1Thymoquinone suppresses migration of Lo Vo human colon cancer cells by reducing prostaglandin E2 induced COX-2 activation显示文摘AIM To identify potential anti-cancer constituents in natural extracts that inhibit cancer cell growth and migration.METHODS Our experiments used high dose thymoquinone(TQ) as an inhibitor to arrest Lo Vo(a human colon adenocarcinoma cell line) cancer cell growth, which was detected by cell proliferation assay and immunoblotting assay. Low dose TQ did not significantly reduce Lo Vo cancer cell growth. Cyclooxygenase 2(COX-2) is an enzyme that is involved in the conversion of arachidonic acid into prostaglandin E2(PGE2) in humans. PGE2 can promote COX-2 protein expression and tumor cell proliferation and was used as a control.RESULTS Our results showed that 20 μmol/L TQ significantly reduced human LoV o colon cancer cell proliferation. TQ treatment reduced the levels of p-PI3 K, p-Akt, p-GSK3β, and β-catenin and thereby inhibited the downstream COX-2 expression. Results also showed that the reduction in COX-2 expression resulted in a reduction in PGE2 levels and the suppression of EP2 and EP4 activation. Further analysis showed that TG treatment inhibited the nuclear translocation of β-catenin in LoV o cancer cells. The levels of the cofactors LEF-1 and TCF-4 were also decreased in the nucleus following TQ treatment in a dose-dependent manner. Treatment with low dose TQ inhibited the COX-2 expression at the transcriptional level and the regulation of COX-2 expression efficiently reduced LoV o cell migration. 2Effect of quercetin against lindane induced alterations in the serum and hepatic tissue lipids in wistar rats显示文摘Objective:To assess the effect of quercetin(flavonoid) against lindane induced alterations in lipid profile of wistar rats.Methods:Rats were administered orally with lindane(100 mg/kg body weight) and quercetin(10 mg/kg body weight) for 30 days.After the end of treatment period lipid profile was estimated in serum and tissue.Results:Elevated levels of serum cholesterol, triglycerides,low density lipoprotein(LDL),very Low Density Lipoprotein(VLDL) and tissue triglycerides,cholesterol with concomitant decrease in serum HDL and tissue phospholipids were decreased in lindane treated rats were found to be significantly decreased in the quercetin and lindane co-treated rats.Conclusions:Our study suggests that quercetin has hypolipidemic effect and offers protection against lindane induced toxicity in liver by restoring the altered levels of lipids.The quercetin cotreatment along with lindane for 30 days reversed these biochemical alterations in lipids induced by lindane.Viswanadha Vijaya Padma Gurusamy Lalitha Nichokon Puthanveedu Shirony Rathinasamy Baskaran 2012Asian Pacific Journal of Tropical Biomedicine2012,2,11:9
3Helicobacter pylori eradication in patients with chronic immune thrombocytopenic purpura显示文摘AIM:To assess the effect of Helicobacter pylori(H.pylori)eradication on platelet counts in patients with chronic immune thrombocytopenic purpura(cITP).METHODS:A total of 36 cITP patients were included in the study.The diagnosis of H.pylori was done by rapid urease test and Giemsa staining of the gastric biopsy specimen.All H.pylori positive patients received standard triple therapy for 14 d and were subjected for repeat endoscopy at 6 wk.Patients who continued to be positive for H.pylori on second endoscopyreceived second line salvage therapy.All the patients were assessed for platelet response at 6 wk,3rd and 6th months.RESULTS:Of the 36 patients,17 were positive for H.pylori infection and eradication was achieved in16 patients.The mean baseline platelet count in the eradicated patients was 88615.38±30117.93/mm3and platelet count after eradication at 6 wk,3 mo and6 mo was 143230.77±52437.51/mm3(P=0.003),152562.50±52892.3/mm3(P=0.0001),150187.50±41796.68/mm3(P=0.0001)respectively and in the negative patients,the mean baseline count was71000.00±33216.46/mm3 and at 6 wk,3rd and 6th month follow up was 137631.58±74364.13/mm3(P=0.001),125578.95±71472.1/mm3(P=0.005),77210.53±56892.28/mm3(P=0.684)respectively.CONCLUSION:Eradication of H.pylori leads to increase in platelet counts in patients with cITP and can be recommended as a complementary treatment with conventional therapy.Ravinder Naik Noonavath Chandrasekharan Padma Lakshmi Tarun Kumar Dutta Vikram Kate 2014World Journal of Gastroenterology2014,20,22:6
4Plant-Based Rasayana Drugs from Ayurveda显示文摘Rasayana tantra is one of the eight specialties of Ayurveda.It is a specialized practice in the form of rejuvenative recipes,dietary regimen,special health promoting behaviour and drugs.Properly administered Rasayana can bestow the human being with several benefits like longevity,memory,intelligence,freedom from diseases,youthful age,excellence of luster,complexion and voice,optimum strength of physique and sense organs,respectability and brilliance.Various types of plant based Rasayana recipes are mentioned in Ayurveda. 6^137Cs tracing dynamics of soil erosion,organic carbon,and total nitrogen in terraced fields and forestland in the Middle Mountains of Nepal显示文摘The Middle Mountains is one of the regions of Nepal most vulnerable to water erosion, where fragile geology, steep topography, anomalous climatic conditions, and intensive human activity have resulted in serious soil erosion and enhanced land degradation. Based on the 137 Cs tracing method, spatial variations in soil erosion, organic carbon, and total nitrogen(TN) in terraced fields lacking field banks and forestland were determined. Soil samples were collected at approximately 5 m and 20 m intervals along terraced field series and forestland transects respectively. Mean 137 Cs inventories of the four soil cores from the reference site was estimated at 574.33 ± 126.22 Bq m-2(1 Bq(i.e., one Becquerel) is equal to 1 disintegration per second(1 dps)). For each terrace, the 137 Cs inventory generally increased fromupper to lower slope positions, accompanied by a decrease in the soil erosion rate. Along the entire terraced toposequence, 137 Cs data showed that abrupt changes in soil erosion rates could occur between the lower part of the upper terrace and the upper part of the immediate terrace within a small distance. This result indicated that tillage erosion is also a dominant erosion type in the sloping farmland of this area. At the same time, we observed a fluctuant decrease in soil erosion rates for the whole terraced toposequence as well as a net deposition at the toe terrace. Although steep terraces(lacking banks and hedgerows) to some extent could act to limit soil sediment accumulation in catchments, soil erosion in the terraced field was determined to be serious. For forestland, with the exception of serious soil erosion that had taken place at the top of slopes due to concentrated flows from a country road situated above the forestland site, spatialvariation in soil erosion was similar to the 'standard' water erosion model. Soil organic carbon(SOC) and TN inventories showed similar spatial patterns to the 137 Cs inventory for both toposequences investigated. However, due to the different dominant erosion processes between the two, we found similar patterns between the <0.002 mm soil particle size fraction(clay sized) and 137 Cs inventories in terraced fields, while different patterns could be found between 137 Cs inventories and the <0.002 mm soil particle size fraction in the forestland site. 9Effect of angiotensin converting enzyme gene I/D polymorphism in South Indian children with nephrotic syndrome显示文摘Nephrotic syndrome is one of the most common childhood kidney diseases. It is mostly found in the age group of 2 to 8 years. Around 10%-15% of nephrotic syndrome cases are non-responders of steroid treatment(SRNS).Angiotensin converting enzyme(ACE)(I/D) gene association studies are important for detecting kidney disease and herein we assessed the association of ACE(I/D) polymorphism with nephrotic syndrome in South Indian children. We recruited 260 nephrotic syndrome(162 boys and 98 girls) and 218(140 boys and 78 girls) control subjects. ACE I/D polymorphism was analyzed by PCR using genotype allele specific primers. In ACE(I/D), we did not find significant association for the ungrouped data of nephrotic syndrome children and the control subjects. Kidney biopsies were done in 86 nephrotic syndrome cases(minimal change disease, n = 51;focal segmental glomerulosclerosis, n = 27;diffuse mesangial proliferation, n = 8). We segregated them into the minimal change disease/focal segmental glomerulosclerosis groups and observed that the ACE’D’ allele was identified with borderline significance in cases of focal segmental glomerulosclerosis and the ’Ⅰ’ allele was assessed as having very weak association in cases of minimal change disease. ’Ⅱ’ genotype was weakly associated with minimal change disease. Gender specific analysis revealed weak association of’ID’ genotype with female nephrotic syndrome in females. Dominant expression of DD genotype was observed in males with nephrotic syndrome. Our finding indicated that ACE(I/D) has moderate association with focal segmental glomerulosclerosis.
