Hes1 Knockdown Exacerbates Ischemic Stroke Following tMCAO by Increasing ER Stress-Dependent Apoptosis via the PERK/ eIF2a/ATF4/CHOP Signaling Pathway显示文摘Apoptosis induced by endoplasmic reticulum(ER)stress plays a crucial role in mediating brain damage after ischemic stroke.Recently,Hes1(hairy and enhancer of split 1)has been implicated in the regulation of ER stress, but whether it plays a functional role after ischemic stroke and the underlying mechanism remain unclear. In this study, using a mouse model of ischemic stroke via transient middle cerebral artery occlusion(tMCAO),we found that Hes1 was induced following brain injury, and that siRNA-mediated knockdown of Hes1 increased the cerebral infarction and worsened the neurological outcome, suggesting that Hes1 knockdown exacerbates ischemic stroke. In addition,mechanistically,Hes1 knockdown promoted apoptosis and activated the PERK/eIF2a/ATF4/CHOP signaling pathway after tMCAO.These results suggest that Hes1 knockdown promotes ER stress-induced apoptosis.Furthermore, inhibition of PERK with the specific inhibitor GSK2606414 markedly attenuated the Hes1 knockdown-induced apoptosis and the increased cerebral infarction as well as the worsened neurological outcome following tMCAO,implying that the protection of Hes1 against ischemic stroke is associated with the amelioration of ER stress via modulating the PERK/eIF2a/ATF4/CHOP signaling pathway.Taken together, these results unveil the detrimental role of Hes1 knockdown after ischemic stroke and further relate it to the regulation of ER stress-induced apoptosis, thus highlighting the importance of targeting ER stress in the treatment of ischemic stroke. | Yueyong Li Yingjun Zhang Huangde Fu Huadong Huang Qifeng Lu Houji Qin Yingning Wu Huatuo Huang Guizhen Mao Zhongheng Wei Pinhu Liao | 2020 | Neuroscience Bulletin2020,36,2: | 12
Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity. However, cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol, plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling, thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover, genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression. While previously established as a hypercholesterolemia target, this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12
Shuganjieyu capsule increases neurotrophic factor expression in a rat model of depression显示文摘Shuganjieyu capsule has been approved for clinical treatment by the State Food and Drug Administration of China since 2008. In the clinic, Shuganjieyu capsule is often used to treat mild to moderate depression. In the rat model of depression established in this study, Shuganjieyu capsule was administered intragastrically daily before stress. Behavioral results confirmed that depressive symptoms lessened after treatment with high-dose(150 mg/kg) Shuganjieyu capsule. Immunohistochemistry results showed that high-dose Shuganjieyu capsule significantly increased phosphorylation levels of phosphorylation cyclic adenosine monophosphate response element binding protein and brain-derived neurotrophic factor expression in the medial prefrontal cortex and hippocampal CA3 area. Overall, our results suggest that in rats, Shuganjieyu capsule effectively reverses depressive-like behaviors by increasing expression levels of neurotrophic factors in the brain. | Jinhua Fu Yingjin Zhang Renrong Wu Yingjun Zheng Xianghui Zhang Mei Yang Jingping Zhao Yong Liu | 2014 | Neural Regeneration Research2014,9,5: | 10
Fabrication of a hydroxyapatite-PDMS microfluidic chip for bone-related cell culture and drug screening显示文摘Bone is an important part of the human body structure and plays a vital role in human health.A microfluidic chip that can simulate the structure and function of bone will provide a platform for bone-related biomedical research.Hydroxyapatite(HA),a bioactive ceramic material, has a similar structure and composition to bone mineralization products. In this study,we used HA as a microfluidic chip component to provide a highly bionic bone environment.HA substrates with different microchannel structures were printed by using ceramic stereolithography(SLA)technology, and the minimum trench width was 50μm.The HA substrate with microchannels was sealed by a thin polydimethylsiloxane(PDMS)layer to make a HA-PDMS microfluidic chip.Cell culture experiments demonstrated that compared with PDMS,HA was more conducive to the proliferation and osteogenic differentiation of the human foetal osteoblast cell line(hFOB).In addition, the concentration gradient of the model drug doxorubicin hydrochloride(DOX)was successfully generated on a Christmas tree structure HA-PDMS chip, and the half maximal inhibitory concentration(IC50)of DOX was determined. The findings of this study indicate that the HA-PDMS microfluidic chip has great potential in the field of high-throughput bonerelated drug screening and bone-related research. | Qiangqiang Tang Xiaoyu Li Chen Lai Lei Li Hongkai Wu Yingjun Wang Xuetao Shi | 2021 | Bioactive Materials2021,6,1: | 5
Theoretical and simulation research of hydrodynamic instabilities in inertial-confinement fusion implosions显示文摘Inertial fusion energy(IFE)has been considered a promising, nearly inexhaustible source of sustainable carbon-free power for the world's energy future. It has long been recognized that the control of hydrodynamic instabilities is of critical importance for ignition and high-gain in the inertial-confinement fusion(ICF)hot-spot ignition scheme. In this mini-review,we summarize the progress of theoretical and simulation research of hydrodynamic instabilities in the ICF central hot-spot implosion in our group over the past decade. In order to obtain sufficient understanding of the growth of hydrodynamic instabilities in ICF,we first decompose the problem into different stages according to the implosion physics processes. The decomposed essential physics processes that are associated with ICF implosions, such as Rayleigh-Taylor instability(RTI),Richtmyer-Meshkov instability(RMI),Kelvin-Helmholtz instability(KHI),convergent geometry effects,as well as perturbation feed-through are reviewed.Analytical models in planar, cylindrical, and spherical geometries have been established to study different physical aspects, including density-gradient,interface-coupling,geometry,and convergent effects. The influence of ablation in the presence of preheating on the RTI has been extensively studied by numerical simulations. The KHI considering the ablation effect has been discussed in detail for the first time.A series of single-mode ablative RTI experiments has been performed on the Shenguang-Ⅱlaser facility. The theoretical and simulation research provides us the physical insights of linear and weakly nonlinear growths, and nonlinear evolutions of the hydrodynamic instabilities in ICF implosions, which has directly supported the research of ICF ignition target design. The ICF hot-spot ignition implosion design that uses several controlling features, based on our current understanding of hydrodynamic instabilities,to address shell implosion stability, has been briefly described, several of which are novel. | LiFeng Wang WenHua Ye XianTu He JunFeng Wu ZhengFeng Fan Chuang Xue HongYu Guo WenYong Miao YongTeng Yuan JiaQin Dong Guo Jia Jing Zhang YingJun Li Jie Liu Min Wang YongKun Ding WeiYan Zhang | 2017 | Science China(Physics,Mechanics & Astronomy)2017,60,5: | 4
Development of matrix solid-phase dispersion method for the extraction of short-chain chlorinated paraffins in human placenta显示文摘Chlorinated paraffins(SCCPs) are widely used worldwide, and they can be released into the environment during their production, transport, usage and disposal, which pose potential risks for human health. In this work, an efficient, reliable and rapid pretreatment method based on matrix solid-phase dispersion(MSPD) was developed for the analysis of short-chain CPs(SCCPs) in human placenta by gas chromatograph-electron capture negative ion low-resolution mass spectrometry(GC-ECNI-LRMS) and gas chromatography–quadrupole time-of-flight mass spectrometry(GC–QTOF-HRMS). The MSPD-relevant parameters including dispersing sorbent,sample-to-sorbent mass ratio, and elution solvent were optimized using the orthogonal test.Silica gel was found to be the optimal dispersing sorbent among the selected matrices. Under the optimal conditions, 44% acidic silica gel can be used as the co-sorbent to remove lipid and eluted by the mixture of hexane and dichloromethane(7:3, V/V). The spiked recoveries of the optimized method were 77.4% and 91.4% for analyzing SCCPs in human placenta by GC-ECNI-LRMS and GC–QTOF-HRMS, and the corresponding relative standard deviations were10.2% and 5.6%, respectively. The method detection limit for the total SCCPs was 36.8 ng/g(dry weight, dw) and 19.2 ng/g(dw) as measured by GC-ECNI-LRMS and GC–QTOF-HRMS,respectively. The concentrations of SCCPs in four human placentas were in the range of | Ying Wang Wei Gao Jing Wu Huijin Liu Yingjun Wang Yawei Wang Guibin Jiang | 2017 | Journal of Environmental Sciences2017,29,12: | 4
A wave of Foxp3^(+) regulatory T cell accumulation in the neonatal liver plays unique roles in maintaining self-tolerance显示文摘Newborn animals require tightly regulated local and systemic immune environments to govern the development and maturation of multiple organs/tissues even though the immune system itself is far from mature during the neonatal period.Regulatory T cells(Tregs)are essential for maintaining immune tolerance/homeostasis and modulating inflammatory responses. The features of Tregs in the neonatal liver under steady-state conditions are not well understood. The present study aimed to investigate the phenotype, functions, and significance of neonatal Tregs in the liver. We found a wave of thymus-derived Treg influx into the liver during 1–2 weeks of age.Depletion of these Tregs between days 7 and 11 after birth rapidly resulted in Th1-type liver inflammation and metabolic disorder.More Tregs in the neonatal liver than in the spleen underwent MHC II-dependent activation and proliferation, and the liver Tregs acquired stronger suppressive functions. The transcriptomic profile of these neonatal liver Tregs showed elevated expression of PPARγand T-bet and features of Tregs that utilize lipid metabolic machinery and are capable of regulating Th1 responses. The accumulation of Tregs with unique features in the neonatal liver is critical to ensure self-tolerance and liver maturation. | Mingyang Li Weijia Zhao Yifan Wang Lixue Jin Gaowen Jin Xiuyuan Sun Wei Wang Ke Wang Xi Xu Jie Hao Rong Jin Wenxian Fu Ying Sun Yingjun Chang Xiaojun Huang Xuyu Zhou Hounan Wu Kunshan Zhang Qing Ge | 2020 | Cellular & Molecular Immunology2020,17,5: | 4
Sustainable management of Chinese grasslands—issues and knowledge显示文摘China has almost 400 Mhm^2 of grasslands,90% of which is considered degraded to varying degrees,on which 16 million herders depend for their livelihoods and many more indirectly, along the value-adding chain.Since 1950, average stocking rates across China have increased 4-fold. National policies have focused over recent decades on finding ways to rehabilitate the degraded grasslands, to sustain livestock production from them, and to improve the livelihoods of herder households, who are among the poorest people in China. A large collaborative program commenced in the early 2000 s to help find solutions to the sustainable management of grasslands. This paper summarizes key findings of many research projects, identifies where knowledge is weak and argues that the successful rehabilitation of grasslands will also require policies that provide incentives and support for herders as they move from a focus on survival to a focus on production of higher quality products, for which consumers are increasingly willing to pay. A key focus is to emphasize the improvement of animal production per head. When this is done, it naturally leads to lower stocking rates, which in turn provides the opportunities for grasslands to recover. Across a range of experiments, farm demonstrations and analyses using models, in Inner Mongolia and Gansu, a consistent result has been that a 50% reduction in stocking rates, improves net household income and starts the process of grassland rehabilitation.Rather than focusing on stocking rates, better management of grasslands could be achieved by maintaining the grasslands above critical values for herbage mass, values that help optimize botanical composition, reduce soil erosion, optimize animal growth rates and aid ecosystem functions. Managing to critical values for herbage mass is likely to be more effective than efforts to calculate sustainable stocking rates. An early summer rest is valuable for aiding grassland rehabilitation and summer productivity, but a total grazing ban(typically for 5 years)may not achieve its aims as evidence shows it may take 10–15 years to achieve a better grassland state. Lessdesirable plant species often increase in degraded grasslands that are rested and grazing can help manage those species. Surveys of herders indicate they have very mixed views on the benefits of total grazing bans that are unlikely to rehabilitate grasslands to an ideal botanical composition. The current objective is to work with grasslands that herders now have and optimize the existing composition.Grazing grasslands in winter results primarily in weight loss by animals and there is now evidence of how winter grazing reduces grassland growth in the next summer. It is better to keep animals in well-built sheds and feed them better, improved feeding through the cold months is required. In addition to the application of results from national programs designed to improve grasslands, it will be important to train herders as they move from survival to production, to foster the development of better markets for their livestock products, to devise better financial support for herder businesses and to revise land tenure arrangements so that herders can expand the area of land they graze on better terms than apply at present. | David KEMP Guodong HAN Fujiang HOU Xiangyang HOU Zhiguo LI Yi SUN Zhongwu WANG Jianping WU Xiaoqing ZHANG Yingjun ZHANG Xuyin GONG | 2018 | Frontiers of Agricultural Science and Engineering2018,5,1: | 3
Concept and Research Framework for Coordinated Situation Awareness and Active Defense of Cyber-physical Power Systems Against Cyber-attacks显示文摘Due to the tight coupling between the cyber and physical sides of a cyber-physical power system(CPPS),the safe and reliable operation of CPPSs is being increasingly impacted by cyber security. This situation poses a challenge to traditional security defense systems, which considers the threat from only one side,i.e.,cyber or physical. To cope with cyberattacks, this paper reaches beyond the traditional one-side security defense systems and proposes the concept of cyber-physical coordinated situation awareness and active defense to improve the ability of CPPSs.An example of a regional frequency control system is used to show the validness and potential of this concept. Then, the research framework is presented for studying and implementing this concept. Finally, key technologies for cyber-physical coordinated situation awareness and active defense against cyber-attacks are introduced. | Ming Ni Manli Li Jun’e Li Yingjun Wu Qi Wang | 2021 | Journal of Modern Power Systems and Clean Energy2021,9,3: | 3

Wind-proof and sand-fixing effects of Artemisia ordosica with different coverages in the Mu Us Sandy Land, northern China显示文摘Wind erosion is a key global environmental problem and has many adverse effects. The Mu Us Sandy Land of northern China is characterized by an arid climate, where vegetation patches and bare sand patches are usually distributed mosaically, and aeolian activities occur frequently.Vegetation plays a significant role in controlling wind erosion.Artemisia ordosica is the most dominant native plant species in the Mu Us Sandy Land.It is urgent to study the wind-proof and sand-fixing effects of Artemisia ordosica in the Mu Us Sandy Land. This study analyzed the wind-proof and sand-fixing effects of Artemisia ordosica based on the field data of wind regimes, aeolian sediment transport, and surface change of Artemisia ordosica plots with four coverages(denoted as site A,site B,site C,and site D)in the Mu Us Sandy Land during the period from 1 June 2018 to 29 June 2019.The coverages of Artemisia ordosica at site A,site B,site C,and site D were 2%,16%,29%,and 69%,respectively.The annual average wind speeds at 2.0 m height above the ground for site A,site B,site C,and site D were 3.47,2.77,2.21,and 1.97 m/s,respectively.The annual drift potentials were 193.80,69.72,15.05,and 6.73 VU at site A,site B,site C,and site D,respectively. The total horizontal aeolian sediment fluxes during the period from 2-3 June 2018 to 6 June 2019 at site A,site B,site C,and site D were 4633.61,896.80,10.54,and 6.14 kg/m,respectively.Site A had the largest surface changes, and the surface changes at site B were significantly weaker than those at site A,whereas the surface changes at site C and site D were minimal. The results indicated that Artemisia ordosica significantly reduced the wind speed, drift potential, aeolian sediment transport, and surface changes. The higher the coverage of Artemisia ordosica is,the more obvious the effects of wind-proof and sand-fixing.Wind erosion would be effectively controlled in the Mu Us Sandy Land if the coverage of Artemisia ordosica is greater than 29%.These results provide a scientific basis for evaluating the ecosystem service function of Artemisia ordosica and the vegetation protection and construction projects in the Mu Us Sandy Land. | PANG Yingjun WU Bo JIA Xiaohong XIE Shengbo | 2022 | Journal of Arid Land2022,14,8: | 2

DESIGN OF NOVEL HIGH PRESSURE-RESISTANT HYDROTHERMAL FLUID SAMPLE VALVE显示文摘Sampling study is an effective exploration method, but the most extreme environments of hydrothermal vents pose considerable engineering challenges for sampling hydrothermal fluids. Moreover, traditional sampler systems with sample valves have difficulty in maintaining samples in situ pressure. However, decompression changes have effect on microorganisms sensitive to such stresses. To address the technical difficulty of collecting samples from hydrothermal vents,a new bidirectional high pressure-resistant sample valve with balanced poppet was designed. The sample valve utilizes a soft high performance plastic 'PEEK' as poppet. The poppet with inapposite dimension is prone to occur to plastic deformation or rupture for high working pressure in experiments. To address this issue, based on the finite element model, simulated results on stress distribution of the poppet with different structure parameters and preload spring force were obtained. The static axial deformations on top of the poppet were experimented. The simulated results agree with the experimental results. The new sample valve seals well and it can withstand high working pressure. | LIU Wei YANG Canjun WU Shijun XIE Yingjun CHEN Ying | 2008 | Chinese Journal of Mechanical Engineering2008,21,1: | 1
