1Super-resolution fluorescence-assisted diffraction computational tomography reveals the threedimensional landscape of the cellular organelle interactome显示文摘The emergence of super-resolution(SR)fluorescence microscopy has rejuvenated the search for new cellular substructures.However,SR fluorescence microscopy achieves high contrast at the expense of a holistic view of the interacting partners and surrounding environment.Thus,we developed SR fluorescence-assisted diffraction computational tomography(SR-FACT),which combines label-free three-dimensional optical diffraction tomography(ODT)with two-dimensional fluorescence Hessian structured illumination microscopy.The ODT module is capable of resolving the mitochondria,lipid droplets,the nuclear membrane,chromosomes,the tubular endoplasmic reticulum,and lysosomes.Using dual-mode correlated live-cell imaging for a prolonged period of time,we observed novel subcellular structures named dark-vacuole bodies,the majority of which originate from densely populated perinuclear regions,and intensively interact with organelles such as the mitochondria and the nuclear membrane before ultimately collapsing into the plasma membrane.This work demonstrates the unique capabilities of SR-FACT,which suggests its wide applicability in cell biology in general.Dashan Dong Xiaoshuai Huang Liuju Li Heng Mao Yanquan Mo Guangyi Zhang Zhe Zhang Jiayu Shen Wei Liu Zeming Wu Guanghui Liu Yanmei Liu Hong Yang Qihuang Gong Kebin Shi Liangyi Chen 2020Light(Science & Applications)2020,9,1:7
2Direct observation of ultrafast plasmonic hot electron transfer in the strong coupling regime显示文摘Achieving strong coupling between plasmonic oscillators can significantly modulate their intrinsic optical properties.Here,we report the direct observation of ultrafast plasmonic hot electron transfer from an Au grating array to an MoS_(2) monolayer in the strong coupling regime between localized surface plasmons(LSPs)and surface plasmon polaritons(SPPs).By means of femtosecond pump-probe spectroscopy,the measured hot electron transfer time is approximately 40 fs with a maximum external quantum yield of 1.65%.Our results suggest that strong coupling between LSPs and SPPs has synergetic effects on the generation of plasmonic hot carriers,where SPPs with a unique nonradiative feature can act as an‘energy recycle bin’to reuse the radiative energy of LSPs and contribute to hot carrier generation.Coherent energy exchange between plasmonic modes in the strong coupling regime can further enhance the vertical electric field and promote the transfer of hot electrons between the Au grating and the MoS_(2) monolayer.Our proposed plasmonic strong coupling configuration overcomes the challenge associated with utilizing hot carriers and is instructive in terms of improving the performance of plasmonic opto-electronic devices.Hangyong Shan Ying Yu Xingli Wang Yang Luo Shuai Zu Bowen Du Tianyang Han Bowen Li Yu Li Jiarui Wu Feng Lin Kebin Shi Beng Kang Tay Zheng Liu Xing Zhu Zheyu Fang 2019Light(Science & Applications)2019,8,1:7
3Robust and high-speed rotation control in optical tweezers by using polarization synthesis based on heterodyne interference显示文摘The rotation control of particles in optical tweezers is often subject to the spin or orbit angular momentum induced optical torque,which is susceptible to the mechanical and morphological properties of individual particle.Here we report on a robust and high-speed rotation control in optical tweezers by using a novel linear polarization synthesis based on optical heterodyne interference between two circularly polarized lights with opposite handedness.The synthesized linear polarization can be rotated in a hopping-free scheme at arbitrary speed determined electronically by the heterodyne frequency between two laser fields.The experimental demonstration of a trapped vaterite particle in water shows that the precisely controlled rotation frequency of 300 Hz can be achieved.The proposed method will find promising applications in optically driven micro-gears,fluidic pumps and rotational micro-rheology.Wei Liu Dashan Dong Hong Yang Qihuang Gong Kebin Shi 2020Opto-Electronic Advances2020,3,8:5

5Frequency-domain diagonal extension imaging显示文摘The pixel size of a charge-coupled device(CCD)camera plays a major role in the image resolution,and the square pixels are attributed to the physical anisotropy of the sampling frequency.We synthesize the high sampling frequency directions from multiple frames acquired with different angles to enhance the resolution by 1.4×over conventional CCD orthogonal sampling.To directly demonstrate the improvement of frequency-domain diagonal extension(FDDE)microscopy,lens-free microscopy is used,as its resolution is dominantly determined by the pixel size.We demonstrate the resolution enhancement with a mouse skin histological specimen and a clinical blood smear sample.Further,FDDE is extended to lens-based photography with an ISO 12233 resolution target.This method paves a new way for enhancing the image resolution for a variety of imaging techniques in which the resolution is primarily limited by the sampling pixel size,for example,microscopy,photography,and spectroscopy.Shan Jiang Meiling Guan Jiamin Wu Guocheng Fang Xinzhu Xu Dayong Jin Zhen Liu Kebin Shi Fan Bai Shu Wang Peng Xi 2020Advanced Photonics2020,2,3:3
6An actively ultrafast tunable giant slow-light effect in ultrathin nonlinear metasurfaces显示文摘A slow-light effect based on metamaterial-induced transparency(MIT)possesses great practical applications for integrated photonic devices.However,to date,only very weak slow-light effects have been obtained in metamaterials because of the intrinsic loss of metal.Moreover,no active control of slow-light has been achieved in metamaterials.Here,we report the realization of a giant slow-light effect on an ultrathin metasurface that consists of periodic arrays of gold nanoprism dimers with a thickness of 40 nm sandwiched between a multilayer-graphene micro-sheet/zinc oxide nanoparticle layer and a monolayer graphene/polycrystalline indium tin oxide layer.The strong field confinement of the plasmonic modes associated with the MIT ensures a tremendous reduction in the group velocity around the transparency window.A group index of more than 4×10^(3) is achieved,which is one order of magnitude greater than that of previous reports.A large tunable wavelength range of 120 nm is achieved around the center of the transparency window when the pump light intensity is only 1.5 kW cm^(-2).The response time is as fast as 42.3 ps.These results demonstrate the potential for the realization of various functional integrated photonic devices based on metasurfaces,such as all-optical buffers and all-optical switches.Cuicui Lu Xiaoyong Hu Kebin Shi Qin Hu Rui Zhu Hong Yang Qihuang Gong 2015Light(Science & Applications)2015,4,1:3

9A comparative study of particle size distribution from two oxygenated fuels and diesel fuel显示文摘Oxygenated fuels are known to reduce particulate matter(PM)emissions from diesel engines.In this study,100%soy methyl ester(SME)biodiesel fuel(B100)and a blend of 10%acetal denoted by A-diesel with diesel fuel were tested as oxygenated fuels.Particle size and number distributions from a diesel engine fueled with oxygenated fuels and base diesel fuel were measured using an Electrical Low Pressure Impactor(ELPI).Measurements were made at ten steady-state operational modes of various loads at two engine speeds.It was found that the geometric mean diameters of particles from SME and Adiesel were lower than that from base diesel fuel.Compared to diesel fuel,SME emitted more ultra-fine particles at rated speed while emitting less ultra-fine particles at maximum speed.Ultra-fine particle number concentrations of A-diesel were much higher than those of base diesel fuel at most test modes.Xiaoyan SHI Kebin HE Jie ZHANG Yongliang MA Yunshan GE Jianwei TAN 2010Frontiers of Environmental Science & Engineering2010,4,1:1

12Porcine methionine sulfoxide reductase B3:molecular cloning,tissue-specific expression profiles,and polymorphisms associated with ear size in Sus scrofa显示文摘Background: In Sus scrofa, methionine sulfoxide reductase B3(MSRB3) is a crucial candidate gene for ear size, and an important conformational trait of pig breeds. However, challenges in MSRB3 c DNA amplification have prevented further identification of MSRB3 allelic variants influencing pig ear size.Results: We cloned a full-length c DNA sequence of porcine MSRB3 by rapid-amplification of c DNA ends. The3,765-bp gene contained a 5'-untranslated region(UTR)(190 bp), a coding region(552 bp), and a 3'-UTR(3,016 bp) and shared 84 %, 84 %, 87 %, 86 %, and 70 % sequence identities with human, orangutan, mouse, chicken, and zebrafish,respectively. The gene encoded a 183-amino acid protein, which shared 88 %, 91 %, 89 %, 86 %, and 67 % identities with human, orangutan, mouse, chicken, and zebrafish, respectively. Tissue expression analysis using q RT-PCR revealed that MSRB3 was expressed in the heart, liver, lung, kidney, spleen, ear, muscle, fat, lymph, skeletal, and hypothalamic tissues. Three single nucleotide polymorphisms(SNPs) were identified in MSRB3: c.-735 C > T in the 5' flanking region,c.2571 T > C in the 3'-UTR, and a synonymous mutation of c.484 T > C in the coding region. The SNPs c.-735 C > T and c.2571 T > C were significantly associated with ear size in a Large White × Minzhu F2 population other than in Beijing Black pigs. Subsequently, at SNP c.-735 C > T, the m RNA of MSRB3 was significantly higher expressed in ears of individuals with the TT genotype(Minzhu) than those with CC(Large White).Conclusions: The porcine MSRB3 owned a 3,765-bp full-length c DNA sequence and was detected to express in ear tissue. Two SNPs of this gene were shown to be significantly associated with ear size in a Large White × Minzhu intercross population instead of Beijing Black pig population. What's more, the individuals with higher m RNA expression of MSRB3 have larger ear sizes. 14Local and regional contributions to PM_(2.5) in the Beijing 2022 Winter Olympics infrastructure areas during haze episodes显示文摘The 2022 Winter Olympics is scheduled to take place in Beijing and Zhangjiakou,which were defined as OIAs(Olympic infirastructure areas)in this study.This study presents the characteristics and source apportionment of PM_(2.5) in the OIAs,China.The entire region of China's Mainland,except for the OIAs,was divided into 9 source regions,including four regions in the BTH(Beijing-Tianjin-Hebei)region,the four provinces surrounding the BTH and the remaining areas.Using CAMx/PSAT,the contributions of the nine regions to the PM_(2.5) concentration in the OIAs were simulated spatially and temporally.The simulated source apportionment results showed that the contribution of regional transportation was 48.78%,and when PM_(2.5) concentration was larger than 75μg/m^(3) central Hebei was the largest contributor with a contribution of 19.18%,followed by Tianjin,northern Hebei,Shanxi,Inner Mongolia,Shandong,southern Hebei,Henan and Liaoning.Furthermore,the contribution from neighboring regions of the OIAs was 47.12%,which was nearly twice that of long-range transportation.Haze episodes were analyzed,and the results presented the importance of regional transportation during severe PM_(2.5) pollution periods.It was also found that they were associated with differences in pollution sources between Zhangjiakou and Beijing.Regional transportation was the main factor affecting PM_(2.5) pollution in Zhangjiakou due to its low local emissions.Stagnant weather with a low planetary boundary layer height and a low wind velocity prevented the local emitted pollutants in Beijing from being transported outside,and as a result,local emissions constituted a larger contribution in Beijing.Yue Wang Mengshuang Shi Zhaofeng Lv Huan Liu Kebin He 2021Frontiers of Environmental Science & Engineering2021,15,6:1
15Genome-wide search for candidate genes determining vertebrae number in pigs显示文摘Longer porcine carcasses may be expected to have more vertebrae. Therefore, vertebrae number in pigs is an economically important trait. To examine the genetic basis of this trait, we genotyped 578 F_2 Large White ?Minzhu pigs using the Porcine SNP60K Bead Chip. A genome-wide association study(GWAS) identified 36 significant single nucleotide polymorphisms(SNPs) on the chromosomes SSC1(294.28–300.32 Mb) and SSC7(102.22–109.39 Mb). A 6.04-Mb region that contained all 13 significant SNPs on SSC1 also contained the gene NR6A1, previously reported to influence the number of vertebrae in pigs. However, the reported putative casual mutation of NR6A1 c.748 C > T showed no genome-wide significant association with the trait, suggesting it was not a causal mutation in our population. The remaining 23 significant SNPs on SSC7 were concentrated in a 7.17-Mb region, which was within a quantitative trait locus interval for number of vertebrae. TMED10 was the closest gene to the most significant SNP and might be a candidate.Haplotype sharing and block analysis refined the QTL to an interval of about 3 Mb containing 29 candidate genes.Of these 29 genes, the previously reported possible casual mutation of VRTN g.19034 A > C was not found to be a causal mutation in our population. Exploration of these genes via additional genetic and functional studies in mammals revealed that TGFβ3 could be a good candidate on SSC7. 19Detour-phased perovskite ultrathin planar lens using direct femtosecond laser writing显示文摘Perovskite-enabled optical devices have drawn intensive interest and have been considered promising candidates for integrated optoelectronic systems.As one of the important photonic functions,optical phase modulation previously was demonstrated with perovskite substrate and complex refractive index engineering with laser scribing.Here we report on the new scheme of achieving efficient phase modulation by combining detour phase design with 40 nm ultrathin perovskite films composed of nanosized crystalline particles.Phase modulation was realized by binary amplitude patterning,which significantly simplifies the fabrication process.Perovskite nanocrystal films exhibit significantly weak ion migration effects under femtosecond laser writing,resulting in smooth edges along the laser ablated area and high diffractive optical quality.Fabrication of a detour-phased perovskite ultrathin planar lens with a diameter of 150μm using femtosecond laser scribing was experimentally demonstrated.A high-performance 3D focus was observed,and the fabrication showed a high tolerance with different laser writing powers.Furthermore,the high-quality imaging capability of perovskite ultrathin planar lenses with a suppressed background was also demonstrated.WENKAI YANG LIGE LIU DASHAN DONG XIN ZHANG HAN LIN YUNKUN WANG HONG YANG YUNAN GAO HAIZHENG ZHONG BAOHUA JIA KEBIN SHI 2022Photonics Research2022,10,12:0
