| 2 | Evaluation of antimicrobial and anticancer activities of three peptides identified from the skin secretion of Hylarana latouchii显示文摘The skins of frogs of the family Ranidae are particularly rich sources of biologically active peptides, among which antimicrobial peptides (AMPs) constitute the major portion. Some of these have attracted the interest of researchers because they possess both antimicrobial and anticancer activities. In this study, with ‘shotgun’ cloning & MS/MS fragmentation, three AMPs, homologues of family brevinin-1 (brevinin-1HL), & temporin (temporin-HLa and temporin-HLb), were discovered from the skin secretion of the broad-folded frog, Hylarana latouchii. They exhibited various degrees of antimicrobial & antibiofilm activities against test microorganisms & hemolysis on horse erythrocytes. It was found that they could induce bacteria death through disrupting cell membranes & binding to bacterial DNA. In addition, they also showed different potencies towards human cancer cell lines. The secondary structure & physicochemical properties of each peptide were investigated to preliminarily reveal their structure–activity relationships. Circular dichroism spectrometry showed that they all adopted a canonical α-helical conformation in membrane-mimetic solvents. Notably, the prepropeptide of brevinin-1HL from H. latouchii was highly identical to that of brevinin-1GHd from Hylarana guentheri, indicating a close relationship between these two species. | 3 | Immunotherapy as adjuvant therapy for a patient with adenosquamous carcinoma of the intrahepatic bile duct:A case report and literature review显示文摘Adenosquamous carcinoma(ASC)is a rare histological type of intrahepatic cholangiocarcinoma,which includes both adenocarcinoma and squamous cell carcinoma.The clinical features,physical examination,routine laboratory tests,and imaging examinations of patients with ASC are nonspecific.ASC is easily misdiagnosed as hepatocellular carcinoma,and patients with ASC always have a poor prognosis.This study reports a patient with ASC who was diagnosed based on pathological results,underwent surgical resection,and received postoperative chemotherapy(gemcitabine plus cisplatin)combined with immunotherapy(sintilimab).During the 1-year follow-up,the patient was in good condition,and no signs of cancer recurrence were noted.This case highlights that surgical resection and chemotherapy combined with immunotherapy may be feasible for patients with ASC. | Jun Feng Aimaiti Yasen Tianxing Dai Runbin Liang Zhihong Liao Ping He Zhihong Lin Guoying Wang | 2023 | Liver Research2023,7,2: | 0 |
| 4 | RFID indoor positioning based on semi-supervised actor-critic co-training显示文摘For large-scale radio frequency identification(RFID) indoor positioning system, the positioning scale is relatively large, with less labeled data and more unlabeled data, and it is easily affected by multipath and white noise. An RFID positioning algorithm based on semi-supervised actor-critic co-training(SACC) was proposed to solve this problem. In this research, the positioning is regarded as Markov decision-making process. Firstly, the actor-critic was combined with random actions and the unlabeled best received signal arrival intensity(RSSI) data was selected by co-training of the semi-supervised. Secondly, the actor and the critic were updated by employing Kronecker-factored approximation calculate(K-FAC) natural gradient. Finally, the target position was obtained by co-locating with labeled RSSI data and the selected unlabeled RSSI data. The proposed method reduced the cost of indoor positioning significantly by decreasing the number of labeled data. Meanwhile, with the increase of the positioning targets, the actor could quickly select unlabeled RSSI data and updates the location model. Experiment shows that, compared with other RFID indoor positioning algorithms, such as twin delayed deep deterministic policy gradient(TD3), deep deterministic policy gradient(DDPG), and actor-critic using Kronecker-factored trust region(ACKTR), the proposed method decreased the average positioning error respectively by 50.226%, 41.916%, and 25.004%. | 6 | Upregulation of TMCO3 Promoting Tumor Progression and Contributing to the Poor Prognosis of Hepatocellular Carcinoma显示文摘Background and Aims:TMCO3,a member of the monovalent cation:proton antiporter-2 family,has been annotated as a Na^(+)/H^(+)antiporter,but its pathophysiological role is still unclear.We aimed to investigate the expression profile,prognostic significance,and oncogenic role of TMCO3 in hepatocellular carcinoma(HCC).Methods:Bioinformatic analyses were conducted using transcriptome data from public databases to determine the expression,prognosis,and functional enrichment of TMCO3 in HCC.TMCO3 expression was further validated in an independent HCC cohort from our institution.The oncogenic role of TMCO3 in HCC was evaluated using in vitro and in vivo experiments.Results:The upregulated expression of TMCO3 was identified and verified in multiple HCC cohorts,and worse overall survival and recurrence-free survival were observed in patients with high TMCO3 expression.The overexpression and knockdown of TMCO3 could affect the proliferation and metastasis of HCC cells,which might be associated with the p53-induced cell cycle regulation and epithelial-mesenchymal transition,respectively.Notably,significant correlations were found between dysregulated TMCO3 and various antitumor agents.Its role in sorafenib sensitivity was further identified by in vitro experiments and the potential mechanism might be related to the regulation of apoptosis.Positive correlations were also identified between upregulation of TMCO3 and the increased infiltration of various immune cells and the elevated expression of multiple immune checkpoint genes in HCC.Conclusions:Upregulated TMCO3 could act as an oncogenic mediator and promote sorafenib resistance in HCC,providing a potential therapeutic target for HCC treatment. | Tianxing Dai Linsen Ye Mingbin Deng Guozhen Lin Rongqiang Liu Haoyuan Yu Wei Liu Yang Yang Guoying Wang | 2022 | Journal of Clinical and Translational Hepatology2022,10,5: | 0 |
