| 1 | Hepatocyte-like cells from directed differentiation of mouse bone marrow cells in vitro显示文摘Aim: To design the effective directed differentiation medium to differentiate bone marrow cells into hepatocyte-like cells. Methods: Bone marrow cells were cultured in the directed differentiation media including fibroblast growth factor-4 (FGF-4) and oncostatin M (OSM). Hepatocyte-like cells from directed differentiation of bone marrow cells were identified through cell morphology, RNA expressions by reverse transcriptase-polymerase chain reaction (RT-PCR), protein expressions by Western blot, and hepatocellular synthesis and metabolism functions by albumin ELISA, Periodic acid-Shift staining and urea assay. Results: Some epithelial-like cells or polygonal cells appeared and increased in the course of the cell directed differentiation. Hepatocyte nucleur factor-3β(HNF-3β), albumin (ALB), cytokeratin 18 (CK18), transthyretin (TTR), glucose-6-phosphate (G-6-Pase), and tyrosine aminotransferase (TAT) mRNA were expressed in the course of the directed differentiation. The directed differentiated cells on d 21 expressed HNF-3β, ALB, and CK18 proteins. The directed differentiated cells produced albumin and synthesized urea in a time-dependent manner. They could also synthesize glycogen. | 2 | Abrupt temperature change and a warming hiatus from 1951 to 2014 in Inner Mongolia, China显示文摘An abrupt temperature change and a warming hiatus have strongly influenced the global climate.This study focused on these changes in Inner Mongolia, China. This study used the central clustering method, Mann-Kendall mutation test and other methods to explore the abrupt temperature change and warming hiatus in three different temperature zones of the study region based on average annual data series.Among the temperature metrics investigated, average minimum temperature(Tnav) shifted the earliest,followed by average temperature(Tnv) and average maximum temperature(Txav). The latest change was observed in summer(1990 s), whereas the earliest was observed in winter(1970 s). Before and after the abrupt temperature change, Tnav fluctuated considerably, whereas there was only a slight change in Txav.Before and after the abrupt temperature change, the winter temperature changed more dramatically than the summer temperature. Before the abrupt temperature change, Tnav in the central region(0.322°C/10 a)and west region(0.48°C/10 a) contributed the most to the increasing temperatures. After the abrupt temperature change, Tnav in winter in the central region(0.519°C/10 a) and in autumn in the west region(0.729°C/10 a) contributed the most to the temperature increases. Overall, in the years in which temperature shifts occurred early, a warming hiatus also appeared early. The three temperature metrics in spring(1991)in the east region were the first to exhibit a warming hiatus. | 4 | Cardiac Na^(+)-Ca^(2+) exchanger 1(ncx1h)is critical for the ventricular cardiomyocyte formation via regulating the expression levels of gata4 and hand2 in zebrafish显示文摘Ca^(2+) signaling is critical for heart development;however,the precise roles and regulatory pathways of Ca^(2+) transport proteins in cardiogenesis remain largely unknown.Sodium-calcium exchanger 1(Ncx1)is responsible for Ca^(2+) efflux in cardiomyocytes.It is involved in cardiogenesis,while the mechanism is unclear.Here,using the forward genetic screening in zebrafish,we identified a novel mutation at a highly-conserved leucine residue in ncx1 gene(mutant^(LDD353)/ncx1h^(L154P))that led to smaller hearts with reduced heart rate and weak contraction.Mechanistically,the number of ventricular but not atrial cardiomyocytes was reduced in ncx1h^(L154P) zebrafish.These defects were mimicked by knockdown or knockout of ncx1h.Moreover,ncx1h^(L154P) had cytosolic and mitochondrial Ca^(2+) overloading and Ca^(2+) transient suppression in cardiomyocytes.Furthermore,ncx1h^(L154P) and ncx1h morphants downregulated cardiac transcription factors hand2 and gata4 in the cardiac regions,while overexpression of hand2 and gata4 partially rescued cardiac defects including the number of ventricular myocytes.These findings demonstrate an essential role of the novel 154th leucine residue in the maintenance of Ncx1 function in zebrafish,and reveal previous unrecognized critical roles of the 154th leucine residue and Ncx1 in the formation of ventricular cardiomyocytes by at least partially regulating the expression levels of gata4 and hand2. | Liming Chu Huimin Yin Lei Gao Li Gao Yu Xia Chiyuan Zhang Yi Chen Tingxi Liu Jijun Huang Kenneth R.Boheler Yong Zhou Huang-Tian Yang | 2021 | Science China(Life Sciences)2021,64,2: | 3 |
| 5 | Influences of landform as a confounding variable on SOM-NDVI association in semiarid Ordos Plateau显示文摘Soil organic matter(SOM) plays an important role in maintaining vegetation cover and thus mitigating land erosion of fragile terrestrial ecosystems such as in the Northern Ordos Plateau of China(NOPC).However,little information is available on whether and how SOM varies spatially as an intrinsic characteristic of landform in NOPC.The objective of this study was to examine the spatial associations of SOM with landform and vegetation cover.The study was conducted in a 23,000-km2 area within NOPC because this area has landforms of mobile dunes(MD),flat dunes(FD),grassy sandy land(GSL),flat sandy bedrocks(FSB),and swamps and salt lakes(SW),which are typical landforms in semiarid ecosystems.SOM was determined using a standard laboratory analysis method for 5 cm topsoil samples collected at 72 locations across the study area.In addition,the 250 m Multitemporal Moderate Resolution Imaging Spectroradiometer(MODIS) imageries taken in the period from August 2006 to August 2010 were used to extract Normalized Difference Vegetation Index(NDVI) which in turn was used as the surrogate of vegetation cover.Classic and geostatistical methods were used to compare SOM concentration across different landforms.The results indicated that an area with a greater value for NDVI(i.e.better vegetation cover) tended to have a higher SOM concentration regardless of the landform types.However,the association between SOM and NDVI varied from one landform to another.The SW and GSL had a highest SOM concentration,while MD had a lowest concentration.For the study area as a whole and the FD,GSL,and MD,SOM was found to be the sole function of NDVI,whereas,for the FSB,SOM was influenced by several intrinsic variables,namely ground surface altitude,slope,and aspect,as well as NDVI.SOM for the SW landform was found to be a function of NDVI.Furthermore,SOM and NDVI exhibited a consistent spatial pattern of increasing from north to south and from west to east.The highest SOM concentration of 3.5% occurred along an east-westward belt,which is adjacent to water pathways,in the mid part of the study area. | YanYun LUO TingXi LIU XiXi WANG LiMin DUAN | 2012 | Journal of Arid Land2012,4,4: | 2 |

Danshensu (β-3, 4-dihydroxyphenyl-lactic acid) has a similar molecular structure and pharmacological action to caffeic acid. This study aimed to validate the protection conferred by Danshensu against excitotoxic effects of maternal intragastric administration of monosodium glutamate at late stages of pregnancy in the developing mouse fetal brain. Behavioral tests, as well as histopathological and immunohistochemical examination of hippocampi were performed in filial mice. Results revealed that maternal intragastric administration of excessive monosodium glutamate (1.0, 2.0, 4.0 g/kg body weight) at a late stage of pregnancy resulted in a series of behavioral disorders (hyperactivity, lesions of learning and memory, and disturbance in cooperation of movement ability under high-altitude stress), histopathological impairment (neuronal edema, degeneration, necrosis, and hyperplasia) and molecular cellular biological changes (upregulated expression of N-methyl-D-aspartate receptor type 1 and neuropeptide Y in the hippocampal region of the brain of the filial mice from mothers treated with monosodium glutamate). Simultaneous administration of sodium Danshensu partially reversed the effects of monosodium glutamate on the above mentioned phenomena. | 18 | Transformation among precipitation,surface water,groundwater,and mine water in the Hailiutu River Basin under mining activity显示文摘Coal mining has changed the hydrogeological conditions of river basins,and studying how the relationship among different types of water body has changed under the influence of coal mining is of great significance for understanding the regional hydrological cycle.We analyzed the temporal and spatial distribution of hydrochemical properties and environmental isotopes in the Hailiutu River Basin(HRB),China with a mixed model.The results showed that:(1)human activity(e.g.,coal mining and agricultural production)causes considerable changes in the hydrochemical properties of surface water in and around the mining areas,and leads to significant increases in the concentrations of Na^(+)and SO_(4)^(2-);(2)precipitation is the main source of water vapour in the HRB.The transformation between surface water and groundwater in the natural watershed is mainly affected by precipitation;and(3)in the mining areas,the average contribution rates of precipitation to the recharge of surface water and groundwater increased by 2.6%-7.9%and 2.7%-9.9%,respectively.Groundwater in the Salawusu Formation constitutes up to 61.3%-72.4%of mine water.Overall,this study is beneficial for quantifying the effects of coal mining on local hydrological cycles.The research results can provide a reference for local water resources management and ecological environment improvement. | LI Qian MA Long LIU Tingxi | 2022 | Journal of Arid Land2022,14,6: | 0 |
| 19 | Boosting efficiency and stability of 2D alternating cation perovskite solar cells via rational surface-modification: Marked passivation efficacy of anion显示文摘Two-dimensional(2D) alternating cation(ACI) perovskite surface defects,especially dominant iodine vacancies(V_Ⅰ) and undercoordinated Pb^(2+),limit the performance of perovskite solar cells(PVSCs).To address the issue,1-butyl-3-methylimidazolium trifluoro-methane-sulfonate(BMIMOTF) and its iodide counterpart(BMIMI) are utilized to modify the perovskite surface respectively.We find that BMIMI can change the perovskite surface,whereas BMIMOTF shows a nondestructive and more effective defect passivation,giving significantly reduced defect density and suppressed charge-carrier nonradiative recombination.This mainly attributes to the marked passivation efficacy of OTF-anion on V_Ⅰ and undercoordinated Pb^(2+),rather than BMIMI^(+) cation.Benefiting from the rational surface-modification of BMMIMOTF,the films exhibit an optimized energy level alignment,enhanced hydrophobicity and suppressed ion migration.Consequently,the BMIMOTF-modified devices achieve an impressive efficiency of 21.38% with a record open-circuit voltage of 1.195 V,which is among the best efficiencies reported for 2D PVSCs,and display greatly enhanced humidity and thermal stability. | Hualin Zheng Xuefeng Peng Tingxi Chen Ting Zhang Shihao Yuan Lei Wang Feng Qian Jiang Huang Xiaodong Liu Zhi David Chen Yanning Zhang Shibin Li | 2023 | Journal of Energy Chemistry2023,,9: | 0 |
