| 1 | Diet high in fructose leads to an overexpression of lipocalin-2 in rat fatty liver显示文摘AIM:To explore lipocalin-2(LCN-2)expression and its possible role and mechanism(s)of production in rat models of diet-inducible fatty liver.METHODS:Fatty liver was triggered in male SpragueDawley rats fed either with liquid Lieber-DeCarli(LDC)or LDC+70%cal fructose(L-HFr)diet for 4 or 8 wk.Chow-nourished animals served as controls.Hepatic expression of LCN-2 and other metabolic and inflammatory mediators was assessed by quantitative reverse transcription polymerase chain reaction and Western blotting.Serum LCN-2,fasting leptin,and lipid profile were evaluated via Enzyme-Linked Immunosorbent Assay,Radioimmunoassay,and colorimetric assays,respectively.The localization of LCN-2 in the liver was detected by using immunofluorescence staining.Furthermore,HE stain was used to evaluate hepatic fatdegeneration and inflammation.RESULTS:Both LDC-fed and L-HFr-fed rat histologically featured fatty liver.In the liver,mRNA transcriptions of Mcp-1,a2-m,Il-8 and Glut5 were increased in the L-HFr group at both time points(P<0.001),while the transcription of Tlr4,Inos,and Tnf-a was significantly up-regulated at week 4.Interestingly,hepatic Lcn-2 expression was 90-fold at week 4 and 507-fold at week 8 higher in L-HFr-subjected rats vs control(P<0.001).In contrast to HDL-cholesterol,systemic levels of LCN-2,fasting leptin and triglycerides were elevated in the L-HFr regimen(P<0.001).Moreover,protein expression of hepatic LCN-2,CD14,phosphoMAPK,caspase-9,cytochrome c and 4-hydroxynonenal was increased in the L-HFr group.Conversely,the hepatic expression of PGC-1a(a mitochondrial-biogenic protein)was reduced in the L-HFr category at week 8.The localization of LCN-2 in the liver was predominantly restricted to MPO+granulocytes.CONCLUSION:Fructose diet up-regulates hepatic LCN-2 expression,which correlates with the increased indicators of oxidative stress and mitochondrial dysfunction.The LCN-2 may be involved in liver protection. | Salamah Mohammad Alwahsh Min Xu Hatice Ali Seyhan Shakil Ahmad Sabine Mihm Giuliano Ramadori Frank Christian Schultze | 2014 | World Journal of Gastroenterology2014,20,7: | 7 |
| 2 | Spatio-temporal variation of land surface temperature and temperature lapse rate over mountainous Kashmir Himalaya显示文摘In this study, Land Surface Temperature(LST) and its lapse rate over the mountainous Kashmir Himalaya was estimated using MODIS data and correlated with the observed in-situ air temperature(Tair) data. Comparison between the MODIS LST and Tair showed a close agreement with the maximum error of the estimate ±1°C and the correlation coefficient >0.90. Analysis of the LST data from 2002-2012 showed an increasing trend at all the selected locations except at a site located in the southeastern part of Kashmir valley. Using the GTOPO30 DEM, MODIS LST data was used to estimate the actual temperature lapse rate(ATLR) along various transects across Kashmir Himalaya, which showed significant variations in space and time ranging from 0.3°C to 1.2°C per 100 m altitude change. This observation is at variance with the standard temperature lapse rate(STLR) of 0.65°C used universally in most of the hydrological and other land surface models. Snowmelt Runoff Model(SRM) was used to determine the efficacy of using the ATLR for simulating the stream flows in one of the glaciated and snow-covered watersheds in Kashmir. The use of ATLR in the SRM model improved the R2 between the observed and predicted streamflows from 0.92 to 0.97.It is hoped that the operational use of satellite-derived LST and ATLR shall improve the understanding and quantification of various processes related to climate, hydrology and ecosystem in the mountainous and data-scarce Himalaya where the use of temperature and ATLR are critical parameters for understanding various land surface and climate processes. | Shakil Ahmad ROMSHOO Mohammd RAFIQ Irfan RASHID | 2018 | Journal of Mountain Science2018,15,3: | 4 |
| 3 | Morphotectonic and Lithostratigraphic Analysis of Intermontane Karewa Basin of Kashmir Himalayas,India显示文摘Morpho-tectonic study plays an important role in deciphering the effects of tectonic activity in the geomorphic evolution of the drainage basins.Romushi watershed forms one of the major watersheds of the intermontane Karewa Basin of Kashmir Valley.The Karewa sediments are characterized by glacio-fluvio-lacustrine deposits capped by the aeolian loess.The geomorphic,morphometric and lithostratigraphic studies of these cap deposits have been carried out to elucidate the effect of tectonics on the geomorphic evolution of Romushi Watershed.Geomorphic mapping was carried out using GPS measurements,DEM at 30m resolution,Topographic Position Index(TPI) model,topographic maps,LANDSAT TM Imagery and field data.Morphometric and morphotectonic analyses in GIS environment were used to calculate various geomorphic indices(Mountain Front Sinuosity Index,Bifurcation Ratio,Asymmetry Factor,River Profile,etc).These indices reveal that the tectonic uplift observed in the region due to Himalayan orogeny coupled with mass movement and aeolian deposition have dominated the landscape evolution of intermontane Karewa Basin of Kashmir throughout the Late Quaternary Period.Additional data from lithostratigraphic measurements were analyzed to understand the geomorphic evolution of intermontane Karewa Basin.The data revealed that the basin has experienced differential uplift and erosion rates from time to time in the geological past.This was corroborated by the results from the morphometric and morphotectonic analysis. | Reyaz Ahmad DAR Rakesh CHANDRA Shakil Ahmad ROMSHOO | 2013 | Journal of Mountain Science2013,10,1: | 4 |
| 4 | Effect of glycyrrhizic acid and 18β-glycyrrhetinic acid on the differentiation of human umbilical cord-mesenchymal stem cells into hepatocytes显示文摘BACKGROUND End-stage liver disease is a global health complication with high prevalence and limited treatment options.Cell-based therapies using mesenchymal stem cells(MSCs)emerged as an alternative approach to support hepatic regeneration.In vitro preconditioning strategies have been employed to strengthen the regenerative and differentiation potential of MSCs towards hepatic lineage.Chemical compounds of the triterpene class;glycyrrhizic acid(GA)and 18β-glycyrrhetinic acid(GT)possess diverse therapeutic properties including hepatoprotection and anti-fibrosis characteristics.They are capable of modulating several signaling pathways that are crucial in hepatic regeneration.Preconditioning with hepato-protective triterpenes may stimulate MSC fate transition towards hepatocytes.AIM To explore the effect of GA and GT on hepatic differentiation of human umbilical cord-MSCs(hUC-MSCs).METHODS hUC-MSCs were isolated and characterized phenotypically by flow cytometry and immunocytochemistry for the expression of MSC-associated surface molecules.Isolated cells were treated with GA,GT,and their combination for 24 h and then analyzed at three time points;day 7,14,and 21.qRT-PCR was performed for the expression of hepatic genes.Expression of hepatic proteins was analyzed by immunocytochemistry at day 21.Periodic acid Schiff staining was performed to determine the functional ability of treated cells.RESULTS The fusiform-shaped morphology of MSCs in the treatment groups in comparison with the untreated control,eventually progressed towards the polygonal morphology of hepatocytes with the passage of time.The temporal transcriptional profile of preconditioned MSCs displayed significant expression of hepatic genes with increasing time of differentiation.Preconditioned cells showed positive expression of hepatocyte-specific proteins.The results were further corroborated by positive periodic acid Schiff staining,indicating the presence of glycogen in their cytoplasm.Moreover,bi-nucleated cells,which is the typical feature of hepatocytes,were also seen in the preconditioned cells.CONCLUSION Preconditioning with glycyrrhizic acid,18β-glycyrrhetinic acid and their combination,successfully differentiates hUC-MSCs into hepatic-like cells.These MSCs may serve as a better therapeutic option for degenerative liver diseases in future. | Abiha Fatima Tuba Shakil Malick Irfan Khan Aisha Ishaque Asmat Salim | 2021 | World Journal of Stem Cells2021,13,10: | 3 |
