2A New Method of Wind Turbine Bearing Fault Diagnosis Based on Multi-Masking Empirical Mode Decomposition and Fuzzy C-Means Clustering显示文摘Based on Multi-Masking Empirical Mode Decomposition (MMEMD) and fuzzy c-means (FCM) clustering, a new method of wind turbine bearing fault diagnosis FCM-MMEMD is proposed, which can determine the fault accurately and timely. First, FCM clustering is employed to classify the data into different clusters, which helps to estimate whether there is a fault and how many fault types there are. If fault signals exist, the fault vibration signals are then demodulated and decomposed into different frequency bands by MMEMD in order to be analyzed further. In order to overcome the mode mixing defect of empirical mode decomposition (EMD), a novel method called MMEMD is proposed. It is an improvement to masking empirical mode decomposition (MEMD). By adding multi-masking signals to the signals to be decomposed in different levels, it can restrain low-frequency components from mixing in highfrequency components effectively in the sifting process and then suppress the mode mixing. It has the advantages of easy implementation and strong ability of suppressing modal mixing. The fault type is determined by Hilbert envelope finally. The results of simulation signal decomposition showed the high performance of MMEMD. 3Classification of four distinct osteoarthritis subtypes with a knee joint tissue transcriptome atlas显示文摘The limited molecular classifications and disease signatures of osteoarthritis(OA)impede the development of prediagnosis and targeted therapeutics for OA patients.To classify and understand the subtypes of OA,we collected three types of tissue including cartilage,subchondral bone,and synovium from multiple clinical centers and constructed an extensive transcriptome atlas of OA patients.By applying unsupervised clustering analysis to the cartilage transcriptome,OA patients were classified into four subtypes with distinct molecular signatures:a glycosaminoglycan metabolic disorder subtype(C1),a collagen metabolic disorder subtype(C2),an activated sensory neuron subtype(C3),and an inflammation subtype(C4).Through ligand-receptor crosstalk analysis of the three knee tissue types,we linked molecular functions with the clinical symptoms of different OA subtypes.For example,the Gene Ontology functional term of vasculature development was enriched in the subchondral bone-cartilage crosstalk of C2 and the cartilage-subchondral bone crosstalk of C4,which might lead to severe osteophytes in C2 patients and apparent joint space narrowing in C4 patients.Based on the marker genes of the four OA subtypes identified in this study,we modeled OA subtypes with two independent published RNA-seq datasets through random forest classification.The findings of this work contradicted traditional OA diagnosis by medical imaging and revealed distinct molecular subtypes in knee OA patients,which may allow for precise diagnosis and treatment of OA.Chunhui Yuan Zongyou Pan Kun Zhao Jun Li Zixuan Sheng Xudong Yao Hua Liu Xiaolei Zhang Yang Yang Dongsheng Yu Yu Zhang Yuzi Xu Zhi-Yong Zhang Tianlong Huang Wanlu Liu Hongwei Ouyang 2020Bone Research2020,8,4:4

5Effect of Oxygen Concentration and Annealing Theatment on the Optical Properties of the Transparent Conductive CdIn_2O_4 Thin Films显示文摘Transparent conductive cadmium indium oxide films (CdIn2O4) were prepared by r.f. reactive sputtering from Cd-In alloy targets under an Ar-O2 atmosphere. Electrical conductivity of the order of 105Ω-1.m-1 and the optical transmission as high as 94% are easily attained by postdeposition annealing treatment. The effects of oxygen concentration in the reactive gas mixture and post-deposition annealing treatment on the optical transmittance as well as optical parameters, such as refractive index (n), extinction coefficient (k), real part (ε’) and imaginary part (ε') of the dielectric constant, were studied in the visible and near-infrared region. 6Mutual dependency between lncRNA LETN and protein NPM1 in controlling the nucleolar structure and functions sustaining cell proliferation显示文摘Fundamental processes such as ribosomal RNA synthesis and chromatin remodeling take place in the nucleolus,which is hyperactive in fast-proliferating cells.The sophisticated regulatory mechanism underlying the dynamic nucleolar structure and functions is yet to be fully explored.The present study uncovers the mutual functional dependency between a previously uncharacterized human long non-coding RNA,which we renamed LETN,and a key nucleolar protein,NPM1.Specifically,being upregulated in multiple types of cancer,LETN resides in the nucleolus via direct binding with NPM1.LETN plays a critical role in facilitating the formation of NPM1 pentamers,which are essential building blocks of the nucleolar granular component and control the nucleolar functions.Repression of LETN or NPM1 led to similar and profound changes of the nucleolar morphology and arrest of the nucleolar functions,which led to proliferation inhibition of human cancer cells and neural progenitor cells.Interestingly,this inter-dependency between LETN and NPM1 is associated with the evolutionarily new variations of NPM1 and the coincidental emergence of LETN in higher primates.We propose that this human-specific protein–lncRNA axis renders an additional yet critical layer of regulation with high physiological relevance in both cancerous and normal developmental processes that require hyperactive nucleoli.Xianteng Wang Xiaolin Hu Wanlu Song Hui Xu Zhengtao Xiao Rongyao Huang Qingran Bai Fan Zhang Yongzhen Chen Yu Liu Jianhuo Fang Xin Li Qin Shen Haitao Zhao Xuerui Yang 2021Cell Research2021,31,6:3

10Investigation of carbonyl amidation and O-methylation during biosynthesis of the pharmacophore pyridyl of antitumor piericidins显示文摘Piericidins are a large family of bacterialα-pyridone antibiotics with antitumor activities such as their anti-renal carcinoma activity exhibited recently in nude mice.The backbones of piericidins are derived fromβ,δ-diketo carboxylic acids,which are offloaded from a modular polyketide synthase(PKS)and putatively undergo a carbonyl amidation beforeα-pyridone ring formation.The tailoring modifications to theα-pyridone structure mainly include the verified hydroxylation and O-methylation of the C-4′position and an unidentified C-5′O-methylation.Here,we describe a piericidin producer,terrestrial Streptomyces conglobatus,which contains a piericidin biosynthetic gene cluster in two different loci.Deletion of the amidotransferase gene pieD resulted in the accumulation of two fatty acids that should be degraded from the nascent carboxylic acid released by the PKS,supporting the carbonyl amidation function of PieD duringα-pyridone ring formation.Deletion of the O-methyltransferase gene pieB1 led to the production of three piericidin analogues lacking C-5′O-methylation,therefore confirming that PieB1 specifically catalyses the tailoring modification.Moreover,bioactivity analysis of the mutant-derived products provided clues regarding the structure-function relationship for antitumor activity.The work addresses two previously unidentified steps involved in pyridyl pharmacophore formation during piericidin biosynthesis,facilitating the rational bioengineering of the biosynthetic pathway towards valuable antitumor agents.Wanlu Li Wenyu Zhang Yijia Cheng Yaoyao Shen Jianzhao Qi Hou-Wen Lin Yongjun Zhou 2022Synthetic and Systems Biotechnology2022,7,3:2
11Piezoresistive effect in carbonnanotube films显示文摘The piezoresistive effect of the pristine carbon nanotube (CNT) films has been studied. Carbon nanotubes were synthesized by hot filament chemical vapor deposition. The piezoresistive effect in the pristine CNT films was stud-ied by a three-point bending test. The gauge factor for the pristine CNT films under 500 microstrains was found to be at least 65 at room temperature, and increased with tem-perature, exceeding that of polycrystalline silicon (30) at 35℃. 12Multi-scale discrepancy adversarial network for cross-corpus speech emotion recognition显示文摘Background One of the most critical issues in human-computer interaction applications is recognizing human emotions based on speech.In recent years,the challenging problem of cross-corpus speech emotion recognition(SER)has generated extensive research.Nevertheless,the domain discrepancy between training data and testing data remains a major challenge to achieving improved system performance.Methods This paper introduces a novel multi-scale discrepancy adversarial(MSDA)network for conducting multiple timescales domain adaptation for cross-corpus SER,i.e.,integrating domain discriminators of hierarchical levels into the emotion recognition framework to mitigate the gap between the source and target domains.Specifically,we extract two kinds of speech features,i.e.,handcraft features and deep features,from three timescales of global,local,and hybrid levels.In each timescale,the domain discriminator and the feature extrator compete against each other to learn features that minimize the discrepancy between the two domains by fooling the discriminator.Results Extensive experiments on cross-corpus and cross-language SER were conducted on a combination dataset that combines one Chinese dataset and two English datasets commonly used in SER.The MSDA is affected by the strong discriminate power provided by the adversarial process,where three discriminators are working in tandem with an emotion classifier.Accordingly,the MSDA achieves the best performance over all other baseline methods.Conclusions The proposed architecture was tested on a combination of one Chinese and two English datasets.The experimental results demonstrate the superiority of our powerful discriminative model for solving cross-corpus SER.Wanlu ZHENG Wenming ZHENG Yuan ZONG 2021Virtual Reality & Intelligent Hardware2021,3,1:2

15Single cell analysis reveals inhibition of angiogenesis attenuates the progression of heterotopic ossification in Mkx^(−/−)mice显示文摘Tendon heterotopic ossification(HO)is characterized by bone formation inside tendon tissue,which severely debilitates people in their daily life.Current therapies fail to promote functional tissue repair largely due to our limited understanding of HO pathogenesis.Here,we investigate the pathological mechanism and propose a potential treatment method for HO.Immunofluorescence assays showed that the Mohawk(MKX)expression level was decreased in human tendon HO tissue,coinciding with spontaneous HO and the upregulated expression of osteochondrogenic and angiogenic genes in the tendons of Mkx^(−/−)mice.Single-cell RNA sequencing analyses of wild-type and Mkx^(−/−)tendons identified three cell types and revealed the excessive activation of osteochondrogenic genes during the tenogenesis of Mkx^(−/−)tendon cells.Single-cell analysis revealed that the gene expression program of angiogenesis,which is strongly associated with bone formation,was activated in all cell types during HO.Moreover,inhibition of angiogenesis by the small-molecule inhibitor BIBF1120 attenuated bone formation and angiogenesis in the Achilles tendons of both Mkx mutant mice and a rat traumatic model of HO.These findings provide new insights into the cellular mechanisms of tendon HO and highlight the inhibition of angiogenesis with BIBF1120 as a potential treatment strategy for HO.Junxin Lin Yuwei Yang Wenyan Zhou Chao Dai Xiao Chen Yuanhao Xie Shan Han Huanhuan Liu Yejun Hu Chenqi Tang Varitsara Bunpetch Dandan Zhang Yishan Chen Xiaohui Zou Di Chen Wanlu Liu Hongwei Ouyang 2022Bone Research2022,10,4:1
