1A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Holocene vegetational and climatic variation in westerly-dominated areas of Central Asia inferred from the Sayram Lake in northern Xinjiang,China显示文摘Changes in the vegetation and climate of the westerly-dominated areas in Central Asia during the Holocene were interpreted using pollen-assemblages and charcoal data from a 300-cm-long sediment core of the Sayram Lake,northern Xinjiang.Accele-rator Mass Spectrometry(AMS) radiocarbon dating methods were applied to bulk organic matter of the samples.Artemisia spp./Chenopodiaceae ratios and results from principal component analysis were used to infer that the lake basin was dominated by desert vegetation before ca.9.6 cal.ka BP,which suggests a warm and dry climate in the early Holocene.Desert steppe/steppe expanded during 9.6-5.5 cal.ka BP,indicating a remarkable increase both in the precipitation and temperature during the mid-Holocene.Desert vegetation dominated between 6.5 and 5.5 cal.ka BP,marking an extreme warmer and drier interval.The steppe/meadow steppe recovered,and temperatures decreased from 5.5 cal.ka BP in the late Holocene,as indicated by the increased abundance of Artemisia and the development of meadows.Holocene temperatures and moisture variations in the Sayram Lake areas were similar to those of adjacent areas.This consistency implies that solar radiation was the main driving factor for regional temperature changes,and that the effect of temperature variations was significant on regional changes in humidity.The evolution of climate and environment in the Sayram Lake areas,which were characterized as dry in the early Holocene and relatively humid in the middle-late Holocene,are clearly different from those in monsoonal areas.Dry conditions in the early Holocene in the Sayram Lake areas were closely related to decreased water vapor advection.These conditions were a result of reduced westerly wind speeds and less evaporation upstream,which in turn were caused by seasonal changes in solar radiation superimposed by strong evaporation following warming and drying local climate.JIANG QingFeng JI JunFeng SHEN Ji Ryo MATSUMOTO TONG GuoBang QIAN Peng REN XueMei YAN DeZhi 2013Science China Earth Sciences2013,56,3:32
4Role of epithelial-mesenchymal transition in gastric cancer initiation and progression显示文摘Gastric cancer is one of the most common malignant tumors worldwide.Due to its intricate initiation and progression mechanisms,early detection and effective treatment of gastric cancer are difficult to achieve.The epithelial-mesenchymal transition(EMT)is characterized as a fundamental process that is critical for embryonic development,wound healing and fibrotic disease.Recent evidence has established that aberrant EMT activation in the human stomach is closely associated with gastric carcinogenesis and tumor progression.EMT activation endows gastric epithelial cells with increased characteristics of mesenchymal cells and reduces their epithelial features.Moreover,mesenchymal cells tend to dedifferentiate and acquire stem cell or tumorigenic phenotypes such as invasion,metastasis and apoptosis resistance as well as drug resistance during EMT progression.There are a number of molecules that indicate the stage of EMT(e.g.,E-cadherin,an epithelial cell biomarker);therefore,certain transcriptional proteins,especially E-cadherin transcriptional repressors,may participate in the regulation of EMT.In addition,EMT regulation may be associated with certain epigenetic mechanisms.The aforementioned molecules can be used as early diagnostic markers for gastric cancer,and EMT regulation can provide potential targets for gastric cancer therapy.Here,we review the role of these aspects of EMT in gastric cancer initiation and development.Zhao Peng Chen-Xiao Wang Er-Hu Fang Guo-Bin Wang Qiang Tong 2014World Journal of Gastroenterology2014,20,18:28
5Remote ischemic perconditioning prevents liver transplantation-induced ischemia/reperfusion injury in rats: Role of ROS/RNS and e NOS显示文摘AIM To investigate the underlying mechanisms of the protective role of remote ischemic perconditioning(RIPerC) in rat liver transplantation. METHODS Sprague-Dawley rats were subjected to sham, orthotopic liver transplantation(OLT), ischemic postconditioning(IPostC) or RIPerC. After 3 h reperfusion, blood samples were taken for measurement of alanine aminotransferase, aspartate aminotransferase, creatinine(Cr) and creatinine kinase-myocardial band(CK-MB). The liver lobes were harvested for the following measurements: reactive oxygen species(ROS), H2O2, mitochondrial membrane potential(ΔΨm) and total nitric oxide(NO). These measurements were determined using an ROS/H2O2, JC1 and Total NOx Assay Kit, respectively. Endothelial NO synthase(e NOS) was analyzed by reverse transcription-polymerase chain reaction(RTPCR) and western blotting, and peroxynitrite was semiquantified by western blotting of 3-nitrotyrosine. RESULTS Compared with the OLT group, the grafts subjected to RIPerC showed significantly improved liver and remote organ functions(P < 0.05). ROS(P < 0.001) including H2O2(P < 0.05) were largely elevated in the OLT group as compared with the sham group, and RIPerC(P < 0.05) reversed this trend. The collapse of ΔΨm induced by OLT ischemia/reperfusion(I/R) injury was significantly attenuated in the RIPerC group(P < 0.001). A marked increase of NO content and phosphoserine eN OS, both in protein and mR NA levels, was observed in liver graft of the RIPer C group as compared with the OLT group(P < 0.05). I/R-induced 3-nitrotyrosine content was significantly reduced in the RIPerC group as compared with the OLT group(P < 0.05). There were no significant differences between the RIPerC and IPostC groups for all the results except Cr. 6Selective Ferroptosis Inhibitor Liproxstatin-1 Attenuates Neurological Deficits and Neuroinflammation After Subarachnoid Hemorrhage显示文摘Ferroptosis is a form of iron-dependent regulated cell death.Evidence of its existence and the effects of its inhibitors on subarachnoid hemorrhage(SAH)is still lacking.In the present study,we found that liproxstatin-1 protected HT22 cells against hemin-induced injury by protecting mitochondrial functions and ameliorating lipid peroxidation.In in vivo experiments,we demonstrated the presence of characteristic shrunken mitochondria in ipsilateral cortical neurons after SAH.Moreover,liproxstatin-1 attenuated the neurological deficits and brain edema,reduced neuronal cell death,and restored the redox equilibrium after SAH.The inhibition of ferroptosis by liproxstatin-1 was associated with the preservation of glutathione peroxidase 4 and the downregulation of acylCoA synthetase long-chain family member 4 as well as cyclooxygenase 2.In addition,liproxstatin-1 decreased the activation of microglia and the release of IL-6,IL-1 b,and TNF-a.These data enhance our understanding of cell death after SAH and shed light on future preclinical studies.Yang Cao Yin Li Chao He Feng Yan Jian-Ru Li Hang-Zhe Xu Jian-Feng Zhuang Hang Zhou Yu-Cong Peng Xiong-Jie Fu Xiao-Yang Lu Yuan Yao Yu-Yu Wei Yun Tong Yi-Fu Zhou Lin Wang 2021Neuroscience Bulletin2021,37,4:16
7Oral berberine improves brain dopa/dopamine levels to ameliorate Parkinson’s disease by regulating gut microbiota显示文摘The phenylalanine-tyrosine-dopa-dopamine pathway provides dopamine to the brain.Iin this process,tyrosine hydroxylase(TH)isthe rate-limiting enzyme that hydroxylates tyrosine and generates levodopa(L-dopa)with tetranydrobiopterin(BH_(4))as a coenzyme.Here,we show that oral berberine(BBR)might supply H^(·) through dihydroberberine(reduced BBR produced by bacterial nitroreductase)and promote the production of BHl from dihydrobiopterin;the increased BH,enhances TH activity,which accelerates the production of L-dopa by the gut bacteria.Oral BBR acts in a way similar to vitamins.The L-dopa produced by theintestinal bacteria enters the brain through the circulation and is transformed to dopamine.To verify the gut-brain dialog activatedby BBR's effect,Enterococcus foecalis or Enterococcus faecium was transplanted into Parkinson's disease(PD)mice.The bacteriasignificantly increased brain dopamine and ameliorated PD manifestation in mice;additionally,combination of BBR with bacteriashowed better therapeutic effect than that with bacteria alone.Moreover,2,4,6-trimethy-pyranylium tetrafluoroborate(TMP-TFB)-derivatized matrix-assisted laser desorption mass spectrometry(MALDI-MS)imaging of dopamine identihed elevated striataldopamine levels in mouse brains with oral Enterococcus,and BBR strengthened the imaging intensity of brain dopamine.Theseresults demonstrated that BBR was an agonist of TH in Enterococcus and could lead to the production of L-dopa in the gut.Furthermore,a study of 28 patients with hyperlipidemia conhrmed that oral BBR increased bloodfecal L-dopa by the intestinalbacteria.Hence,BBR might improve the brain function by upregulating the biosynthesis of-dopa in the gut microbiota through avitamin-like effect.Yan Wang Qian Tong Shu-Rong Ma Zhen-Xiong Zhao Li-Bin Pan Lin Cong Pei Han Ran Peng Hang Yu Yuan Lin Tian-Le Gao Jia-Wen Shou Xiao-Yang Li Xian-Feng Zhang Zheng-Wei Zhang Jie Fu Bao-Ying Wen Jin-Bo Yu Xuetao Cao Jian-Dong Jiang 2021Signal Transduction and Targeted Therapy2021,6,3:16

9Prognostic factors in the surgical treatment of caudate lobe hepatocellular carcinoma显示文摘AIM:To evaluate the short-and long-term outcomes of liver resection for caudate lobe hepatocellular carcinoma (HCC).METHODS:We retrospectively analyzed 114 consecutive patients with HCC,originating from the caudate lobe,who underwent resection between January 2001 and January 2007.Univariate and multivariate analyses were performed on several clinicopathologic variables to determine the factors affecting long-term outcome and intrahepatic recurrence.RESULTS:Overall mortality and morbidity were 0% and 18%,respectively.After a median follow-up of 31 mo (interquartile range,11-66 mo),tumor recurrence had occurred in 76 patients (66.7%).The 1-,3-,and 5-year disease-free survival rates were 65.7%,38.1%,and 18.4%,respectively.The 1-,3-,and 5-year overall survival rates were 76.1%,54.7%,and 31.8%,respectively.Univariate analysis showed that subsegmental location of the tumor (45.7% vs 16.2%,P=0.01),liver cirrhosis (12.3% vs 47.9%,P=0.03),surgical margin (18.5% vs 54.6%,P=0.04),vascular invasion (37.9% vs 23.2%,P=0.04) and extended caudate resection (42.1% vs 15.4%,P=0.04) were related to poorer long-term survival.Multivariate analysis showed that only subsegmental location of the tumor,liver cirrhosis and surgical margin were significant independent prognostic factors.CONCLUSION:Hepatectomy was an effective treatment for HCC in the caudate lobe.The subsegmental location of the tumor,liver cirrhosis and surgical margin affected long-term survival.Peng Liu,Jia-Mei Yang,Wen-Yang Niu,Tong Kan,Feng Xie,Dian-Qi Li,Ye Wang,Yan-Ming Zhou,Department of Special Treatment and Liver Transplantation,Eastern Hepatobiliary Surgery Hospital,Second Military Medical University,Shanghai 200438,China Peng Liu,Department of Hepatobiliary Surgery,Navy General Hospital,Beijing 100037,China 2010World Journal of Gastroenterology2010,16,9:15

12The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 13A Synthetic Photorespiratory Shortcut Enhances Photosynthesis to Boost Biomass and Grain Yield in Rice显示文摘Several photorespiratory bypasses have been introduced into plants and shown to improve photosynthesis by increasing chloroplastic C02 concentrations or optimizing energy balance.We recently reported that an engineered GOC bypass could increase photosynthesis and productivity in rice.However,the grain yield of GOC plants was unstable,fluctuating in different cultivation seasons because of varying seed setting rates.In this study,we designed a synthetic photorespiratory shortcut(the GCGT bypass)consisting of genes en-coding Oryza sativa glycolate oxidase and Escherichia coli catalase,glyoxylate carboligase,and tartronic semialdehyde reductase.The GCGT bypass was guided by an optimized chloroplast transit peptide that targeted rice chloroplasts and redirected 75% of carbon from glycolate metabolism to the Calvin cycle,identical to the native photorespiration pathway.GCGT transgenic plants exhibited significantly increased biomass production and grain yield,which were mainly attributed to enhanced photosynthesis due to increased chloroplastic C02 concentrations.Despite the increases in biomass production and grain yield,GCGT transgenic plants showed a reduced seed setting rate,a phenotype previously reported for the GOC plants.Integrative transcriptomic,physiological,and biochemical assays revealed that photosynthetic car-bohydrates were not transported to grains in an efficient manner,thereby reducing the seed setting rate.Taken together,our results demonstrate that the GCGT photorespiratory shortcut confers higher yield by promoting photosynthesis in rice,mainly through increasing chloroplastic C02 concentrations.Li-Min Wang Bo-Ran Shen Bo-Di Li Chuan-Ling Zhang Min Lin Pan-Pan Tong Li-Li Cui Zhi-Sheng Zhang Xin-Xiang Peng 2020Molecular Plant2020,13,12:11

16Efficient microwave-assisted photocatalytic degradation of endocrine disruptor dimethyl phthalate over composite catalyst ZrO_x /ZnO显示文摘A highly active ZrO x /ZnO catalyst for microwave-assisted photocatalytic (MW/PC) degradation of endocrine disruptor dimethyl phthalate (DMP) has been prepared via cetyltrimethylammonium assisted hydrothermal method. The ZrO x /ZnO was characterized by XPS, XRD, UV-Vis, BET and SEM techniques. The XPS result showed that Zr oxides with different valences (+2, +3, +4) co-existed in ZrO x /ZnO. By using the ZrO x /ZnO (0.1 g), the TOC removal efficiency of DMP (100 mL of 50 mg/L) was 88% after 30 min reaction, which was about 15% higher than P25 TiO 2 . It was found that the removal process of DMP by MW/PC followed pseudo first-order kinetics in all cases, and ZrO x /ZnO significantly accelerated the degradation of DMP. The degradation half-life time of DMP was shortened 45% compared with P25 TiO 2 . A possible catalytic mechanism was proposed based on microwave response and interfacial charge transfer. ZrO x /ZnO could be reused for six times without obvious decrease in catalytic activity. 18Depositional architecture of the late Ordovician drowned carbonate platform margin and its responses to sea-level fluctuation in the northern slope of the Tazhong region, Tarim Basin显示文摘The Tazhong Uplift of the late Ordovician is a drowned rimmed carbonate platform. The carbonate rock of the late Ordovician Lianglitage Formation in the northern slope of the Tazhong region is one of the significant petroliferous intervals. Based on petrofacies, depositional cycles, natural gammaray spectrometry and carbon/oxygen isotope data from the Lianglitage Formation, one 2nd-order, three 3rd-order and several 4th-order sequences have been recognized, and the late Ordovician relative sealevel fluctuation curve has been established. The sequences O 3 l-1 and O 3 l-2 on the platform are composed of highstand and transgressive systems tracts, but lack the lowstand systems tract. The sequence O 3 l-3 is a drowning sequence. The sequence O 3 l-1 overlapped the eroded slope and pinched out to the northwest and landward. The highstand systems tract in the sequence O 3 l-2 consists of low-angle sigmoid and high-angle shingled progradation configuration. Major sedimentary facies of the Lianglitage Formation include reef and shoal in the platform margin and lagoon, which can be subdivided into coral-sponge-stromatoporoid reef complex, sand shoal, lime mud mound, and intershoal sea. Reefs, sand shoals and their complex are potential reservoir facies. The reefs and sand shoals in the sequence O 3 l-1 developed in the upper of its highstand systems tract. In the sequence O 3 l-2, the highstand systems tract with an internal prograding configuration is a response to the lateral shifting of the complex of reef and sand shoal. The transgressive systems tract, in particular the sand shoals, developed widely on the slope of the platform margin and interior. The reefs in the sequence O 3 l-3 migrated towards high positions and formed retrograding reefs in the western platform and low relief in the platform interior. Basinward lateral migration of the reefs and pure carbonate rock both characterize highstand systems tract and show that the rise of the relative sea-level was very slow. Shingled prograding stacking pattern of the 4th-order sequences and reefs grow horizontally, which represents the late stage of highstand systems tract and implies relative sealevel stillstand. Reefs migrating towards high land and impure carbonate rock both indicate transgressive systems tract and suggest that the relative sea-level rose fast. Erosional truncation and epidiagenetic karstification represent a falling relative sea-level. The relative sea-level fluctuation and antecedent palaeotopography control the development and distribution of reef complexes and unconformity karst zones. 19The gut microbial metabolite, 3,4-dihydroxyphenylpropionic acid, alleviates hepatic ischemia/reperfusion injury via mitigation of macrophage pro-inflammatory activity in mice显示文摘Hepatic ischemia/reperfusion injury(HIRI) is a serious complication that occurs following shock and/or liver surgery. Gut microbiota and their metabolites are key upstream modulators of development of liver injury. Herein, we investigated the potential contribution of gut microbes to HIRI.Ischemia/reperfusion surgery was performed to establish a murine model of HIRI. 16 S r RNA gene sequencing and metabolomics were used for microbial analysis. Transcriptomics and proteomics analysis were employed to study the host cell responses. Our results establish HIRI was significantly increased when surgery occurred in the evening(ZT12, 20:00) when compared with the morning(ZT0, 08:00);however, antibiotic pretreatment reduced this diurnal variation. The abundance of a microbial metabolite3,4-dihydroxyphenylpropionic acid was significantly higher in ZT0 when compared with ZT12 in the gut and this compound significantly protected mice against HIRI. Furthermore, 3,4-dihydroxyphenylpropionic acid suppressed the macrophage pro-inflammatory response in vivo and in vitro. This metabolite inhibits histone deacetylase activity by reducing its phosphorylation. Histone deacetylase inhibition suppressed macrophage pro-inflammatory activation and diminished the diurnal variation of HIRI. Our findings revealed a novel protective microbial metabolite against HIRI in mice. 20Daytime HONO formation in the suburban area of the megacity Beijing, China显示文摘Nitrous acid(HONO),as a primary precursor of OH radicals,has been considered one of the most important nitrogencontaining species in the atmosphere.Up to 30%of primary OH radical production is attributed to the photolysis of HONO.However,the major HONO formation mechanisms are still under discussion.During the Campaigns of Air Quality Research in Beijing and Surrounding Region(CAREBeijing2006)campaign,comprehensive measurements were carried out in the megacity Beijing,where the chemical budget of HONO was fully constrained.The average diurnal HONO concentration varied from 0.33 to 1.2 ppbv.The net OH production rate from HONO,POH(HONO)net,was on average(from 05:00 to 19:00)7.1×106 molecule/(cm3 s),2.7 times higher than from O3 photolysis.This production rate demonstrates the important role of HONO in the atmospheric chemistry of megacity Beijing.An unknown HONO source(Punknown)with an average of 7.3×106molecule/(cm3 s)was derived from the budget analysis during daytime.Punknown provided four times more HONO than the reaction of NO with OH did.The diurnal variation of Punknown showed an apparent photo-enhanced feature with a maximum around 12:00,which was consistent with previous studies at forest and rural sites.Laboratory studies proposed new mechanisms to recruit NO2 and J(NO2)in order to explain a photo-enhancement of of Punknown.In this study,these mechanisms were validated against the observation-constraint Punknown.The reaction of exited NO2 accounted for only 6%of Punknown,and Punknown poorly correlated with[NO2](R=0.26)and J(NO2)[NO2](R=0.35).These results challenged the role of NO2 as a major precursor of the missing HONO source.YANG Qiang SU Hang LI Xin CHENG YaFang LU KeDing CHENG Peng GU JianWei GUO Song HU Min ZENG LiMin ZHU Tong ZHANG YuanHang 2014Science China Chemistry2014,57,7:8
