| 1 | Hepatitis B virus infection of transplanted human hepatocytes causes a biochemical and histological hepatitis in immunocompetent rats显示文摘AIM: To characterize the host response to hepatitis B virus (HBV) infection in human hepatocytes transplanted into immunocompetent rodent rats tolerized by, and transplanted with primary human hepatocytes.METHODS: One week after the transplantation, rats were inoculated with HBV, and viral gene expression, replication,and host response was monitored.RESULTS: HBV DNA was detectable in serum for at least 60 days. HBsAg levels rose steadily for 3 weeks postinoculation and then plateaued at a level of about 0.6 pg/mi. HBV RNA was also found in liver at levels that remained constant through the time course. Immunofluorescence revealed clusters of hepatocytes that stained positive for HBcAg. The presence of HBV covalently closed circular DNA (cccDNA) in liver was demonstrated using nuclease digestion of single-stranded DNA followed by PCR. Serum ALT levels rose and reached a peak level of 180 IU/L on day 18, but remained elevated for 60 days. | 3 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 7 | Particle size dependent sinterability and magnetic properties of recycled HDDR Nd-Fe-B powders consolidated with spark plasma sintering显示文摘The dependence of the magnetic properties on the particle size of recycled HDDR Nd-Fe-B powders was investigated,with the aim to assess the reprocessing potential of the end-of-life scrap magnets via spark plasma sintering(SPS).The as received recycled HDDR powder has coercivity(Hci)=830 kA/m and particles in the range from 30 to 700 μm(average 220 μm).After burr milling,the average particle size is reduced to 120 μm and subsequently the Hci of fine(milled) powder was 595 kA/m.Spark plasma sintering was exploited to consolidate the nanograined HDDR powders and limit the abnormal grain coarsening.The optimal SPS-ing of coarse HDDR powder at 750℃for 1 min produces fully dense magnets with Hci=950±100 kA/m which further increases to 1200 kA/m via thermal treatment at 750℃for 15 min.The burr milled fine HDDR powder under similar SPS conditions and after thermal treatment results in Hci=940 kA/m.The fine powder is further sieved down from 630 to less than 50 μm mesh size,to evaluate the possible reduction in Hci in relation to the particle size.The gain in oxygen content doubles for <50 μm sized particles as compared with coarser fractions(>200 μm).The XRD analysis for fractionated powder indicates an increase in Nd2O3 phase peaks in the finer(<100 μm)fractions.Similarly,the Hci reduces from 820 kA/m in the coarse particles(>200 μm) to 460 kA/m in the fine sized particles(<100μm).SPS was done on each HDDR powder fraction under the optimal conditions to measure the variation in Hci and density.The Hci of SPS-ed coarse fraction(>200 μm) is higher than 930 kA/m and it falls abruptly to just 70 kA/m for the fine sized particles(<100 μm).The thermal treatment further improves the Hci to>1000 kA/m only up to 100 μm sized fractions with>90% sintered density.The full densification(>99%) is observed only in the coarse fractions.The loss of coercivity and lack of sinterability in the fine sized particles(<100 μm) are attributed to a very high oxygen content.This implies that during recycling,if good magnetic properties are to be maintained or even increase the HDDR powder particles can be sized down only up to≥100 μm. | Awais Ikram Farhan Mehmood Richard Stuart Sheridan Muhammad Awais Allan Walton Anas Eldosouky Saso Sturm Spomenka Kobe Kristina Zuzek Rozman | 2020 | Journal of Rare Earths2020,38,1: | 5 |
| 8 | Investigation of factors influencing roof stability at a Western U.S.longwall coal mine显示文摘The coal mine roof rating(CMRR) was developed to bridge the gap between geological variation in underground coal mines and engineering design. The CMRR accounts for the compressive strength of the immediate roof, the shear strength and intensity of any discontinuities present, and the moisture sensitivity of the immediate roof. The CMRR has been widely used and validated in Eastern US coal mines, but it has seen limited application in the Western US. This study focuses on roof behavior at a Western coal mine(Mine A). Mine A shows significant lateral geological variation, along with localized faulting and a laterally extensive sandstone channel network. The CMRR is not used to predict roof instability at the mine. It is, therefore, hypothesized that there are other factors that are correlated with roof instability in underground coal mines that could potentially also be considered in conjunction with the CMRR.This hypothesis was tested by collecting 30 CMRR measurements at Mine A. At each measurement location, a binary record of the roof condition(stable or unstable) was made, and other parameters such as depth of cover, presence of faulting, and sandstone channels were also recorded. ANOVA tests showed that the CMRR values and the roof conditions were not strongly correlated, indicating that the CMRR input criteria are not fully predictive of roof stability at this mine. The CMRR values showed statistically significant correlations(p less than 0.05) with faulting as well as with location at an intersection. | 16 | A DNA delivery system containing listeriolys in Oresults in enhanced hepatocytedirected gene expression显示文摘AIM To determine whether incorporation of the pH dependent bacterial toxin listeriolysin O (LLO) into the DNA carrier system could increase the endosomal escape of internalized DNA and result gene expression. METHODS A multi component delivery system was prepared consisting of asialoglycoprotein (ASG), poly L lysine (PL), and LLO. Two marker genes, luciferase and β galactosidase in plasmids were complexed and administered in vitro to Huh7[ASG receptor (+) ] and SK Hep1[ASG receptor (-) ] cells. Purity, hemolytic activity, gene expression, specificity, and toxicity were evaluated. RESULTS An LLO containing conjugate retained cell targeting specificity and membranolytic activity. In ASG receptor (+) cells, luciferase gene expression was enhanced by more than 7 fold over that of conjugates without the incorporation of listeriolysin O. No significant expression occurred in ASG receptor (-) cells. Enhancement of β galactosidase gene expression was less, but still significantly increased over controls. There was no detectable toxicity at concentrations shown to be effective in transfection studies. | 18 | Numerical analyses of pillar behavior with variation in yield criterion,dilatancy, rock heterogeneity and length to width ratio显示文摘With recent advances in numerical modeling, design of underground structures increasingly relies on numerical modeling-based analysis approaches. While modeling tools like the discrete element method(DEM) and the combined finite-discrete element method(FDEM) are useful for investigating small-scale damage processes, continuum models remain the primary practical tool for most field-scale problems.The results obtained from such models are significantly dependent on the selection of an appropriate yield criterion and dilation angle. Towards improving its capabilities in handling mining-related problems, the authors have previously developed a new yield criterion(called progressive S-shaped criterion). The focus of the current study is to demonstrate its use in modeling rock pillars through a comparative analysis against four other yield criteria. In addition to the progressive S-shaped criterion,only one out of the four other criteria predicted a trend in strength consistent with an empirical pillar strength database compiled from the literature. Given the closely-knit relationship between yield criteria and dilation angle in controlling the overall damage process, a separate comparison was conducted using a mobilized dilation model, a zero degree dilation angle and a constant non-zero dilation angle. This study also investigates the impact of meso-scale heterogeneity in mechanical properties on the overall model response by assigning probability distributions to the input parameters. The comparisons revealed that an isotropic model using a combination of progressive S-shaped criterion and mobilized dilation angle model is sufficient in capturing the behaviors of rock pillars.
