| 1 | The Epitope Study on the SARS-CoV Nucleocapsid Protein显示文摘The nucleocapsid protein (N protein) has been found to be an antigenic protein in a number of coronaviruses. Whether the N protein in severe acute respiratory syndrome-associated coronavirus (SARS-CoV) is antigenic remains to be elucidated. Using Western blot and Enzyme-linked Immunosorbent Assay (ELISA),the recombinant N proteins and the synthesized peptides derived from the N protein were screened in sera from SARS patients. All patient sera in this study displayed strong positive immunoreactivities against the recombinant N proteins,whereas normal sera gave negative immunoresponses to these proteins, indicating that the N protein of SARS-CoV is an antigenic protein. Furthermore, the epitope sites in the N protein were determined by competition experiments, in which the recombinant proteins or the synthesized peptides competed against the SARS-CoV proteins to bind to the antibodies raised in SARS sera. One epitope site located at the C-terminus was confirmed as the most antigenic region in this protein. A detailed screening of peptide with ELISA demonstrated that the amino sequence from Codons 371 to 407 was the epitope site at the C-terminus of the N protein. | 4 | Preparation, characterization, antimicrobial and cytotoxicity studies of copper/zincloaded montmorillonite显示文摘Background: A series of modified montmorillonites(Mt) including zinc-loaded Mt(Zn-Mt), copper-loaded Mt(Cu-Mt), copper/zinc-loaded Mt with different Cu/Zn ratio(Cu/Zn-Mt-1, Cu/Zn-Mt-2, Cu/Zn-Mt-3) were prepared by an ion-exchange reaction, and characterized using X-ray diffraction(XRD), fourier transformed infrared spectroscopy(FTIR) and transmission electron microscopy(TEM). The specific surface areas, antimicrobial activity and cytotoxicity of the modified Mt were investigated.Results: In the modified Mt, hydrated Cu ions and Zn ions were exchanged in the interlayer space of Mt and the particles were irregular shapes. The results showed that Cu/Zn-Mt enhanced antibacterial and antifungal activity compared with Zn-Mt and Cu-Mt possibly due to the synergistic effect between Cu and Zn. Among the Cu/Zn-Mt with different Cu/Zn raitos, Cu/Zn-Mt with a Cu/Zn ratio of 0.98 or 0.51 showed higher antimicrobial activity against gram-negative bacteria(Escherichia coli), gram-positive bacteria(Staphylococcus aureus), fungi(Candida albicans).Moreover, the antimicrobial activity of Cu/Zn-Mt was correlated with its specific surface area. | 5 | A novel PDZ domain-containing gene is essential for male sex differentiation and maintenance in yellow catfish(Pelteobagrus fulvidraco)显示文摘The sex-determining genes are found to be variable among different fish species. Yellow catfish(Pelteobagrus fulvidraco) is an important aquaculture fish species in China with XX/XY sex-determining type. Recently, YY super-male yellow catfish has been successfully produced by combining hormonalinduced sex reversal method with sex chromosome-linked markers. Here, we identified a novel PDZ domain-containing gene in yellow catfish designated as pfpdz1, in whose intron the sex-linked marker was located. The coding sequence of pfpdz1 in Y chromosome was identical to that in X chromosome except a missense SNP(A/T) that changes an amino acid(E8V) in the N-terminal region. Pfpdz1 displayed male-specific expression during sex differentiation. Overexpression of pfpdz1 using additive transgenesis induces XX ovary to differentiate into testis-like tissue, while the targeted inactivation of pfpdz1 in Y chromosome using CRISPR/Cas9-mediated mutagenesis triggers ovarian differentiation. Furthermore,we demonstrated that pfpdz1 initiates testicular differentiation through upregulating expression of amh, dmrt1 and sox9a1, as well as downregulating expression of cyp19a1, foxl2 and wnt4. | 6 | Electrodynamic Tethered Deorbit System Dynamics and Performance Analysis显示文摘Electrodynamic tethered deorbit technology is a novel way to remove abandoned spacecrafts like upper stages or unusable satellites. This paper investigates and analyses the deorbit performance and mission applicability of the electrodynamic tethered system. To do so, the electrodynamic tethered deorbit dynamics with multi-perturbation is firstly formulated, where the Earth magnetic field, the atmospheric drag, and the Earth oblateness effect are considered. Then, the key system parameters, including payload mass, tether length and tether type, are analyzed by numerical simulations to investigate their influences on the deorbit performance and to give the setting principles for choosing system parameters. | 8 | Nlrc3-like is required for microglia maintenance in zebrafish显示文摘Microglia are tissue-resident macrophages residing in the central nervous system (CNS) and play critical roles in removing cellular debris and infectious agents as well as regulating neurogenesis and neuronal activities. Yet, the molecular basis underlying the establishment of microglia pool and the maintenance of their homeostasis in the CNS remain largely undefined. Here we report the identification and characterization of a mutant zebrafish, which harbors a point mutation in the nucleotide-binding oligomerization domain (NOD) like receptor gene nlrc3-like, resulting in the loss of microglia in a temperature sensitive manner. Temperature shift assay reveals that the late onset of nlrc3-like deficiency leads to excessive microglia cell death. Further analysis shows that the excessive microglia death in nlrc3-like deficient mutants is attributed, at least in part, to aberrant activation of canonical inflammasome pathway. | 9 | A Strategy for Searching Antigenic Regions in the SARS-CoV Spike Protein显示文摘In the face of the worldwide threat of severe acute respiratory syndrome (SARS)to human life, some of the most urgent challenges are to develop fast and accurate analytical methods for early diagnosis of this disease as well as to create a safe anti-viral vaccine for prevention. To these ends, we investigated the antigenicity of the spike protein (S protein), a major structural protein in the SARS-coronavirus (SARS-CoV). Based upon the theoretical analysis for hydrophobicity of the S protein, 18 peptides were synthesized. Using Enzyme-Linked Immunosorbent Assay (ELISA), these peptides were screened in the sera from SARS patients. According to these results, two fragments of the S gene were amplified by PCR and cloned into pET-32a. Both S fragments were expressed in the BL-21 strain and further purified with an affinity chromatography. These recombinant S fragments were confirmed to have positive cross-reactions with SARS sera, either by Western blot or by ELISA. Our results demonstrated that the potential epitope regions were located at Codons 469-882 in the S protein, and one epitope site was located at Codons 599-620. | 10 | Prevalence of Congenital Heart Disease at Extremely High Altitudes in Tibet and the Establishment of a New Free Treatment Model显示文摘Background: This study aimed to illustrate the prevalence of CHD by screening children in extremely high-altitudeareas (over 4000 m to even 5000 m above sea level) and explore an aid model for early diagnosis and treatmentfor the Tibetan population. Methods: A total of 2242 students from different schools in Ngamring County,Xigaze City, Tibet from September 2019 to September 2020 were selected for screening. The students were examinedthrough the inquiry of their current medical history and family history, cardiac auscultation and a physicalexamination, in order to screen out the suspected cases of CHD, and then the suspected cases were confirmed bycardiac color ultrasound examinations. After that, positive patients were be transferred to Shandong Province forfree treatment. Results: The prevalence of CHD among children in Ngamring County was 3.70% (83 cases),which is the highest incidence rate ever reported. The most common type of CHD was patent ductus arteriosus(PDA), accounting for 55.42% of the total number of cases screened, followed by atrial septal defect (ASD) andventricular septal defect (VSD), accounting for 28.92% and 12.05%, respectively. The prevalence of CHD in childrenat high altitude was significantly higher than that in children at low altitude. | 11 | Asian,regional,and national burdens of respiratory tract cancers and associated risk factors from 1990 to 2019:A systematic analysis for the global burden of disease study 2019显示文摘Background:Respiratory cancer is the leading cause of cancer-related deaths worldwide,but its statistics vary between the East and West.This study aimed to estimate the burdens of tracheal,bronchus,and lung(TBL)cancer and larynx cancer and their attributable risks from 1990 to 2019 in Asia,and at regional and national levels.Methods:This research evaluated the incidence,mortality,years lived with disability,years of life lost,and disability-adjusted life years(DALYs)for respiratory tract cancers using the Global Burden of Diseases,Injuries,and Risk Factors Study(GBD)2019 database.Age-standardized rates were calculated for TBL cancer from 1990 to 2019,adjusted for smoking and socio-demographic index(SDI).Deaths from TBL cancer and larynx cancer attributable to each risk factor were estimated for 33 Asian countries.Results:The age-standardized incidence and death rates for TBL cancer in Asia declined from 2010 to 2019,while the incidence rate of larynx cancer increased.Smoking was the leading specific risk factor for deaths from both TBL and larynx cancers.The burden of TBL cancer in Asian countries was influenced by SDI and smoking,particularly among males in Central Asia.Deaths,DALYs,and incidences of larynx cancer in East Asia had not changed significantly over the past 30 years,but showed slight downward trends in males and both sexes combined,and an upward trend in females in recent years.Conclusions:The past decade saw increases in numbers of incident cases and deaths from TBL cancer and larynx cancer in Asia.SDI and smoking were the main factors influencing the disease burden of TBL cancer in Asian countries.This study highlights the need for tailored cancer control programs to address the burden of respiratory tract cancers in different Asian countries. | Ran Zhong Xiuyu Cai Jianfu Li Peiling Chen Runchen Wang Xinting Li Feng Li Shan Xiong Caichen Li Huiting Wang Yang Xiang Shuting Zhan Ziwen Yu Haixuan Wang Chunyan Li Lixuan Lin Wenhai Fu Xin Zheng Shen Zhao Kang Zhang Jianxing He | 2023 | Chinese Medical Journal Pulmonary and Critical Care Medicine2023,1,4: | 1 |
| 12 | Evaluation of antimicrobial and anticancer activities of three peptides identified from the skin secretion of Hylarana latouchii显示文摘The skins of frogs of the family Ranidae are particularly rich sources of biologically active peptides, among which antimicrobial peptides (AMPs) constitute the major portion. Some of these have attracted the interest of researchers because they possess both antimicrobial and anticancer activities. In this study, with ‘shotgun’ cloning & MS/MS fragmentation, three AMPs, homologues of family brevinin-1 (brevinin-1HL), & temporin (temporin-HLa and temporin-HLb), were discovered from the skin secretion of the broad-folded frog, Hylarana latouchii. They exhibited various degrees of antimicrobial & antibiofilm activities against test microorganisms & hemolysis on horse erythrocytes. It was found that they could induce bacteria death through disrupting cell membranes & binding to bacterial DNA. In addition, they also showed different potencies towards human cancer cell lines. The secondary structure & physicochemical properties of each peptide were investigated to preliminarily reveal their structure–activity relationships. Circular dichroism spectrometry showed that they all adopted a canonical α-helical conformation in membrane-mimetic solvents. Notably, the prepropeptide of brevinin-1HL from H. latouchii was highly identical to that of brevinin-1GHd from Hylarana guentheri, indicating a close relationship between these two species. | 15 | Water molecules and oxygen-vacancy modulation of vanadium pentoxide with fast kinetics toward ultrahigh power density and durable flexible all-solid-state zinc ion battery显示文摘Aqueous zinc ion battery(ZIB)with many virtues such as high safety,cost-effective,and good environmental compatibility is a large-scale energy storage technology with great application potential.Nevertheless,its application is severely hindered by the slow diffusion of zinc ions in desirable cathode materials.Herein,a technique of water-incorporation coupled with oxygen-vacancy modulation is exploited to improve the zinc ions diffusion kinetics in vanadium pentoxide(V_(2)O_5)cathode for ZIB.The incorporated water molecules replace lattice oxygen in V_(2)O_5,and function as pillars to expand interlayer distance.So the structural stability can be enhanced,and the zinc ions diffusion kinetics might also be promoted during the repeated intercalation/deintercalation.Meanwhile,the lattice water molecules can effectively enhance conductivity due to the electronic density modulation effect.Consequently,the modulated V_(2)O_5(H-V_(2)O_5)cathode behaves with superior rate capacity and stable durability,achieving 234 mA h g^(-1)over 9000 cycles even at 20 A g^(-1).Furthermore,a flexible all-solid-state(ASS)ZIB has been constructed,exhibiting an admirable energy density of 196.6 Wh kg^(-1)and impressive power density of 20.4 kW kg^(-1)as well as excellent long-term lifespan.Importantly,the assembled flexible ASS ZIB would be able to work in a large temperature span(from-20 to 70℃).Additionally,we also uncover the energy storage mechanism of the H-V_(2)O_5 electrode,offering a novel approach for creating high-kinetics cathodes for multivalent ion storage. | Wenda Qiu Yunlei Tian Shuting Lin Aihua Lei Zhangqi Geng Kaitao Huang Jiancong Chen Fuchun Huang Huajie Feng Xihong Lu | 2023 | Journal of Energy Chemistry2023,,10: | 0 |
| 16 | High rate and ultralong life flexible all-solid-state zinc ion battery based on electron density modulated NiCo_(2)O_(4) nanosheets显示文摘The development of zinc ion batteries (ZIBs) with large capacity,high rate,and durable cathode material is a crucial and urgent task.Ni Co_(2)O_(4)(NCO) has received ever-growing interest as a potential cathode material for ZIBs,owing to the high theoretical capacity,rich source,cost-effective,and versatile redox nature.However,due to the slow dynamics of the NCO electrodes,its practical application in highperformance systems is severely limited.Herein,we report an electron density modulated NCO nanosheets (N-NCO NSs) with high-kinetics Zn^(2+)-storage capability as an additive-free cathode for flexible all-solid-state (ASS) ZIBs.By virtue of the enhanced electronic conductivity,improved reaction kinetics,and increased active sites,the optimized N-NCO NSs electrode delivers a high capacity of 357.7 m Ah g^(-1)at 1.0 A g^(-1)and a superior rate capacity of 201.4 m Ah g^(-1)at 20 A g^(-1).More importantly,a flexible ASS ZIBs device is manufactured using a solid polymer electrolyte of a poly (vinylidene fluoride hexafluoropropylene)(PVDF-HFP) film.The flexible ASS ZIBs device shows superb durability with 80.2%capacity retention after 20,000 cycles and works well in the range of-20–70℃.Furthermore,the flexible ASS ZIBs achieves an impressive energy density as high as 578.1 W h kg^(-1)with a peak power density of 33.6 k W kg^(-1),substantially outperforming those latest ZIBs.This work could provide valuable insights for constructing high-kinetics and high-capability cathodes with long-term stability for flexible ASS ZIBs. | Wenda Qiu Yunlei Tian Zhenchao Lin Shuting Lin Zhangqi Geng Kaitao Huang Aihua Lei Fuchun Huang Huajie Feng Fengze Ding Yu Li Xihong Lu | 2022 | Journal of Energy Chemistry2022,31,7: | 0 |
