| 3 | A Data-Driven Clustering Recommendation Method for Single-Cell RNA-Sequencing Data显示文摘Recently,the emergence of single-cell RNA-sequencing(scRNA-seq)technology makes it possible to solve biological problems at the single-cell resolution.One of the critical steps in cellular heterogeneity analysis is the cell type identification.Diverse scRNA-seq clustering methods have been proposed to partition cells into clusters.Among all the methods,hierarchical clustering and spectral clustering are the most popular approaches in the downstream clustering analysis with different preprocessing strategies such as similarity learning,dropout imputation,and dimensionality reduction.In this study,we carry out a comprehensive analysis by combining different strategies with these two categories of clustering methods on scRNA-seq datasets under different biological conditions.The analysis results show that the methods with spectral clustering tend to perform better on datasets with continuous shapes in two-dimension,while those with hierarchical clustering achieve better results on datasets with obvious boundaries between clusters in two-dimension.Motivated by this finding,a new strategy,called QRS,is developed to quantitatively evaluate the latent representative shape of a dataset to distinguish whether it has clear boundaries or not.Finally,a data-driven clustering recommendation method,called DDCR,is proposed to recommend hierarchical clustering or spectral clustering for scRNA-seq data.We perform DDCR on two typical single cell clustering methods,SC3 and RAFSIL,and the results show that DDCR recommends a more suitable downstream clustering method for different scRNA-seq datasets and obtains more robust and accurate results. | Yu Tian Ruiqing Zheng Zhenlan Liang Suning Li Fang-Xiang Wu Min Li | 2021 | Tsinghua Science and Technology2021，26，5： | 3 |

| 8 | Effects of harvesting method and date on yield loss and seed quality of rapeseed显示文摘China is one of the major rapeseed production countries in the world,but harvesting mechanization was still backward,and high harvest loss was a key inhibiting factor for rapeseed production.To obtain optimum harvesting date for winter rape in the Yangtze River Valley of China,artificial simulated combine harvesting and artificial two-stage harvesting were adopted to find correlations between harvesting date,rape variety,seed oil content,protein content,grain moisture rate,rapeseed straw moisture content,rapeseed unthreshing rate,grain drop loss rate,thousand seeds weight(TSW),and harvesting economic coefficient(HEC).Analysis of variance showed that rapeseed oil content,protein content and TSW were correlated with rape variety;HEC was correlated with harvesting method and rape variety.Rape variety was the dominant factor of rapeseed oil content,protein content,TSW and HEC.Grain moisture rate,un-threshing rate,grain drop loss rate and straw moisture content were correlated with harvesting method and date.Harvesting date was the dominant factor of rapeseed moisture rate,un-threshing rate,grain drop loss rate and straw moisture rate.Single factor tests further proved that harvesting date had less impact on rapeseed oil content,protein content,TSW and HEC.It showed little correlation with rapeseed quality except grain moisture rate.The optimum harvesting date could be known by change in straw moisture content.The combined harvesting should be carried out during the grain moisture content of 15%-20%with TSW stabilized in the highest level.The twostage harvesting cutting should be carried out at the grain moisture content of 35%-40%.This research offered a reference to harvesting method and date for rape cultivated in the Yangtze River Valley. | Min Zhang Chongyou Wu Mei Jin Senlin Mu Suning Liang Qing Tang | 2019 | Oil Crop Science2019，4，3： | 3 |

| 11 | Probing thermally-induced structural evolution during the synthesis of layered Li-, Na-, or K-containing 3d transition-metal oxides显示文摘Layered alkali-containing 3d transition-metal oxides are of the utmost importance in the use of electrode materials for advanced energy storage applications such as Li-,Na-,or K-ion batteries.A significant challenge in the field of materials chemistry is understanding the dynamics of the chemical reactions between alkali-free precursors and alkali species during the synthesis of these compounds.In this study,in situ high-resolution synchrotron-based X-ray diffraction was applied to reveal the Li/Na/K-ion insertion-induced structural transformation mechanism during high-temperature solid-state reaction.The in situ diffraction results demonstrate that the chemical reaction pathway strongly depends on the alkali-free precursor type,which is a structural matrix enabling phase transi-tions.Quantitative phase analysis identifies for the first time the decomposition of lithium sources as the most critical factor for the formation of metastable intermediates or impurities during the entire process of Li-rich layered Li[Li_(0.2)Ni_(0.2)Mn_(0.6)]O_(2) formation.Since the alkali ions have different ionic radii,Na/K ions tend to be located on prismatic sites in the defective layered structure(Na_(2/3-x)[Ni_(0.25)Mn_(0.75)]O_(2) or K_(2/3-x)[Ni_(0.25)Mn_(0.75)]O_(2))during calcination,whereas the Li ions prefer to be localized on the tetrahedral and/or octahedral sites,forming O-type structures. | Weibo Hua Xiaoxia Yang Nicola PMCasati Laijun Liu Suning Wang Volodymyr Baran Michael Knapp Helmut Ehrenberg Sylvio Indris | 2022 | eScience2022，2，2： | 2 |
