-mediated pyroptosis are attributed to SIC.Interestingly,HSP70 ameliorates sepsis-induced myocardial dysfunction by improving mitochondrial dysfunction and inhibiting the acti-vation of NLRP3 inflammasome-mediated pyroptosis,and such a result may provide approaches for novel therapies for SIC. | Chenlu Song Yiqiu Zhang Qing Pei Li Zheng Meiyu Wang Youzhen Shi Shan Wu Wei Ni Xiujun Fu Yinbo Peng Wen Zhang Min Yao | 2022 | Burns & Trauma2022,10,1: | 2 |
| 20 | The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP.
