| 1 | Jianpijiedu Fang improves survival of hepatocarcinoma mice by affecting phosphatase and tensin homolog, phosphoinositide 3-kinase, and focal adhesion kinase显示文摘OBJECTIVE: To investigate the effect of Jianpijiedu Fang (JPJDF) on phosphatase and tensin homolog (PTEN), phosphoinositide 3-kinase (PI3K), and focal adhesion kinase (FAK), and on the survival of hepatocellular carcinoma (HCC) nude mice. METHODS: Forty male nude mice were randomly divided into 4 groups. Human HCC tissue was implanted in the livers of three groups. After 24 h, the three groups were treated respectively with JPJDF (37.5 g/kg), saline (20 mL/kg) and Tegafur (FT-207, 160 mg/kg) once a day for 10 weeks. The control group without implanting the tissue was concurrently treated with saline (20 mL/kg). The survival data and body weight of all mice were recorded, and expression levels of PTEN, PI3K and FAK in normal tissue and cancer tissue of the livers were eval-uated with immunohistochemical method. RESULTS: The cumulative survival rate of the mice in the JPJDF group was higher than those of the other groups. The rate of weight loss was the lowest in JPJDF group. The survivability and weight loss rate in FT-207 group were the poorest in all groups. The expression intensity of PTEN was higher in normal tissues than in cancer tissues, and lower in the normal tissues of HCC models than in that of mice without HCC. The PTEN expression intensity in normal tissue and cancer tissue from mice treated with FT-207 were lower than that from the mice treated with JPJDF or saline.The expression intensity of PI3K was higher in cancer tissue than in normal tissue. The PI3K expression intensity was the lowest in normal tissue and cancer tissue from mice treated with JPJDF, and the intensity from mice treated with FT-207 was the highest. In mice treated with JPJDF, the expression intensity of FAK was higher in the normal tissue and lower in the cancer tissue than those of the other treatment groups. | 3 | Subcutaneous and intramuscular fat transcriptomes show large differences in network organization and associations with adipose traits in pigs显示文摘Subcutaneous fat(SCF)and intramuscular fat(IMF)deposition is relevant to health in humans,as well as meat production and quality in pigs.In this study,we generated RNA sequence data for 122 SCF,120 IMF,and 87 longissimus dorsi muscle(LDM)samples using 155 F6 pigs from a specially designed heterogeneous population generated by intercrossing four highly selected European commercial breeds and four indigenous Chinese pig breeds.The phenotypes including waist back fat thickness and intramuscular fat content were also measured in the 155 F6 pigs.We found that the genes in SCF and IMF differed largely in both expression levels and network connectivity,and highlighted network modules that exhibited strongest gain of connectivity in SCF and IMF,containing genes that were associated with the immune process and DNA double-strand repair,respectively.We identified 215 SCF genes related to kinase inhibitor activity,mitochondrial fission,and angiogenesis,and 90 IMF genes related to lipolysis and fat cell differentiation,displayed a tissue-specific association with back fat thickness and IMF content,respectively.We found that cis-expression QTL for trait-associated genes in the two adipose tissues tended to have tissuedependent predictability for the two adipose traits.Alternative splicing of genes was also found to be associated with SCF or IMF deposition,but the association was much less extensive than that based on expression levels.This study provides a better understanding of SCF and IMF gene transcription and network organization and identified critical genes and network modules that displayed tissue-specific associations with subcutaneous and intramuscular fat deposition.These features are helpful for designing breeding programs to genetically improve the two adipose traits in a balanced way. | Yifeng Zhang Yingchun Sun Zhongzi Wu Xinwei Xiong Junjie Zhang Junwu Ma Shijun Xiao Lusheng Huang Bin Yang | 2021 | Science China(Life Sciences)2021,64,10: | 5 |
| 4 | Contribution of Baroreflex Afferent Pathway to NPY-Mediated Regulation of Blood Pressure in Rats显示文摘Neuropeptide Y (NPY), a metabolism-related cardiovascular factor, plays a crucial role in blood pressure(BP) regulation via peripheral and central pathways. The expression of NPY receptors (Y1R/Y2R) specific to baroreflex afferents impacts on the sexually dimorphic neural control of circulation. This study was designed to investigate the expression profiles of NPY receptors in the nodose ganglion (NG) and nucleus tractus solitary (NTS) under hypertensive conditions. To this end, rats with hypertension induced by NG-nitro-L-arginine methylester (L-NAME) or high fructose drinking (HFD), and spontaneously hypertensive rats (SHRs) were used to explore the effects/mechanisms of NPY on BP using functional, molecular, and electrophysiological approaches. The data showed that BP was elevated along with baroreceptor sensitivity dysfunction in model rats;Y1R was up-or down-regulated in the NG or NTS of male and female HFD/L-NAME groups,while Y2R was only down-regulated in the HFD groups as well as in the NG of the male L-NAME group. In SHRs,Y1R and Y2R were both down-regulated in the NTS, and not in the NG. In addition to NPY-mediated energy homeostasis, leptin-melanocortin activation may be essential for metabolic disturbance-related hypertension. We found that leptin and a-melanocyte stimulating hormone (aMSH) receptors were aberrantly down-regulated in HFD rats. In addition, a-MSH concentrations were reduced and NPY concentrations were elevated in the serum and NTS at 60 and 90 min after acute leptin infusion. Electrophysiological recordings showed that the decay time-constant and area under the curve of excitatory post-synaptic currents were decreased by Y1R activation in A-types, whereas, both were increased by Y2R activation in Ah-or C-types. | 5 | Deer Body Adaptive Threshold Segmentation Algorithm Based on Color Space显示文摘In large-scale deer farming image analysis,K-means or maximum between-class variance(Otsu)algorithms can be used to distinguish the deer from the background.However,in an actual breeding environment,the barbed wire or chain-link fencing has a certain isolating effect on the deer which greatly interferes with the identification of the individual deer.Also,when the target and background grey values are similar,the multiple background targets cannot be completely separated.To better identify the posture and behaviour of deer in a deer shed,we used digital image processing to separate the deer from the background.To address the problems mentioned above,this paper proposes an adaptive threshold segmentation algorithm based on color space.First,the original image is pre-processed and optimized.On this basis,the data are enhanced and contrasted.Next,color space is used to extract the several backgrounds through various color channels,then the adaptive space segmentation of the extracted part of the color space is performed.Based on the segmentation effect of the traditional Otsu algorithm,we designed a comparative experiment that divided the four postures of turning,getting up,lying,and standing,and successfully separated multiple target deer from the background.Experimental results show that compared with K-means,Otsu and hue saturation value(HSV)+K-means,this method is better in performance and accuracy for adaptive segmentation of deer in artificial breeding scenes and can be used to separate artificially cultivated deer from their backgrounds.Both the subjective and objective aspects achieved good segmentation results.This article lays a foundation for the effective identification of abnormal behaviour in sika deer. | Yuheng Sun Ye Mu Qin Feng Tianli Hu He Gong Shijun Li Jing Zhou | 2020 | Computers, Materials & Continua2020,,8: | 5 |

| 7 | Effect of polypeptide 2B1 on condition of dampness pattern in rats in terms of Traditional Chinese Medicine显示文摘OBJECTIVE: This study investigated how polypeptide 2B1 is involved in regulating and governing dampness in rat models with dampness pattern defined in terms ofTraditional Chinese Medicine.METHODS: We randomly divided 48 SPF10-week-old male Sprague-Dawley(SD) rats into a normal group, normal + Aristolochic acid I(AA-I) for5 min group, normal + AA-I for 60 min group,dampness pattern group(DS-Group), dampness pattern + AA-I for 5 min group, and dampness pattern + AA-I for 60 min group. Groups were then treated accordingly. The initial lithium insertion of crystal NiSi2 can reach up to 600 mAh·g-1, but large irreversible capacity occurrs simultaneously for serious structure transformation and the irreversible phase forms. XRD and XPS were employed to detect the crystal structure and composition changes produced by lithium insertion. The lithium insertion-extraction behavior of NiSi2 electrode is similar to that of silicon after the first discharge. | 9 | Correlation between pathogenesis of dampness syndrome and Interleukin-2,Interleukin-8 in rats显示文摘OBJECTIVE: To explore the pathogenesis of dampness syndrome by detecting the changes of interleukin-2 (IL-2) and interleukin-8 (IL-8) levels. METHODS: Female Sprague Dawley rats were divided randomly into five groups according to the random number table: a normal group (GroupⅠ), an external dampness group (GroupⅡ), an internal dampness group (Group Ⅲ), and an external and internal dampness group (GroupⅣ). Twenty days after the model made, IL-2 and IL-8 levels were detected by radioimmunoassay method. RESULTS: The IL-2 and IL-8 levels among groups were significant (F=3.102, P<0.05; F=2.657, P<0.05, respectively). The level of IL-2 in Group Ⅱ and Group Ⅲ were higher than that in GroupⅠ(P<0.05, P<0.01, respectively), especially higher in the Group Ⅲ compared with GroupⅡ (P<0.05). The lev- el of IL-8 in Group Ⅲ were higher than those inGroupⅠ, GroupⅡ and GroupⅣ (P<0.05, P<0.01, P< 0.05, respectively). In the GroupⅢ, the 24-hour wa ter and body weight were higher than that in the GroupⅣ (all P<0.05), and spontaneous movemen frequency was higher than those in GroupⅡ and GroupⅣ (P<0.05). | 10 | Effects of Root Zone Temperature on Growth and Photosynthetic Parameters of Grafted Cucumber显示文摘Taking Cucurbita maxima and Cucurbita moschata as root stocks,and‘Jinyou No 3'cucumber as scion,the effects of different root zone temperature conditions optimal temperature( CK)( 18-20 ℃),suboptimal temperature( 13-15 ℃) and low temperature( 8-10 ℃) on the growth and photosynthesis indexes were studied. The results showed that,compared with optimal temperature( CK),suboptimal temperature and low temperature produced a significant inhibition of growth on cucumbers. The plant height,stem diameter,leaf area,number of leaves and dry weight of aboveground part were all reduced,dry weight of underground part and root shoot ratio all increased,while the inhibition was more significant at low temperature. Low and suboptimal temperature conditions also reduced SPAD value,net photosynthetic rate,transpiration rate,intercellular CO_2 concentration and stomatal conductance of the grafted cucumber.
