| 2 | IL-10-producing regulatory B cells restrain the T follicular helper cell response in primary Sjögren’s syndrome显示文摘Increased numbers of T follicular helper(Tfh)cells have been implicated in the development of autoimmune diseases including primary Sjögren’s syndrome(pSS),but how the Tfh cell response is regulated during autoimmune pathogenesis remains largely unclear.Here,we first found negative correlations between IL-10^(+)regulatory B(Breg)cell numbers and Tfh cell responses and disease activity in patients with pSS and mice with experimental Sjögren’s syndrome(ESS).Moreover,we detected high expression of IL-10 receptor on Tfh cells and their precursors in both humans and mice.In culture,IL-10 suppressed human and murine Tfh cell differentiation by promoting STAT5 phosphorylation.By using an adoptive transfer approach and two-photon live imaging,we found significantly increased numbers of Tfh cells with enhanced T cell homing into B cell follicles in the draining cervical lymph nodes of RAG-2−/−mice transferred with IL-10-deficient B cells during ESS development compared with those of RAG-2−/−mice transferred with wild-type B cells.In ESS mice,CD19^(+)CD1d^(hi)CD5^(+)Breg cells with decreased IL-10 production exhibited severely impaired suppressive effects on T cell proliferation.Consistently,CD19^(+)CD24^(+)CD38^(hi) Breg cells from pSS patients showed significantly reduced IL-10 production with defective inhibitory function in the suppression of autologous Tfh cell expansion.Furthermore,the adoptive transfer of IL-10-producing Breg cells markedly suppressed the Tfh cell response and ameliorated ESS progression in ESS mice.Together,these findings demonstrate a critical role for IL-10-producing Breg cells in restraining the effector Tfh cell response during pSS development. | Xiang Lin Xiaohui Wang Fan Xiao Kongyang Ma Lixiong Liu Xiaoqi Wang Dong Xu Fei Wang Xiaofei Shi Dongzhou Liu Yan Zhao Liwei Lu | 2019 | Cellular & Molecular Immunology2019,16,12: | 13 |

Although many progresses have been achieved, it is still highly desirable to develop novel strategies for controllable synthesis of the well-defined ultrathin MOF nanosheets. Herein we report a polyvinylpyrrolidone (PVP)-assisted route to synthesize the ultrathin Ni-MOF nanosheets characteristic of 1.5 nm in thickness, in which PVP is reacted with 2-aminoterephthalic acid (H2BDC-NH2) via formation of C=N bon d, followed by coord inatio n with Ni2+ io ns to form the ultrathi n MOF n anosheets. | 5 | Piezo1 channel activation in response to mechanobiological acoustic radiation force in osteoblastic cells显示文摘Mechanobiological stimuli,such as low-intensity pulsed ultrasound(LIPUS),have been shown to promote bone regeneration and fresh fracture repair,but the fundamental biophysical mechanisms involved remain elusive.Here,we propose that a mechanosensitive ion channel of Piezo1 plays a pivotal role in the noninvasive ultrasound-induced mechanical transduction pathway to trigger downstream cellular signal processes.This study aims to investigate the expression and role of Piezo1 in MC3T3-E1 cells after LIPUS treatment.Immunofluorescence analysis shows that Piezo1 was present on MC3T3-E1 cells and could be ablated by shRNA transfection.MC3T3-E1 cell migration and proliferation were significantly increased by LIPUS stimulation,and knockdown of Piezo1 restricted the increase in cell migration and proliferation.After labeling with Fluo-8,MC3T3-E1 cells exhibited fluorescence intensity traces with several high peaks compared with the baseline during LIPUS stimulation.No obvious change in the fluorescence intensity tendency was observed after LIPUS stimulation in shRNA-Piezo1 cells,which was similar to the results in the GsMTx4-treated group.The phosphorylation ratio of ERK1/2 in MC3T3-E1 cells was significantly increased(P<0.01)after LIPUS stimulation.In addition,Phalloidin-iFluor-labeled F-actin filaments immediately accumulated in the perinuclear region after LIPUS stimulation,continued for 5 min,and then returned to their initial levels at 30 min.These results suggest that Piezo1 can transduce LIPUS-induced mechanical signals into intracellular calcium.The influx of Ca2+serves as a second messenger to activate ERK1/2 phosphorylation and perinuclear F-actin filament polymerization,which regulate the proliferation of MC3T3-E1 cells. | Guangdao Zhang Xiaofei Li Lin Wu Yi-Xian Qin | 2021 | Bone Research2021,9,2: | 9 |
| 6 | NMDA Receptor Antagonist MK801 Protects Against 1-Bromopropane-Induced Cognitive Dysfunction显示文摘Occupational exposure to 1-bromopropane(1-BP) induces learning and memory deficits. However, no therapeutic strategies are currently available. Accumulating evidence has suggested that N-methyl-D-aspartate receptors(NMDARs) and neuroinflammation are involved in the cognitive impairments in neurodegenerative diseases. In this study we aimed to investigate whether the noncompetitive NMDAR antagonist MK801 protects against 1-BPinduced cognitive dysfunction. Male Wistar rats were administered with MK801(0.1 mg/kg) prior to 1-BP intoxication(800 mg/kg). Their cognitive performance was evaluated by the Morris water maze test. The brains of rats were dissected for biochemical, neuropathological,and immunological analyses. We found that the spatial learning and memory were significantly impaired in the1-BP group, and this was associated with neurodegeneration in both the hippocampus(especially CA1 and CA3)and cortex. Besides, the protein levels of phosphorylated NMDARs were increased after 1-BP exposure. MK801 ameliorated the 1-BP-induced cognitive impairments and degeneration of neurons in the hippocampus and cortex.Mechanistically, MK801 abrogated the 1-BP-induced disruption of excitatory and inhibitory amino-acid balance and NMDAR abnormalities. Subsequently, MK801 inhibited the microglial activation and release of pro-inflammatory cytokines in 1-BP-treated rats. | 10 | Knockdown of long non-coding RNA LCPAT1 inhibits autophagy in lung cancer显示文摘Objective: Long non-coding RNAs(lnc RNAs) are involved in numerous biological processes in lung cancer cells. In our previous studies, we identified a lnc RNA, ENST00000439577, which is highly expressed in lung carcinomas, and termed it lung cancer progression-associated transcript 1(LCPAT1). To characterize the role of LCPAT1 in lung cancer, we conducted the current study.Methods: Expression of LCPAT1 and autophagy-associated markers in tumor tissues and lung cancer cell lines was determined by real-time quantitative polymerase chain reaction(q PCR). Hematoxylin and eosin(HE) staining, q PCR, Western blot, and immunohistochemistry were performed to evaluate xenografted tumor tissues. Autophagy induced by rapamycin was detected by Western blot and immunofluorescence in lung cancer cell lines.Results: Expression of LCPAT1 and microtubule-associated protein 1 light chain 3 beta(LC3B) was positively correlated in lung cancer. Knockdown of LCPAT1 inhibited tumor growth and suppressed cell autophagy in vivo. | 13 | Growth structures and growth strata of the Qianjiadian Basin in the western Yanshan fold and thrust belt, North China显示文摘The northwestward subduction of the Izanagi Plate beneath the eastern Eurasian Plate during the Late Mesozoic caused a series of compressional deformation events in the Yanshan fold and thrust belt(YFTB), but the tectonic deformation timing, tectonic properties and relationship between tectonic uplift and sediment accumulation in the intermontane basins continue to be debated. For this reason, the sedimentology, sediment provenance, and basin structure of the Qianjiadian Basin(QJB) in the northern Beijing region during the Late Jurassic and Early Cretaceous were studied in detail. The results suggest that a fault-propagation fold-type(FPFT) growth structure and growth strata developed on the western edge of the QJB and that the top part of Member 2 and Member 3 of the Tuchengzi Formation are growth strata controlled by the limb rotation mechanism. In two small thrust faults in the QJB, the Mesoproterozoic Xiamaling Formation is thrust over the Tuchengzi Formation, and these faults may have controlled the development of the fault-bend fold-type growth strata. An analysis of the 'source-to-sink' process suggests that the hanging wall succession of the Shangyi-Pingquan fault(SPF) was the main source area of the Late Jurassic to early Early Cretaceous strata in the QJB. A zircon206 Pb/238 U age of 140.8±2.4 Ma for the volcanic rocks at the bottom of the FPFT growth strata represents the timing of the initiation of FPFT growth structure development. The discovery of the FPFT growth structure and growth strata in the QJB indicate that the QJB was an intermontane flexural basin controlled by fold and thrust structures during the early Early Cretaceous. | 15 | Preventing abnormal NF-κB activation and autoimmunity by Otub1-mediated p100 stabilization显示文摘NF-κB, a family of transcription factors regulating diverse biological processes including immune responses, is activated by canonical and noncanonical pathways based on degradation of IκBα and processing of the IκB-like protein p100, respectively. Although p100 responds to noncanonical NF-κB stimuli for processing, it does not undergo degradation, but rather becomes accumulated, along with canonical NF-κB activation. We show here that the stability of p100 is tightly controlled by a deubiquitinase, Otub1. Otub1 deficiency not only promotes signal-induced p100 processing and noncanonical NF-κB activation but also causes steady-state p100 degradation, leading to aberrant NF-κB activation in the canonical pathway. B-cell-conditional deletion of Otub1 results in B-cell hyperplasia, antibody hyper-production, and lupus-like autoimmunity. Otub1-deficient B cells display aberrantly activated phenotypes and overproduce the cytokine IL-6, contributing to autoimmunity induction. | 19 | Response of PM_(2.5)-bound elemental species to emission variations and associated health risk assessment during the COVID-19 pandemic in a coastal megacity显示文摘The coronavirus(COVID-19)pandemic is disrupting the world from many aspects.In this study,the impact of emission variations on PM_(2.5)-bound elemental species and health risks associated to inhalation exposure has been analyzed based on real-time measurements at a remote coastal site in Shanghai during the pandemic.Most trace elemental species decreased significantly and displayed almost no diel peaks during the lockdown.After the lockdown,they rebounded rapidly,of which V and Ni even exceeded the levels before the lockdown,suggesting the recovery of both inland and shipping activities.Five sources were identified based on receptor modeling.Coal combustion accounted for more than 70%of the measured elemental concentrations before and during the lockdown.Shipping emissions,fugitive/mineral dust,and waste incineration all showed elevated contributions after the lockdown.The total non-carcinogenic risk(HQ)for the target elements exceeded the risk threshold for both children and adults with chloride as the predominant species contributing to HQ.Whereas,the total carcinogenic risk(TR)for adults was above the acceptable level and much higher than that for children.Waste incineration was the largest contributor to HQ,while manufacture processing and coal combustion were the main sources of TR.Lockdown control measures were beneficial for lowering the carcinogenic risk while unexpectedly increased the non-carcinogenic risk.From the perspective of health effects,priorities of control measures should be given to waste incineration,manufacture processing,and coal combustion.A balanced way should be reached between both lowering the levels of air pollutants and their health risks. | Guochen Wang Kan Huang Qingyan Fu Jia Chen Juntao Huo Qianbiao Zhao Yusen Duan Yanfen Lin Fan Yang Wenjie Zhang Hao Li Jian Xu Xiaofei Qin Na Zhao Congrui Deng | 2022 | Journal of Environmental Sciences2022,34,12: | 2 |
| 20 | Multiple Emitters Localization by UAV with Nested Linear Array:System Scheme and 2D-DOA Estimation Algorithm显示文摘Unmanned Aerial Vehicle(UAV)equipped with uniform linear array has been applied to multiple emitters localization.Meanwhile,nested linear array enables to enhance localization resolution and achieve under-determined Direction of Arrival(DOA)estimation.In this paper,we propose a new system structure for emitters localization that combines the UAV with nested linear array,which is capable of significantly increasing the positioning accuracy of interested targets.Specifically,a localization scheme is designed to obtain the paired two-dimensional DOA(2D-DOA,i.e.azimuth and elevation angles)estimates of emitters by nested linear array with UAV.Furthermore,we propose an improved DOA estimation algorithm for emitters localization that utilizes Discrete Fourier Transform(DFT)method to obtain coarse DOA estimates,subsequently,achieve the fine DOA estimates by sparse representation.The proposed algorithm has lower computational complexity because the coarse DOA estimates enable to shrink the range of over-complete dictionary of sparse representation.In addition,compared to traditional uniform linear array,improved 2D-DOA estimation performance of emitters can be obtained with a nested linear array.Extensive simulation results testify the effectiveness of the proposed method. | Xinping Lin Xiaofei Zhang Lang He Wang Zheng | 2020 | China Communications2020,17,3: | 2 |
