1Gut-brain connection: The neuroprotective effects of the anti-diabetic drug liraglutide显示文摘Long-acting glucagon-like peptide-1(GLP-1) analogues marketed for type 2 diabetes(T2D) treatment have been showing positive and protective effects in several different tissues, including pancreas, heart or even brain. This gut secreted hormone plays a potent insulinotropic activity and an important role in maintaining glucose homeostasis. Furthermore, growing evidences suggest the occurrence of several commonalities between T2 D and neurodegenerative diseases, insulin resistance being pointed as a main cause for cognitive decline and increased risk to develop dementia. In this regard, it has also been suggested that stimulation of brain insulin signaling may have a protective role against cognitive deficits. As GLP-1 receptors(GLP-1R) are expressed throughout the central nervous system and GLP-1 may cross the blood-brain-barrier, an emerging hypothesis suggests that they may be promising therapeutic targets against brain dysfunctional insulin signaling-related pathologies. Importantly, GLP-1 actions depend not only on the direct effect mediated by its receptor activation, but also on the gut-brain axis involving an exchange of signals between both tissues via the vagal nerve, thereby regulating numerous physiological functions(e.g., energy homeostasis, glucose-dependent insulin secretion, as well as appetite and weight control). Amongst the incretin/GLP-1 mimetics class of anti-T2 D drugs with an increasingly described neuroprotective potential, the already marketed liraglutide emerged as a GLP-1R agonist highly resistant to dipeptidyl peptidase-4 degradation(thereby having an increased half-life) and whose systemic GLP-1R activity is comparable to that of native GLP-1. Importantly, several preclinical studies showed anti-apoptotic, anti-inflammatory, anti-oxidant and neuroprotective effects of liraglutide against T2 D, stroke and Alzheimer disease(AD), whereas several clinical trials, demonstrated some surprising benefits of liraglutide on weight loss, microglia inhibition, behavior and cognition, and in AD biomarkers. 2Equilibrium, thermodynamic, and kinetic of Cr(VI) adsorption using a modified and unmodified bentonite clay显示文摘The capacities of natural and modified Brazilian bentonite samples as adsorbents to remove hexavalent metal chromium were investigated under several conditions in batch and column methods. The raw material, Ca-bentonite, was modified by anchorament of 3-aminopropyltrietoxisilane(APS) and 3,2-aminoethylaminopropyltrimetoxisilane(AEAPS) in the surface of bentonite sample. This type of new occurrence of bentonite is suitable as a raw material for adsorption process. Adsorption behavior of three bentonite types was strongly depending on pH of adsorbate solution, contact time adsorbent/adsorbate,and initial concentration of Cr(VI). The results were confirmed by column method and reveals that the adsorption process of materials accorded by the Redlich-Peterson, Sips, Dubinin-Radushkevich, and Langmuir isotherm models. The exothermic enthalpic values reflected a favorable energetic process for chromium ions anchored in the material surfaces. 4Murine model to study brain,behavior and immunity during hepatic encephalopathy显示文摘AIM:To propose an alternative model of hepatic encephalopathy(HE) in mice,resembling the human features of the disease.METHODS:Mice received two consecutive intraperitoneal injections of thioacetamide(TAA) at low dosage(300 mg/kg).Liver injury was assessed by serum transaminase levels(ALT) and liver histology(hematoxylin and eosin).Neutrophil infiltration was estimated by confocal liver intravital microscopy.Coagulopathy was evaluated using prolonged prothrombin and partial thromboplastin time.Hemodynamic parameters were measured through tail cuff.Ammonia levels were quantified in serum and brain samples.Electroencephalography(EEG) and psychomotor activity score were performed to show brain function.Brain edema was evaluated using magnetic resonance imaging.RESULTS:Mice submitted to the TAA regime developed massive liver injury,as shown by elevation of serum ALT levels and a high degree of liver necrosis.An intense hepatic neutrophil accumulation occurred in response to TAA-induced liver injury.This led to mice mortality and weight loss,which was associated with severe coagulopathy.Furthermore,TAA-treated mice presented with increased serum and cerebral levels of ammonia,in parallel with alterations in EEG spectrum and discrete brain edema,as shown by magnetic resonance imaging.In agreement with this,neuropsychomotor abnormalities ensued 36 h after TAA,fulfilling several HE features observed in humans.In this context of liver injury and neurological dysfunction,we observed lung inflammation and alterations in blood pressure and heart rate that were indicative of multiple organ dysfunction syndrome.CONCLUSION:In summary,we describe a new murine model of hepatic encephalopathy comprising multiple features of the disease in humans,which may provide new insights for treatment.Lindisley Ferreira Gomides Pedro Elias Marques Bruno Engler Faleiros Rafaela Vaz Pereira Sylvia Stella Amaral Thais Reis Lage Gustavo Henrique Souza Resende Patricia Alves Maia Guidine Giselle Foureaux Fabíola Mara Ribeiro Fabiana Paiva Martins Marco Antonio Peliky Fontes Anderson José Ferreira Remo Castro Russo Mauro Martins Teixeira Márcio Flávio Moraes Antonio Lúcio Teixeira Gustavo Batista Menezes 2014World Journal of Hepatology2014,6,4:2
