| 1 | Isolation and Characterization of Fucoidans from Five Brown Algae and Evaluation of Their Antioxidant Activity显示文摘In this study, we evaluated the chemical property and antioxidant activity of fucoidans isolated from brown algae, Laminaria japonica(LJF), Lessonia nigrescens(LNF), Lessonia trabeculata(LTF), Ascophyllum mackaii(AMF), and Ecklonia maxima(EMF). LJF was less in sulfate content(14.16%) and more in galactose and mannose content(1.08 and 0.68) than the documented early. EMF contained 20%–30% of sulfate and fucose, 0.97 in molar ratio which was lower than that of sulfate to other four fucoidans(1.21–1.41). AMF(162 kDa) and EMF(150 kDa) were the first two largest in molecular weight, which were followed by LJP(126 kDa), LNF(113 kDa) and LTF(105 kDa). The fucoidans isolated these algae showed a wide range of antioxidant activity in vitro. It was found that the reducing power of the isolated fucoidans was positively correlated with their sulfate content and molecular weight. In addition, LNF and LTF at low concentrations exhibited high superoxide and hydroxyl radical scavenging activity. | 3 | Pharmacokinetics after single and multiple intravaginal application of clindamycin phosphate vaginal gel in Chinese female volunteers显示文摘Objective:To investigate the pharmacokinetics of clindamycin phosphate vaginal gel in healthy Chinese female volunteers.Methods:Ten healthy Chinese female volun- teers were intravaginally given with 5.0g of clindamycin phosphate vaginal gel (equivalent to 100mg of clindamycin) once for single dose treatment,and 5.0g,once a day for 3 days,for mul- tiple dose treatment.The serum concentration of clindamycin were determined by HPLC-MS method and its pharmacokinetic parameters of clindamycin were calculated by DAS 1.0 soft- ware.Results:The main pharmacokinetic parameters of clindamycin for single dose and multiple doses were as follows:t_(1/2) were (15.30±2.62) hours and (14.78±2.49) hours,Tmax were (4.88±0.94) hours and (4.70±0.59) hours,Cmax were (38.30±22.77) ng/ml and (44.87±26.71) ng/ml,AUC0_(-∞) were (783.45±351.19) ng·ml^(-1)·h^(-1) and (1015.68±456.95) ng·ml^(-1)·h^(-1),respectively.Conclusion:The Cmax of clindamycin phosphate vaginal gel after a single dose and multiple doses are obviously lower and t_(1,2) are longer than that of clindamycin phosphate oral preparations,which suggests that clindamycin phosphate vaginal gel acts locally and can be slowly absorbed to circulation for systemic actions. | Wang Benjie Li Yunxia Yuan Guiyan Bu Fanlong Guo Ruichen | 2007 | 现代妇产科进展2007,16,2: | 1 |

| 6 | Visible light cross-linking and bioactive peptides loaded integrated hydrogel with sequential release to accelerate wound healing complicated by bacterial infection显示文摘The effective management of bacterial infections that are resistant to multiple drugs remains a substantial clinical challenge.The eradication of drug-resistant bacteria and subsequent promotion of angiogenesis are imperative for the regeneration of the infected wounds.Here,a novel and facile peptide containing injectable hydrogel with sustained antibacterial and angiogenic capabilities is developed.The antibacterial peptide that consists of 11 residues(CM11,WKLFKKILKVL)is loaded onto acrylate-modified gelatin through charge interactions.A vascular endothelial growth factor mimetic peptide KLT(KLTWQELYQLKYKGI)with a GCG(Gly-Cys-Gly)modification at the N-terminal is covalently coupled through a visible light-induced thiol-ene reaction.In this reaction,the acrylate gelatin undergoes cross-linkage within seconds.Based on the physical/chemical double crosslinking strategy,the bioactive peptides achieve sustained and sequential release.The results show that the hydrogel significantly inhibits methicillin-resistant Staphylococcus aureus(MRSA)growth through the rapid release of CM11 peptides at early stage;it forms obvious growth inhibition zones against pathogenic bacterial strains.Moreover,cell counting kit-8 assay and scratch test confirm that the CM11/KLT-functionalized hydrogels promote cell proliferation and migration through the later release of KLT peptides.In a mouse skin wound infected with self-luminous MRSA,the CM11/KLT-functionalized hydrogels enhance wound healing,with rapidly bacterial infection reduction,lower expression of inflammatory factors,and neovascularization promotion.These results suggest that the rationally designed,sustained and sequential release CM11/KLT-functionalized hydrogels have huge potential in promoting the healing of multi-drug resistant bacterial infected wounds. | Guiyan Wang Ning Yuan Jun Zhang Man Qin Suwei Dong Yuguang Wang | 2024 | Nano Research2024,17,3: | 0 |
