1Different contributions of STAT3, ERK1/2, and PI3-K signaling to cardiomyocyte hypertrophy by cardiotrophin-1显示文摘AIM: To assess the contribution of signal transducer and activator of transcription 3(JAK-STAT3) pathway, extracellular signal-regulated kinases1/2 (ERK1/2) pathway, and phosphatidylinositol 3-kinase (PI3-K) pathway to cardiomyocytes hypertrophy induced by cardiotrophin-1 (CT-1), a new member of interleukin-6 (IL-6) family of cytokines. METHODS: STAT3, ERK1/2, and PI3-K were assessed by Western blot analysis. Activity of ERK1/2 was also confirmed by in-gel kinase assay. Hypertrophy of cardiomyocyte was evaluated by [^3H]leucine incorporation and cellular protein-to-DNA ratio. RESULTS: CT-1 simultaneously activated phosphorylation of STAT3, ERK1/2, and PI3-K in rat cardiomyocytes. Parthenolide, an inhibitor of STAT, suppressed CT-1-induced [^3H]leucine incorporation by 88.3% and protein-to-DNA ratio by 75.0%. U0126, an MEK1/2 inhibitor, increased CT-1-induced the phosphorylation of STAT3 in a dose-dependent manner and, consistently, augmented CT-1-induced increase in [^3H]leucine incorporation and cellular protein-to-DNA ratio by 17.6% and 16.3%, respectively. Wortmannin, a PI3-K inhibitor, did not influence CT-1-induced [^3H]leucine incorporation and cellular protein-to-DNA ratio. CONCLUSION: The hypertrophic effect of CT-1 was essentially mediated by STAT3, independent of PI3-K, and negatively regulated by ERK1/2 via inhibiting the phosphorylation of STAT3. 3Novel hydrophobic associated polymer based nano-silica composite with core–shell structure for intelligent drilling fluid under ultra-high temperature and ultra-high pressure显示文摘Micro-nano-based drilling fluid has attracted a strong interest due to its attractive properties, and micro-nano composite materials have great potential for developing intelligent drilling fluid. In this study a novel hydrophobic associated polymer based nano-silica composite with core–shell structure was prepared and characterized by PSD, SEM, TEM and ESEM. The results showed that the composite, as a micro-nano drilling fluid additive, possessed excellent properties such as thermal stability, rheology, fluid loss and lubricity. 5Epigenetic regulation of phosphodiesterase 4d in restrictive cardiomyopathy mice with cTnI mutations显示文摘Epigenetic regulations play an important role in disease development.In this study,we have investigated epigenetic regulations in restrictive cardiomyopathy mice with cTnI 193 His mutation.Our results demonstrated that phosphodiesterase(PDEs)4d was down-regulated in the heart of these mice.Further studies showed that the epigenetic modifications were associated with enhanced acetylation of histone 3 lysine 4 and lysines 9,whereas tri-methylation of histone 3 lysine 4,were decreased in histones near the PDE4d gene promoter regions.The binding levels of histone transmethylase SMYD1 and histone deacetylase HDAC1 were increased in the gene promoter regions in cTnI193 His transgenic hearts.Using immune-fluorescent labeling we found an evidence of cTnI existence in the nucleus of cardiomyocytes and Western blotting further confirmed that both wild type and mutated cTnI could be detected in the cell nucleus of the hearts.Furthermore,an interaction between cTnI and SMYD1,or cTnI and HDAC1 was observed.Overexpression of the mutated cTnI in cultured cardiomyocytes reduced the expression of PDE4d.Our data suggest that the decrease of PDE4d expression in RCM mice caused by cTnI mutations may be related to epigenetic regulation,i.e.,histone acetylation and methylation,and cTnI might be involved in this procedure via an interaction with HDAC1 and SMAD1 in the hearts.Weian Zhao Xiaoqi Wu Zhiyuan Wang Bo Pan Lifei Liu Lingjuan Liu Xupei Huang Jie Tian 2020Science China(Life Sciences)2020,63,4:4
6ERK1/2 contributes negative regulation to STAT3 activity in HSS-transfected HepG2 cells显示文摘Signal transducer and activator of transcription 3 (STAT3) is a recently characterized transcription factor which is essential to liver regeneration. We have previously reported that hepatic stimulator substance (HSS), a novel growthpromoting substance, phosphorylated the epidermal growth factor (EGF) receptors and activated downstream RasMAP kinase (extracellular signal-regulated kinases, ERK1/2) cascade. However, whether HSS signal is related to STAT3pathway remains unclear. The present study is aiming to explore the regulatory effect of activation of ERK1/2 evoked by HSS on STAT3 phosphorylation and STAT3 signaling. Human hepatoma cell line HepG2 was stably transfected with HSS cDNA and HSS expression was measured by Northern blot. The results showed that the transfection of HSS into HepG2 resulted in remarkable increase in cellular proliferation as compared with the non-transfected cells, and it was further proved that the cellular proliferation in the HSS-transfected cells was related to ERK1/2 activation. Treatment of the cells with 50 μM of PD98059, an ERK1/2 specific upstream inhibitor, resulted in ERK1/2 inactivation completely.Inhibition of ERK1/2 allowed the tyrosine of STAT3 to be phosphorylated in a dose-dependent manner to PD98059.Furthermore, transient transfection of STAT3 mutant (STAT3S727A) into HSS-bearing cells could remarkably reverse the inhibitory effect of ERK1/2 on STAT3 phosphorylation. 7An acid-triggered porphyrin-based block copolymer for enhanced photodynamic antibacterial efficacy显示文摘Bacterial infection, especially multidrug-resistant(MDR) bacterial infection has threatened public health drastically. Here, we fabricate an 'acid-triggered' nanoplatform for enhanced photodynamic antibacterial activity by reducing the aggregation of photosensitizers(PSs) in bacterial acidic microenvironment. Specifically, a functional amphiphilic block copolymer was first synthesized by using a pH-sensitive monomer, 2-(diisopropylamino) ethyl methacrylate(DPA) and porphyrin-based methacrylate(TPPC6MA) with poly(oligo(ethylene glycol) methyl ether methacrylate)(POEGMA) as the macromolecular chain transfer agent, and POEGMA-b-[PDPA-co-PTPPC6MA] block copolymer was further self-assembled into spherical nanoparticles(PDPA-TPP). PDPA-TPP nanoparticles possess an effective electrostatic adherence to negatively charged bacterial cell membrane, since they could rapidly achieve positive charge in acidic bacterial media. Meanwhile, the acid-triggered dissociation of PDPA-TPP nanoparticles could reduce the aggregation caused quenching(ACQ) of the photosensitizers, leading to around 5 folds increase of the singlet oxygen(1O2) quantum yield. In vitro results demonstrated that the 'acid-triggered' PDPA-TPP nanoparticles could kill most of MDR S. aureus(Gram-positive) and MDR E. coli(Gram-negative) by enhanced photodynamic therapy, and they could resist wound infection and accelerate wound healing effectively in vivo. Furthermore, PDPA-TPP nanoparticles could well disperse the biofilm and almost kill all the biofilm-containing bacteria. 8Retrovirus-mediated herpes simplex virus thymidine kinase gene therapy approach for hepatocellular carcinoma显示文摘The therapeutic effect of herpes simplex virus thymi-dine kinase/ganciclovir (HSV-tk/GCV) system on hepa-tocellular carcinoma was studied in this experimeflt. Thetk-containing retroviral recombinants were used to infecthepatoma cells (BEL-7402) and the cells were treated withganciclovir (0-1000 pg/ml). The results showed that HSV-tk gene could be efficielltly transferred in Vitro into hep-atoma cells and stably expressed. The growth potentialof the tk-containing cells was significantly inhibited byGCV (P<0.01) as compared to the non-tk-containing cells.The antitumor effect of HSV-tk/GCV system was also pro-duced ex vivo in tk-containing tumor of nude mice as char-acterized by a marked decrease in tumor growth after GCVtreatment contrary to a progressive enlargement of non-tk-containing tumors. Although the histological examinationdemonstrated that the efficiency of the gene transfer wasless than 30%, the killing effect of HSV-tk/GCV systemon hepatocellular carcinoma was still significantIy gener-ated.
