| 1 | Bone marrow-derived mesenchymal stem cells protect against experimental liver fibrosis in rats显示文摘AIM: Recent reports have shown the capacity of mesenchymal stem cells (MSCs) to differentiate into hepatocytes in vitro and in vivo. MSCs administration could repair injured liver, lung, or heart through reducing inflammation, collagen deposition, and remodeling. These results provide a clue to treatment of liver fibrosis. The aim of this study was to investigate the effect of infusion of bone marrow (BM)-derived MSCs on the experimental liver fibrosis in rats.METHODS: MSCs isolated from BM in male Fischer 344 rats were infused to female Wistar rats induced with carbon tetrachloride (CCl4) or dimethylnitrosamine (DMN).There were two random groups on the 42nd d of CCl4:CCl4/MSCs, to infuse a dose of MSCs alone; CCl4/saline,to infuse the same volume of saline as control. There were another three random groups after exposure to DMN: DMN10/MSCs, to infuse the same dose of MSCs on d 10; DMN10/saline, to infuse the same volume of saline on d 10; DMN20/MSCs, to infuse the same dose of MSCson d 20. The morphological and behavioral changes ofrats were monitored everyday. After 4-6 wk of MSCs administration, all rats were killed and fibrosis index were assessed by histopathology and radioimmunoassay. Smooth muscle alpha-actin (alpha-SMA) of liver were tested by immunohistochemistry and quantified by IBAS 2.5 software. Male rats sex determination region on the Y chromosome (sry) gene were explored by PCR.RESULTS: Compared to controls, infusion of MSCsreduced the mortality rates of incidence in CCl4-induced model (10% vs 20%) and in DMN-induced model (2040% vs 90%).The amount of collagen deposition and alpha-SMA staining was about 40-50% lower in liver of rats with MSCs than that of rats without MSCs. The similar results were observed in fibrosis index. And the effect of the inhibition of fibrogenesis was greater in DMN10/MSCs than in DMN20/MSCs. The particle flow behavior and transport processes at different screw rotating speeds and filling levels were investigated in this study. The spatial distributions of particle velocity were predicted. The predicted mass flow rate increased w让h increasing screw rotating speed and filling level. The contact forces and granular temperatures of particles were also calculated. The simulation results showed that the translational granular particle temperatures were higher than the rotational granular particle temperatures. In add让ion, the configurational temperatures of particles were calculated from simulated instantaneous particle overlaps, and results indicated that deformation of elastic particles contributed to the rate of energy dissipation. | 4 | Mechanisms and field test of solution mining by self-resonating cavitating water jets显示文摘Rapid solution mining is the key to cavern construction in salt formations. Rapid solution mining technology with self-resonating cavitating water jets is described in this paper. It can generate three main physical effects: helical flow dissolution, self-resonating cavitating jet erosion, and ultrasonic waves. A self-resonating cavitating nozzle was also designed with the principles based on theory of fluid transients and hydro-acoustics. Under ambient pressure, the experimental results show that the impulse amplitude of pressure reaches a peak at a standoff distance of 5-13 times the nozzle outlet diameter and the cutting ability of self-resonating cavitating jets is twice that of conventional jets under the same conditions. | 5 | In Vitro Characterizations of PLLA/β-TCP Porous Matrix Materials and RMSC-PLLA-β-TCP Composite Scaffolds显示文摘To develop a novel degradable poly (L-lactic acid)/β-tricalcium phosphate (PLLA/β-TCP) bioactive materials for bone tissue engineering, β-TCP powder was produced by a new wet process. Porous scaffolds were prepared by three steps, I.e. Solvent casting, compression molding and leaching stage. Factors influencing the compressive strength and the degradation behavior of the porous scaffold, e.g. Weight fraction of pore forming agent-sodium chloride (NaCl), weight ratio of PLLA: β-TCP, the particle size ofβ-TCP and the porosity, were discussed in details. Rat marrow stromal cells (RMSC) were incorporated into the composite by tissue engineering approach. Biological and osteogenesis potential of the composite scaffold were determined with MTT assay, alkaline phosphatase (ALP) activity and bone osteocalcin (OCN) content evaluation. Results show that PLLA/β-TCP bioactive porous scaffold has good mechanical and pore structure with adjustable compressive strength needed for surgery. RMSCs seeding on porous PLLA/β-TCP composite behaves good seeding efficacy, biocompatibility and osteoinductive potential. Osteoprogenitor cells could well penetrate into the material matrix and begin cell proliferation and osteogenic differentiation. Osseous matrix could be formed on the surface of the composite after culturing in vitro. | 6 | Key technologies and practice for gas field storage facility construction of complex geological conditions in China显示文摘In view of complex geological characteristics and alternating loading conditions associated with cyclic large amount of gas injection and withdrawal in underground gas storage(UGS) of China, a series of key gas storage construction technologies were established, mainly including UGS site selection and evaluation, key index design, well drilling and completion, surface engineering and operational risk warning and assessment, etc. The effect of field application was discussed and summarized. Firstly, trap dynamic sealing capacity evaluation technology for conversion of UGS from the fault depleted or partially depleted gas reservoirs. A key index design method mainly based on the effective gas storage capacity design for water flooded heterogeneous gas reservoirs was proposed. To effectively guide the engineering construction of UGS, the safe well drilling, high quality cementing and high pressure and large flow surface injection and production engineering optimization suitable for long-term alternate loading condition and ultra-deep and ultra-low temperature formation were developed. The core surface equipment like high pressure gas injection compressor can be manufactured by our own. Last, the full-system operational risk warning and assessment technology for UGS was set up. The above 5 key technologies have been utilized in site selection, development scheme design, engineering construction and annual operations of 6 UGS groups, e.g. the Hutubi UGS in Xinjiang. | 19 | Evaluation of power regeneration in primary suspension for a railway vehicle显示文摘To improve the fuel economy of rail vehicles,this study presents the feasibility of using power regenerating dampers(PRDs)in the primary suspension systems of railway vehicles and evaluates the potential and recoverable power that can be obtained.PRDs are configured as hydraulic electromagnetic-based railway primary vertical dampers and evaluated in parallel and series modes(with and without a viscous damper).Hydraulic configuration converts the linear behavior of the track into a unidirectional rotation of the generator,and the electromagnetic configuration provides a controllable damping force to the primary suspension system.In several case studies,generic railway vehicle primary suspension systems that are configured to include a PRD in the two configuration modes are modeled using computer simulations.The simulations are performed on measured tracks with typical irregularities for a generic UK passenger route.The performance of the modified vehicle is evaluated with respect to key performance indicators,including regenerated power,ride comfort,and running safety.Results indicate that PRDs can simultaneously replace conventional primary vertical dampers,regenerate power,and exhibit desirable dynamic performance.A peak power efficiency of 79.87%is theoretically obtained in series mode on a top-quality German Intercity Express track(Track 270)at a vehicle speed of 160 mile/h(~257 km/h). | Ruichen WANG Zhiwei WANG | 2020 | Frontiers of Mechanical Engineering2020,15,2: | 1 |
| 20 | Pharmacokinetics after single and multiple intravaginal application of clindamycin phosphate vaginal gel in Chinese female volunteers显示文摘Objective:To investigate the pharmacokinetics of clindamycin phosphate vaginal gel in healthy Chinese female volunteers.Methods:Ten healthy Chinese female volun- teers were intravaginally given with 5.0g of clindamycin phosphate vaginal gel (equivalent to 100mg of clindamycin) once for single dose treatment,and 5.0g,once a day for 3 days,for mul- tiple dose treatment.The serum concentration of clindamycin were determined by HPLC-MS method and its pharmacokinetic parameters of clindamycin were calculated by DAS 1.0 soft- ware.Results:The main pharmacokinetic parameters of clindamycin for single dose and multiple doses were as follows:t_(1/2) were (15.30±2.62) hours and (14.78±2.49) hours,Tmax were (4.88±0.94) hours and (4.70±0.59) hours,Cmax were (38.30±22.77) ng/ml and (44.87±26.71) ng/ml,AUC0_(-∞) were (783.45±351.19) ng·ml^(-1)·h^(-1) and (1015.68±456.95) ng·ml^(-1)·h^(-1),respectively.Conclusion:The Cmax of clindamycin phosphate vaginal gel after a single dose and multiple doses are obviously lower and t_(1,2) are longer than that of clindamycin phosphate oral preparations,which suggests that clindamycin phosphate vaginal gel acts locally and can be slowly absorbed to circulation for systemic actions. | Wang Benjie Li Yunxia Yuan Guiyan Bu Fanlong Guo Ruichen | 2007 | 现代妇产科进展2007,16,2: | 1 |
