2Dendritic Cells Transduced with SOCS1 Gene Exhibit Regulatory DC Properties and Prolong Allograft Survival显示文摘SOCS1 is a key regulator of cytokine signaling and is important for maintaining balance in the immune system. It is thought to participate in negative feedback loops in cytokine signaling and may be an important signal for the regulation of dendritic cell (DC) maturation. However, it remains unclear whether DCs transduced with SOCS1 exhibit characteristics of regulatory DCs and induce allogeneic T-cell hyporesponsiveness. In this study, we constructed adenoviral vector coding SOCS1 (Ad-SOCS1) that can efficiently increase SOCS1 gene expression in bone marrow-derived dendritic cells. DCs transduced with Ad-SOCS1 (DC-SOCS1) expressed low levels of costimulatory and MHC molecules, were resistant to maturation and activation stimulation, induced allogeneic T-cell hyporesponsiveness, and promoted the generation of regulatory-like T cells in vitro. DC-SOCS1 pretreatment significantly prolonged the survival of allografts and led to a substantial increase in the generation of regulatory T cells. 3Hyperthermic intraperitoneal chemotherapy for gastric cancer with peritoneal metastasis:A multicenter propensity scorematched cohort study显示文摘Objective:Systemic chemotherapy has limited efficacy in the treatment of peritoneal metastasis(PM)in gastric cancer(GC).Hyperthermic intraperitoneal chemotherapy(HIPEC)combined with complete cytoreductive surgery(CRS)has shown promising outcomes but remains controversial.The present study aimed to evaluate the safety and efficacy of HIPEC without CRS in GC patients with PM.Methods:This retrospective propensity score-matched multicenter cohort study included GC patients with PM treated with either chemotherapy alone(Cx group)or with HIPEC combined with chemotherapy(HIPEC-Cx group)in four Chinese high-volume gastric medical centers between 2010 and 2017.The primary outcomes were median survival time(MST)and 3-year overall survival(OS).Propensity score matching was performed to compensate for controlling potential confounding effects and selection bias.Results:Of 663 eligible patients,498 were matched.The MST in the Cx and HIPEC-Cx groups was 10.8 and 15.9 months,respectively[hazard ratio(HR)=0.71,95%confidence interval(95%CI),0.58-0.88;P=0.002].The 3-year OS rate was 10.1%(95%CI,5.4%-14.8%)and 18.4%(95%CI,12.3%-24.5%)in the Cx and HIPEC-Cx groups,respectively(P=0.017).The complication rates were comparable.The time to first flatus and length of hospital stay for patients undergoing HIPEC combined with chemotherapy was longer than that of chemotherapy alone(4.6±2.4 d vs.2.7±1.8 d,P<0.001;14.2±5.8 d vs.11.4±7.7 d,P<0.001),respectively.The median follow-up period was 33.2 months.Conclusions:Compared with standard systemic chemotherapy,HIPEC combined with chemotherapy revealed a statistically significant survival benefit for GC patients with PM,without compromising patient safety.Ziying Lei Jiahong Wang Zhi Li Baozhong Li Jiali Luo Xuejun Wang Jin Wang MingchenBa Hongsheng Tang Qingjun He Quanxing Liao Xiansheng Yang Tianpei Guan Han Liang Shuzhong Cui 2020Chinese Journal of Cancer Research2020,32,6:11
4Effect of Mg/Al atom ratio of support on catalytic performance of Co-Mo/MgO-Al_2O_3 catalyst for water gas shift reaction显示文摘Co-Mo-based catalysts supported on mixed oxide supports MgO-Al2O3 with different Mg/Al atom ratios for water gas shift reaction were studied by means of TPR, Raman, XPS and ESR. It was found that the octahedral Mo species in oxidized Co-Mo/MgO(x)-Al2O3 catalyst and the contents of Mo5+, Mo4+, S2- and S22- species in the functioning catalysts increased with increasing the Mg/Al atom ratio of the support under the studied experimental conditions. This is favorable for the formation of the active Co-Mo-S phase of the catalysts. 6A multi-institutional retrospective study of hyperthermic plus intravesical chemotherapy versus intravesical chemotherapy treatment alone in intermediate and high risk nonmuscle-invasive bladder cancer显示文摘Objective:To compare the efficacy and safety of hyperthermic intravesical chemotherapy(HIVEC)and intravesical chemotherapy(IVEC)in patients with intermediate and high risk nonmuscle-invasive bladder cancer(NMIBC)after transurethral resection.Methods:We included 560 patients diagnosed with primary or recurrent NMIBC between April 2009 and December 2015 at 1 of 6 tertiary centers.We matched 364 intermediate or high risk cases and divided them into 2 groups:the HIVEC+IVEC group[chemohyperthermia(CHT)composed of 3 consecutive sessions followed by intravesical instillation without hyperthermia]and the IVEC group(intravesical instillation without hyperthermia).The data were recorded in the database.The primary endpoint was 2-year recurrence-free survival(RFS)in all NMIBC patients(n=364),whereas the secondary endpoints were the assessment of radical cystectomy(RC)and 5-year overall survival(OS).Results:There was a significant difference in the 2-year RFS between the two groups in all patients(n=364;HIVEC+IVEC:82.42%vs.IVEC:74.18%,P=0.038).Compared with the IVEC group,the HIVEC+IVEC group had a lower incidence of RC(P=0.0274).However,the 5-year OS was the same between the 2 groups(P=0.1434).Adverse events(AEs)occurred in 32.7%of all patients,but none of the events was serious(grades 3–4).No difference in the incidence or severity of AEs between each treatment modality was observed.Conclusions:This retrospective study showed that HIVEC+IVEC had a higher 2-year RFS and a lower incidence of RC than IVEC therapy in intermediate and high risk NMIBC patients.Both treatments were well-tolerated in a similar manner.Qiang Ruan Degang Ding Bin Wang Chaohong He Xuequn Ren Zhenhua Feng Zhigang Pang Jin Wang Xiangliang Zhang Hongsheng Tang Jiahong Wang Qingjun He Ziying Lei Quanxing Liao Jiali Luo Shuzhong Cui 2021Cancer Biology & Medicine2021,18,1:4

Comparing with Amberlite XAD-4 and NDA-150, the equilibrium adsorption for phenol on the JN-3 from aqueous solutions was tested, perfect adsorption capacity was shown. Pb2+ can be also removed by JN-3 because of the chelate interaction between sulfhydryl groups and metal ions. 16Prehistoric damaging earthquake promoted the decline of'Heluo Ancient State'in Early China显示文摘The emergence of the'Ancient State'is considered an important stage in the discourse on human civilizations.Based on its complex and hierarchical social structure,the Shuanghuaishu site of Henan Province is regarded as an'Ancient state',specifically the'Heluo Ancient State'.The site is situated in the middle reaches of the Yellow River region and played a key role in prehistoric China.However,the reasons behind its decline are unclear.Here,we conducted an integrated investigation of the archaeological excavations and archaeoseismic analyses at the Shuanghuaishu site.We identified paleoseismic remains within paleo-artificial moats,which were formed owing to off-fault structures of soil liquefaction,hillslope landslides,and related ground fractures,along with shake-related wall disruption and building collapse in the residential zone.Charcoal dating was conducted to constrain the age for this earthquake event,which was determined to be approximately 5.0 cal kyr BP.Furthermore,based on the practical curve for earthquake-induced liquefaction,the preliminary estimate of the magnitude of this paleo seismic event is greater than M_(s)5.9-6.0.We propose that the Shuanghuaishu site experienced a recession after the earthquake event,which may have influenced the continuous development of the'Heluo Ancient State'as a regional center settlement.Xiu HU Peng LU Youli LI Duowen MO Wanfa GU Qingri LIU Ailun GUO Panpan CHEN Hui WANG Yan TIAN Xu WANG Yayi HU Jiqin ZHANG Zhen WANG Quanxing LUO Junjie XU 2023Science China Earth Sciences2023,66,5:0
17Glutamine deprivation induces ferroptosis in pancreatic cancer cells显示文摘Ferroptosis is a type of programmed cell death closely related to amino acid metabolism.Pancreatic cancer cells have a strong dependence on glutamine,which serves as a carbon and nitrogen substrate to sustain rapid growth.Glutamine also aids in self-protection mechanisms.However,the effect of glutamine on ferroptosis in pancreatic cancer remains largely unknown.Here,we aim to explore the association between ferroptosis and glutamine deprivation in pancreatic cancer.The growth of pancreatic cancer cells in culture media with or without glutamine is evaluated using Cell Counting Kit-8.Reactive oxygen species(ROS)are measured by 2′,7′-dichlorodihydrofluorescein diacetate staining.Ferroptosis is assessed by BODIPY-C11 dye using confocal microscopy and flow cytometry.Amino acid concentrations are measured using ultrahigh-performance liquid chromatography-tandem mass spectrometry.Isotope-labelled metabolic flux analysis is performed to track the metabolic flow of glutamine.Additionally,RNA sequencing is performed to analyse the genetic alterations.Glutamine deprivation inhibits pancreatic cancer growth and induces ferroptosis both in vitro and in vivo.Additionally,glutamine decreases ROS formation via glutathione production in pancreatic cancer cells.Interestingly,glutamine inhibitors(diazooxonorleucine and azaserine)promotes ROS formation and ferroptosis in pancreatic cancer cells.Furthermore,ferrostatin,a ferroptosis inhibitor,rescues ferroptosis in pancreatic cancer cells.Glutamine deprivation leads to changes in molecular pathways,including cytokine-cytokine receptor interaction pathways(CCL5,CCR4,LTA,CXCR4,IL-6R,and IL-7R).Thus,exogenous glutamine is required for the detoxification of ROS in pancreatic cancer cells,thereby preventing ferroptosis.Zhiwen Xiao Shengming Deng He Liu Ruijie Wang Yu Liu Zhengjie Dai Wenchao Gu Quanxing Ni Xianjun Yu Chen Liu Guopei Luo 2023Acta Biochimica et Biophysica Sinica2023,55,8:0
18ICOS-Ig combined with CsA induces long term survival of cardiac allografts in mouse显示文摘Objective: To study the synergistic effect of ICOS-Ig combined with cyclosporine (CsA) on mouse heart transplantation and explore its therapeutic potential. Methods: ICOS-Ig fusion protein was generated by fusing the extracellular portion of human ICOS and Fc portion of human IgG. To investigate the effect of ICOS-Ig on T-cell proliferation in vitro, ICOS-Ig or IgG was added to the primary MLR cultures (BALB/c spleen T cells as responder cells and irradiated C57BL/6 spleen cells as stimulator cells). The cells responsiveness rates were detected by 3H-TdR methods. Then the T cells of each group in primary MLR were cultured as responder cells for secondary MLR, and irradiated C57BL/6 (donor) or C3H (third party) spleen cells as stimulator cells. To study the effect of ICOS-Ig on T-cell proliferation in vivo, CFSE-labeled C57BL/6 spleen cells were transferred to irradiated BALB/c mice. Mice were then treated with IgG, ICOS-Ig or CsA. Seventy two hours after transfer, the spleen cells of the mice were harvested for the detection of CD4+CFSE+ and CD8+CFSE+ by FACS. C57BL/6 mouse underwent transplantation of the hearts of BALB/c mouse and were then randomly divided into five equal groups: no treatment group, control IgG treated group (250 μg i.p. d2, 4, 6), ICOS-Ig treated group (250 μg i.p. d2, 4, 6), CsA treated group (10 mg/kg i.p. d0-6), ICOS-Ig combined with CsA group. The cardiac allograft survival was monitored by daily palpation. Results: In primary MLR, ICOS-Ig inhibited T-cell proliferation, (inhibition ratio 58±8.2% in 50 μg/ml). In secondary MLR, ICOS-Ig specifically inhibited donor spleen cells, which suggested ICOS-Ig could induce donor-specific hyporesponsiveness. In the CFSE dye assay, CD4+CFSE+ and CD8+CFSE+ in ICOS-Ig and CsA group was stronger than those in control group, which showed ICOS-Ig and CsA could inhibit the proliferation of allo-reactive T cells in vivo. In mouse heart transplantation model, survival was significantly prolonged in animals treated with ICOS-Ig or CsA as compared with controls. Moreover, ICOS-Ig combined with CsA group had even longer engraftment (>100 d) than ICOS-Ig or CsA used alone. In histological examination, it was found that there were congestions and edemas in no treatment and IgG treated recipients, together with a lot of inflammatory cells infiltrated. Allogeneic hearts from ICOS-Ig and/or CsA immunized recipients revealed milder histological changes. It was revealed in mechanical analysis that splenic T cells from recipients also exhibited depressed mixed leukocyte reactions (MLR) and cytotoxic lymphocyte reactions (CTL). Conclusion: These data suggest that ICOS-Ig combined with CsA induces a long-term survival of mouse cardiac allografts, whereas monotherapy is less effective in this regard.
