1Endothelial CDS2 deficiency causes VEGFA-mediated vascular regression and tumor inhibition显示文摘The response of endothelial cells to signaling stimulation is critical for vascular morphogenesis,homeostasis and function.Vascular endothelial growth factor-a(VEGFA)has been commonly recognized as a pro-angiogenic factor in vertebrate developmental,physiological and pathological conditions for decades.Here we report a novel finding that genetic ablation of CDP-diacylglycerol synthetase-2(CDS2),a metabolic enzyme that controls phosphoinositide recycling,switches the output of VEGFA signaling from promoting angiogenesis to unexpectedly inducing vessel regression.Live imaging analysis uncovered the presence of reverse migration of the angiogenic endothelium in cds2 mutant zebrafish upon VEGFA stimulation,and endothelium regression also occurred in postnatal retina and implanted tumor models in mice.In tumor models,CDS2 deficiency enhanced the level of tumorsecreted VEGFA,which in-turn trapped tumors into a VEGFA-induced vessel regression situation,leading to suppression of tumor growth.Mechanistically,VEGFA stimulation reduced phosphatidylinositol(4,5)-bisphosphate(PIP2)availability in the absence of CDS2-controlled-phosphoinositide metabolism,subsequently causing phosphatidylinositol(3,4,5)-triphosphate(PIP3)deficiency and F0X01 activation to trigger regression of CDS2-null endothelium.Thus,our data indicate that the effect of VEGFA on vasculature is context-dependent and can be converted from angiogenesis to vascular regression.Wencao Zhao Le Cao Hanru Ying Wenjuan Zhang Dantong Li Xiaolong Ziiu Wenzhi Xue Shuang Wu Mengye Cao Cong Fu Haonan Qi Yimei Hao Yun-Chi Tang Jun Qin Tao PZhong Xiaoxi Lin Luyang Yu Xuri Li Lin Li Dianqing Wu Weijun Pan 2019Cell Research2019,29,11:9
2Mapping QTLs for seed yield and drought susceptibility index in soybean (Glycine max L.) across different environments显示文摘Drought stress has long been a major constraint in maintaining yield stability of soybean(Glycine max(L.) Merr.) in rainfed ecosystems.The identification of consistent quantitative trait loci(QTL) involving seed yield per plant(YP) and drought susceptibility index(DSI) in a population across different environments would therefore be important in molecular marker-assisted breeding of soybean cultivars suitable for rainfed regions.The YP of a recombinant line population of 184 F2:7:11 lines from a cross of Kefeng1 and Nan-nong1138-2 was studied under water-stressed(WS) and well-watered(WW) conditions in field(F) and greenhouse(G) trials,and DSI for yield was calculated in two trials.Nineteen QTLs associated with YP-WS and YP-WW,and 10 QTLs associated with DSI,were identified.Comparison of these QTL locations with previous findings showed that the majority of these regions control one or more traits related to yield and other agronomic traits.One QTL on molecular linkage group(MLG) K for YP-F,and two QTLs on MLG C2 for YP-G,remained constant across different water regimes.The regions on MLG C2 for YP-WW-F and MLG H for YP-WS-F had a pleiotropic effect on DSI-F,and MLG A1 for YP-WS-G had a pleiotropic effect on DSIG.The identification of consistent QTLs for YP and DSI across different environments will significantly improve the efficiency of selecting for drought tolerance in soybean.Weijun Du Min Wang Sanxiong Fu Deyue Yu 2009Journal of Genetics and Genomics2009,36,12:8

4Detection of Quantitative Trait Loci for Yield and Drought Tolerance Traits in Soybean Using a Recombinant Inbred Line Population显示文摘To investigate the genetic basis of drought tolerance in soybean(Glycine max L.Merr.) a recombinant inbred population with 184 F2:7:11 lines developed from a cross between Kefeng1(drought tolerant) and Nannong1138-2(drought sensitive) were tested under water-stressed and well-watered conditions in field and greenhouse trials.Traits measured included leaf wilting coefficient,excised leaf water loss and relative water content as indicators of plant water status and seed yield.A total of 40 quantitative trait loci(QTLs) were identified:17 for leaf water status traits under drought stress and 23 for seed yield under well-watered and drought-stressed conditions in both field and greenhouse trials.Two seed yield QTLs were detected under both well-watered and drought-stressed conditions in the field on molecular linkage group H and D1b,while two seed yield QTLs on molecular linkage group C2 were found under greenhouse conditions.Several QTLs for traits associated with plant water status were identified in both field and greenhouse trials,including two leaf wilting coefficient QTLs on molecular linkage group A2 and one excised leaf water loss QTL on molecular linkage group H.Phenotypic correlations of traits suggested several QTLs had pleiotropic or location-linked associations.These results will help to elucidate the genetic basis of drought tolerance in soybean,and could be incorporated into a marker-assisted selection breeding program to develop high-yielding soybean cultivars with improved tolerance to drought stress.Weijun Du Deyue Yu Sanxiong Fu 2009Journal of Integrative Plant Biology2009,51,9:7

8Limited Spatial Transferability of the Relationships Between Kriging Variance and Soil Sampling Spacing in Some Grasslands of Ireland:Implications for Sampling Design显示文摘Sampling plays an important role in acquiring precise soil information required in modern agricultural production worldwide, which determines both the cost and quality of final soil mapping products. For sampling design, it has been proposed possibile to transfer the relationships between kriging variance and sampling grid spacing from an area with existing information to other areas with similar soil-forming environments. However, this approach is challenged in practice because of two problems: i) different population vaxiograms among similar areas and ii) sampling errors in estimated variograms. This study evaluated the effects of these two problems on the transferability of the relationships between kriging variance and sampling grid spacing, by using spatial data simulated with three variograms and soil samples collected from four grasslands in Ireland with similar soil-forming environments. Results showed that the variograms suggested by different samples collected with the same grid spacing in the same or similar areas were different, leading to a range of mean kriging variance (MKV) for each grid spacing. With increasing grid spacing, the variation of MKV for a specific grid spacing increased and deviated more from the MKV generated using the population variograms. 10Integrated Fertilization Regimes Boost Heavy Metals Accumulation and Biomass of Sedum alfredii Hance显示文摘The hyperaccumulator Sedum alfredii Hance(S.alfredii)may be employed for zinc(Zn)and cadmium(Cd)-polluted soil remediation.However,the low phytoremediation efficiency,related to the low biomass production,limits its use with that purpose.In this experiment,nitrogen(N),phosphorus(P),and potassium(K)fertilizers,and organic manure were applied to investigate the phytoremediation ability of S.alfredii.Hydroponic and pot experiments were conducted using Zn-Cd polluted soil.The hydroponic experiment indicated that appropriate fertilizer application could increase(p<0.05)the amount of accumulated Zn and Cd in S.alfredii.When N supply ranged from 0.5 to 2.5 mmol L^(−1),it could improve growth and accumulation of Zn and Cd in whole plants of S.alfredii.The 1 mmol L^(−1) N was an optimal N dosage for shoot biomass production and Cd accumulation in shoots,while the 2.5 mmol L^(−1) was an optimal N dosage for Zn accumulation in shoots.Both low(<0.05 mmol L^(−1))and high(>0.8 mmol L^(−1))P supply decreased growth,and Zn/Cd accumulation in whole plants of the studied species.The 0.1 mmol L^(−1) P was an optimal dosage for S.alfredii biomass production and Zn/Cd accumulation in shoots.The supply levels within the range from 0.3 to 1 mmol L^(−1) K could significantly improve the biomass production of S.alfredii and its capability to accumulate Zn and Cd in the biomass.The 0.5 mmol L^(−1) K was an optimal dosage for the whole biomass production and Zn accumulation in shoots,while the 1 mmol L^(−1) was an optimal K dosage for Zn accumulation in shoots,which was 17.2%higher than the control.Moreover,the soil pot experiment showed that the combination of organic(fermented manure)and inorganic fertilizers made significant effects on the Zn and Cd-polluted soil remediation by S.alfredii.These effects varied,however,with the application of different proportions of N,P,K and organic matter.The Zn accumulation by S.alfredii reached the highest efficiency ability under the highest fertilizer mixing rate(N:50 mg kg^(−1),P:40 mg kg^(−1),K:100 mg kg^(−1),organic matter:1%).Even more,S.alfredii showed the strongest ability to accumulate Cd with a lower fertilizer mixing rate(N:25mg kg^(−1),P:20mg kg^(−1),K:50 mg kg^(−1),organic matter:0.5%).Kangning Zhou Yingying Zhang Jiasen Wu Chunying Dou Zihao Ye Zhengqian Ye Weijun Fu 2021Phyton-International Journal of Experimental Botany2021,90,4:2
11溃疡性结肠炎内镜严重度指数(UCEIS)预测溃疡性结肠炎患者行储袋成形术的截点值:多中心长期随访研究结果显示文摘背景:全结直肠切除回肠储袋肛管吻合(IPAA)是溃疡性结肠炎(UC)患者的首选外科治疗术式。溃疡性结肠炎内镜严重度指数(UCEIS)用以预测UC患者的IPAA手术需求,目前尚缺乏数据。本研究旨在建立明确UCEIS预测UC患者IPAA手术需求的截点值。方法:对1986年6月至2020年3月间两个中心收治的UC患者进行回顾性分析。基于入院后首次结肠镜检查结果进行UCEIS评分。采用受试者操作特征曲线分析确定UCEIS评分预测IPAA需求的截点值。结果:293例UC患者纳入研究,中位UCEIS为4分。13年随访期间,80例(28.3%)患者接受外科治疗,其中75例(93.8%)行IPAA手术,5例(6.2%)行次全结肠切除加永久性造口。采用6分作为截点值,UCEIS对IPAA手术需求的预测价值最高,曲线下面积为0.769(P<0.001),灵敏度和特异度分别为72.0%和81.8%。UCEIS≥6是需要行IPAA手术(P<0.001)和恶性转化(P=0.010)的独立预测因素。UCEIS≥6的患者无IPAA生存时间显著短缩(P<0.001)。结论:UCEIS≥6或可作为IPAA手术决策的依据,推荐对这些UC患者行手术治疗以减少恶性转化的发生率。Weimin Xu Weijun Ou Jihong Fu Yubei Gu Long Cui Jie Zhong Peng Du 2021Gastroenterology Report2021,9,5:1

15Design, fabrication, and characterization of a highly nonlinear few-mode fiber显示文摘We present the design, fabrication, and characterization of a highly nonlinear few-mode fiber(HNL-FMF) with an intermodal nonlinear coefficient of 2.8 W · km-1, which to the best of our knowledge is the highest reported to date. The graded-index circular core fiber supports two mode groups(MGs) with six eigenmodes and is highly doped with germanium. This breaks the mode degeneracy within the higher-order MG, leading to different group velocities among corresponding eigenmodes. Thus, the HNL-FMF can support multiple intermodal four-wave mixing processes between the two MGs at the same time. 16A small molecule UPR modulator for diabetes identified by high throughput screening显示文摘Unfolded protein response(UPR) is a stress response that is specific to the endoplasmic reticulum(ER).UPR is activated upon accumulation of unfolded(or misfolded) proteins in the ER's lumen to restore protein folding capacity by increasing the synthesis of chaperones.In addition,UPR also enhances degradation of unfolded proteins and reduces global protein synthesis to alleviate additional accumulation of unfolded proteins in the ER.Herein,we describe a cell-based ultra-high throughput screening(uHTS) campaign that identifies a small molecule that can modulate UPR and ER stress in cellular and in vivo disease models.Using asialoglycoprotein receptor 1(ASGR) fused with Cypridina luciferase(CLuc) as reporter assay for folding capacity,we have screened a million small molecule library and identified APC655 as a potent activator of protein folding,that appears to act by promoting chaperone expression.Furthermore,APC655 improved pancreatic β cell viability and insulin secretion under ER stress conditions induced by thapsigargin or cytokines.APC655 was also effective in preserving β cell function and decreasing lipid accumulation in the liver of the leptin-deficient(ob/ob) mouse model.These results demonstrate a successful uHTS campaign that identified a modulator of UPR,which can provide a novel candidate for potential therapeutic development for a host of metabolic diseases.Valeria Marrocco Tuan Tran Siying Zhu Seung Hyuk Choi Ana M.Gamo Sijia Li Qiangwei Fu Marta Diez Cunado Jason Roland Mitch Hull Van Nguyen-Tran Sean Joseph Arnab K.Chatterjee Nikki Rogers Matthew S.Tremblay Weijun Shen 2021Acta Pharmaceutica Sinica B2021,11,12:1

18Distributed curvature sensing based on a bending loss-resistant ring-core fiber显示文摘A theoretical and experimental study on curvature sensing using a Brillouin optical time-domain analyzer based on the ring-core fiber(RCF)is reported.The Brillouin gain spectrum of the RCF is investigated,and the Brillouin frequency shift(BFS)dependence on temperature and strain is calibrated.We theoretically analyze the fiber bending-induced BFS and peak Brillouin gain variation for the RCF through a numerical simulation method,and the RCF is revealed to have a high curvature sensitivity.Distributed curvature sensing is successfully demonstrated,with the bending radius ranging from 0.5 to 1.5 cm,corresponding to a BFS variation from 32.90 to7.81 MHz.The RCF takes advantage of great bending loss resistance,and the maximum macrobending loss at the extreme bending radius of 0.5 cm is less than 0.01 d B/turn.Besides,the peak Brillouin gain of the RCF is discovered to vary significantly in response to fiber bending,which is expected to be another parameter for distributed curvature determination.The results imply that the RCF is a promising candidate for highly sensitive distributed curvature measurement,especially in sharp bending circumstances.LI SHEN HAO WU CAN ZHAO LEI SHEN RUI ZHANG WEIJUN TONG SONGNIAN FU MING TANG 2020Photonics Research2020,8,2:1

20Development and validation of prognostic implications of chromosome abnormalities algorithm for newly diagnosed multiple myeloma显示文摘Fluorescence in situ hybridization(FISH)evaluation is essential for initial risk stratification in multiple myeloma(MM).The presence of specific cytogenetic abnormalities(CA)confers a heterogeneity impact on prognosis.However,the cutoff values among different centers are not uniform.Therefore,we conduct this study to better predict the prognosis of newly diagnosed MM patients based on FISH results.The Kaps method was used to calculate the chromosomal abnormal cutoff values.A total of 533 participants were included in the study.The best cutoff value of overall survival were as follows:17p20.1%,13q85%,1q21+39%,t(11;14)55.5%,t(14;16)87%,and t(4;14)53.5%.The survival analysis showed that 17pand 1q21+were the independent factors affecting both OS and progress free survival(PFS)among CA.The analysis based on the cutoff value obtained by Kaps suggested that 13q,t(14;16),17p,and 1q21+were independent factors affecting OS among CA;t(14;16),17p,and 1q21+were independent factors affecting PFS among CA.The prognostic model was constructed by the Kaps method with the Harrell concordance index(c-index)at 0.719(95%CI,0.683–0.756;corrected 0.707),which was higher than that calculated by the European Myeloma Network criteria(0.714;95%CI,0.678–0.751;corrected 0.696).In conclusion,chromosomal abnormalities in different proportions and combinations can affect the prognosis ofMMpatients.Therefore,effective criteria should be formulated to evaluate the prognosis ofMMpatients better.Tiancheng Luo Wanting Qiang Jing Lu Haiyan He Jin Liu Lu Li Hua Jiang Weijun Fu Juan Du 2021Blood Science2021,3,3:0
