| 1 | A national report from China Liver Transplant Registry: steroid avoidance after liver transplantation for hepatocellular carcinoma显示文摘Objective:We aimed to evaluate the efficacy and safety of steroid-free immunosuppression after liver transplantation(LT)for hepatocellular carcinoma(HCC).Methods:We retrospectively analyzed HCC recipients without steroids after LT(SF group,n=368)based on the China Liver Transplant Registry(CLTR)database.These recipients were matched 1:2 with patients using steroids(S group,n=736)for the same period after LT for HCC,according to propensity scores.Results:Multivariate analysis indicates that recipients with younger age[odds ratio(OR),1.053;P=0.011],preoperative hepatitis B virus(HBV)DNA≥1,000 copies/mL(OR,2.597;P=0.004)and beyond Milan criteria(OR,4.255;P<0.001)were identified as the risk factors associated with tumor recurrence in steroid avoidance recipients after LT.The patients fulfilling the Milan criteria in the SF group presented higher overall and tumor-free survival rates than those in the S group(P<0.05).Multivariate analysis revealed that recipient beyond Milan criteria was an independent prognostic factor for overall survival(OR,1.690;P<0.001)and tumor-free survival(OR,2.066;P<0.001).The incidences of new-onset diabetes mellitus(21.20% vs.33.29%,P<0.001),new-onset hypertension(10.05%vs.18.61%,P<0.001)and hyperlipidemia(4.08% vs.7.20%,P=0.042)were significantly lower in the SF group.Conclusions:Steroid-free immunosuppression could be safe and feasible for HBV-related HCC patients in LT.Age,HBV DNA level and Milan criteria maybe risk factors associated with tumor recurrence in steroid avoidance recipients.Recipient beyond Milan criteria was an independent prognostic factor and recipient fulfilling Milan criteria can benefit the most from steroid-free immunosuppression. | Qiang Wei Feng Gao Runzhou Zhuang Qi Ling Qinghong Ke Jian Wu Tian Shen Mangli Zhang Min Zhang Xiao Xu Shusen Zheng | 2017 | Chinese Journal of Cancer Research2017,29,5: | 25 |

| 3 | The Use of Postoperative Serum HS-AFP and GGTⅡfor Judgment of the Prognosis for Hepatocellular Carcinoma patients显示文摘OBJECTIVE To investigate the clinical value of hepatoma-specific alpha -fetoprotein (HS-AFP) and gamma-glutamyltransferaseⅡ(GGTⅡ) for judgment of postoperative prognosis of patients with hepatocellular carcinoma (HCC). METHODS HS-AFP was separated and determined using native polyacrylamide electrophoresis with a discontinuous buffer system and Western blots. GGTⅡwas separated with native polyacrylamide electrophoresis with a discontinuous buffer system and detected by enzyme staining. Forty cases with HCC underwent serial determination of HS-AFP and GGTⅡbefore and after radical excision. The correlations were analyzed between the two indices and survival time. RESULTS In the 40 cases with HCC, before radical excision the positive rates of HS-AFP and GGTⅡwere 57.5% and 67.5% respectively, with the positive rate of combined HS-AFP and GGTⅡreaching 80.0%. After operation, the recurrence and metastasis rate in the groups with positive HS-AFP and GGTⅡwere 90.9% and 58.8% respectively, while in the groups with negative HS-AFP and GGTⅡthe rates were 20.7% and 26.1% respectively. Recurrence and metastasis occurred in all cases with both postoperative positive HS-AFP and GGTⅡbut only in 9.5% of the cases in whom both postoperative HS-AFP and GGTⅡwere negative. Univariate analysis revealed that postoperative HS-AFP and GGTⅡwere related to the prognosis in HCC. | 6 | The PtoTCP20-miR396d-PtoGRF15 module regulates secondary vascular development in Populus显示文摘Secondary vascular development is a key biological characteristic of woody plants and the basis of wood formation.Our understanding of gene expression regulation and dynamic changes in microRNAs(miRNAs)during secondary vascular development is still limited.Here we present an integrated analysis of the miRNA and mRNA transcriptome of six phase-specific tissues-the shoot apex,procambium,primary vascular tissue,cambium,secondary phloem,and secondary xylem-in Populus tomentosa.Several novel regulatory modules,including the PtoTCP20-miR396d-PtoGRF15 module,were identified during secondary vascular development in Populus.A series of biochemical and molecular experiments confirmed that PtoTCP20 activated transcription of the miR396d precursor gene and that miR396d targeted PtoGRF15 to downregulate its expression.Plants overexpressing miR396d(35S:miR396d)showed enhanced secondary growth and increased xylem production.Conversely,during the transition from primary to secondary vascular development,plants with downregulated PtoTCP20expression(PtoTCP20-SRDX),downregulated miR396 expression(35S:STTM396),and PtoGRF15 overexpression(35S:PtoGRF15)showed delayed secondary growth.Novel regulatory modules were identified by integrated analysis of the miRNA and mRNA transcriptome,and the regulatory role of the PtoTCP20-miR396d-PtoGRF15 signaling cascade in secondary vascular development was validated in Populus,providing information to support improvements in forest cultivation and wood properties. | Lingyan Wang Jie Hou Huimin Xu Yufei Zhang Runzhou Huang Donghui Wang Xin-Qiang He | 2023 | Plant Communications2023,4,2: | 1 |

| 11 | Expression of TRAIL and its receptors in primary hepatic carcinoma and apoptosis-inducing effect of HrsTRAIL on hepatoma cell line HepG2显示文摘Objective: To investigate the expression of tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) and its receptors in primary hepatic carcinoma(PHC) and the apoptosis-inducing effect on hepatoma cell line HepG2. Methods: TRAIL and its receptors were detected by semiquantitive RT-PCR in 30 PHC and para-carcinoma tissues and two hepatoma cell lines of HepG2 and SMMC-7721. HepG2 cells were treated with human recombinant soluble TRAIL protein (HrsTRAIL) and then the viability of HepG2 cells was measured by microculture tetrazolium dye(MTT) assay and apoptosis index was demonstrated by fluorescence-activated cell sorting(FACS). Results:TRAIL and its receptors were detectable in all PHC and para-carcinoma tissues and hepatoma cell line HepG2. TRAIL, death receptor 4(DR4), DR5, and decoy receptor 2(DcR2) but not DcR1 were detectable in hepatoma cell line SMMC-7721. The expression patterns of TRAIL receptors in HepG2 were quite similar to PHC specimens. The semiquantitive results showed that the expression level of TRAIL and DcR were lower but DR was higher in hepatoma tissues than in para-carcinoma tissues. In PHC tissues, the expressions of DR were higher than DcR, while there was no difference in para-carcinoma tissues. HrsTRAIL had potent antitumor activity in a time- and dose-dependent manner. After co-incubations of the HepG2 cells in the presence of HrsTRAIL at concentration 1 000 ng/ml for 24 hours, the viability of HepG2 cells decreased to 45% and the apoptosis index reached 51%. Conclusion:TRAIL and its receptors were expressed in both PHC tissues and para-carcinoma tissues but the expression levels were different. The lower expression of TRAIL in PHC tissues suggested that insufficient apoptosis occured in the development of PHC. High expression of DR in PHC tissues may be a self-defense mechanism and may afford a theory of HrsTRAIL therapy for PHC. | 12 | Experimental Study on the Film Cooling Characteristics of Three Complex Tip Structures显示文摘The turbine blades of aircrafts must be properly cooled to prevent engine failure.Thus,to investigate the influence of the tip structure on the film cooling effect,pressure-sensitive paint test technology was used to determine the adiabatic film cooling effectiveness in this study.The experiment was completed in a cascade comprising three straight blades.The effects of the blowing ratio,density ratio,tip clearance,and tip structure on film cooling efficiency were analyzed.The experimental results demonstrated that,as the blowing ratio increased,the film coverage area and film cooling efficiency increased under most experimental conditions.However,the film cooling efficiency was found to initially increase,and subsequently decrease,as the blowing ratio increased.The respective influences of the density ratio and tip clearance on the film cooling efficiency were found to be significant.The density ratio experiments revealed that a high-density ratio can result in better film coverage than the low-density-ratio air.The tip clearance experimental results indicated that a small tip clearance promotes an increase in film cooling efficiency;this is because the small tip clearance negatively affects the main stream leakage flow,which can reduce the film coverage area.Under the conditions of the Base case 2 configuration,a blowing ratio of 2.1,and a tip clearance of 0.6%h,the average film cooling efficiency of the blade tip was 0.22.Among the three blade tip structures applied in this study,Base case 2 demonstrated higher film cooling efficiency than the other two blade tip structures under the conditions of the same blowing ratio,tip clearance,and density ratio. | LIN Juqiang LI Haiwang YOU Ruquan LIU Runzhou TAO Zhi LIU Song | 2023 | Journal of Thermal Science2023,32,4: | 0 |
| 13 | Cost-Utility Analysis of Screening for Diabetic Retinopathy in China显示文摘Background.Diabetic retinopathy(DR)has been primarily indicated to cause vision impairment and blindness,while no studies have focused on the cost-utility of telemedicine-based and community screening programs for DR in China,especially in rural and urban areas,respectively.Methods.We developed a Markov model to calculate the cost-utility of screening programs for DR in DM patients in rural and urban settings from the societal perspective.The incremental cost-utility ratio(ICUR)was calculated for the assessment.Results.In the rural setting,the community screening program obtained 1 QALY with a cost of$4179(95%CI 3859 to 5343),and the telemedicine screening program had an ICUR of$2323(95%CI 1023 to 3903)compared with no screening,both of which satisfied the criterion of a significantly cost-effective health intervention.Likewise,community screening programs in urban areas generated an ICUR of$3812(95%CI 2906 to 4167)per QALY gained,with telemedicine screening at an ICUR of$2437(95%CI 1242 to 3520)compared with no screening,and both were also cost-effective.By further comparison,compared to community screening programs,telemedicine screening yielded an ICUR of 1212(95%CI 896 to 1590)per incremental QALY gained in rural setting and 1141(95%CI 859 to 1403)in urban setting,which both meet the criterion for a significantly cost-effective health intervention.Conclusions.Both telemedicine and community screening for DR in rural and urban settings were cost-effective in China,and telemedicine screening programs were more cost-effective. | Yue Zhang Weiling Bai Ruyue Li Yifan Du Runzhou Sun Tao Li Hong Kang Ziwei Yang Jianjun Tang Ningli Wang Hanruo Liu | 2022 | Health Data Science2022,,1: | 0 |

| 15 | Clinical Value of Hepatoma-Specific Alpha-Fetoprotein in the Diagnosis of Hepatocellular Carcinoma显示文摘OBJECTIVE To study the clinical value of hepatoma-specific alpha-feto- protein (HS-AFP) in the diagnosis and differential diagnosis of hepato- cellular carcinoma (HCC). METHODS A method of vertical slab polyacrylamide gel electrophoresis with discontinuous buffer system was developed to separate AFP sub- types. After separation, the AFP subtypes were transferred to nitrocellu- lose and reacted first with rabbit anti-human AFP and then with goat an- ti-rabbit IgG -HRP. Finally, AFP subtypes were visualized by reacting with 3,3’-diaminobenzidine tetrahydrochloride. A HS-AFP band was de- termined in 82 cases with HCC and 95 cases with benign liver diseases. The correlations between the positive rates of HS-AFP and serum AFP concentration, tumor size as well as portal vein metastasis were analyzed. RESULTS Serum AFP in the cases with various liver diseases was sepa- rated into one to several bands. The fastest band on electrophoresis (F- AFP) was found in all patients, while the band at the cathodal site (HS- AFP) was detected predominantly in HCC but rarely in benign liver dis- eases. The positive rate of HS-AFP in HCC was 74.4%, which was sig- nificantly higher than that in benign liver diseases (9.1%, 7.3% and 10.0% in liver cirrhosis, chronic hepatitis and acute hepatitis respectively). HS- AFP was detected in 3 out of 9 HCC cases with AFP<50 μg/L , but in none of 22 cases of benign liver diseases with the same AFP concentra- tion. HS-AFP correlated with serum AFP concentration and tumor size to some extent, but not with portal vein metastasis. | 17 | Correlation between Thymidylate Synthase Genotype and Susceptibility to Gastric Carcinoma显示文摘OBJECTIVE To investigate the correlation betweenpolymorphism of the 5'-untranslated region (5'-UTR) ofthymidylate synthase genes,as well as the lifestyle,and thesusceptibility of gastric carcinoma.METHODS A case-control study,with 60 cases of gastriccarcinoma and 170 cases of general risk population-based controlsfrom Nantong,Jiangsu province,China,was conducted.Theepidemiological data,such as living habits of the cancer patients,were collected.DNA of peripheral blood leukocytes was obtainedfrom all of the subjects.The TS 5'-UTR tandem repeat genotypewas detected using polymerase chain reaction (PCR).RESULTS There were three TS 5'-UTR genotypes in the groupof gastric cancer cases (2R/2R,2R/3R and 3R/3R) and six TS5'-UTR genotypes in the group of the controls (2R/2R,2R/3R,3R/3R,2R/4R,2R/5R and 3R/4R).The genotypic frequencies wererespectively 5.0%,43.3% and 51.7% in the gastric cancer group.Compared with the parameters in the control group,i.e.,4.7%,31.7%,60.6%,1.2%,1.2% and 0.6%.There were no significantdifferences between the two groups.Compared with the 3R/3R-genotype individuals who where non-smokers,drank alcoholtwice or less each week,drank tea and did not intake pickled food(PF),the risk of gastric cancer significantly went up in the 2R/2Ror 2R/3R-genotype people who had habits of smoking,drinkingalcohol more than twice each week,no tea drinking but withfrequent intake of PF.The adjusted ORs were as follows,3.79 (95%CI:2.45~8.64),3.41 (95% CI:1.21~8.47),5.99 (95% CI:3.01~14.7),and 3.61 (95% CI:1.81~8.78).CONCLUSION There is an obvious correlation between thepolymorphisms of TS 5'-UTR genotypes and the lifestyle ofindividuals in the development of gastric carcinoma. | Lei Yang Mingbing Xiao Runzhou Ni Qinghe Tan Jinzhi Wei Jianhong Wang Bojian Ge | 2008 | Chinese Journal of Clinical Oncology2008,5,6: | 0 |

| 19 | Transcriptome-wide analysis of mRNA N^(6)-methyladenosine modification in the embryonic development of Spodoptera frugiperda显示文摘N^(6)-methyladenosine(m^(6)A)RNA is the most abundant modification of mRNA,and has been demonstrated in regulating various post-transcriptional processes.Many studies have shown that m^(6)A methylation plays key roles in sex determination,neuronal functions,and embryonic development in Drosophila and mammals.Here,we analyzed transcriptome-wide profile of m^(6)A modification in the embryonic development of the destructive agricultural pest Spodoptera frugiperda.We found that the 2 key mRNA m^(6)A methyltransferases SfrMETTL3 and SfrMETTL14 have high homologies with other insects and mammals,suggesting that SfrMETTL3 and SfrMETTL14 may have conserved function among different species.From methylated RNA immunoprecipitation sequencing analysis,we obtained 46869 m^(6)A peaks representing 8587 transcripts in the 2-h embryos after oviposition,and 41389 m^(6)A peaks representing 9230 transcripts in the 24-h embryos.In addition,5995 m^(6)A peaks were differentially expressed including 3752 upregulated and 2243 downregulated peaks.Functional analysis with Gene Ontology and Kyoto Encyclopedia of Genes and Genomes suggested that differentially expressed m^(6)A peak-modified genes were enriched in cell and organ development between the 2-and 24-h embryos.By conjoint analysis of methylated RNA immunoprecipitation-seq and RNA-seq data,we found that RNA m^(6)A methylation may regulate the transcriptional levels of genes related to tissue and organ development from 2-to 24-h embryos.Our study reveals the role of RNA m^(6)A epigenetic regulation in the embryonic development of S.frugiperda,and provides new insights for the embryonic development of insects. | Yaqing Chen Yushan Lai Runzhou Liu Lin Yao Xiao-Qiang Yu Xiaoyun Wang | 2023 | Insect Science2023,30,5: | 0 |
| 20 | Nonlinear sealing force of a seawater balance valve used in an 11000-meter manned submersible显示文摘Balance valve is a core component of the 11000-meter manned submersible“struggle,”and its sealing performance is crucial and challenging when the maximum pressure difference is 118 MPa.The increasing sealing force improves the sealing performance and increases the system’s energy consumption at the same time.A hybrid analytical–numerical–experimental(ANE)model is proposed to obtain the minimum sealing force,ensuring no leakage at the valve port and reducing energy consumption as much as possible.The effects of roundness error,environmental pressure,and materials on the minimum sealing force are considered in the ANE model.The basic form of minimum sealing force equations is established,and the remaining unknown coefficients of the equations are obtained by the finite element method(FEM).The accuracy of the equation is evaluated by comparing the independent FEM data to the equation data.Results of the comparison show good agreement,and the difference between the independent FEM data and equation data is within 3%when the environmental pressure is 0–118 MPa.Finally,the minimum sealing force equation is applied in a balance valve to be experimented using a deep-sea simulation device.The balance valve designed through the minimum sealing force equation is leak-free in the experiment.Thus,the minimum sealing force equation is suitable for the ultrahigh pressure balance valve and has guiding significance for evaluating the sealing performance of ultrahigh pressure balance valves. | Zhenyao WANG Yinshui LIU Qian CHENG Runzhou XU Yunxiang MA Defa WU | 2023 | Frontiers of Mechanical Engineering2023,18,1: | 0 |
