| 1 | Telbivudine vs tenofovir in hepatitis B e antigen-negative chronic hepatitis B patients: OPTIMA roadmap study显示文摘AIM To make efficacy and safety comparison of telbivudineraodmap and tenofovir-roadmap in hepatitis B e antigen(HBe Ag)-negative chronic hepatitis B(CHB) patients.METHODS This was the first prospective, randomised, two-arm, open-label, non-inferiority study in HBe Ag-negative CHB patients that compared telbivudine and tenofovir administered as per roadmap concept. Patients were treated up to 24 wk and, depending on virologic response, continued the same therapy or received addon therapy up to 104 wk. Eligible patients received an additional 52 wk of treatment in the extension period(i.e., up to 156 wk). Patients who developed virologic breakthrough(VB) while on monotherapy also received add-on therapy. The primary efficacy endpoint was the rate of patients achieving hepatitis B virus(HBV) DNA < 300 copies/m L at week 52. Secondary efficacy endpoints included the rates of HBV DNA < 300 and < 169 copies/m L, HBV DNA change from baseline, alanine aminotransferase normalisation, hepatitis B surface antigen(HBs Ag) loss, HBs Ag seroconversion, VB, and emergence of resistance at various timepoints throughout the study. Safety and estimated glomerular filtration rate(e GFR) were also analysed.RESULTS A total of 241 patients were randomised. Non-inferiority of telbivudine arm to tenofovir arm was demonstrated at week 52(± 7 d window), with over 91% of patients in each treatment arm achieving HBV DNA level < 300 copies/m L. Both arms were similar in terms of key secondary efficacy variables at weeks 104 and 156. The percentage of patients achieving HBV DNA < 300 copies/m L remained high and was similar in the telbivudine and tenofovir arms at both weeks 104 and 156. Over 82% of patients in both arms achieved alanine aminotransferase normalisation at week 52, and this percentage remained high at weeks 104 and 156. Telbivudine treatment progressively reduced serum HBs Ag levels from baseline while no change was reported in quantitative HBs Ag during therapy with tenofovir. Both treaments showed acceptable safety profiles. The telbivudine arm showed e GFR improvement unlike the tenofovir arm.CONCLUSION Efficacy was shown for both telbivudine-roadmap and tenofovir-roadmap regimens in HBe Ag-negative CHB patients over 156 wk. | 2 | Nrf2 activator corosolic acid meliorates alloxan induced diabetic nephropathy in mice显示文摘Objective: To determine whether corosolic acid(CA) targeting nuclear protein expression of Nrf2 activation can be used to attenuate renal damage and preserve renal function in alloxan diabetic mice.Methods: A mouse model with diabetic nephropathy was established to examine the Nrf2 expression. Mice were randomly divided into control, diabetic control, and CA groups treated at 0.4 mg/kg, 2 mg/kg and 10 mg/kg p.o. for 8 weeks. Diabetes was induced in mice by single intraperitoneal injection of alloxan 200 mg/kg in all groups except the control. The mice with fasting blood glucose level over 200 mg/d L were considered as diabetic and were employed in the study. After 4 th and 8 th weeks, urine samples were collected(using metabolic cages) to measure protein and urea. Animals were euthanized, and serum samples were collected to estimate the glucose, creatinine, total protein, urea and blood urea nitrogen. Kidney was isolated at the end of experiment for histology to evaluate anti-oxidant parameters. Immunohistochemistry was performed to examine the Nrf2 expression.Results: CA treatment showed dose dependent reduction in level of biochemical parameters in serum and urine. CA group(10 mg/kg) showed significantly higher body weight and reduced kidney weight. Histopathological examination revealed reduced inflammation, collagen deposition and glomerulosclerosis in renal tissue. | 3 | β-半乳糖苷酶的固定化及其在制备低乳糖牛奶中的应用显示文摘利用海藻酸钠和壳聚糖两种固定化载体对三种β-半乳糖苷酶(Maxilact酶、Lactozym酶和来源于米曲霉的β-半乳糖苷酶)进行固定化,研究温度和p H对酶活力的影响,游离酶和固定化酶水解牛奶中乳糖制备低乳糖牛奶,以及固定化酶的重复利用性。结果表明:与游离酶相比较,固定化酶在最适反应温度、最适反应p H、乳糖水解和重复利用性方面均有提高,表现出良好的稳定性。相比之下,壳聚糖固定化酶比海藻酸钠固定化酶的效果好,其中壳聚糖固定化Maxilact酶效果更为突出,该酶的最适反应温度为60℃,最适反应p H为7.0,在相同加酶量的条件下水解牛奶2 h后,乳糖水解率达99.93%,重复利用5次后,乳糖水解率仍能达到99.28%,重复利用性高,可以减少成本。此次研究为利用固定化酶工业化生产低乳糖牛奶奠定了一定的基础。 | 李冠龙 刘晓兰 KATROLIA Priti | 2018 | 食品工业2018,39,9: | 6 |
| 4 | Biomedical Perspective of Electrochemical Nanobiosensor显示文摘Electrochemical biosensor holds great promise in the biomedical area due to its enhanced specificity, sensitivity, label-free nature and cost effectiveness for rapid point-of-care detection of diseases at bedside. In this review, we are focusing on the working principle of electrochemical biosensor and how it can be employed in detecting biomarkers of fatal diseases like cancer, AIDS, hepatitis and cardiovascular diseases. Recent advances in the development of implantable biosensors and exploration of nanomaterials in fabrication of electrodes with increasing the sensitivity of biosensor for quick and easy detection of biomolecules have been elucidated in detail. Electrochemical-based detection of heavy metal ions which cause harmful effect on human health has been discussed. | 5 | Human immunodeficiency virus/acquired immune deficiency syndrome: Using drug from mathematical perceptive显示文摘Entry of acquired immune deficiency syndrome virus into the host immune cell involves the participation of various components of host and viral cell unit. These components may be categorized as attachment of the viral surface envelope protein subunit, gp120, to the CD4+ receptor and chemokine coreceptors, CCR5 and CXCR4, present on T cell surface. The viral fusion protein, gp41, the second cleaved subunit of Env undergoes reconfiguration and the membrane fusion reaction itself. Since the CD4+ T cell population is actively involved; the ultimate outcome of human immunodeficiency virus infection is total collapse of the host immune system. Mathematical modeling of the stages in viral membrane protein-host cell receptor-coreceptor interaction and the effect of antibody vaccine on the viral entry into the susceptible host cell has been carried out using as impulsive differential equations. We have studied the effect of antibody vaccination and determined analytically the threshold value of drug dosage and dosing interval for optimum levels of infection. | 14 | Saponin synthesis and cotton growth is antagonistically regulated by solar UV-B radiation显示文摘Background: Earlier we have reported that the exclusion of solar UV-B increased the growth and yield of cotton plants as compared with ambient UV-B. The UV-B radiation effects on the saponins and the impact of ambient and reduced UV-B on the accumulation of saponins has not been investigated yet. Thus a field experiment was conducted to study the influence of solar UV-B on the growth response and saponin synthesis in cotton(Gossypium hirsutum) var. Vikram plants by the exclusion of UV-B. The cotton plants were grown in specially designed iron chambers, wrapped with filters that excluded UV-B(<315 nm), or transmitted ambient UV-B.Results: Exclusion of ambient UV-B enhanced the growth of cotton plants in terms of plant height and leaf area.Greater plant height in UV-B excluded plants was due to elongated internode and more number of nodes.Enhancement in growth was accompanied by a decrease in the accumulation of saponins, which was quantified by the spectrophotometric, TLC and HPLC methods. Solvent extraction of saponins from the internodes and leaves were used for the bioassay of Amaranthus hypocotyl growth and expansion of cucumber cotyledons. | 20 | Saturation Effects on Immunopathogenic Mechanism of Psoriasis:A theoretical approach显示文摘Psoriasis vulgaris is a common chronic autoimmune inflammatory skin disease characterized by T-cell mediated hyper proliferation of keratinocytes.This type of skin disorder is characterized by macroscopic and also microscopic skin alterations.It was observed that,psoriatic lesions are sharply demarcated,red and slightly raised lesions along with silver whitish scales.The striking feature of T-cell mediated chronic psoriatic lesions are the epidermal infiltration of predominantly oligoclonal CD8^+T-cells and probably also of CD4^+T-cells in the dermis.In this research article we proposed a mathematical model for the chronic disease,psoriasis,consisting a set of differential equations, involving T-Lymphocytes cells,Dendritic cells and the epidermal Keratinocytes,incorporating half saturation constant through the T lymphocyte cells and also in that sense,it has been infiltrated in the activation process of keratinocytes in the course of the plaque of psoriasis.Cell biological research on psoriasis has been established the suppression of epidermal T-cell density.Here we are interested to know how the cell biological system changes for introducing the half saturation constant in presence of suppression taking place DCs on the model dynamics.Our analytical and numerical result reveal that due to control effect,the macrophages population density does not impede for the interaction between T-cell and keratinocytes.T-cell and keratinocyte population ultimately become stable after a certain period of time span. | Priti Kumar Roy Abhirup Datta Amar Nath Chatterjee | 2011 | 应用泛函分析学报2011,13,3: | 1 |
