| 2 | Conversion therapy and suitable timing for subsequent salvage surgery for initially unresectable hepatocellular carcinoma: What is new?显示文摘AIM To review the conversion therapy for initially unre-sectable hepatocellular carcinoma(HCC) patients and the suitable timing for subsequent salvage surgery. METHODS A Pub Med search was undertaken from 1987 to 2017 to identify articles using the keywords including 'unresectable' 'hepatocellular carcinoma', 'hepate-ctomy', 'conversion therapy', 'resection', 'salvage surgery' and 'downstaging'. Additional studies were investigated through a manual search of the references from the articles. The exclusion criteria were duplicates, case reports, case series, videos, contents unrelated to the topic, comments, and editorial essays. The main and widely used conversion therapies and the suitable timing for subsequent salvage surgery were discussed in detail. Two members of our group independently performed the literature search and data extraction. RESULTS Liver volume measurements [future liver remnant(FLR)/total liver volume or residual liver volume/bodyweight ratio] and function tests(scoring systems and liver stiffness) were often performed in order to justify whether patients were suitable candidates for surgery. Successful conversion therapy was usually defined as downstaging the tumor, increasing FLR and providing subsequent salvage surgery, without increasing com-plications, morbidity or mortality. The requirementsfor performing salvage surgery after transcatheter arterial chemoembolization were the achievement of a partial remission in radiology, the disappearance of the portal vein thrombosis, and the lack of extrahepatic metastasis. Patients with a standardized FLR(sF LR) > 20% were good candidates for surgery after portal vein embolization, while other predictive parameters like growth rate, kinetic growth rate were treated as an effective supplementary. There was probably not enough evidence to provide a standard operation time after associating liver partition and portal vein ligation for staged hepatectomy or yttrium-90 microsphere radioembolization. The indications of any combinations of conversion therapies and the subsequent salvage surgery time still need to be carefully and comprehen-sively evaluated. | 5 | FluxExplorer: A general platform for modeling and analyses of metabolic net-works based on stoichiometry显示文摘Stoichiometry-based analyses of meta- bolic networks have aroused significant interest of systems biology researchers in recent years. It is necessary to develop a more convenient modeling platform on which users can reconstruct their network models using completely graphical operations, and explore them with powerful analyzing modules to get a better understanding of the properties of metabolic systems. Herein, an in silico platform, FluxExplorer, for metabolic modeling and analyses based on stoichiometry has been developed as a publicly available tool for systems biology research. This platform integrates various analytic approaches, in- cluding flux balance analysis, minimization of meta- bolic adjustment, extreme pathways analysis, shadow prices analysis, and singular value decom- position, providing a thorough characterization of the metabolic system. Using a graphic modeling process, metabolic networks can be reconstructed and modi- fied intuitively and conveniently. The inconsistencies of a model with respect to the FBA principles can be proved automatically. In addition, this platform sup- ports systems biology markup language (SBML). FluxExplorer has been applied to rebuild a metabolic network in mammalian mitochondria, producing meaningful results. | 7 | Interface structures and mechanical properties of corrugated + flat rolled and traditional rolled Mg/Al clad plates显示文摘The corrugated + flat rolling(CFR) and traditional rolling(TR) methods were used to prepare Mg/Al clad plates using AZ31 B Mg and 5052 Al plates, and the interface morphologies and mechanical properties of the resulting clad plates were compared. Examination of the microstructures of the plates showed that the TRed Mg/Al clad plate presented a straight interface, while a corrugated interface containing fractured intermetallic particulates was observed for the CFRed plate due to the inhomogeneous strain induced by the corrugated roller. During the CFR process, the corrugated roller can accelerate the rupture of the substrate work-hardening layers and facilitate the mutual extrusion of fresh metals to enhance the interface bonding. Compared with the traditional basal texture of the Mg alloy, the CFR process can change the texture morphology, thereby enhancing the plastic deformation ability of the Mg plate. Tensile tests showed that the CFRed Mg/Al clad plate exhibited a higher ultimate tensile strength(UTS, 316 MPa), which was~ 8% higher than that of the TRed plate(293 MPa). In addition, the bending curve of the CFRed clad plate was smooth and there was no stress sudden drop phenomenon in the bending process. | 8 | Integrating full-length transcriptomics and metabolomics reveals the regulatory mechanisms underlying yellow pigmentation in tree peony(Paeonia suffruticosa Andr.)flowers显示文摘Tree peony(Paeonia suffruticosa Andr.)is a popular ornamental plant in China due to its showy and colorful flowers.However,yellow-colored flowers are rare in both wild species and domesticated cultivars.The molecular mechanisms underlying yellow pigmentation remain poorly understood.Here,petal tissues of two tree peony cultivars,“High Noon”(yellow flowers)and“Roufurong”(purple–red flowers),were sampled at five developmental stages(S1–S5)from early flower buds to full blooms.Five petal color indices(brightness,redness,yellowness,chroma,and hue angle)and the contents of ten different flavonoids were determined.Compared to“Roufurong,”which accumulated abundant anthocyanins at S3–S5,the yellow-colored“High Noon”displayed relatively higher contents of tetrahydroxychalcone(THC),flavones,and flavonols but no anthocyanin production.The contents of THC,flavones,and flavonols in“High Noon”peaked at S3 and dropped gradually as the flower bloomed,consistent with the color index patterns.Furthermore,RNA-seq analyses at S3 showed that structural genes such as PsC4Hs,PsDFRs,and PsUFGTs in the flavonoid biosynthesis pathway were downregulated in“High Noon,”whereas most PsFLSs,PsF3Hs,and PsF3’Hs were upregulated.Five transcription factor(TF)genes related to flavonoid biosynthesis were also upregulated in“High Noon.”One of these TFs,PsMYB111,was overexpressed in tobacco,which led to increased flavonols but decreased anthocyanins.Dual-luciferase assays further confirmed that PsMYB111 upregulated PsFLS.These results improve our understanding of yellow pigmentation in tree peony and provide a guide for future molecular-assisted breeding experiments in tree peony with novel flower colors. | Xiaoning Luo Daoyang Sun Shu Wang Sha Luo Yaqi Fu Lixin Niu Qianqian Shi Yanlong Zhang | 2021 | Horticulture Research2021,8,1: | 5 |

| 11 | Metal-Free C_(3)N_(4) with plentiful nitrogen vacancy and increased specific surface area for electrocatalytic nitrogen reduction显示文摘As a substitute for synthetic ammonia under mild condition, electrocatalytic nitrogen reduction reaction(NRR) provides a hopeful approach for the development of ammonia. Nevertheless, the current development of NRR electrocatalysts is far from enough and a systematic research is needed to gain a better improvement. This article presents that 2 D C_(3)N_(4)-NV with a large specific surface area and abundant nitrogen vacancies is prepared by a simple and feasible method, and used as a metal-free catalyst for electrocatalytic NRR. Experiment result and density functional theory(DFT) calculation reveal that nitrogen vacancies in 2 D C_(3)N_(4)-NV can act as an efficient active site for catalytic NRR, which is conducive to capturing and activating N_(2), lowering Gibbs free energy(DG) in reaction and inhibiting hydrogen evolution reaction(HER) at the same time. In addition, the larger specific surface area also makes more active site exposed, which is good for the contact between the electrolyte and the active site, thus enhancing its NRR activity. They have been developed from herpes simplex virus 1(HSV-1) strain H129(H129), and have been successfully applied to map diverse neural circuits.Initially, the anterograde polysynaptic tracer H129-G4 was used by many groups. We then developed the first monosynaptic tracer, H129-dTK-tdT, which was highly successful, yet improvements are needed. Now, by inserting another tdTomato expression cassette into the H129-dTK-tdT genome, we have created H129-dTK-T2, an updated version of H129-dTK-tdT that has improved labeling intensity. To help scientists produce and apply our H129-derived viral tracers, here we provide the protocol describing our detailed and standardized procedures. Commonly-encountered technical problems and their solutions are also discussed in detail. | 14 | Clinical practice guidelines for treating headache with Traditional Chinese Medicine: quality assessment with the appraisal of guidelines for research and evaluation Ⅱ instrument显示文摘OBJECTIVE: To critically appraise the methodological quality of clinical practice guidelines for headache produced over the last two decades, including those covering specific interventions using Traditional Chinese Medicine.METHODS: The guidelines on headache disorders were obtained by searching a number of databases, including Pub Med, EMBASE, Web of Science,Chinese Biomedical Literature Database, China National Knowledge Infrastructure Database, China Science and Technology Journal Database, and Wanfang database, three guideline-related databases [Guideline-International Network, National Guideline Clearinghouse, and Medlive], and the records of organizations that develop guidelines. The publication date was limited to the period from January 1996 to June 2015. The search terms 'headache', 'headache disorders', 'cephalalgia', 'migraine', 'tension-type headache', 'practice guideline', 'consensus ', 'statement', 'regulation', and 'recommendation' were used in the 'Me SH' and 'Free-text' fields. The guidelines were independently appraised by four researchers using the Appraisal of Guidelines for Research and Evaluation Ⅱ instrument.RESULTS: A total of 23 guidelines published between 1998 and 2014 were reviewed. The overall consistency of the four appraisers was good [interclass correlation coefficient 0.84; 95% confidence interval(CI) 0.82-0.86]. The mean(standard deviation) scores for scope and purpose, stakeholder involvement, rigor of development, clarity of presentation, applicability, and editorial independence were 52.1(18.0), 39.5(17.1), 33.4(21.0), 49.8(21.9),23.8(19.3), and 24.2(23.7). Only two guidelines were recommended, 12 were recommended with modification, and nine were not recommended.CONCLUSION: Physical Traditional Chinese Medicine therapies were recommended to treat headache. The overall quality of headache guidelines was low in China, but evidence-based guidelines are gradually becoming mainstream. The adsorption efficiency of nanometer-size TiO2 colloid for ultra-trace Cd(II) could reach above 96% in a short time when the pH value was between 5 and 6. Other problems were also studied,such as adsorption capacity,nanometer-size TiO2 colloid dosage,effect of coexistent ions. The detection limit(3σ) and the relative standard deviation (R.S.D) of this method were 4.46?10-3 μg/L and 1.30%(n=7),respectively. | 18 | Visualizing DC morphology and T cell motility to characterize DC-T cell encounters in mouse lymph nodes under mTOR inhibition显示文摘Mammalian target of rapamycin(mTOR),a serine/threonine kinase orchestrating cellular metabolism,is a crucial immune system regulator.However,it remains unclear how mTOR regulates dendritic cell(DC) function in vivo,especially DC-T cell encounters,a critical step for initiating adaptive immune responses.We dynamically visualized DC-T contacts in mouse lymph node using confocal microscopy and established an encounter model to characterize the effect of mTOR inhibition on DC-T cell encounters using DC morphology.Quantitative data showed mTOR inhibition via rapamycin altered DC shape,with an increased form factor(30.17%) and decreased cellular surface area(20.36%) and perimeter(22.43%),which were associated with Cdc42 protein downregulation(52.71%).Additionally,DCs adopted a similar morphological change with Cdc42 inhibition via ZCL278 as that observed with mTOR inhibition.These morphologically altered DCs displayed low encounter rates with T cells.Time-lapse imaging data of T cell motility supported the simulated result of the encounter model,where antigen-specific T cells appeared to reduce arrest in the lymph nodes of rapamycin-pretreated mice relative to the untreated group.Therefore,mTOR inhibition altered DC morphology in vivo and decreased the DC-T cell encounter rate,as well as Cdc42 inhibition.By establishing an encounter model,our study provides an intuitive approach for the early prediction of DC function through morphological quantification of form factor and area. | Qiaoya Lin Zheng Liu Meijie Luo Hao Zheng Sha Qiao Chenlu Han Deqiang Deng Zhan Fan Yafang Lu Zhihong Zhang Qingming Luo | 2019 | Science China(Life Sciences)2019,62,9: | 2 |
