| 1 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. | 3 | Inertia Match of a 3-RRR Reconfigurable Planar Parallel Manipulator显示文摘Inertia match of the parallel manipulator means the ratio of the inertial load of the parallel manipulator converted to each actuator shaft and the moment of inertia of the actuator is kept within a reasonable range. Currently there are many studies on parallel manipulators, but few mention inertia parameters and inertia match of parallel manipulators. This paper focuses on the inertia characteristics of the 3-RRR reconfigurable planar parallel manipulator. On the basis of the inverse dynamic formulations deduced with the principle of virtual work, the inertia matrix of the 3-RRR planar parallel manipulator in the actuator space is obtained in algebraic form. Then, by unifying the dimension and averaging diagonal elements of the inertia matrix, the equivalent inertia of the parallel manipulator, which is the inertial load of the parallel manipulator converted to each actuator shaft, is determined. By transforming the inertia problem of the 3-RRR parallel manipulator into that of the serial multi-bar manipulator, the practicality of the equivalent inertia deduced by inverse dynamics is demonstrated. According to the physical meaning of the inertia equation, the manipulator is divided in to three parts. Further analysis is carried out on the contribution of each part to the equivalent inertia and their distributions in the required workspace, revealing that the passive links cannot ignored in calculating the equivalent inertia of the parallel manipulator. Finally, the inertia match for the 3-RRR reconfigurable parallel manipulator under three configurations is accomplished, and reducers are selected. The equivalent inertia calculation and the inertial match results illustrate that the inertia math is a necessary step to the design of the parallel manipulator, and inertia parameters dramatically affect dynamic performances of parallel manipulators. | 4 | Atlas Based Kinematic Optimum Design of the Stewart Parallel Manipulator显示文摘Optimum design is a key approach to make full use of potential advantages of a parallel manipulator. The optimum design of multi-parameter parallel manipulators(more than three design parameters), such as Stewart manipulator, relies on analysis based and algorithm based optimum design methods, which fall to be accurate or intuitive. To solve this problem and achieve both accurate and intuition, atlas based optimum design of a general Stewart parallel manipulator is established, with rational selection of design parameters. Based on the defined spherical usable workspace(SUW), primary kinematic performance indices of the Stewart manipulator, involving workspace and condition number are introduced and analyzed. Then, corresponding performance atlases are drawn with the established non-dimensional design space, and impact of joint distribution angles on the manipulator performance is analyzed and illustrated. At last, an example on atlas based optimum design of the Stewart manipulator is accomplished to illustrate the optimum design process, considering the end-effector posture. Deduced atlases can be flexibly applied to both quantitative and qualitative analysis to get the desired optimal design for the Stewart manipulator with respect to related performance requirements. | 5 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. The modular leg of RPKMs is designed and the RPKMs can be built by changing the setting of modules. A compositive kinematic model is developed accordingly. The inverse kinematics and Jacobian of these two Tricept units are analyzed. Considering workspace volume and dexterity, the effects of geometric size of some modules on the two Tricept units are discussed. In the end, comparison results of these two Tricept units are given. | 8 | Human paraoxonase gene cluster overexpression alleviates angiotensin II-induced cardiac hypertrophy in mice显示文摘Cardiac hypertrophy is the strongest predictor of the development of heart failure, and anti-hypertrophic treatment holds the key to improving the clinical syndrome and increasing the survival rates for heart failure. The paraoxonase(PON) gene cluster(PC) protects against atherosclerosis and coronary artery diseases. However, the role of PC in the heart is largely unknown. To evaluate the roles of PC in cardiac hypertrophy, transgenic mice carrying the intact human PON1, PON2, and PON3 genes and their flanking sequences were studied. We demonstrated that the PC transgene(PC-Tg) protected mice from cardiac hypertrophy induced by Ang II; these mice had reduced heart weight/body weight ratios, decreased left ventricular wall thicknesses and increased fractional shortening compared with wild-type(WT) control. The same protective tendency was also observed with an Apoe^(-/-)background. Mechanically, PC-Tg normalized the disequilibrium of matrix metalloproteinases(MMPs)/tissue inhibitors of MMPs(TIMPs) in hypertrophic hearts, which might contribute to the protective role of PC-Tg in cardiac fibrosis and, thus, protect against cardiac remodeling.
