1Update in version 2021 of CSCO guidelines for colorectal cancer from version 2020显示文摘Colorectal cancer(CRC) is one of the major cancers threatening life and health of Chinese residents. According to the latest data released by the National Cancer Center in2015(1), there were 387,000 new cases and 187,100 deaths of CRC in China, accounting for 9.87% and 8.01% of all malignancies. The first version of the Chinese Society of Clinical Oncology(CSCO) guidelines was released in April2017, and has been updated annually based on the latest clinical research data and the availability of new drugs(2-4). 2Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. 3Sleep Deprivation Selectively Down-Regulates Astrocytic 5-HT2B Receptors and Triggers Depressive-Like Behaviors via Stimulating P2X7 Receptors in Mice显示文摘Chronic loss of sleep damages health and disturbs the quality of life.Long-lasting sleep deprivation(SD)as well as sleep abnormalities are substantial risk factors for major depressive disorder,although the underlying mechanisms are not clear.Here,we showed that chronic SD in mice promotes a gradual elevation of extracellular ATP,which activates astroglial P2X7 receptors(P2X7Rs).Activated P2X7Rs,in turn,selectively down-regulated the expression of 5-HT2B receptors(5-HT2BRs)in astrocytes.Stimulation of P2X7Rs induced by SD selectively suppressed the phosphorylation of AKT and FoxO3 a in astrocytes,but not in neurons.The overexpression of FoxO3a in astrocytes inhibited the expression of 5-HT2BRs.Down-regulation of 5-HT2BsRs instigated by SD suppressed the activation of STAT3 and relieved the inhibition of Ca2+-dependent phospholipase A2.This latter cascade promoted the release of arachidonic acid and prostaglandin E2.The depression-like behaviors induced by SD were alleviated in P2X7R-KO mice.Our study reveals the mechanism underlying chronic SD-induced depression-like behaviors and suggests 5-HT2BRs as a key target for exploring therapeutic strategies aimed at the depression evoked by sleep disorders.Maosheng Xia Zexiong Li Shuai Li Shanshan Liang Xiaowei Li Beina Chen Manman Zhang Chengyi Dong Alexei Verkhratsky Dawei Guan Baoman Li 2020Neuroscience Bulletin2020,36,11:10

5Incoherent imaging through highly nonstatic and optically thick turbid media based on neural network显示文摘Imaging through nonstatic scattering media is one of the major challenges in optics, and encountered in imaging through dense fog, turbid water, and many other situations. Here, we propose a method to achieve single-shot incoherent imaging through highly nonstatic and optically thick turbid media by using an end-to-end deep neural network. In this study, we use fat emulsion suspensions in a glass tank as a turbid medium and an additional incoherent light to introduce strong interference noise. We calibrate that the optical thickness of the tank of turbid media is as high as 16, and the signal-to-interference ratio is as low as -17 dB. 6Joint Occurrence Period of Wind Speed and Wave Height Based on Both Service Term and Risk Probability显示文摘Return periods calculated for different environmental conditions are key parameters for ocean platform design.Many codes for offshore structure design give no consideration about the correlativity among multi-loads and over-estimate design values.This frequently leads to not only higher investment but also distortion of structural reliability analysis.The definition of design return period in existing codes and industry criteria in China are summarized.Then joint return periods of different ocean environmental parameters are determined from the view of service term and danger risk.Based on a bivariate equivalent maximum entropy distribution,joint design parameters are estimated for the concomitant wave height and wind speed at a site in the Bohai Sea.The calculated results show that even if the return period of each environmental factor,such as wave height or wind speed,is small,their combinations can lead to larger joint return periods.Proper design criteria for joint return period associated with concomitant environmental conditions will reduce structural size and lead to lower investment of ocean platforms for the exploitation of marginal oil field.DONG Sheng FAN Dunqiu TAO Shanshan 2012Journal of Ocean University of China2012,11,4:5
7Clinical features, survival and prognostic factors of primary testicular diffuse large B-cell lymphoma显示文摘Objective: To assess the clinical features, survival and prognostic factors of primary testicular diffuse large B-cell lymphoma(DLBCL).Methods: A retrospective study of 37 patients with primary testicular DLBCL was carried out from November 2003 to May 2012. Their clinical features, survival and prognostic factors were analyzed.Results: During a median follow-up period of 39.8 months(5.4-93.0 months), the median progression-free survival(PFS) was 26.2 months(95% CI: 0-65 months) and the 3-year overall survival(OS) rate was 78.4%. Within the whole cohort, the factors significantly associated with a superior PFS were limited stage(stage I/II), lactate dehydrogenase(LDH) ≤245 U/L, international prognostic index(IPI) ≤1, primary tumor diameter <7.5 cm, and patients who had complete response(CR) and received doxorubicin-contained chemotherapy(P<0.05). There was a trend toward superior outcome for patients who received combined therapy(surgery/chemotherapy/radiotherapy)(P=0.055). Patients who had CR, primary tumor diameter <7.5 cm and IPI score ≤1 were significantly associated with longer PFS at multivariate analysis.Conclusions: Primary testicular DLBCL had poorer survival. CR, primary tumor diameter and IPI were independent prognostic factors. 8Effect of Jianpiyiqi granule on ocular myasthenia gravis in children prepared with a formula from Traditional Chinese Medicine显示文摘OBJECTIVE: To investigate the effect of Jianpiyiqi granule, prepared according to a Traditional Chinese Medicine(TCM) formula, on the ocular type of myasthenia gravis(MG) caused by a defect in synaptic transmission at the neuromuscular junction.METHODS: A total of 155 children with ocular MG were recruited from January 2008 to January 2015.All individuals received ineffective glucocorticoid treatment prior to admission, and were given Jianpiyiqi granules(two doses per day) for 12 months.Plasma levels of acetylcholine receptor autoantibodies(ACh R-Ab), cytokines, immune parameters and clinical score were analyzed.RESULTS: After intervention with Jianpiyiqi granule for 12 months, the levels of cytokines [including interleukin(IL)-6, IL-12, IL-17], immune parameters[including immunoglobulin G(Ig G), Ig M, free triiodothyronine, free thyroxine] and ACh R-Ab were sig-nificantly decreased(P = 0.042, P = 0.049, P = 0.011,P = 0.02, P = 0.04, P = 0.03, P = 0.01; F = 21.60, P =0.000). In contrast, IL-4 levels were significantly increased(P = 0.049). The absolute clinical score after treatment declined significantly compared with before treatment(F = 33.24, P = 0.000). The effective cure rate and the total effective rate gradually increased to a maximum of 149(96.2%) and 151(97.4%) in MG patients after 12 months of treatment, respectively.CONCLUSION: The Jianpiyiqi granule treatment lowered ACh R-Ab levels and improved cytokine and other immune parameter levels, which suggests that the granule could be an ancillary treatment for ocular MG in children caused by a defect in synaptic transmission at the neuromuscular junction.Qi Guoyan Liu Peng Gu Shanshan Yang Hongxia Dong Huimin Xue Yinping 2017Journal of Traditional Chinese Medicine2017,37,4:4
9The genome of Magnolia biondii Pamp. provides insights into the evolution of Magnoliales and biosynthesis of terpenoids显示文摘Magnolia biondii Pamp.(Magnoliaceae,magnoliids)is a phylogenetically,economically,and medicinally important ornamental tree species widely grown and cultivated in the north-temperate regions of China.Determining the genome sequence of M.biondii would help resolve the phylogenetic uncertainty of magnoliids and improve the understanding of individual trait evolution within the Magnolia genus.We assembled a chromosome-level reference genome of M.biondii using~67,~175,and~154Gb of raw DNA sequences generated via Pacific Biosciences single-molecule real-time sequencing,10X Genomics Chromium,and Hi-C scaffolding strategies,respectively.The final genome assembly was~2.22Gb,with a contig N50 value of 269.11 kb and a BUSCO complete gene percentage of 91.90%.Approximately 89.17%of the genome was organized into 19 chromosomes,resulting in a scaffold N50 of 92.86Mb.The genome contained 47,547 protein-coding genes,accounting for 23.47%of the genome length,whereas 66.48%of the genome length consisted of repetitive elements.We confirmed a WGD event that occurred very close to the time of the split between the Magnoliales and Laurales.Functional enrichment of the Magnolia-specific and expanded gene families highlighted genes involved in the biosynthesis of secondary metabolites,plant–pathogen interactions,and responses to stimuli,which may improve the ecological fitness and biological adaptability of the lineage.Phylogenomic analyses revealed a sister relationship of magnoliids and Chloranthaceae,which are sister to a clade comprising monocots and eudicots.The genome sequence of M.biondii could lead to trait improvement,germplasm conservation,and evolutionary studies on the rapid radiation of early angiosperms.Shanshan Dong Min Liu Yang Liu Fei Chen Ting Yang Lu Chen Xingtan Zhang Xing Guo Dongming Fang Linzhou Li Tian Deng Zhangxiu Yao Xiaoan Lang Yiqing Gong Ernest Wu Yaling Wang Yamei Shen Xun Gong Huan Liu Shouzhou Zhang 2021Horticulture Research2021,8,1:4
10Metagenome analysis reveals potential microbial functions in topsoil of wheat-maize rotation system with five-year application of fertilizers显示文摘Fertilization mode affects soil quality and ecological health.The effects of four fertilization regimens on lignocellulose content,readily degradable carbohydrate decomposition,and potential microbial functions in the topsoil of a wheat-maize rotation system between 2012 and 2017 were investigated.The fertilization regimens of control(control NFNB),high chemical fertilizer(HCF),high biochar plus low chemical fertilizer(HBLCF),and biochar-based fertilizer(BBF)were compared on soil fundamental properties,microbial structure,and potential function in soil carbohydrate degradation based on metagenome analysis.The diversity of carbohydrate-active enzyme genes in the topsoil microbial consortia in the four trials was primarily distributed within the ten ecologically most dominant phyla.Application of BBF was associated with the lowest decline in total nitrogen and P2O5(2012-2017:6.5%and 28.1%,respectively)and the most effective carbohydrate decomposition(2015-2017:67.0%for cellulose and 59.9%for readily degradable carbohydrate).Carbohydrate transport and metabolism accounted for 6.0%of reads assigned functional classification under the BBF regimen.These findings reveal the ecologically functional diversity of topsoil microorganisms and suggest BBF application as a promising strategy for sustainable agriculture and beneficial to soil health.Yujun Shen Lixin Zhao Haibo Meng Hongsheng Cheng Jingtao Ding Jiarui Wang Shanshan Dong Xi Zhang Liqiu Song Shengwei Zheng 2019International Journal of Agricultural and Biological Engineering2019,12,6:3

12Addition of rituximab is not associated with survival benefit compared with CHOP alone for patients with stage I diffuse large B-cell lymphoma显示文摘Background:The role of rituximab in combination with CHOP regimen in patients with stage I diffuse large B-cell lymphoma(DLBCL) remains to be defined.We aimed to compare CHOP plus rituximab(R-CHOP) with CHOP alone and determine the value of radiotherapy in these patients.Methods:Between 2003 and 2009,140 untreated patients with stage I DLBCL were retrospectively analyzed in this study.Results:Seventy-eight patients were treated in R-CHOP group and 62 in CHOP group.Ninety-one patients received additional radiotherapy at the end of chemotherapy.The different treatment groups were well-balanced with respect to baseline characteristics.Complete response(CR) rate was 77% both in R-CHOP and CHOP groups(P=0.945).After a median follow-up period of 56 months,patients received R-CHOP regimen had similar 5-year progression-free survival(PFS)(76%vs.85%;log-rank P=0.215)and 5-year overall survival(OS)(90%vs.96%;log-rank P=0.175) compared with those with CHOP alone.Patients with radiotherapy had significantly increased 5-year PFS compared with those who had chemotherapy alone(86%vs.71%;log-rank P=0.005).At multivariate analysis,patients who had CR(P=0.008) and received radiotherapy(P=0.003) were significantly associated with superior PFS.Conclusions:CHOP alone could be as effective as R-CHOP regimen and additional radiotherapy would be necessary for stage I or stage I non-bulky DLBCL patients.Bo Jia Yuankai Shi Suyi Kang Sheng Yang Shaoxuan Hu Yexiong Li Mei Dong Weihu Wang Jianliang Yang Liqiang Zhou Peng Liu Shengyu Zhou Yan Qin Lin Gui Changgong Zhang Hua Lin Shanshan Chen Lin Wang Xiaohui He 2015Chinese Journal of Cancer Research2015,27,5:2

15Multifaceted functions of STING in human health and disease:from molecular mechanism to targeted strategy显示文摘Since the discovery of Stimulator of Interferon Genes(STING)as an important pivot for cytosolic DNA sensation and interferon(IFN)induction,intensive efforts have been endeavored to clarify the molecular mechanism of its activation,its physiological function as a ubiquitously expressed protein,and to explore its potential as a therapeutic target in a wide range of immune-related diseases.With its orthodox ligand 2’3’-cyclic GMP–AMP(2’3’-cGAMP)and the upstream sensor 2’3’-cGAMP synthase(cGAS)to be found,STING acquires its central functionality in the best-studied signaling cascade,namely the cGAS–STING–IFN pathway.However,recently updated research through structural research,genetic screening,and biochemical assay greatly extends the current knowledge of STING biology.A second ligand pocket was recently discovered in the transmembrane domain for a synthetic agonist.On its downstream outputs,accumulating studies sketch primordial and multifaceted roles of STING beyond its cytokine-inducing function,such as autophagy,cell death,metabolic modulation,endoplasmic reticulum(ER)stress,and RNA virus restriction.Furthermore,with the expansion of the STING interactome,the details of STING trafficking also get clearer.After retrospecting the brief history of viral interference and the milestone events since the discovery of STING,we present a vivid panorama of STING biology taking into account the details of the biochemical assay and structural information,especially its versatile outputs and functions beyond IFN induction.We also summarize the roles of STING in the pathogenesis of various diseases and highlight the development of small-molecular compounds targeting STING for disease treatment in combination with the latest research.Finally,we discuss the open questions imperative to answer.Zili Zhang Haifeng Zhou Xiaohu Ouyang Yalan Dong Alexey Sarapultsev Shanshan Luo Desheng Hu 2023Signal Transduction and Targeted Therapy2023,8,1:2

18Long-Term Statistics of Extreme Tsunami Height at Crescent City显示文摘Historically, Crescent City is one of the most vulnerable communities impacted by tsunamis along the west coast of the United States, largely attributed to its offshore geography. Trans-ocean tsunamis usually produce large wave runup at Crescent Harbor resulting in catastrophic damages, property loss and human death. How to determine the return values of tsunami height using relatively short-term observation data is of great significance to assess the tsunami hazards and improve engineering design along the coast of Crescent City. In the present study, the extreme tsunami heights observed along the coast of Crescent City from 1938 to 2015 are fitted using six different probabilistic distributions, namely, the Gumbel distribution, the Weibull distribution, the maximum entropy distribution, the lognormal distribution, the generalized extreme value distribution and the generalized Pareto distribution. The maximum likelihood method is applied to estimate the parameters of all above distributions. Both Kolmogorov-Smirnov test and root mean square error method are utilized for goodness-of-fit test and the better fitting distribution is selected. Assuming that the occurrence frequency of tsunami in each year follows the Poisson distribution, the Poisson compound extreme value distribution can be used to fit the annual maximum tsunami amplitude, and then the point and interval estimations of return tsunami heights are calculated for structural design. 19Secure Data Aggregation Approach Based on Monitoring in Wireless Sensor Networks显示文摘In order to avoid internal attacks during data aggregation in wireless sensor networks, a grid-based network architecture fit for monitoring is designed and the algorithms for network division, initialization and grid tree construction are presented. The characteristics of on-off attacks are first studied and monitoring mechanisms are then designed for sensor nodes. A Fast Detection and Slow Recovery (FDSR) algorithm is proposed to prevent on-off attacks by observing the behaviors of the nodes and computing reputations. A recovery mechanism is designed to isolate malicious nodes by identifying the new roles of nodes and updating the grid tree. In the experiments, some situations of on-off attacks are simulated and the results are compared with other approaches. 20Mining and fine-tuning sugar uptake system for titer improvement of milbemycins in Streptomyces bingchenggensis显示文摘Dramatic decrease of sugar uptake is a general phenomenon in Streptomyces at stationary phase,when antibiotics are extensively produced.Milbemycins produced by Streptomyces bingchenggensis are a group of valuable macrolide biopesticides,while the low yield and titer impede their broad applications in agricultural field.Considering that inadequate sugar uptake generally hinders titer improvement of desired products,we mined the underlying sugar uptake systems and fine-tuned their expression in this work.First,we screened the candidates at both genomic and transcriptomic level in S.bingchenggensis.Then,two ATP-binding cassette transporters named TP2 and TP5 were characterized to improve milbemycin titer and yield significantly.Next,the appropriate native temporal promoters were selected and used to tune the expression of TP2 and TP5,resulting in a maximal milbemycin A3/A4 titer increase by 36.9%to 3321 mg/L.Finally,TP2 and TP5 were broadly finetuned in another two macrolide biopesticide producers Streptomyces avermitilis and Streptomyces cyaneogriseus,leading to a maximal titer improvement of 34.1%and 52.6%for avermectin B1a and nemadectin,respectively.This work provides useful transporter tools and corresponding engineering strategy for Streptomyces.Pinjiao Jin Shanshan Li Yanyan Zhang Liyang Chu Hairong He Zhuoxu Dong Wensheng Xiang 2020Synthetic and Systems Biotechnology2020,5,3:1
