2Identification of a novel mutation in POU3F4 for prenatal diagnosis in a Chinese family with X-linked nonsyndromic hearing loss显示文摘We present the clinical and genetic findings for a Chinese family with X-linked non-syndromic hearing loss in which the affected males showed congenital profound sensorineural hearing impairment. In two affected brothers, the computer tomography of temporal bone showed bilateral dilation of the internal auditory canal with fistulous communication between the lateral canal and the basal cochlear turn, which is consistent with the typical DFNX2 phenotype. A missense mutation (c.647G→A) in the POU3F4 gene caused a substitu- tion from glycine to glutamic acid at position 216 (p.G216E), and this mutation was found to consistently cosegregate with the deafness phenotype in the family. The mutation resulted in the loss of function of the POU3F4 by decreasing the affinity between the protein and DNA, as shown in silico by the structural analysis. Prenatal diagnosis of pregnant proband of this family revealed the c.647G→A muta- tion in DNA extracted from the amniotic fluid surrounding the fetus. 3Phenotypic identification of peanut germplasm for resistance to southern stem rot显示文摘Southern stem rot,caused by Sclerotium rolfsii Sacc.,is a destructive soil fungal disease of peanut in China and other countries.To evaluate resistant germplasm,a total of 256 peanut accessions were investigated on their resistance to southern stem rot in 3 environments by artificial inoculation.Variance analysis indicated that disease index was significantly influenced by environment,genotype and genotypeenvironment interactions.Peanut accessions of var.vulgaris type exhibited higher resistance to southern stem rot.Disease index was significantly negatively correlated with linoleic acid content,while positively correlated with oleic acid content.Six resistant accessions were identified,including Hua 28,Shandongzai,ICG 6326,Quanhua 7,Quanhua 9 and Guihua 836,with their disease indexes under 40 and mortality were less than 30%in the three environments.The identified resistant accessions showed the great potential to be applied in resistant peanut breeding,and would be good genetic resources for enhancing the resistance to southern stem rot.Pengmin Fan Wanduo Song Yanping Kang Liyun Wan Yong Lei Dongxin Huai Yuning Chen Xin Wang Huifang Jiang Liying Yan Boshou Liao 2020Oil Crop Science2020,5,4:3
4Assembly of carbon nanodots in graphene-based composite for flexible electro-thermal heater with ultrahigh efficiency显示文摘A high-efficiency electro-thermal heater requires simultaneously high electrical and thermal conductivities to generate and dissipate Joule heat efficiently.A low in put voltage is essential to en sure the heaters safe applications.However,the low voltage gen erally leads to low saturated temperature and heati ng rate and hence a low thermal efficie ncy.How to reduce the in put voltage while maintai ning a high electro-thermal efficiency is still a challenge.Herein,a highly electrical and thermal conductive film was 8nstructed using a graphene-based composite which has an internal three-dimensional(3D)conductive network.In the 3D framework,cellulose nanocrystalline(CNC)phase with chiral liquid crystal manner presents in the form of alig ned helix betwee n the graphe ne oxide(GO)layers.Carbon nano dots(CDs)are assembled泊side the composite as con ductive nano fillers.Subseque nt an neali ng and compressi on results in the formati on of the assembled GO-CNC-CDs film.The carb on ized CNC nano rods(CNR)with the helical alignment act as irrplane and through-pla ne conn ecti ons of n eighbori ng reduced GO(rGO)nano sheets,forming a con ductive network in the composite film.The CDs with ultrafast electr ons tran sfer rates provide additi onal electro ns and phonons tran sport paths for the composite.As a result,the obtai ned graphe ne?based composite film(rGO-CNR-CDs)exhibited a high thermal conductivity of 1,978.6 W m^-1·K^-1 and electrical conductivity of 2,053.4 S·cm^-1,respectively.The composite film showed an outstanding electro-thermal heating efficiency with the saturated temperature of 315℃and maximum heating rate of 44.9℃·s^-1 at a very low in put voltage of 10 V.The freestandi ng graphe ne composite film with the delicate nano structure desig n has a great pote ntial to be integrated into electro-thermal devices.Xin Meng Tianxing Chen Yao Li Siyuan Liu Hui Pan Yuning Ma Zhixin Chen Yanping Zhang Shenmin Zhu 2019Nano Research2019,12,10:3
5High-temperature superconductivity in sulfur hydride evidenced by alternating-current magnetic susceptibility显示文摘The search for high-temperature superconductivity is one of the research frontiers in physics. In the sulfur hydride system, an extremely high T_c(~200 K) has been recently developed at pressure. However, the Meissner effect measurement above megabar pressures is still a great challenge. Here, we report the superconductivity identification of sulfur hydride at pressure, employing an in situ alternating-current magnetic susceptibility technique. We determine the superconducting phase diagram, finding that superconductivity suddenly appears at 117 GPa and Tcreaches 183 K at 149 GPa before decreasing monotonically with increasing pressure. By means of theoretical calculations, we elucidate the variation of T_c in the low-pressure region in terms of the changing stoichiometry of sulfur hydride and the further decrease in Tcowing to a drop in the electron–phonon interaction parameter λ. 6Effects of water stress on quality and sugar metabolism in'Gala'apple fruit显示文摘Sugar plays an important role in apple fruit development,appearance and quality as well as contributing to a plant’s water stress response.Trehalose and the trehalose biosynthetic metabolic pathways are part of the sugar signaling system in plants,which are important regulator of water stress response in apple.The effect of water stress treatments applied to apple trees and the corresponding effects of ABA on developmental fruit quality were examined for indicators of fruit quality during fruit development.The results indicated that the severe water stress treatment(W2)occurring after the last stage of fruit cell division caused a decrease in the color and size of fruit.The moderate water stress(W1)occurring after the last stage of fruit cell enlargement(S2)caused an increase in the content of fructose and sorbitol while the apple fruit shape was not affected.These changes in sugar are related to the activity of sugar metabolic enzymes.While the enzymatic activity of vacuolar acid invertase(vAINV)was higher,that of sucrose-phosphate synthase(SPS)was lower in water stress treated fruit throughout the developmental period.This indicates that enhanced sucrose degradation and reduced sucrose synthesis leads to an overall reduced sucrose content during times of drought.Thus,water stress reduced sucrose content.Whereas the content of endogenous trehalose and ABA were the highest in water stress treated fruit.A moderate water stress(W1)imposed on apple trees via water restriction(60%–65%of field capacity)after the fruit cell enlargement phase of fruit development yielded sweeter fruit of higher economic value.Hongxia Tao Hanqing Sun Yufei Wang Xin Wang Yanping Guo 2023Horticultural Plant Journal2023,9,1:2
7A durable 4-1BB-based CD19 CAR-T cell for treatment of relapsed or refractory non-Hodgkin lymphoma显示文摘Objective:Previous studies reported that 4-1BB-based CD19 chimeric antigen receptor(CAR)-T cells were more beneficial for the clinical outcomes than CD28-based CAR-T cells,especially the lower incidence rate of severe adverse events.However,the median progression-free survival(mPFS)of 4-1BB-based product Kymriah was shorter than that of CD28-based Yescarta(2.9 months vs.5.9 months),suggesting that Kymriah was limited in the long-term efficacy.Thus,a safe and durable 4-1BB-based CD19 CAR-T needs to be developed.Methods:We designed a CD19-targeted CAR-T(named as IM19)which consisted of an FMC63 scFv,4-1BB and CD3ζintracellular domain and was manufactured into a memory T-enriched formulation.A phase I/II clinical trial was launched to evaluate the clinical outcomes of IM19 in relapsed or refractory(r/r)B cell non-Hodgkin lymphoma(B-NHL).Dose-escalation investigation(at a dose of 5×10^(5)/kg,1×10^(6)/kg and 3×106/kg)was performed in 22 r/r B-NHL patients.All patients received a single infusion of IM19 after 3-day conditional regimen.Results:At month 3,the overall response rate(ORR)was 59.1%,the complete response rate(CRR)was 50.0%.The mPFS was 6 months and the 1-year overall survival rate was 77.8%.Cytokine release syndrome(CRS)occurred in 13 patients(59.1%),with 54.5%of grade 1−2 CRS.Only one patient(4.5%)experienced grade 3 CRS and grade 3 neurotoxicity.Conclusions:These results demonstrated the safety and durable efficacy of a 4-1BB-based CD19 CAR-T,IM19,which is promising for further development and clinical investigation.Zhitao Ying Ting He Shanzhao Jin Xiaopei Wang Wen Zheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Weiping Liu Lijuan Deng Yanping Ding Xuelian Hu Bing Bu Xin’an Lu Yuqin Song Jun Zhu 2022Chinese Journal of Cancer Research2022,34,1:2

9Rationally designed Ta_(3)N_(5)/BiOCl S-scheme heterojunction with oxygen vacancies for elimination of tetracycline antibiotic and Cr(Ⅵ):Performance,toxicity evaluation and mechanism insight显示文摘S-scheme heterojunction photocatalysts have been the“stars”in the field of photocatalysis.Herein,a novel S-scheme heterojunction of Ta_(3)N_(5)/BiOCl with oxygen vacancies(OVs)was fabricated via a facile method.The charge separation and transport mechanism of this Ta_(3)N_(5)/BiOCl S-scheme heterojunction was verified by the analyses of band energy structures,active species,photoelectric behaviors and DFT theoretical calculation.Compared with Ta_(3)N_(5)and BiOCl,the Ta_(3)N_(5)/BiOCl unveils substantially upgraded photocatalytic property under visible light,and the photocatalytic efficiency for removal of tetracycline(TC)and hexavalent chromium(Cr(VI))reaches 89.6%and 91.6%,respectively.The substantial enhancement of the photocatalytic activity is attributed to the synergistic effect of the S-scheme hetero-structure and oxygen vacancies,which improves the visible-light absorption,while promoting the spatial separation of charge carriers with the optimum redox capacity,thereby boosting the production of active species for catalytic reactions.The TC degradation pathway is deduced and the toxicity evolution of TC is appraised using the QSAR method.In a nutshell,this work gives a deep understanding of the photocatalytic mechanism based on Ta_(3)N_(5)/BiOCl as well as presents a newfangled thought for developing highly efficient S-scheme heterojunction photocatalysts for water decontamination.Shijie Li Mingjie Cai Chunchun Wang Yanping Liu Neng Li Peng Zhang Xin Li 2022Journal of Materials Science & Technology2022,,28:2

11Human cytomegalovirus miR-US5-1 inhibits viral replication by targeting Geminin mRNA显示文摘Viruses commonly create favorable cellular conditions for their survival through multiple mechanisms. Micro RNAs(mi RNAs), which function as post-transcriptional regulators, are utilized by human cytomegalovirus(HCMV) in its infection and pathogenesis. In the present study, the DNA replication inhibitor Geminin(GMNN) was identified to be a direct target of hcmv-mi R-US5-1. Overexpression of hcmv-mi R-US5-1 could block the accumulation of GMNN during HCMV infection, and the decrease of GMNN expression caused by hcmv-mi R-US5-1 or GMNN specific si RNA reduced HCMV DNA copies in U373 cells. Meanwhile, ectopic expression of hcmv-mi R-US5-1 and consequent lower expression of GMNN influenced host cell cycle and proliferation. 13HPV16 E6蛋白与hDaxx的相互作用及其对HeLa细胞凋亡的影响(英文)显示文摘Objective: To study the interaction of human papillomavirus type 16 (HPV16 E6) protein and human death domain associated protein (hDaxx) and its effect on apoptosis of HeLa cells to provide the experimental basis for exploring the oncogenic mechanism of HPV16 E6 protein. Methods: Recombinant vector of pGADT7/E6 or pGBKT7/hDaxx was con- structed. The interaction of E6 protein and hDaxx was detected by yeast two-hybrid system. Their expression in yeast was detected by Western blotting. The eukaryotic plasmids of E6 and hDaxx were co-transfected into HeLa cells. Apoptosis was induced by 5-FU. The apoptotic rate was measured by flow cytometry (FCM). Results: E6 protein had intracellular interaction with hDaxx. The apoptotic rate was rising with the increase in the transfection quantity of pcDNA3.1 (-) / hDaxx in pcDNA3.1 (-) /E6 and pcDNA3.1 (-) / hDaxx co-transfected cells. The difference was significant ( P < 0. 01). Conclusion: There is intracellular interaction between HPV16 E6 protein and hDaxx. The over-expression of hDaxx can increase the sensitivity of E6 protein positive HeLa cells to 5-FU. The effect was in a dose dependent manner. 18Early-Holocene Paleo-Tropical Cyclone Activity Inferred from a Sedimentary Sequence in South Yellow Sea,East Asia显示文摘Although tropical cyclones play a critical role in global climate changes,their long-term variations in the past are not well documented.In this article,a sediment core from the South Yellow Sea was studied in order to reveal the influence of tropical cyclones on depositional processes.Integrating the results of radiocarbon dating and sediment grain-size analysis,we show that the studied sequence was deposited during the Holocene and the sedimentary dynamics were stable and at a relatively low level,with a median grain-size range of 5.3-8.7μm.It is found that coarse particles were likely transported by highly dynamic depositional events.Based on the findings,a record of paleo-tropical cyclones was derived for the Early Holocene,and several intervals with a reduced influence of tropical cyclones were identified.In addition,it reveals a good agreement between the grain-size results and the changes in solar activity,monsoonal intensity,and the El Ni?o-Southern Oscillation.Overall,it can be concluded that the influence of tropical cyclones on the sedimentary evolution of the muddy zone of the South Yellow Sea was substantial during the Early Holocene on centennial timescales,and that solar maxima may control the intensity of tropical cyclones via strengthening the walker circulation over the tropical Pacific.Geng Liu Xibin Han Yanping Chen Jun Li Lehui Song Xin Zhou Bangqi Hu Liang Yi 2022Journal of Earth Science2022,33,3:1
