1PREVALENCE AND RISK FACTORS OF PERIPHERAL ARTERIAL DISEASE IN DIABETIC PATIENTS OVER 50 YEARS OLD IN CHINA显示文摘Objective To investigate the prevalence of peripheral arterial disease (PAD) in China type 2 diabetic patients and to demonstrate the relationships between putative risk factors and PAD. Methods In total 1 397 type 2 diabetic patients aged 50 years and older were enrolled and determined ankle-brachial index (ABI) and brachial-ankle pulse wave velocity (baPWV) in 15 Class III Grade A hospitals in 7 major cities of China. Results Mean patient age was 63.7±8.2 years and mean duration of diabetes mellitus was 9.39±7.4 years. Two hundreds and seventy-two (19.47%) patients were diagnosed as PAD by ABI <0.9, 122 (18.37%) in male and 150 (20.46%) in female. PAD patients had a significantly longer duration of diabetes mellitus, higher hemoglobin Alc, and a significantly lower mean body mass index than non-PAD ones. Aging, smoking, and systolic blood pressure were found to be positively related with the prevalence of PAD. In terms of lipid profiles, no variable was found to relate with PAD. Notably, baPWV showed as the same significant guiding index for PAD, almost matched with ABI. Conclusions PAD is a common complication in China type 2 diabetic patients. 2Effects of Salvia miltiorrhiza on intestinal microflora in rats with ischemia/reperfusion liver injury显示文摘BACKGROUND: Hepatic ischemia/reperfusion injury may induce intestinal microflora imbalance. Salvia miltiorrhiza is effective in promoting blood circulation and counteracting peroxidation in tissues. The aim of the present study was to determine the effects of Salvia miltiorrhiza on intestinal mi- croflora, endotoxemia, and bacterial translocation in rats with hepatic I/R injury. METHODS: Sprague-Dawley rats in specific pathogen free grade were divided into 3 groups: group I(n =6) for sham operation: groups ( n = 7) for liver ische- mia for 20 minutes and reperfusion for 22 hours. Group was also pretreated with 4 ml/day of Salvia miltiorrhiza solu- tion (250 mg/kg) by daily gavage for 7 days. The levels of serum alanine aminotransferase (ALT), aspartate amino- transferase (AST), malondialdehyde ( MDA) and supero- xide dismutase ( SOD ) in liver tissues, serum endotoxin, intestinal bacterial counts, intestinal mucosal histology and bacterial translocation were studied. RESULTS: The levels of ALT, AST, plasma endotoxin and MDA in liver tissues were decreased more markedly in group (57.57 ± 18.08 U/L, 147.57 ±40.84 U/L, 0.42 ± 0.144 EU/ml and 0. 52 ±0.19 nmol/mg-prot respectively) in group 295.9±216.92 U/L, 0.80± 0.262 EU/ml and 0.72±0.12 nmol/mg-prot; P <0.05-0.01 respectively). Liver SOD activity was increased more sig- nificantly in group (318.47±64.62 U/mg-prot) than in group U/mg-prot, P<0.05). The counts of Bifidobacteria and Bacteroides increased more significantly in group than in group but were similar to those in group I. Bacterial translocation to the kidney in group was 50% (5/10), whereas no bacterial translocation to the kidney occurred in the other two groups (P <0. 01). 3Increase of TNFα-stimulated Osteoarthritic Chondrocytes Apoptosis and Decrease of Matrix Metalloproteinases 9 by NF-κB Inhibition显示文摘Objective To investigate the in vitro effect of caffeic acid phenethyl ester(CAPE),a NF-κB inhibitor,on the apoptosis of osteoarthritic(OA)chondrocytes and on the regulation of the gelatinases matrix metalloproteinase 2(MMP-2)and matrix metalloproteinase 9(MMP-9).Methods Annexin V-FITC/propidium iodide(PI)labeling and western blotting were used to observe and determine the apoptosis in TNFα-stimulated primary cultured osteoarthritic chondrocytes.Also,gelatin zymography was applied to examine MMP-2 and MMP-9 activities in supernatants.Results It was confirmed by both flow cytometry and western blotting that chondrocytes from OA patients have an apoptotic background.Use of CAPE in combination with 10 ng/mL of TNFα for 24 h facilitated the apoptosis.MMP-9 in the supernatant could be autoactivated(from proMMP-9 to active MMP-9),and the physiologic calcium concentration(2.5 mmol/L)could delay the autoactivation of MMP-9.The activities of MMP-2 and MMP-9 in the fresh supernatant increased significantly in response to stimulation by 10 ng/mL of TNFα for 24 h.The stimulatory effect of TNFα just on proMMP-9 was counteracted significantly by CAPE.Conclusion NF-κB could prevent chondrocytes apoptosis though its activation was attributed to the increase of proMMP-9 activity induced by TNFα(a pro-apoptotic factor).Therefore,therapeutic NF-κB inhibitor was a 'double-edged swords' to the apoptosis of chondrocytes and the secretion of MMP-9.WANG Yan LI De Ling ZHANG Xin Bo DUAN Yuan Hui WU Zhi Hong HAO Dong Sheng CHEN Bao Sheng QIU Gui Xing 2013Biomedical and Environmental Sciences2013,26,4:13

6BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. Thus, BubR1-mediated CENP-E phosphorylation produces a temporal switch that enables transition from lateral to end-on microtubule capture and organization of microtubules into stable midzone arrays.Yuejia Huang Lin Lin Xing Liu Sheng Ye Phil Y. 7Distribution characteristics of poly-and perfluoroalkyl substances in the Yangtze River Delta显示文摘In this work, a method was developed and optimized for the analysis of polyfluoroalkyl and/or perfluoroalkyl substances(PFASs) content in surface water and sediment samples with high instrumental response and good separation. Surface water and sediment samples were collected from the Yangtze River Delta(YRD) to analyze the distribution characteristics of perfluoroalkyl carboxylic acids(PFCAs), perfluoroalkane sulfonic acids(PFSAs), perfluoroalkyl phosphonic acids(PFPAs), perfluoroalkyl phosphinic acids(PFPiAs), and polyfluoroalkyl phosphoric acid diesters(di PAPs). The results showed that the total concentrations of PFCAs and PFSAs in YRD varied from 31 to 902 ng/L. PFCAs(≥ 11 carbons) and PFSAs(≥ 10 carbons atoms) were not detected in any surface water samples. The mean concentrations of all PFCAs and PFSAs in surface water from the sampling areas decreased in the following order:Yangtze river(191 ng/L) ≈ Taihu lake(189 ng/L) > Huangpu river(122 ng/L) ≈ Qiantang river(120 ng/L) > Jiaxing urban river(100 ng/L). Strong significant(p < 0.05) correlations between the concentrations of many of the compounds were found in the sampling areas, suggesting a common source for these compounds. Only perfluorooctanoic acid(PFOA) was observed in all sediment samples, at concentrations varying from 0.02 to 1.35 ng/g. 9Co-doped IT-M0S2 nanosheets embedded in N,S-doped carbon nanobowls for high-rate and ultra-stable sodium-ion batteries显示文摘Despite various 2H-MoS/carbon hybrid nanostructures have been constructed and committed to improve the performance for sodium-ion batteries(SIBs),they still show the limited cycle stability due to the relatively large volumetric expansion during the charge-discharge process Herein,we report the construction of cobalt-doped few-layered 1T-MoS2 nanosheets embedded in N,S-doped carbon(CMS/NSC)nanobowls derived from metal-organic framework(MOF)precursor via a simple in situ sulfurization process.This unique hierarchical structure enables the uniformly dispersed Co-doped 1T-MoS2 nanosheets intimately couple with the highly conductive carbon nanobowls,thus efficiently preventing the aggregation.In particular,the Co-doping plays a crucial role in maintaining the integrity of structure for MoS2 during cycling tests,confirmed by first-principles calculations.Compared with pristine MoS2,the volume deformation of Co-doped MoS2 can be shrunk by a prominent value of 52%during cycling.Furthermore,the few-layered MoS2 nanosheets with 1T metalic phase endow higher conductivity,and thus can surpass its counterpart 2H semiconducting phase in battery performance.By virtue of the synergistic effect of stable structure,appropriate doping and high conductivity,the resulting CMS/NSC hybrid shows superior rate capability and cycle stability.The capacity of CMS/NSC can still be 235.9 mAh·g^-1 even at 25 A·g^-1,which is 51.3%of the capacity at 0.2 A·g^-1.Moreover,the capacity can still remain 218.6 mAh·g^-1 even over 8,240 cycles at 5 Ag·g^-1 with a low decay of 0.0044%per cycle,one of the best performances among the reportec MoS2-based anode materials for SIBs.Peihao Li Yong Yang Sheng Gong Fan Lv Wei Wang Yiju Li Mingchuan Luo Yi Xing Qian Wang Shaojun Guo 2019Nano Research2019,12,9:6

12The Changing Trends of HIV/AIDS in An Ethnic Minority Region of China: Modeling the Epidemic in Liangshan Prefecture, Sichuan Province显示文摘Objective This study was to investigate the HIV current situation in Liangshan prefecture, in order to predict prevalence and transmission trends. Methods Region-specific population, behavior, serosurveillence, and policy/program data (from 1995 to 2010) were gathered from various local and national organizations and applied to the Asian Epidemic Model (AEM) and used to derive estimates of future HIV prevalence, epidemic trends, and outcomes of intervention strategies. Results The AEM projections for 2020 included increased number of people living with HIV (PLHIV; to 136 617), increased HIV prevalence (2.51%), and 8037 deaths from acquired immunodeficiency syndrome (AIDS) in this region. However, the overall HIV incidence rate (per 10 000) was projected to decline from 27 in 2015 to 22 in 2020, largely due to a predicted decrease in HIV infection rate (per 10 000) from 658 in 2013 to 621 in 2020 among intravenous drug users. In contrast, the cases of HIV infection per 10 000 was projected to increase from 420 in 2010 to 503 in 2020 among men who have sex with men, and from 8 in 2010 to 15 in 2020 among the general population. The predominant risk factor for HIV transmission over the next decade in Liangshan was casual sex. Community-based outreach strategies to reduce injected drug use and casual sex, and to promote condom use, were predicted as effective interventions to decrease HIV transmission. 14Performance improvement in multi-ship imaging for ScanSAR based on sparse representation显示文摘There is always a compromise between unambiguous wide-swath imaging and high cross-range resolution owing to the constraint of minimum antenna area for conventional single-channel spaceborne synthetic aperture radar(SAR)imaging.To overcome the inherent systemic limitation,multi-channel SAR imaging has been developed.Nevertheless,this still suffers from various problems such as high system complexity.To simplify the system structure,a novel algorithm for high resolution multi-ship ScanSAR imaging based on sparse representation is proposed in this paper,where the SAR imaging model is established via maximum a posterior estimation by utilizing the sparsity prior of multi-ship targets.In the scheme,a wide swath is generated in the ScanSAR mode by continuously switching the radar footprint between subswaths.Meanwhile,high crossrange resolution is realized from sparse subapertures by exploiting the sparsity feature of multi-ship imaging.In particular,the SAR observation operator is constructed approximately as the inverse of conventional SAR imaging and then high resolution SAR imaging including range cell migration compensation is achieved by solving the optimization.Compared with multi-channel SAR imaging,the system complexity is effectively reduced in the ScanSAR mode.In addition,enhancement of the cross-range resolution is realized by incorporating the sparsity prior with sparse subapertures.As a result,the amount of data is effectively reduced.Experiments based on measured data have been carried out to confirm the effectiveness and validity of the proposed algorithm.XU Gang SHENG JiaLian ZHANG Lei XING MengDao 2012Science China(Information Sciences)2012,55,8:4

16Nematode-Encoded RALF Peptide Mimics Facilitate Parasitism of Plants through the FERONIA Receptor Kinase显示文摘The molecular mechanism by which plants defend against plant root-knot nematodes(RKNs)is largely unknown.The plant receptor kinase FERONIA and its peptide ligands,rapid alkalinization factors(RALFs),regulate plant immune responses and cell expansion,which are two important factors for successful RKN parasitism.In this study,we found that mutation of FERONIA in Arabidopsis thaliana resulted in plants showing low susceptibility to the RKN Meloidogyne incognita.To identify the underlying mechanisms associated with this phenomenon,we identified 18 novel RALF-likes from multiple species of RKNs and showed that two RALF-likes(i.e.,MiRALF1 and MiRALF3)from M.incognita were expressed in the esophageal gland with high expression during the parasitic stages of nematode development.These nematode RALF-likes also possess the typical activities of plant RALFs and can directly bind to the extracellular domain of FERONIA to modulate specific steps of nematode parasitism-related immune responses and cell expansion.Genetically,both MiRALF1/3 and FERONIA are required for RKN parasitism in Arabidopsis and rice.Collectively,our study suggests that nematode-encoded RALFs facilitate parasitism via plant-encoded FERONIA and provides a novel paradigm for studying host-pathogen interactions.Xin Zhang Huan Peng Sirui Zhu Junjie Xing Xin Li Zhaozhong Zhu Jingyuan Zheng Long Wang Bingqian Wang Jia Chen Zhenhua Ming Ke Yao Jinzhuo Jian Sheng Luan Devin Coleman-Derr Hongdong Liao Yousong Peng Deliang Peng Feng Yu 2020Molecular Plant2020,13,10:3
17Ameliorating end-product inhibition to improve cadaverine production in engineered Escherichia coli and its application in the synthesis of bio-based diisocyanates显示文摘Cadaverine is an important C5 platform chemical with a wide range of industrial applications.However,the cadaverine inhibition on the fermenting strain limited its industrial efficiency of the strain.In this study,we report an engineered Escherichia coli strain with high cadaverine productivity that was generated by developing a robust host coupled with metabolic engineering to mitigate cadaverine inhibition.First,a lysine producing E.coli was treated with a combination of radiation(ultraviolet and visible spectrum)and ARTP(atmospheric and room temperature plasma)mutagenesis to obtain a robust host with high cadaverine tolerance.Three mutant targets including HokD,PhnI and PuuR are identified for improved cadaverine tolerance.Further transcriptome analysis suggested that cadaverine suppressed the synthesis of ATP and lysine precursor.Accordingly,the related genes involved in glycolysis and lysine precursor,as well as cadaverine exporter was engineered to release the cadaverine inhibition.The final engineered strain was fed-batch cultured and a titer of 58.7 g/L cadaverine was achieved with a yield of 0.396 g/g,both of which were the highest level reported to date in E.coli.The bio-based cadaverine was purified to>99.6%purity,and successfully used for the synthesis of polyurethane precursor 1,5-pentamethylene diisocyanate(PDI)through the approach of carbamate decomposition.Xin Wang Xing Guo Jing Wang Hui Li Feng He Sheng Xu Kequan Chen Pingkai Ouyang 2021Synthetic and Systems Biotechnology2021,6,4:3

19A Diagnostic Tool for Identification of Etiologies of Fever of Unknown Origin in Adult Patients显示文摘The diagnosis and treatment of fever of unknown origin (FUO) are huge challenges to clinicians.Separating the etiologies of FUO into infectious and non-infectious disease is conducive to clinical physicians not only on making decisions rapidly concerning the prescription of suitable antibiotics but also on further analysis of the final diagnosis.In order to develop and validate a diagnostic tool to efficiently distinguish the etiologies of adult FUO patients as infectious or non-infectious disease,FUO patients from the departments of infectious disease and internal medicine in three Chinese tertiary hospitals were enrolled retrospectively and prospectively.By using polynomial logistic regression analysis,the diagnostic formula and the associated scoring system were developed.The variables included in this diagnostic formula were from clinical evaluations and common laboratory examinations.The proposed tool could discriminate infectious and noninfectious causes of FUO with an area under receiver operating characteristic curve (AUC) of 0.83,sensitivity of 0.80 and specificity of 0.75.This diagnosis tool could predict the infectious and non-infectious causes of FUO in the validation cohort with an AUC of 0.79,sensitivity of 0.79 and specificity of 0.70.The results suggested that this diagnostic tool could be a reliable tool to discriminate between infectious and non-infectious causes of FUO.Man-zhi ZHAO Qiu-rong RUAN Ming-you XING Sheng WEI Dong XU Zhu-hua WU Lin ZHU Ji-ling ZHU Cai-feng ZHENG Shi LIU Zu-jiang YU Jun-ying QI Jian-xin SONG 2019Current Medical Science2019,39,4:3
