Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 5 | XingNaoJing injections protect against cerebral ischemia/reperfusion injury and alleviate blood-brain barrier disruption in rats, through an underlying mechanism of NLRP3 inflammasomes suppression显示文摘The aim of this study was to explore the neuroprotective effect and mechanism of XingNaoJing injections(XNJ) on cerebral ischemia injury and blood-brain barrier(BBB) disruption. Middle cerebral artery occlusion(MCAO) method was applicated to establish the model of cerebral ischemia/reperfusion(I/R) injury in rats. BBB permeability after I/R injury was assessed with the leaking amount of Evans Blue and the expression of occludin and ZO-1. The expression of NOD-like receptor family, pyrin domain containing(NLRP3) was checked to explore the inhibition of inflammation by XNJ. The results showed that XNJ could significantly increase the survival percent, decrease the infarct area and ameliorate neurological deficits and brain damage after I/R injury. Leaking amount of Evans Blue was reduced by XNJ, and the expression of tight junction protein, occludin and ZO-1 was also up-regulated by XNJ, which showed a role of protection on BBB disruption. The expression of NLRP3 was inhibited after exposure of XNJ, which was associated with inhibition of the inflammatory response. | 10 | The revised Atlanta criteria 2012 altered the classification,severity assessment and management of acute pancreatitis显示文摘BACKGROUND:The Atlanta criteria for acute pancreatitis(AP) has been revised recently.This study was to evaluate its practical value in classification of AP,the severity assessment and management.METHODS:The clinical features,severity classification,outcome and risk factors for mortality of 3212 AP patients who had been admitted in Ruijin Hospital from 2004 to 2011 were analyzed based on the revised Atlanta criteria(RAC) and the original Atlanta criteria(OAC).RESULTS:Compared to the OAC group,the incidence of severe acute pancreatitis(SAP) was decreased by approximately one half(13.9% vs 28.2%) in the RAC group.The RAC presented a lower sensitivity but higher specificity,and its predictive value for severity and poor outcome was higher than those of the OAC.The proportion of SAP diagnosis and ICU admission in the early phase in the RAC group was significantly lower than that in the OAC group(P<0.05).Based on the RAC,the risk factors for death among SAP patients were older age,high CT severity index(CTSI),renal failure,cardiovascular failure,acute necrotic collection and walled-off necrosis.Compared to the OAC,the acute physiology and chronic health evaluation II(APACHE II) score,Ranson score,idiopathic etiology,respiratory failure and laparotomy debridement were not risk factors of death in contrast to walled-off necrosis.Interestingly,hypertriglyceridemia-related SAP had good outcomes in both groups.CONCLUSIONS:The RAC showed a higher predictive value for severity and poorer outcome than the OAC.However,the RAC resulted in fewer ICU admissions in the early phase due to its lower sensitivity for diagnosis of SAP.Among SAP cases,older age,high CTSI,renal and cardiovascular failure,complications of acute necrotic collection and walled-off necrosis were independent risk factors for mortality. | Jie Huang Hong-Ping Qu Yun-Feng Zheng Xu-Wei Song Lei Li Zhi-Wei Xu En-Qiang Mao Er-Zhen Chen | 2016 | Hepatobiliary & Pancreatic Diseases International2016,15,3: | 15 |
| 11 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. | 12 | Effects of viral infection and microbial diversity on patients with sepsis:A retrospective study based on metagenomic next-generation sequencing显示文摘BACKGROUND: The study aims to investigate the performance of a metagenomic next-generationsequencing (NGS)-based diagnostic technique for the identifi cation of potential bacterial and viral infectionsand eff ects of concomitant viral infection on the survival rate of intensive care unit (ICU) sepsis patients.METHODS: A total of 74 ICU patients with sepsis who were admitted to our institution from February1, 2018 to June 30, 2019 were enrolled. Separate blood samples were collected from patients for bloodcultures and metagenomic NGS when the patients’ body temperature was higher than 38 °C. Patients’demographic data, including gender, age, ICU duration, ICU scores, and laboratory results, were recorded.The correlations between pathogen types and sepsis severity and survival rate were evaluated.RESULTS: NGS produced higher positive results (105 of 118;88.98%) than blood cultures(18 of 118;15.25%) over the whole study period. Concomitant viral infection correlated closelywith sepsis severity and had the negative effect on the survival of patients with sepsis. However,correlation analysis indicated that the bacterial variety did not correlate with the severity of sepsis.CONCLUSIONS: Concurrent viral load correlates closely with the severity of sepsis and thesurvival rate of the ICU sepsis patients. | 14 | Chemical screen identifies a geroprotective role of quercetin in premature aging显示文摘Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. Grain yields of proso millet genotypes were significantly influenced by environment(E), genotype(G) and their interaction(G×E)(P<0.1%). G×E interaction effect was six times higher than G effect in non-waxy group and seven times in waxy group. N04-339 in non-waxy and Neimi 6(NM6) in waxy showed higher grain yields and stability compared with other genotypes. Also, Neimi 9(NM9, a non-waxy cultivar) and 90322-2-33(a waxy cultivar) showed higher adaptability in 7 and in 11 locations, respectively. For non-waxy, Dalat, Inner Mongolia(E2) and Wuzhai, Shanxi(E5) were the best sites among all the locations for maximizing the variance among candidate cultivars, and Yanchi, Ningxia(E10) had the best representativeness. Wuzhai, Shanxi(e9) and Yanchi, Ningxia(e14) were the best representative locations, and Baicheng, Jilin(e2) was better discriminating location than others for waxy genotypes. | 18 | Central obesity and nonalcoholic fatty liver disease risk after adjusting for body mass index显示文摘AIM:To investigate whether central obesity is associated with nonalcoholic fatty liver disease(NAFLD) formation after adjusting for general obesity.METHODS:The online databases Pub Med,EMBASE,and ISI Web of Science were searched for studies estimating the influence of central obesity on NAFLD occurrence published through April 2014.Studies that did not adjust for body mass index(BMI) were excluded.In addition,the independent effect of BMI was also assessed with the included studies.The pooled effect sizes and 95% confidence intervals(CIs) were calculated using random- or fixed-effects models based on the degree of heterogeneity.Furthermore,subgroup analyses,meta-regression,sensitivity analyses,and publication bias were performed.RESULTS:Twenty eligible studies were identified.The summary odds ratio(OR) values per-unit increase in waist circumference(WC) and BMI for NAFLD formation were 1.07(95%CI:1.03-1.10,I2 = 73.9%,n = 11 studies) and 1.25(95%CI:1.13-1.38,I2 = 88.7%,n = 11 studies),respectively.When the indices were expressed as binary variables(with the non-obesity group as reference),the pooled OR in WC,waist-tohip ratio,and BMI were 2.34(95%CI:1.83-3.00,I2 = 41.8%,n = 7 studies),4.06(95%CI:1.53-10.79,I2 = 65.7%,n = 3 studies),and 2.85(95%CI:1.60-5.08,I2 = 57.8%,n = 5 studies),respectively.Using the same studies as the latter(n = 5),pooled OR in WC was 3.14(95%CI:2.07-4.77),which is greater than that in BMI.CONCLUSION:Central obesity may pose a greater threat to national health than general obesity,although both are independently associated with increased risk of NAFLD. | Qing Pang Jing-Yao Zhang Si-Dong Song Kai Qu Xin-Sen Xu Su-Shun Liu Chang Liu | 2015 | World Journal of Gastroenterology2015,21,5: | 13 |

| 20 | Ore-forming event and geodynamic setting of molybdenum deposit at Wenquan in Gansu Province,Western Qinling显示文摘The Wenquan molybdenum deposit is associated with a Triassic granite in this area.The Wenquan granite is enriched in LILE and LREE,poor in HFSE,and has significantly higher contents of alkali(K2O+Na2O) and Sr,Ba than those of the island arc volcanic rocks.These geochemical characteristics are similar to post-collisional granites in high K calc-alkaline series.Studies of major elements,trace elements,REEs and chronology of the Wenquan pluton show that,in the geodynamic transition stage of continent-continent convergence to extension,the partial melting of the enriched lithospheric mantle generated the basaltic magma and triggered the partial melting of the thickened lower crust which produceded the acidic magma,and the Wenquan pluton was formed by mixing of the two magmas.Molybdenite Re-Os isotopic dating gave Os model ages of 212.7±2.6 Ma to 215.1±2.6 Ma with a weighted mean of 214.1±1.1 Ma,and an Re-Os isochron age of 214.4±7.1 Ma.These ages are close to K-Ar ages(223 to 226 Ma) and a SHRIMP zircon U-Pb age(223±7 Ma) for the Wenquan granite within the error range,but relatively younger.This implies that the Mo mineralization occurred in a late stage of the magmatic intrusion,and the metallogenesis took place in the transition stage from syn-collision to post-collision in the tectonic setting of the Qinling Orogenic Belt(QOB) after continental collision be-tween the North China Block(NCB) and the South China Block(SCB).This process is also corre-sponding to the geological events of metamorphism and deformation in South Qinling,closure of the Mian-Lue oceanic basin,and exhumation of the Dabie-Sulu ultrahigh-pressure metamorphic rocks.The large-scale continent-continent collision between NCB and SCB in the middle Triassic triggered sig-nificant crustal thickening and exhumation of subducted slab.In the late Triassic,the tectonic setting was transformed to the transition stage from collision to extension.Materials from the asthenospheric mantle would ascend into the root of the lower crust,which could induce partial melting of the lower crust and generate Mo-enriched granitic magma.The ore-forming elements enriched in the fluid derived from the condensation and fraction of the magma resulted in the Mo mineralization.The Mo deposits in the QOB are mainly formed in two episodes,namely 220± Ma and 140± Ma.The two episodes of metal-logenesis were developed in the tectonic transition settings from compression to extension,but they were in the different stages of the tectonic evolution.The occurrence of the Wenquan Mo-bearing plu-ton indicates that the Triassic tectonic-magmatic belt of Western Qinling is another favorable region for Mo mineralization in the QOB.Therefore,it is significant to pay more attention to evaluation of the ore-forming potentiality in the Triassic granites in Western Qinling. | ZHU LaiMin DING ZhenJu YAO ShuZhen ZHANG GuoWei SONG ShiGang QU WenJun GUO Bo LEE Ben | 2009 | Chinese Science Bulletin2009,54,13: | 12 |
