1A comprehensive classification method for VOC emission sources to tackle air pollution based on VOC species reactivity and emission amounts显示文摘In China, volatile organic compound(VOC) control directives have been continuously released and implemented for important sources and regions to tackle air pollution. The corresponding control requirements were based on VOC emission amounts(EA), but never considered the significant differentiation of VOC species in terms of atmospheric chemical reactivity. This will adversely influence the effect of VOC reduction on air quality improvement. Therefore,this study attempted to develop a comprehensive classification method for typical VOC sources in the Beijing–Tianjin–Hebei region(BTH), by combining the VOC emission amounts with the chemical reactivities of VOC species. Firstly, we obtained the VOC chemical profiles by measuring 5 key sources in the BTH region and referencing another 10 key sources, and estimated the ozone formation potential(OFP) per ton VOC emission for these sources by using the maximum incremental reactivity(MIR) index as the characteristic of source reactivity(SR). Then, we applied the data normalization method to respectively convert EA and SR to normalized EA(NEA) and normalized SR(NSR) for various sources in the BTH region.Finally, the control index(CI) was calculated, and these sources were further classified into four grades based on the normalized CI(NCI). 5A generalized power iteration method for solving quadratic problem on the Stiefel manifold显示文摘In this paper, we first propose a novel generalized power iteration(GPI) method to solve the quadratic problem on the Stiefel manifold(QPSM) as minWTW =ITr(WTAW-2WTB) along with the theoretical analysis. 10WX20120108,a novel IAP antagonist,induces tumor cell autophagy via activating ROS-FOXO pathway显示文摘Recently,inhibitor of apoptosis proteins(IAPs)and some IAP antagonists were found to regulate autophagy,but the underlying mechanisms remain unclear.WX20120108 is an analogue of GDC-0152(a known IAP antagonist)and displays more potent anti-tumor and autophagy-regulating activity in tumor cells,we investigated the regulatory mechanisms underlying WX20120108-induced autophagy.Using molecular docking and fluorescence polarization anisotropy(FPA)competitive assay,we first demonstrated that WX20120108,acting as an IAP antagonist,bound to the XIAP-BIR3,XIAP BIR2-BIR3,cIAP1 BIR3,and cIAP2 BIR3 domains with high affinities.In six cancer cell lines,WX20120108 inhibited the cell proliferation with potencies two to ten-fold higher than that of GDC-0152.In HeLa and MDA-MB-231 cells,WX20120108 induced caspase-dependent apoptosis and activated TNFα-dependent extrinsic apoptosis.On the other hand,WX20120108 induced autophagy in HeLa and MDA-MB-231 cells in dose-and time-dependent manners.We revealed that WX20120108 selectively activated Foxo3,evidenced by Foxo3 nuclear translocation in both gene modified cell line and HeLa cells,as well as the upregulated expression of Foxo3-targeted genes(Bnip3,Pik3c3,Atg5,and Atg4b),which played a key role in autophagy initiation.WX20120108-induced autophagy was significantly suppressed when Foxo3 gene was silenced.WX20120108 dose-dependently increased the generation of reactive oxygen species(ROS)in HeLa cells,and WX20120108-induced Foxo3 activation was completely blocked in the presence of catalase,a known ROS scavenger.However,WX20120108-induced ROS generation was not affected by cIAP1/2 or XIAP gene silencing.In conclusion,WX20120108-induced autophagy relies on activating ROS-Foxo3 pathway,which is independent of IAPs.This finding provides a new insight into the mechanism of IAP antagonist-mediated regulation of autophagy.Rui Ding Xin Wang Wei Chen Zhi Li Ai-li Wei Qing-bin Wang Ai-hua Nie Li-li Wang 2019Acta Pharmacologica Sinica2019,40,11:4
11Research on variety characteristics of China's Mainland troposphere based on CMONOC显示文摘In this research, we processed the GPS and meteorological data from about 220 stations of Crustal Movement Observation Network of China(CMONOC) observed in 2014 and derived the Zenith Total Delay(ZTD) map in both spatial and temporal dimension. The results of ZTD have high accurate and reliable as IGS and all sites with varying locations show obvious variety characteristics of Chinese mainland. Meanwhile, the precipitable water vapor(PWV) correlation coefficients between GPS observation and upper air sounding is close to 1, and the comparison of GPS-derived PWV and observed PWV from meteorological sites indicating GPS observation data generated in CMONOC project applied to the weather forecast research is feasible. In addition, based on all stations covered the whole Chinese land area and using interpolation algorithms, we make contour plots of PWV distribution per hour. We observe obvious feature that the precipitable water in north and western area is less than south and east area all over this year. 12Novel approach to and implementation of design and analysis of armored face conveyor power train显示文摘An armored face conveyor(AFC) is a key piece of equipment for a fully mechanized long-wall mining system and is currently the only means for transporting bulk material in hard coal mines. To date, the AFC power train design has mainly been based on heuristics obtained via experience, coupled with simple calculations, which cannot take the dynamic behaviors and coupling effects of the components into consideration. Therefore, model-based and simulation-driven design is preferred. In this paper, a new design and analysis methodology for an AFC power train is presented to achieve the optimal dynamic characteristics and transmission performance. A preliminary design procedure for a power train is first introduced. Then, a system-level hydro-mechatronic model of the power train is built to evaluate and optimize the preliminary scheme. Sub-models, including those for the motors, fluid couplers, gearboxes, and chain, are obtained according to their individual disciplines and assembled to form the system-level model. The chain sub-system is discretized into multiple finite elements. Governing equations are established for each element based on the Newton Euler approach and assembled according to the topological structure of the chain system. In order to make the new approach applicable for engineers, a design and analysis software is developed, with a graphical user interface that involves the whole design process. MATLAB/SIMULINK is used as the computational engine, and Visual C++ is adopted to develop the interactive software framework. 13Gene cloning and induced expression pattern of IRF4 and IRF10 in the Asian swamp eel(Monopterus albus)显示文摘The Asian swamp eel(Monopterus albus) is one of the most economically important freshwater fish in East Asia,but data on the immune genes of M.albus are scarce compared to other commercially important fish.A better understanding of the eel's immune responses may help in developing strategies for disease management,potentially improving yields and mitigating losses.In mammals,interferon regulatory factors(IRFs) play a vital role in both the innate and adaptive immune system; though among teleosts IRF4 and IRF10 have seldom been studied.In this study,we characterized IRF4 and IRF10 from M.albus(maIRF4 and maIRF10) and found that maIRF4 cDNA consists of 1 716 nucleotides encoding a 451 amino acid(aa) protein,while maIRF10 consists of 1 744 nucleotides including an open reading frame(ORF) of 1 236 nt encoding 411 aa.The maIRF10 gene was constitutively expressed at high levels in a variety of tissues,while maIRF4 showed a very limited expression pattern.Expression of maIRF4 and maIRF10 in head kidney,and spleen tissues was significantly up-regulated from 12 h to 48 h post-stimulation with polyinosinic: polycytidylic acid(poly I:C),lipopolysaccharide(LPS) and a common pathogenic bacteria Aeromonas hydrophila.These results suggest that IRF4 and IRF10 play roles in immune responses to both viral and bacterial infections in M.albus.Qiao-Qing XU Dai-Qin YANG Rui TUO Jing WAN Ming-Xian CHANG Pin NIE 2014Zoological Research2014,35,5:3
14Near-field surface deformation during the April 20,2013,Ms7.0 Lushan earthquake measured by 1-Hz GNSS显示文摘The April 20,2013,Ms7.0 Lushan earthquake was successfully recorded by closely spaced Continuous Global Positioning System (CGPS) stations owned by the Crustal Movement Observation Network of China (CMONC).The 1-Hz GNSS data from eight CGPS stations,which are located between 30 km and 200 km from the hypocenter,were processed within quasi-real-time.The near-field surface deformation indicated the following characteristics:the near-field movements were limited to several centimeters;the peak of the deformation wave was significantly larger than the static permanent offset;at the beginning of the event,the north wall of the fault moved to the southeast as the south wall moved to the southwest;station SCTQ,which was the closest station to the hypocenter at 30 km,had the largest static permanent displacement of 2 cm;the peaks of the deformation waves were 1.5 cm,5 cm and 3 cm,to the east,the south and vertically upward,respectively;and the peaks of velocity and acceleration,derived from the deformation,were 3.4 cm/s and 5.3 cm/s2,respectively.Liu Gang Zhao Bin Zhang Rui Huang Yong Wang Jun Nie Zhaosheng Qiao Xuejun Tan Kai 2013Geodesy and Geodynamics2013,4,2:3
15Molecular mechanism of panaxydol on promoting axonal growth in PC12 cells显示文摘Nerve growth factor(NGF) promotes axonal growth in PC12 cells primarily by regulating the RTK-RAS-MEK-ERK pathway. Panaxydol, a polyacetylene isolated from Panax notoginseng, can mimic the effects of NGF. Panaxydol promotes neurite outgrowth in PC12 cells, but its molecular mechanism remains unclear. Indeed, although alkynol compounds such as panaxydol can increase intracellular cyclic adenosine 3′,5′-monophosphate(cAMP) levels and the ERK inhibitor U0126 inhibits alkynol-induced axonal growth, how pathways downstream of cAMP activate ERK have not been investigated. This study observed the molecular mechanism of panaxydol-, NGF-and forskolin-induced PC12 cell axon growth using specific signaling pathway inhibitors. The results demonstrated that although the RTK inhibitor SU5416 obviously inhibited the growth-promoting effect of NGF, it could not inhibit the promoting effect of panaxydol on axonal growth of PC12 cells. The adenylate cyclase inhibitor SQ22536 and cAMP-dependent protein kinase inhibitor RpcAMPS could suppress the promoting effect of forskolin and panaxydol on axonal growth. The ERK inhibitor U0126 inhibited axonal growth induced by all three factors. However, the PKA inhibitor H89 inhibited the promoting effect of forskolin on axonal growth but could not suppress the promoting effect of panaxydol. A western blot assay was used to determine the effects of stimulating factors and inhibitors on ERK phosphorylation levels. The results revealed that NGF activates the ERK pathway through tyrosine receptors to induce axonal growth of PC12 cells. In contrast, panaxydol and forskolin increased cellular cAMP levels and were inhibited by adenylyl cyclase inhibitors. The protein kinase A inhibitor H89 completely inhibited forskolin-induced axonal outgrowth and ERK phosphorylation, but could not inhibit panaxydol-induced axonal growth and ERK phosphorylation. These results indicated that panaxydol promoted axonal growth of PC12 cells through different pathways downstream of cAMP. Considering that exchange protein directly activated by cAMP 1(Epac1) plays an important role in mediating cAMP signaling pathways, RNA interference experiments targeting the Epac1 gene were employed. The results verified that Epac1 could mediate the axonal growth signaling pathway induced by panaxydol. 16Postoperative secondary aggravation of obstructive sleep apneahypopnea syndrome and hypoxemia with bilateral carotid body tumor:A case report显示文摘BACKGROUND Carotid body tumor(CBT)is a chemoreceptor tumor located in the carotid body,accounting for approximately 0.22%of head and neck tumors.Surgery is the main treatment method for the disease.CASE SUMMARY We reviewed the diagnosis and treatment of one patient who had postoperative secondary aggravation of obstructive sleep apnea–hypopnea syndrome(OSAHS)and hypoxia after surgical resection of bilateral CBTs.This patient was admitted,and relevant laboratory and imaging examinations,and polysomnography(PSG)were performed.After the definitive diagnosis,continuous positive airway pressure(CPAP)treatment was given,which achieved good efficacy.CONCLUSION This case suggested that aggravation of OSAHS and hypoxemia is possibly caused by the postoperative complications after bilateral CBTs,and diagnosis by PSG and CPAP treatment are helpful for this patient.Xi Yang Xiao-Guang He Dong-Hui Jiang Chun Feng Rui Nie 2020World Journal of Clinical Cases2020,8,23:2
17Gonadotropin treatment for male partial congenital hypogonadotropic hypogonadism in Chinese patients显示文摘Partial congenital hypogonadotropic hypogonadism(PCHH)is caused by an insufficiency in,but not a complete lack of,gonadotropin secretion.This leads to reduced testosterone production,mild testicular enlargement,and partial pubertal development.No studies have shown the productivity of spermatogenesis in patients with PCHH.We compared the outcomes of gonadotropin-induced spermatogenesis between patients with PCHH and those with complete congenital hypogonadotropic hypogonadism(CCHH).This retrospective study included 587 patients with CHH who were treated in Peking Union Medical College Hospital(Beijing,China)from January 2008 to September 2016.A total of 465 cases were excluded from data analysis for testosterone or gonadotropin-releasing hormone treatment,cryptorchidism,poor compliance,or incomplete medical data.We defined male patients with PCHH as those with a testicular volume of≥4 ml and patients with a testicular volume of<4 ml as CCHH.A total of 122 compliant,noncryptorchid patients with PCHH or CCHH received combined human chorionic gonadotropin and human menopausal gonadotropin and were monitored for 24 months.Testicular size,serum luteinizing hormone levels,follicle-stimulating hormone levels,serum total testosterone levels,and sperm count were recorded at each visit.After gonadotropin therapy,patients with PCHH had a higher spermatogenesis rate(92.3%)than did patients with CCHH(74.7%).During 24-month combined gonadotropin treatment,the PCHH group took significantly less time to begin producing sperm compared with the CCHH group(median time:11.7 vs 17.8 months,P<0.05).In conclusion,after combined gonadotropin treatment,patients with PCHH have a higher spermatogenesis success rate and sperm concentrations and require shorter treatment periods for sperm production.Ming Hao Min Nie Bing-Qing Yu Yin-Jie Gao Xi Wang Wan-Lu Ma Qi-Bin Huang Rui Zhang Jiang-Feng Mao Xue-Yan Wu 2020Asian Journal of Andrology2020,22,4:2

19FX5,a non-steroidal glucocorticoid receptor antagonist,ameliorates diabetic cognitive impairment in mice显示文摘Diabetic cognitive impairment(DCI)is a common diabetic complication characterized by learning and memory deficits.In diabetic patients,hyperactivated hypothalamic-pituitary-adrenal(HPA)axis leads to abnormal increase of glucocorticoids(GCs),which causes the damage of hippocampal neurons and cognitive impairment.In this study we investigated the cognition-improving effects of a non-steroidal glucocorticoid receptor(GR)antagonist 5-chloro-N-[4-chloro-3-(trifluoromethyl)phenyl]thiophene-2-sulfonamide(FX5)in diabetic mice.Four weeks after T1DM or T2DM was induced,the mice were administered FX5(20,40 mg·kg^(-1)·d^(-1),i.g.)for 8 weeks.Cognitive impairment was assessed in open field test,novel object recognition test,Y-maze test,and Morris water maze test.We showed that FX5 administration significantly ameliorated the cognitive impairments in both type 1 and 2 diabetic mice.Similar cognitive improvement was observed in diabetic mice following brain GR-specific knockdown by injecting AAV-si-GR.Moreover,AAV-si-GR injection occluded the cognition-improving effects of FX5,suggesting that FX5 functioning as a non-steroidal GR antagonist.In PA-treated primary neurons(as DCI model in vitro),we demonstrated that FX5(2,5,10μM)dose-dependently ameliorated synaptic impairment via upregulating GR/BDNF/TrkB/CREB pathway,protected against neuronal apoptosis through repressing GR/PI3K/AKT/GSK3β-mediated tauopathy and subsequent endoplasmic reticulum stress.In LPS-treated primary microglia,FX5 dose-dependently inhibited inflammation through GR/NF-κB/NLRP3/ASC/Caspase-1 pathway.These beneficial effects were also observed in the hippocampus of diabetic mice following FX5 administration.Collectively,we have elucidated the mechanisms underlying the beneficial effects of non-steroidal GR antagonist FX5 on DCI and highlighted the potential of FX5 in the treatment of the disease.Dan-yang Zhu Jian Lu Rui Xu Juan-zhen Yang Xiang-rui Meng Xing-nan Ou-Yang Qiu-ying Yan Rui-fang Nie Tong Zhao Yi-di Chen Yin Lu Yi-nan Zhang Wen-jun Li Xu Shen 2022Acta Pharmacologica Sinica2022,43,10:2
