1Significant value of 18F-FDG-PET/CT in diagnosing small cervical lymph node metastases in patients with nasopharyngeal carcinoma treated with intensity-modulated radiotherapy显示文摘Background: Little is known about the nature of metaistasis to small cervical lymph nodes(SCLNS) in the patients with nasopharyngeal carcinoma(NPC)examined by using 18-fluoro-2-deoxy-glucose(^(18)F-FDG) positron emission tomography/computed tomography(PET/CT).The present study aimed to evaluate the diagnostic values of PET/CT in identifying metastasis in SCLNs in NPC patients.Methods: Magnetic resonance images(MRI) and PET/CT scans for 470 patients with newly diagnosed, non-distant metastatic NPC were analyzed. Metastatic rates of SCLNs were defined by the positive number of SCLNs on PET/CT scans and total number of SCLNs on MRI scans. Receiver operating characteristic curve was applied to compare PET/CT-determined stage with MRI-determined stage.Results: In total, 2082 SCLNs were identified, with 808(38.8%) ≥ 5 and < 6 mm in diameter(group A), 526(25.3%)≥ 6 and < 7 mm in diameter(group B),374(18.0%)≥ 7 and < 8 mm in diameter(group C), 237(11.4%) ≥8 and<9 mm in diameter(group D),and 137(6.5%) ≥ 9 and <10 mm in diameter(group E).The overall metastatic rates examined by using PET/CT for groups A, B,C,D, and E were 3.5%, 8.0%, 31.3%, 60.0%, and 83.9%, respectively(P< 0.001). In level IV/Vb, the metastatic rate for nodes ≥ 8 mm was 84.6%. PET/CT examination resulted in modification of N category and overall stage for 135(28.7%) and 46(9.8%) patients, respectively. 3Reduction process and zinc removal from composite briquettes composed of dust and sludge from a steel enterprise显示文摘In this study, composite briquettes were prepared using gravity dust and converter sludge as the main materials; these briquettes were subsequently reduced in a tube furnace at 1000–1300°C for 5–30 min under a nitrogen atmosphere. The effects of reaction temperature, reaction time, and carbon content on the metallization and dezincification ratios of the composite briquettes were studied. The reduced composite briquettes were characterized by X-ray diffraction(XRD), scanning electron microscopy(SEM), and energy-dispersive spectroscopy(EDS). The results show that the gravity dust and converter sludge are combined into the composite briquettes and a reasonable combination not only improves the performance of the composite briquettes, but also leads to the reduction with no or little reductant and flux. As the reaction temperature is increased and the reaction time is extended, the metallization and dezincification ratios of the composite briquettes increase gradually. When the composite briquettes are roasted at 1300°C for 30 min, the metallization ratio and dezincification ratio reaches 91.35% and 99.25%, respectively, indicating that most of the iron oxide is reduced and the zinc is almost completely removed. 4A Precise RFID Indoor Localization System with Sensor Network Assistance显示文摘Indoor localization is very critical for medical care applications, e.g., the patient localization or tracking inside the building of the hospital. Traditional Radio Frequency Identification(RFID) technologies are very popular in this area since their cost is very low. In such technologies, each tag acts as the transmitter and the Radio Signal Strength Indicator(RSSI) information is measured from the readers. However, RSSI information suffers severely from the multi- path phenomenon. As a result, if in a very large area, the localization accuracy will be affected seriously. In order to solve this problem, we introduce Wireless Sensor Networks(WSNs) with only a few nodes, each of which acts as both transmitter and receiver. In such networks, the change of signal strength(referred as dynamic of RSSI) is leveraged to select a cluster of reference tags as candidates. Then the fi nal target location is estimated by using the RSSI relationships between the target tag and candidate reference tags. Thus, the localization accuracy and scalability are able to be improved. We proposed two algorithms, SA-LANDMARC, and COCKTAIL. Experiments show that the localization accuracy of the two algorithms can reach 0.7m and 0.45 m, respectively. 5Deep-learning-based extraction of the animal migration patterns from weather radar images显示文摘Continental coverage and year-round operation of the weather radar networks provide an unprecedented opportunity for studying large-scale airborne migration.The broad and local-scale airborne information collected by these infrastructures can answer many ecological questions.However,extracting and interpreting the biological information from such massive weather radar data remains an intractable problem.Recently,many big-data problems have been solved using the deep learning technology.In this study,the biological information in the weather radar data is identified using the advanced deep learning method.The proposed method consists of two main parts,i.e.,a rendering and casting procedure and an image segmentation procedure based on a convolutional neural network.The biological data are automatically extracted by rendering and mapping,image segmentation,and result masking.By analyzing the typical radar data from single and multiple stations,we partly reveal the intensity and speed of the migration pattern.We present the first feasibility study of the extraction of local and large-scale biological phenomena from the Chinese weather radar network data.Kai CUI Cheng HU Rui WANG Yi SUI Huafeng MAO Huayu LI 2020Science China(Information Sciences)2020,63,4:11
6Controlled synthesis of single cobalt atom catalysts via a facile one-pot pyrolysis for efficient oxygen reduction and hydrogen evolution reactions显示文摘Metal-nitrogen doped carbon catalysts(M-N/C) with abundantly accessible M-Nxsites, particularly single metal atom M-N/C(SAM-N/C), have been developed as a substitute for expensive Pt-based catalysts.These catalysts are used to increase the efficiency of otherwise sluggish oxygen reduction reactions(ORR) and hydrogen evolution reactions(HER). However, although the agglomerated metal nanoparticles are usually easy to form, they are very difficult to remove due to the protective surface-coating carbon layers, a factor that significantly hampers SAM-N/C fabrication. Herein, we report a one-step pyrolysis approach to successfully fabricate single cobalt atom Co-N/C(SACo-N/C) by using a Co2+-SCN-coordination compound as the metal precursor. Thanks to the decomposition of Co2+-SCN-compound at lower temperature than that of carbon layer deposition, Co-rich particles grow up to larger ones before carbon layers formation. Even though encapsulated by the carbon layers, it is difficult for the large Co-rich particle to be completely sealed. And thus, it makes the Co atoms possible to escape from incomplete carbon layer, to coordinate with nitrogen atoms, and to form SACo-N/C catalysts. This SACo-N/C exhibits excellent performances for both ORR(half-wave potential of 0.878 V) and HER(overpotential at 10 mA/cm2 of178 m V), and is thus a potential replacement for Pt-based catalysts. When SACo-N/C is integrated into a Zn-O2 battery, battery with high open-circuit voltage(1.536 V) has high peak power density(266 mW/cm2)and large gravimetric energy density(755 mA h/gZn) at current densities of 100 mA/cm2. 7Three-dimensional printing: review of application in medicine and hepatic surgery显示文摘Three-dimensional(3D) printing(3DP) is a rapid prototyping technology that has gained increasing recognition in many different fields. Inherent accuracy and low-cost property enable applicability of 3DP in many areas, such as manufacturing, aerospace,medical, and industrial design. Recently, 3DP has gained considerable attention in the medical field. The image data can be quickly turned into physical objects by using 3DP technology. These objects are being used across a variety of surgical specialties. The shortage of cadaver specimens is a major problem in medical education. However, this concern has been solved with the emergence of 3DP model. Custom-made items can be produced by using 3DP technology. This innovation allows 3DP use in preoperative planning and surgical training. Learning is difficult among medical students because of the complex anatomical structures of the liver. Thus, 3D visualization is a useful tool in anatomy teaching and hepatic surgical training. However,conventional models do not capture haptic qualities. 3DP can produce highly accurate and complex physical models. Many types of human or animal differentiated cells can be printed successfully with the development of 3D bio-printing technology. This progress represents a valuable breakthrough that exhibits many potential uses, such as research on drug metabolism or liver disease mechanism. 10Effects of mild hypothermia on the ROS and expression of caspase-3 m RNA and LC3 of hippocampus nerve cells in rats after cardiopulmonary resuscitation显示文摘BACKGROUND: Cardiac arrest(CA) is a common and serious event in emergency medicine. Despite recent improvements in resuscitation techniques, the survival rate of patients with CA is unchanged. The present study was undertaken to observe the effect of mild hypothermia(MH) on the reactive oxygen species(ROS) and the effect of neurological function and related mechanisms.METHODS: Sixty-five healthy male Sprague Dawley(SD) adult rats were randomly(random number) divided into 2 groups: blank control group(n=5) and CPR group(n=60). CA was induced by asphyxia. The surviving rats were randomly(random number) divided into two groups: normothermia CPR group(NT) and hypothermia CPR group(HT). Normothermia of 37 °C was maintained in the NT group after return of spontaneous circulation(ROSC), hypothermal intervention of 32 °C was carried out in the HT group for 4 hours immediately after ROSC. Both the NT and HT groups were then randomly divided into 2 subgroups 12 hours and 24 hours after ROSC(NT-12, NT-24, HT-12, HT-24 subgroups). During observation, the neurological defi cit scores(NDSs) was recorded, then the bilateral hippocampi were obtained from rats' head, and monoplast suspension of fresh hippocampus tissue was made immediately to determine the level of intracellular ROS by flow cytometry. Transmission electron microscope was used to observe the ultramicro changes of cellular nucleus and mitochondria. Reverse transcription-polymerase chain reaction(RT-PCR) was used to determine the expression of caspase-3 m RNA, and western-blotting(WB) was used to determine the level of LC3 in frozen hippocampus tissue. Measured data were analyzed with paired sample t test and One-Way ANOVA.RESULTS: Of 60 rats with CA, 44(73%) were successfully resuscitated and 33(55%) survived until the end of the experiment. The NDSs of rats in the NT and HT groups were more signifi cantly reduced than those in the BC group(F=8.107, P<0.05), whereas the NDSs of rats in the HT-12 and HT-24 subgroups were significantly increased in comparison with those NDSs of rats in the NT-12 and NT-24 subgroups, respectively(t=9.692, P<0.001; t=14.374, P<0.001). The ROS in hippocampus nerve cells in the NT and HT groups signifi cantly increased compared to the BC group(F=16.824, P<0.05), whereas the ROS in the HT-12 and HT-24 subgroups significantly reduced compared with that ROS in the NT-12 and NT-24 subgroups, respectively(t=9.836, P<0.001; t=7.499, P<0.001). The expression of caspase-3 m RNA in hippocampus nerve cells in the NT and HT groups were signifi cantly increased compared to the BC group(F=24.527, P<0.05), whereas the expression of caspase-3 m RNA in rats of the HT-12 and HT-24 subgroups was signifi cantly reduced compared to the NT-12 and NT-24 subgroups, respectively(t=6.935, P<0.001; t=4.317, P<0.001). The expression of LC3B-II/I in hippocampus nerve cells of rats in the NT and HT groups signifi cantlyincreased compared to the BC group(F=6.584, P<0.05), whereas the expression of LC3B-II/I in rats of the HT-12 and HT-24 subgroups significantly reduced compared to the NT-12 and NT-24 subgroups, respectively(t=10.836, P<0.001; t=2.653, P=0.02). 11Adaptable hydrogel with reversible linkages for regenerative medicine:Dynamic mechanical microenvironment for cells显示文摘Hydrogels are three-dimensional platforms that serve as substitutes for native extracellular matrix.These materials are starting to play important roles in regenerative medicine because of their similarities to native matrix in water content and flexibility.It would be very advantagoues for researchers to be able to regulate cell behavior and fate with specific hydrogels that have tunable mechanical properties as biophysical cues.Recent developments in dynamic chemistry have yielded designs of adaptable hydrogels that mimic dynamic nature of extracellular matrix.The current review provides a comprehensive overview for adaptable hydrogel in regenerative medicine as follows.First,we outline strategies to design adaptable hydrogel network with reversible linkages according to previous findings in supramolecular chemistry and dynamic covalent chemistry.Next,we describe the mechanism of dynamic mechanical microenvironment influence cell behaviors and fate,including how stress relaxation influences on cell behavior and how mechanosignals regulate matrix remodeling.Finally,we highlight techniques such as bioprinting which utilize adaptable hydrogel in regenerative medicine.We conclude by discussing the limitations and challenges for adaptable hydrogel,and we present perspectives for future studies.Zongrui Tong Lulu Jin Joaquim Miguel Oliveira Rui LReis Qi Zhong Zhengwei Mao Changyou Gao 2021Bioactive Materials2021,6,5:8
12Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 14Steering control strategy guide by two preview vision cues显示文摘Vision cues play an important role in states feedback in motion control.However,the existing driver steering models consider little about vision cues utilized by human drivers during their steering procedure.This paper presents a novel steering control strategy based on two preview points(far point and near point).The far point is used to compensate the steering wheel by predicting the upcoming curvature change with respect to the lane,while the near point as vision feedback,which is used to tune the steering wheel by estimating the errors of vehicle states and lane center.To obtain much smoother lateral acceleration during steering,a forward internal model is established using a second-order yaw dynamics system that captures the influence of yaw angular acceleration caused by steering wheel angle.The input parameter of the second-order system is the vision cues of both the near and far points,and the output parameters are the ideal yaw angle and yaw rate.To calculate suitable the steering wheel angle,an adaptive controller is designed using fuzzy sliding technology,which is used as the input of the vehicle system dynamics.Numerical simulation results show that the proposed method performs better than the existing driver steering models in case of imitating human drivers' behavior,and exhibits excellent adaption to the lane curvature change.SHEN Huan LING Rui MAO JianGuo LI ShunMing 2012Science China(Technological Sciences)2012,55,9:7
15Correlation between the expressions of gastrin, somatostatin and cyclin and cyclin-depend kinase in colorectal cancer显示文摘AIM: To explore the correlation between the expressions of gastrin (GAS), somatostatin (SS) and cyclin, cyclindependent kinase (CDK) in colorectal cancer, and to detect the specific regulatory sites where gastrointestinal hormone regulates cell proliferation.METHODS: Seventy-nine resected large intestine carcinomatous specimens were randomly selected.Immunohistochemical staining for GAS, SS, cyclin D1, cyclin E, cyclin A, cyclin B1, CDK2 and CDK4 was performed according to the standard streptavidinbiotin-peroxidase (S-P) method. According to the semiquantitative integral evaluation, SS and GAS were divided into high, middle and low groups. Cyclin D1, cyclin E,cyclin A, cyclin B1, CDK2, CDK4 expressions in the three GAS and SS groups were assessed.RESULTS: The positive expression rate of cyclin D1 was significantly higher in high (78.6%, 11/14) and middle GAS groups (73.9%, 17/23) than in low GAS group (45.2%, 19/42) (P<0.05,χ2 high vs low = 4.691; P<0.05, χ2 middle vs low =4.945). The positive expression rate of cyclin A was significantly higher in high (100%, 14/14)and middle GAS groups (82.6%, 19/23) than in low GAS group (54.8%, 23/42) (P<0.01, χ2 high vs low = 9.586;P<0.05, χ middle vs low = 5.040). The positive expression rate of CDK2 was significantly higher in high (92.9%, 13/14)and middle GAS groups (87.0%, 20/23) than in low GAS group (50.0%, 21/42) (P<0.01, χ2 high vs low = 8.086;P<0.01, χ2 middle vs low = 8.715). The positive expression rate of CDK4 was significantly higher in high (78.6%, 11/14)and middle GAS groups (78.3%, 18/23) than in low GAS group (42.9%, 18/42) (P<0.05, χ2 high vs low = 5.364;P<0.01, χ2 middle vs low = 7.539). The positive expression rate of cyclin E was prominently higher in low SS group (53.3%, 24/45) than in high (9.1%, 1/11) and middle (21.7%, 5/23) SS groups (P<0.05, χ2 high vs low = 5.325;P<0.05, χ2 middle vs low = 6.212). The positive expression rate of CDK2 was significantly higher in low SS group (77.8%, 35/45) than in high SS group (27.3%, 3/11)(P<0.01, χ2 high vs low = 8.151). There was a significant positive correlation between the integral ratio of GAS to SS and the semi-quantitative integral of cyclin D1, cyclin E, cyclin A, CDK2, CDK4 (P<0.05, D1rs = 0.252; P<0.01,Ers = 0.387; P<0.01, Ars = 0.466; P<0.01, K2rs = 0.519;P<0.01, K4rs = 0.434).CONCLUSION: The regulation and control of gastrin, SS in colorectal cancer cell growth may be directly related to the abnormal expressions of cyclins D1, A, E, and CDK2,CDK4. 16Integrated analysis of high-throughput sequencing data shows abscisic acidresponsive genes and miRNAs in strawberry receptacle fruit ripening显示文摘The perception and signal transduction of the plant hormone abscisic acid(ABA)are crucial for strawberry fruit ripening,but the underlying mechanism of how ABA regulates ripening-related genes has not been well understood.By employing high-throughput sequencing technology,we comprehensively analyzed transcriptomic and miRNA expression profiles simultaneously in ABA-and nordihydroguaiaretic acid(NDGA,an ABA biosynthesis blocker)-treated strawberry fruits with temporal resolution.The results revealed that ABA regulated many genes in different pathways,including hormone signal transduction and the biosynthesis of secondary metabolites.Transcription factor genes belonging to WRKY and heat shock factor(HSF)families might play key roles in regulating the expression of ABA inducible genes,whereas the KNOTTED1-like homeobox protein and Squamosa Promoter-Binding-like protein 18 might be responsible for ABA-downregulated genes.Additionally,20 known and six novel differentially expressed miRNAs might be important regulators that assist ABA in regulating target genes that are involved in versatile physiological processes,such as hormone balance regulation,pigments formation and cell wall degradation.Furthermore,degradome analysis showed that one novel miRNA,Fa_novel6,could degrade its target gene HERCULES1,which likely contributed to fruit size determination during strawberry ripening.These results expanded our understanding of how ABA drives the strawberry fruit ripening process as well as the role of miRNAs in this process.Dongdong Li Wangshu Mou Rui Xia Li Li Christopher Zawora Tiejin Ying Linchun Mao Zhongchi Liu Zisheng Luo 2019Horticulture Research2019,6,1:6
17Treatment of malignant digestive tract obstruction by combined intraluminal stent installation and intra-arterial drug infusion显示文摘AIM To study the palliative treatment of malignant obstrution of digestive tract with placement of intraluminal stent combined with intra-arterial infusion of chemotherapeutic drugs.METHODS A total of 281 cases of digestive tract malignant obstruction were given per oral (esophagus,stomach, duodenum and jejunum), per anal (colon and rectum ) and percutaneous transhepatic ( biliary )installation of metallic stent. Among them, 205 cases received drug infusion by cannulation of tumor supplying artery with Seldingers technique.RESULTS Altogether 350 stents were installed in 281 cases, obstructive symptoms were relieved or ameliorated after installation. Occurrence of restenotic obstruction was 8 - 43 weeks among those with intraarterial drug infusion, which was later than 4 - 26 weeks in the group with only stent installation. 19Functional identification of the terpene synthase family involved in diterpenoid alkaloids biosynthesis in Aconitum carmichaelii显示文摘Aconitum carmichaelii is a high-value medicinal herb widely used across China,Japan,and other Asian countries.Aconitine-type diterpene alkaloids(DAs)are the characteristic compounds in Aconitum.Although six transcriptomes,based on short-read next generation sequencing technology,have been reported from the Aconitum species,the terpene synthase(TPS)corresponding to DAs biosynthesis remains unidentified.We apply a combination of Pacbio isoform sequencing and RNA sequencing to provide a comprehensive view of the A.carmichaelii transcriptome.Nineteen TPSs and five alternative splicing isoforms belonging to TPS-b,TPS-c,and TPS-e/f subfamilies were identified.In vitro enzyme reaction analysis functional identified two sesqui-TPSs and twelve di TPSs.Seven of the TPS-c subfamily genes reacted with GGPP to produce the intermediate ent-copalyl diphosphate.Five Ac KSLs separately reacted with ent-CPP to produce ent-kaurene,ent-atiserene,and ent-13-epi-sandaracopimaradie:a new diterpene found in Aconitum.Ac TPSs gene expression in conjunction DAs content analysis in different tissues validated that ent-CPP is the sole precursor to all DAs biosynthesis,with Ac KSL1,Ac KSL2 s and Ac KSL3-1 responsible for C20 atisine and napelline type DAs biosynthesis,respectively.These data clarified the molecular basis for the C20-DAs biosynthetic pathway in A.carmichaelii and pave the way for further exploration of C19-DAs biosynthesis in the Aconitum species.Liuying Mao Baolong Jin Lingli Chen Mei Tian Rui Ma Biwei Yin Haiyan Zhang Juan Guo Jinfu Tang Tong Chen Changjiangsheng Lai Guanghong Cui Luqi Huang 2021Acta Pharmaceutica Sinica B2021,11,10:6
20Overexpression of p42.3 promotes cell growth and tumorigenicity in hepatocellular carcinoma显示文摘AIM:To investigate the association of p42.3 expression with clinicopathological characteristics and the biological function of p42.3 in human hepatocellular carcinoma(HCC).METHODS:We used reverse transcription-polymerase chain reaction(RT-PCR),quantitative real-time RT-PCR and western blotting to detect p42.3 mRNA and protein expression in hepatic cell lines.We examined primary HCC samples and matched adjacent normal tissue by immunohistochemistry to investigate the correlation between p42.3 expression and clinicopathological features.HepG2 cells were transfected with a pIRES2EGFP-p42.3 expression vector to examine the function of the p42.3 gene.Transfected cells were analyzed for their viability and malignant transformation abilities by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay,colony formation assay,and tumorigenicity assay in nude mice.RESULTS:p42.3 is differentially expressed in primary HCC tumors and cell lines.Approximately 69.6%(96/138) of cells were p42.3-positive in hepatic tumor tissues,while 30.7%(35/114) were p42.3-positive in tumor-adjacent normal tissues.Clinicopathological characteristics of the HCC specimens revealed a significant correlation between p42.3 expression and tumor differentiation(P = 0.031).However,p42.3 positivity was not related to tumor tumor-node-metastasis classification,hepatitis B virus status,or hepatoma type.Regarding p42.3 overexpression in stably transfected HepG2 cells,we discovered significant enhancement of cancer cell growth and colony formation in vitro,and significantly enhanced tumorigenicity in nude mice.Western blot analysis of cell cycle proteins revealed that enhanced p42.3 levels promote upregulation of proliferating cell nuclear antigen,cyclin B1 and mitotic arrest deficient 2.CONCLUSION:p42.3 promotes tumorigenicity and tumor growth in HCC and may be a potential target for future clinical cancer therapeutics.Wei Sun Wei-Wei Dong Lin-Lin Mao Wen-Mei Li Jian-Tao Cui Rui Xing You-Yong Lu 2013World Journal of Gastroenterology2013,19,19:5
