| 1 | Dendritic Cells Transduced with SOCS1 Gene Exhibit Regulatory DC Properties and Prolong Allograft Survival显示文摘SOCS1 is a key regulator of cytokine signaling and is important for maintaining balance in the immune system. It is thought to participate in negative feedback loops in cytokine signaling and may be an important signal for the regulation of dendritic cell (DC) maturation. However, it remains unclear whether DCs transduced with SOCS1 exhibit characteristics of regulatory DCs and induce allogeneic T-cell hyporesponsiveness. In this study, we constructed adenoviral vector coding SOCS1 (Ad-SOCS1) that can efficiently increase SOCS1 gene expression in bone marrow-derived dendritic cells. DCs transduced with Ad-SOCS1 (DC-SOCS1) expressed low levels of costimulatory and MHC molecules, were resistant to maturation and activation stimulation, induced allogeneic T-cell hyporesponsiveness, and promoted the generation of regulatory-like T cells in vitro. DC-SOCS1 pretreatment significantly prolonged the survival of allografts and led to a substantial increase in the generation of regulatory T cells. | 4 | Silicon intensity Mach–Zehnder modulator for single lane 100 Gb/s applications显示文摘In this paper, a substrate removing technique in a silicon Mach–Zehnder modulator(MZM) is proposed and demonstrated to improve modulation bandwidth. Based on the novel and optimized traveling wave electrodes,the electrode transmission loss is reduced, and the electro-optical group index and 50 Ω impedance matching are improved, simultaneously. A 2 mm long substrate removed silicon MZM with the measured and extrapolated 3 dB electro-optical bandwidth of >50 GHz and 60 GHz at the-8 V bias voltage is designed and fabricated.Open optical eye diagrams of up to 90 GBaud∕s NRZ and 56 GBaud∕s four-level pulse amplitude modulation(PAM-4) are experimentally obtained without additional optical or digital compensations. Based on this silicon MZM, the performance in a short-reach transmission system is further investigated. | 5 | Types,Characteristics and Significances of Migrating Pathways of Gas-bearing Fluids in the Shenhu Area,Northern Continental Slope of the South China Sea显示文摘The first marine gas hydrate expedition in China has been conducted by Guangzhou Marine Geological Survey in the Shenhu Area, northern continental slope of the South China Sea. Previous study has analyzed the P-T conditions, geophysical anomalies and saturation calculations of these gas hydrates, but has not documented in detail the migration of gas-bearing fluids in the study area. Based on the interpretations of 2D/3D seismic data, this work identified two types of migration pathways for gas-bearing fluids in the Shenhu area, i.e., vertical and lateral pathways. The vertical pathways(largescale faults, gas chimneys and mud diapirs) presented as steep seismic reflection anomalies, which could be traced downward to the Eocene source rocks and may penetrate into the Late Miocene strata. The deeper gases/fluids might be allowed migrating into the shallower strata through these vertical conduits. However, the distributions showed distinct differences between these pathways. Large-scale faults developed only in the north and northeast of the Shenhu area, while in the drilling area gas chimneys were the sole vertical migration pathways. Since the Pliocene, normal faults, detachment faults and favorable sediments have constituted the lateral pathways in the Shenhu gas hydrate drilling area. Although these lateral pathways were connected with gas chimneys, they exerted different effects on hydrate formation and accumulation. Gas-bearing fluids migrated upward along gas chimneys might further migrate laterally because of the normal faults, thereby enlarging the range of the chimneys. Linking gas chimneys with the seafloor, the detachment faults might act as conduits for escaping gases/fluids. Re-deposited sediments developed at the early stage of the Quaternary were located within the gas hydrate stability zone, so hydrates would be enriched in these favorable sediments. Compared with the migration pathways(large-scale faults and mud diapirs) in the LW3-1 deep-sea oil/gas field, the migration efficiency of the vertical pathways(composed of gas chimneys) in the gas hydrate drilling area might be relatively low. Description and qualitative discrimination of migration pathways in the Shenhu gas hydrate drilling area are helpful to further understand the relationship between good-quality deep source rocks and shallow, mainly biogenicallyproduced, hydrates. As the main source rocks of the Baiyun sag, lacustrine mudstones in the Wenchang and Enping Formations may provide thermogenic methane. Gas chimneys with relatively low migration efficiency created the vertical pathways. Caused by the Dongsha tectonic movement, the release of overpressured fluids might reduce the vertical migration rates of the thermogenic methane. The thick bathyal/abyssal fine-grained sediments since the Late Miocene provided migration media with low permeability. These preconditions may cause carbon isotopic fractionation ofthermogenic methane during long-distance vertical migrations. Heat shock transcription factors(HSFs) have been proven to be the most important transcriptional factor family in the plant response to heat. In this study, a total of 19 putative HSF genes(VHSFs) were identified from the 12 × Vitis vinifera Pinot Noir PN4002 genome, and their chromosomal location, phylogenetic tree, exon-intron structure and conserved motifs were also analyzed. In addition, these HSFs transcriptional profiles in Vitis vinifera 'Jingxiu'(weak heat tolerance) and Vitis davidii 'Tangwei'(strong heat tolerance) under heat stress(47℃) and heat acclimation(38℃) were presented. Only VHSF01, VHSF05, VHSF15 and VHSF18 among 19 VHSFs showed different transcriptional models in these two genotypes under the same treatment, suggesting that these four VHSFs may be important factors which result in a thermotolerance difference between 'Jingxiu' and 'Tangwei'. | 10 | Effects of culm carbohydrate partitioning on basal stem strength in a high-yielding rice population显示文摘Rice culm carbohydrate transport can simultaneously affect grain filling and stem lodging resistance by regulating non-structural carbohydrate(NSC) and structural carbohydrate(SC) contents. However, the relationship between carbohydrate transposition and culm strength is not well documented. Accordingly, a high-yielding hybrid rice cultivar(Y Liangyou 2) was tested under different N fertilization regimes at two locations, Taoyuan(a special high-yield eco-site), Yunnan province and Danyang(a representative eco-site of the middle and lower Yangtze), Jiangsu province, China. Significantly higher grain yield and basal stem strength were found at Taoyuan than Danyang under all N rates throughout the two-year experiment. At heading stage, soluble sugars, starch, cellulose and lignin contents of the basal culm at Taoyuan were significantly 132.0%, 73.7%, 1.2%, and 62.7% higher than those at Danyang, respectively. At 20 days after heading, soluble sugars and starch content at Taoyuan decreased significantly compared to Danyang, but lignin content remained higher. Culm carbohydrate transport to kernels at Taoyuan was significantly greater than that at Danyang, and the proportion of soluble sugars and starch was correspondingly 62.9%lower. However, the proportion of lignin and cellulose was 22.7% higher at Taoyuan than that at Danyang. Soluble sugars and starch partitioning were significantly reduced under an increased nitrogen application rate, but SC partitioning was little affected. | 14 | Methane emissions from rice fields under continuous straw return in the middle-lower reaches of the Yangtze River显示文摘A three-year experiment was conducted in the middle-lower reaches of the Yangtze River in China to study the influence of continuous wheat straw return during the rice season and continuous rice straw return in wheat on methane (CH 4 ) emissions from rice fields in which, the rice-wheat rotation system is the most dominant planting pattern. The field experiment was initiated in October 2009 and has continued since the wheat-growing season of that year. The analyses for the present study were conducted in the second (2011) and third (2012) rice growing seasons. Four treatments, namely, the continuous return of wheat straw and rice straw in every season (WR), of rice straw but no wheat straw return (R), of wheat straw but no rice straw return (W) and a control with no straw return (CK), were laid out in a randomized split-plot design. The total seasonal CH 4 emissions ranged from 107.4 to 491.7 kg/ha (2011) and 160.3 to 909.6 kg/ha (2012). The increase in CH 4 emissions for treatments WR and W were 289% and 230% in the second year and 185% and 225% in the third year, respectively, in relation to CK. We observed less methane emissions in the treatment R than in CK by 14%-43%, but not statistically significant. Treatment R could increase rice productivity while no more CH 4 emission occurs. A nonlinear mechanism model of the vehicle height adjustment system was established based on vehicle system dynamics and thermodynamic theory for variable-mass gas charge/discharge system. In order to model both the continuous/discrete dynamics of vehicle height adjustment process and the on-off statuses switching of solenoid valves, the framework of mixed logical dynamical(MLD) modelling was used. On the basis of the vehicle height adjustment control strategy, the MLD model of the adjustment process was built by introducing auxiliary logical variables and auxiliary continuous variables. | 17 | Fate of Basal N Under Split Fertilization in Rice with ^(15)N Isotope Tracer显示文摘Split fertilization strategy is popularly adopted in rice to synchronize soil nitrogen(N) supply and crop N demand. Attention has been paid more on mid-season topdressing N, but limited on basal N. A clearer understanding of the basal N fate under split fertilization is crucial for determining rational basal N split ratio to improve the yield and reduce the loss to environment. A two-year field experiment with two N rates of 150 and 300 kg Nha^(-1), two split ratios of basal N, 40% and 25%, and two rice varieties,Wuyunjing 23(japonica) and Y-liangyou 2(super hybrid indica), was conducted. Labelled ^(15) N urea was supplied in micro-plots as basal fertilizer to determine the plant uptake, translocation, soil residual, and loss of basal N fertilizer. The results showed that basal N absorbed by rice was only 1.6%–11.5% before tillering fertilization(8–10 d after transplanting), 6.5%–21.4% from tillering fertilization to panicle fertilization, and little(0.1%–4.4%) after panicle fertilization. The recovery efficiency of basal N for the entire rice growth stage was low and ranged from 18.7% to 24.8%, not significantly affected by cultivars or N treatments. Soil residual basal N accounted for 10.3%–36.4% and decreased with increasing total N rate and basal N ratio, regardless of variety and year. 43.8%–70.4% of basal N was lost into the environment based on the N balance. Basal N loss was significantly linearly positive related with the basal N rate and obviously enhanced by the increasing basal N ratio for both varieties in both 2012 and 2013. The N use efficiency and yield was significantly improved when decreasing the basal N ratio from 40% to 25%. | 19 | Periodontitis may induce gut microbiota dysbiosis via salivary microbiota显示文摘The aim of this study was to identify whether periodontitis induces gut microbiota dysbiosis via invasion by salivary microbes.First,faecal and salivary samples were collected from periodontally healthy participants(PH group,n=16)and patients with severe periodontitis(SP group,n=21)and analysed by 16S ribosomal RNA sequencing.Significant differences were observed in both the faecal and salivary microbiota between the PH and SP groups.Notably,more saliva-sourced microbes were observed in the faecal samples of the SP group.Then,the remaining salivary microbes were transplanted into C57BL6/J mice(the C-PH group and the C-SP group),and it was found that the composition of the gut microbiota of the C-SP group was significantly different from that of the C-PH group,with Porphyromonadaceae and Fusobacterium being significantly enriched in the C-SP group.In the colon,the C-SP group showed significantly reduced crypt depth and zonula occludens-1 expression.The m RNA expression levels of pro-inflammatory cytokines,chemokines and tight junction proteins were significantly higher in the C-SP group.To further investigate whether salivary bacteria could persist in the intestine,the salivary microbiota was stained with carboxyfluorescein diacetate succinimidyl ester and transplanted into mice.We found that salivary microbes from both the PH group and the SP group could persist in the gut for at least 24 h.Thus,our data demonstrate that periodontitis may induce gut microbiota dysbiosis through the influx of salivary microbes. | Jun Bao Lili Li Yangheng Zhang Min Wang Faming Chen Shaohua Ge Bin Chen Fuhua Yan | 2022 | International Journal of Oral Science2022,14,3: | 9 |
| 20 | Dynamic tensile behaviour and crack propagation of coal under coupled static-dynamic loading显示文摘The fracture behaviour and crack propagation features of coal under coupled static-dynamic loading conditions are important when evaluating the dynamic failure of coal.In this study,coupled static-dynamic loading tests are conducted on Brazilian disc(BD)coal specimens using a modified split Hopkinson pressure bar(SHPB).The effects of the static axial pre-stress and loading rate on the dynamic tensile strength and crack propagation characteristics of BD coal specimens are studied.The average dynamic indirect tensile strength of coal specimens increases first and then decreases with the static axial pre-stress increasing.When no static axial pre-stress is applied,or the static axial pre-stress is 30%of the static tensile strength,the dynamic indirect tensile strength of coal specimens shows an increase trend as the loading rate increases.When the static axial pre-stress is 60%of the static tensile strength,the dynamic indirect tensile strength shows a fluctuant trend as the loading rate increases.According to the crack propagation process of coal specimens recorded by high-speed camera,the impact velocity influences the mode of crack propagation,while the static axial pre-stress influences the direction of crack propagation.The failure of coal specimens is a coupled tensile-shear failure under high impact velocity.When there is no static axial pre-stress,tensile cracks occur in the vertical loading direction.When the static axial pre-stress is applied,the number of cracks perpendicular to the loading direction decreases,and more cracks occur in the parallel loading direction. | Xianjie Hao Weisheng Du Yixin Zhao Zhuowen Sun Qian Zhang Shaohua Wang Haiqing Qiao | 2020 | International Journal of Mining Science and Technology2020,30,5: | 8 |
