1Prevention and treatment technologies of railway tunnel water inrush and mud gushing in China显示文摘Water inrush and mud gushing are one of the biggest hazards in tunnel construction. Unfavorable geological sections can be observed in almost all railway tunnels under construction or to be constructed, and vary in extent. Furthermore, due to the different heights of mountains and the lengths of tunnels, the locations of the unfavorable geological sections cannot be fully determined before construction, which increases the risk of water inrush and mud gushing. Based on numerous cases of water inrush and mud gushing in railway tunnels, the paper tries to classify water inrush and mud gushing in railway tunnels in view of the conditions of the surrounding rocks and meteorological factors associated with tunnel excavation. In addition, the causes of water inrush and mud gushing in combination of macroand micromechanisms are summarized, and site-specifc treatment method is put forward. The treatment methods include choosing a method of advance geological forecast according to risk degrees of different sections in the tunnel, determining the items of predictions, and choosing the appropriate methods, i.e. draining-oriented method, blocking-oriented method or draining-and-blocking method. The treatment technologies of railway water inrush and mud gushing are also summarized, including energy relief and pressure relief technology, advance grouting technology, and advance jet grouting technology associated with their key technical features and applicable conditions. 2Notoginsenoside Ft1 acts as a TGR5 agonist but FXR antagonist to alleviate high fat diet-induced obesity and insulin resistance in mice显示文摘Obesity and its associated complications are highly related to a current public health crisis around the world.A growing body of evidence has indicated that G-protein coupled bile acid(BA) receptor TGR5(also known as Gpbar-1) is a potential drug target to treat obesity and associated metabolic disorders.We have identified notoginsenoside Ftl(Ftl) from Panax notoginseng as an agonist of TGR5 in vitro.However,the pharmacological effects of Ftl on diet-induced obese(DIO) mice and the underlying mechanisms are still elusive.Here we show that Ftl(100 mg/100 diet) increased adipose lipolysis,promoted fat browning in inguinal adipose tissue and induced glucagon-like peptide-1(GLP-1) secretion in the ileum of wild type but not Tgr5^(-/-) obese mice.In addition,Ftl elevated serum free and taurineconjugated bile acids(BAs) by antagonizing Fxr transcriptional activities in the ileum to activate Tgr5 in the adipose tissues.The metabolic benefits of Ftl were abolished in Cyp27 al^(-/-) mice which have much lower BA levels.These results identify Ftl as a single compound with opposite activities on two key BA receptors to alleviate high fat diet-induced obesity and insulin resistance in mice.Lili Ding Qiaoling Yang Eryun Zhang Yangmeng Wang Siming Sun Yingbo Yang Tong Tian Zhengcai Ju Linshan Jiang Xunjiang Wang Zhengtao Wang Wendong Huang Li Yang 2021Acta Pharmaceutica Sinica B2021,11,6:10
3Bile acid signaling and bariatric surgery显示文摘The rapid worldwide rise in obesity rates over the past few decades imposes an urgent need to develop effective strategies for treating obesity and associated metabolic complications.Bariatric surgical procedures,such as Roux-en-Y gastric bypass(RYGB)and vertical sleeve gastrectomy(VSG),currently provide the most effective treatment for obesity and type 2 diabetes(T2D),as well as for non-alcoholic steatohepatitis(NASH).However,the underlying mechanisms of the beneficial effects of bariatric surgery remain elusive.Recent studies have identified bile acids as potential signaling molecules involved in the beneficial effects of bariatric surgery.This review focuses on the most recent studies on the roles of bile acids and bile acid receptors Farnesoid X receptor(FXR)and G protein-coupled bile acid receptor 5(TGR5)in bariatric surgery.We also discuss the possibility of modulating bile acid signaling as a pharmacological therapeutic approach to treating obesity and its associated metabolic complications.Jingyan Tian Silvia Huang Siming Sun Lili Ding Eryun Zhang Yanjun Liu Wendong Huang 2017Liver Research2017,1,4:2

5Epidemiology and clinical characteristics of burns in China's Mainland from 2009 to 2018显示文摘Background:Burn injuries place a heavy burden on the global healthcare system.However,there is still a lack of nationwide studies on the epidemiological characteristics of burn patients in China's Mainland.The present study aims to accurately analyze the clinical characteristics of burn patients by collecting data in China's Mainland from 2009 to 2018,which will provide effective strategies for healthcare systems and the government in China's Mainland.Methods:Patients admitted for burn injuries to 196 hospitals in 31 provinces,autonomous regions and municipalities in China's Mainland from 2009 to 2018 were included.The data collected included sex,age,month distribution,etiology,region,clinical outcome,injury anatomical location,total burn surface area and mortality.SPSS 19.0 software was used to analyze the data.Results:From 2009 to 2018,the burn patients were 333,995(0.76%),which included 222,480(66.61%)males and 111,515(33.39%)females.From 2009 to 2018,the number of individuals admit-ted to hospitals for burns showed a downward trend year by year.The burn patients accounted for the highest proportion of inpatients in 0-10 years(38.10%),followed by 40-50 years(13.14%).The highest cure ratio of burn inpatients was in the 20-30 age group(31394,71.53%).Among 31 provinces,autonomous regions and municipalities,the province with the highest proportion of total inpatients caused by burns was Inner Mongolia(4.61%),followed by Zhejiang(3.17%),Hainan(2.88%)and Xinjiang(2.64%).Summer(29.16%)was the season with the highest incidence of burn patients admitted to hospitals,followed by spring(25.6%).Scalding(60.19%)was the most frequent kind of burn treated,followed by fire(20.45%).The patients had multiple burn sites(68.89%)most often,followed by burns on the lower limbs(10.91%).From 0%to 10%total body surface area(TBSA)accounted for the highest ratio(37.19%),followed by 90-100%TBSA(21.74%).Conclusions:The present study is the first to describe the associated situation and trends of burn patients in China's Mainland from 2009 to 2018.Our findings will serve as the latest clinical evidence for healthcare planning and prevention efforts in China and other countries.Jie Yang Guanglei Tian Jianchao Liu Huajuan Bai Shuxu Yang Mingzi Ran Hongyu Li Kui Ma Siming Yang Xiaobing Fu 2022Burns & Trauma2022,10,1:1
63D Bioprinted Skin Substitutes for Accelerated Wound Healing and Reduced Scar显示文摘The shortage of skin for grafting continues to be a major problem in the treatment of serious skin injuries.3D bioprinting provides a new way to solve this problem.However,current 3D printed skin is less effective in treatment of large wounds because of severe shrinkage and scarring.In this study,bionically designed bilayer skin was fabricated using an extrusion-based bioprinter and a gelatin/sodium alginate/gelatin methacrylate hydrogel with excellent physical and biological properties.Full-thickness skin wounds were created in the back of nude mice and treated with bioprinted skin or hydrogel.Bioprinted skin accelerated wound healing,reduced wound contraction and scarring,and facilitated wound skin epithelialization compared with the bioprinted hydrogel or untreated wound.The skin from the wound was collected 28 days after grafting for histology and immunofluorescence analysis.The thickness of the dermis and epidermis of the bioprinted skin was similar to that of nude mice.Microvascular formation in the dermis and dense keratinocytes in the epidermis of the bioprinted skin were observed.This study provides a potential treatment strategy for reducing skin contraction and scar in large skin wounds.Qin Lian Tian Jiao Tingze Zhao Huichao Wang Siming Yang Dichen Li 2021Journal of Bionic Engineering2021,18,4:1
7Construction of Lactuca sativa Plastid Transformation Vector and High-level Expression of gfp Gene in Escherichia coli显示文摘Using genomic DNA of bolting-tolerant lettuce as a template,flanking fragments of lettuce plastid rpo A gene were amplified and cloned by PCR. Targeting the sites of these two fragments,homologous recombinant fragments of exogenous gene were integrated to construct lettuce plastid expression vector p Brpo AGFP,which harbored the expression cassette Prrn-gfp-aad A-Tpsb A. The results showed that the amplified flanking fragments were 1.2 and 1.1 kb in size. After sequencing,restriction digestion,ligation and transformation,lettuce plastid expression vector containing expression cassette Prrn-gfp-aad A-Tpsb A was constructed and confirmed by SDS-PAGE electrophoresis. The results of SDS-PAGE electrophoresis indicated that gfp gene was efficiently expressed under the regulation of plasmid specific promoter Prrn and terminator Tpsb A. GFP accounted for 45. 6% of total soluble proteins; inclusion bodies accounted for 47.5 % of bacterial proteins,which reached relatively high expression levels. 8Cloning and Sequence Analysis of Polygalacturonase(PG) Promoter in Tomato显示文摘In this study,1 500 bp PG gene promoter was amplified from leaves of tomato cultivar‘Zhongshu No.4'.The bioinformatic analysis of PG promoter sequences was conducted.PG promoter elements were predicted and analyzed by PLANTCARE and PLACE.The results showed that tomato PG promoter contained multiple cis-acting regulatory elements such as typical basic elements TATA-Box and CAAT-Box,light responsive elements 3-AF1 binding site,AT1-motif,ATCTmotif,Box 4,Box I,GA-motif,GT1-motif,Sp1 and MRE,heat stress-responsive element HSE,ethylene-responsive element ERE,meristem-specific regulatory element CCGTCC-box,endosperm expression-related regulatory elements GCN4_motif and Skn-1_motif,defense and stress responsiveness element TC-rich repeats,and circadian control-related element circadian,indicating that the expression of tomato PG gene is related to light,temperature,hormone,stress and other factors.This study laid the foundation for subsequent research about regulation of PG gene expression in plants.Siming HOU Liying ZHOU Lulu BU Chunlei YANG Ting GAO Tian TIAN Zheng'an YANG 2016Agricultural Biotechnology2016,5,2:0
