1Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. 2Pre-stack inversion for caved carbonate reservoir prediction:A case study from Tarim Basin,China显示文摘The major storage space types in the carbonate reservoir in the Ordovician in the TZ45 area are secondary dissolution caves.For the prediction of caved carbonate reservoir,post-stack methods are commonly used in the oilfield at present since pre-stack inversion is always limited by poor seismic data quality and insufficient logging data.In this paper,based on amplitude preserved seismic data processing and rock-physics analysis,pre-stack inversion is employed to predict the caved carbonate reservoir in TZ45 area by seriously controlling the quality of inversion procedures.These procedures mainly include angle-gather conversion,partial stack,wavelet estimation,low-frequency model building and inversion residual analysis.The amplitude-preserved data processing method can achieve high quality data based on the principle that they are very consistent with the synthetics.Besides,the foundation of pre-stack inversion and reservoir prediction criterion can be established by the connection between reservoir property and seismic reflection through rock-physics analysis.Finally,the inversion result is consistent with drilling wells in most cases.It is concluded that integrated with amplitude-preserved processing and rock-physics,pre-stack inversion can be effectively applied in the caved carbonate reservoir prediction.Zhang Yuanyin Sam Zandong Sun Yang Haijun Wang Haiyang HanJianfa Gao Hongliang Luo Chunshu Jing Bing 2011Petroleum Science2011,8,4:9
3A NAC transcription factor, NOR-like1, is a new positive regulator of tomato fruit ripening显示文摘Ripening of the model fruit tomato(Solanum lycopersicum)is controlled by a transcription factor network including NAC(NAM,ATAF1/2,and CUC2)domain proteins such as No-ripening(NOR),SlNAC1,and SlNAC4,but very little is known about the NAC targets or how they regulate ripening.Here,we conducted a systematic search of fruit-expressed NAC genes and showed that silencing NOR-like1(Solyc07g063420)using virus-induced gene silencing(VIGS)inhibited specific aspects of ripening.Ripening initiation was delayed by 14 days when NOR-like1 function was inactivated by CRISPR/Cas9 and fruits showed obviously reduced ethylene production,retarded softening and chlorophyll loss,and reduced lycopene accumulation.RNA-sequencing profiling and gene promoter analysis suggested that genes involved in ethylene biosynthesis(SlACS2,SlACS4),color formation(SlGgpps2,SlSGR1),and cell wall metabolism(SlPG2a,SlPL,SlCEL2,and SlEXP1)are direct targets of NOR-like1.Electrophoretic mobility shift assays(EMSA),chromatin immunoprecipitation-quantitative PCR(ChIP-qPCR),and dual-luciferase reporter assay(DLR)confirmed that NOR-like1 bound to the promoters of these genes both in vitro and in vivo,and activated their expression.Our findings demonstrate that NOR-like1 is a new positive regulator of tomato fruit ripening,with an important role in the transcriptional regulatory network.Ying Gao Wei Wei Xiaodan Zhao Xiaoli Tan Zhongqi Fan Yiping Zhang Yuan Jing Lanhuan Meng Benzhong Zhu Hongliang Zhu Jianye Chen Cai-Zhong Jiang Donald Grierson Yunbo Luo Da-Qi Fu 2018Horticulture Research2018,5,1:9
4Overview of China's Antarctic research progress 1984–2016显示文摘It is more than 30 years since the first Chinese National Antarctic Research Expedition(CHINARE) landed in Antarctica in 1984, representing China's initiation in polar research. This review briefly summarizes the Chinese Antarctic scientific research and output accomplished over the past 30 years. The developments and progress in Antarctic research and the enhancement of international scientific cooperation achieved through the implementation of the CHINARE program have been remarkable. Since the 1980 s, four permanent Chinese Antarctic research stations have been established successively and 33 CHINAREs have been completed. 5Radiofrequency ablation combined with transcatheter arterial chemoembolization therapy versus surgical resection for Barcelona-Clinic Liver Cancer(BCLC) A hepatocellular carcinoma: a meta-analysis显示文摘Purpose: The objective of our study was to compare the effectiveness of the combination of transcatheter arterial chemoembolization(TACE) and radiofrequency ablation(RFA) with that of surgical resection(SR) in Barcelona-Clinic Liver Cancer(BCLC) A hepatocellular carcinoma. Materials and Methods: PubMed, Medline, Embase, and Cochrane Library were searched for comparisons of the two therapies from January 2006 to December 2017. Overall survival rate(OS), recurrence-free survival rate(RFS), complications, and the average length of hospital stay were compared and analyzed. Review Manager v. 5.2 from the Cochrane Collaboration was used for statistical analyses. Results: Seven case-control studies and one randomized controlled trial were identified, of which 717 were treated with a combination of TACE and RFA and 785 were treated with SR. Meta-analysis data revealed that TACE plus RFA had significantly better effectiveness on 1.0-y OS(OR = 0.50, p =.009). The major complications(ORcomplications = 1.88, p =.02) after the combined therapy were significantly lower than those after SR. There were three studies that reported the average length of hospital stay. The hospital stay for the SR group vs the combined therapy group was 19.8 ± 8.4 d vs 7.4 ± 2.2 d, respectively(p <.0001); 18.7 ± 4.9 d vs 11.5 ± 6.9 d, respectively(p <.0001); and 16.6 ± 6.7 d vs 8.5 ± 4.1 d, respectively(p <.0001). There was no significant difference in 3.0-or 5.0-y OS and 1.0-, 3.0-, or 5.0-y RFS. 7Tomato DCL2b is required for the biosynthesis of 22-nt small RNAs, the resulting secondary siRNAs, and the host defense against ToMV显示文摘The tomato encode four functional DCL families,of which DCL2 is poorly studied.Here,we generated loss-of-function mutants for a tomato DCL2 gene,dcl2b,and we identified its major role in defending against tomato mosaic virus in relation to both natural and manual infections.Genome-wide small RNA expression profiling revealed that DCL2b was required for the processing 22-nt small RNAs,including a few species of miRNAs.Interestingly,these DCL2b-dependent 22-nt miRNAs functioned similarly to the DCL1-produced 22-nt miRNAs in Arabidopsis and could serve as triggers to generate a class of secondary siRNAs.In particular,the majority of secondary siRNAs were derived from plant defense genes when the plants were challenged with viruses.We also examined differentially expressed genes in dcl2b through RNA-seq and observed that numerous genes were associated with mitochondrial metabolism and hormone signaling under virus-free conditions.Notably,when the loss-of-function dcl2b mutant was challenged with tomato mosaic virus,a group of defense response genes was activated,whereas the genes related to lipid metabolism were suppressed.Together,our findings provided new insights into the roles of tomato DCL2b in small RNA biogenesis and in antiviral defense.Tian Wang Zhiqi Deng Xi Zhang Hongzheng Wang Yu Wang Xiuying Liu Songyu Liu Feng Xu Tao Li Daqi Fu Benzhong Zhu Yunbo Luo Hongliang Zhu 2018Horticulture Research2018,5,1:4
8AFF3-DNA methylation interplay in maintaining the mono-allelic expression pattern of XIST in terminally differentiated cells显示文摘X chromosome inactivation and genomic imprinting are two classic epigenetic regulatory processes that cause mono-allelic gene expression.In female mammals,mono-allelic expression of the long non-coding RNA gene X-inactive specific transcript(XIST)is essential for initiation of X chromosome inactivation upon differentiation.We have previously demonstrated that the central factor of super elongation complex-like 3(SEC-L3),AFF3,is enriched at gamete differentially methylated regions(DMRs)of the imprinted loci and regulates the imprinted gene expression.Here,we found that AFF3 can also bind to the DMR downstream of the XIST promoter.Knockdown of AFF3 leads to de-repression of the inactive allele of X IST in terminally differentiated cells.In addition,the binding of AFF3 to the XIST DMR relies on DNA methylation and also regulates DNA methylation level at DMR region.However,the KAP1-H3K9 methylation machineries,which regulate the imprinted loci,might not play major roles in maintaining the mono-aUelic expression pattern of XIST in these cells.Thus,our results suggest that the differential mechanisms involved in the X IST DMR and gDMR regulation,which both require AFF3 and DNA methylation.Yue Zhang Chao Wang Xiaoxu Liu Qian Yang Hongliang Ji Mengjun Yang Manman Xu Yunyan Zhou Wei Xie Zhuojuan Luo Chengqi Lin 2019Journal of Molecular Cell Biology2019,11,9:3
9Lycopene Accumulation Affects the Biosynthesis of Some Carotenoid-related Volatiles Independent of Ethylene in Tomato显示文摘For elucidating the regulatory mechanism of ethylene on carotenoid-related volatiles (open chain) compounds and the relationship between lycopene and carotenoid-related volatiles, transgenic tomato fruits in which ACC synthase was suppressed were used. The transgenic tomato fruit showed a significant reduction of lycopene and aroma volatiles with low ethylene production. 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol and geranylacetone, which were suspected to be lycopene degradation products, were lower than those in wild type tomato fruits. In order to identify whether lycopene accumulation effects the biosynthesis of some carotenoid-related volatiles independent of ethylene in tomato or not, the capability of both wild type and transgenic tomato fruits discs to convert lycopene into carotenoid-related volatiles was evaluated. The data showed that external lycopene could convert into 6-methyl-5-hepten-2-one and 6-methyl-5-hepten-2-ol in vivo, indicating that the strong inhibition of ethylene production had no effect on enzymes in the biosynthesis pathway of some carotenoid-related volatiles. Therefore, in ACS-suppression transgenic tomato fruits, the low levels of 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol was due to decreased lycopene accumulation, not ethylene production. 15Simultaneously improving the EMI shielding performances and mechanical properties of CF/PEKK composites via MXene interfacial modification显示文摘In this study, two-dimensional MXene (Ti3 C2 Tx ) was employed to modify the interface of carbon fiber-reinforced polyetherketoneketone (CF/PEKK) composites, in order to simultaneously improve the electromagnetic interference (EMI) shielding performances and mechanical properties. The obtained CF/PEKK composites possessed outstanding EMI and mechanical performances, as anticipated. Specifically, the CF/PEKK composites modified with MXene at 1 mg mL–1 exhibited an excellent EMI shielding effectiveness of 65.2 dB in the X-band, a 103.1% enhancement compared with the unmodified CF/PEKK composites. The attractive EMI shielding performances of CF/PEKK composites originated from enhanced ohmic losses and multiple reflections of electromagnetic waves with the help of the MXene and CF layers. In addition, CF/PEKK composites achieved the best mechanical properties by optimizing the dispersion concentration of MXene to 0.1 mg mL–1 . The flexural strength, flexural modulus, and interlaminar shear strength of CF/PEKK composites reached 1127 MPa, 81 GPa, and 89 MPa, which were 28.5%, 9.5%, and 29.7% higher than that of the unmodified CF/PEKK composites, respectively. 16Pressure-induced robust emission in a zero-dimensional hybrid metal halide (C_(9)NH_(20))_(6)Pb_(3)Br_(12)显示文摘Zero-dimensional(0D)hybrid metal halides are under intensive investigation owing to their unique physical properties,such as the broadband emission from highly localized excitons that is promising for white-emitting lighting.However,fundamental understanding of emission variations and structure–property relationships is still limited.Here,by using pressure processing,we obtain robust exciton emission in 0D(C_(9)NH_(20))_(6)Pb_(3)Br_(12) at room temperature that can survive to 80 GPa,the recorded highest value among all the hybrid metal halides.In situ experimental characterization and first-principles calculations reveal that the pressure-induced emission is mainly caused by the largely suppressed phonon-assisted nonradiative pathway.Lattice compression leads to phonon hardening,which considerably weakens the exciton–phonon interaction and thus enhances the emission.The robust emission is attributed to the unique structure of separated spring-like[Pb_(3)Br_(12)]^(6−)trimers,which leads to the outstanding stability of the optically active inorganic units.Our findings not only reveal abnormally robust emission in a 0D metal halide,but also provide new insight into the design and optimization of local structures of trimers and oligomers in lowdimensional hybrid materials.Mengting Chen Songhao Guo Kejun Bu Sujin Lee Hui Luo Yiming Wang Bingyan Liu Zhipeng Yan Hongliang Dong Wenge Yang Biwu Ma Xujie Lü 2021Matter and Radiation at Extremes2021,6,5:1
17Computational identification of microRNAs and their targets in wheat (Triticum aestivum L.)显示文摘microRNAs (miRNAs) are a class of endogenous, non-coding, short (~21 nt) RNAs directly involved in regulating gene expression at the post-transcriptional level. Previous reports have noted that plant miRNAs in the plant kingdom are highly conserved, which provides the foundation for identification of conserved miRNAs in other plant species through homology alignment. Conserved miRNAs in wheat are identified using EST (Expressed Sequence Tags) and GSS analysis. All previously known miRNAs in other plant species were blasted against wheat EST and GSS sequences to select novel miRNAs in wheat by a series of filtering criteria. From a total of 37 conserved miRNAs belonging to 18 miRNA families 10 conserved miRNAs comprising 4 families were reported in wheat. MiR395 is found to be a special family, because three members belonging to the same miR395 family are clustered together, similar to animal miRNAs. 20Research on intelligent judgment method of natural gas hydrate drilling risk显示文摘There are many emergency risks in the process of natural gas hydrate(NGH)drilling.In order to ensure the safe and efficient exploitation of NGH,it is urgent to establish an intelligent judgment method for the risks in the process of NGH drilling.In this paper,the response relationship between monitoring parameters and risk categories of NGH while drilling is established.Based on fuzzy analytic hierarchy process(FAHP),the comprehensive weights of 10 risk monitoring parameters are obtained,including gas production,wellbore instability,hydrate ice barrier,drill string fracture,sticking,bit balling,drilling tool piercement,gas seepage,seabed subsidence and seabed landslide.Besides,the comprehensive judgment weight matrix is constructed,and the reasonable fluctuation range of monitoring parameters is formed.Thus,the intelligent judgment method of NGH drilling risk is established.The intelligent judgment and alarm of NGH drilling risks can be realized quickly and accurately by this method,namely,it can monitor the risks in the process of operation and guarantee the construction safety of NGH drilling.Haitao Li Zhaolong Ge Na Wei Wantong Sun Yao Zhang Jin Xue Lin Jiang Jun Pei Bing Liao Hongliang Cao Ruixu Zhang Shuangli Li Qixia Lu Jinyan Luo 2021Petroleum2021,7,4:1
