| 1 | 不同沙棘籽提取物的抗氧化和抗菌活性研究显示文摘使用Soxhlet提取装置,用氯仿、乙酸乙酯、丙酮、甲醇4种不同溶剂对沙棘籽分别进行8小时萃取。粗提物被用来进行抗氧化和抗菌活性筛选。在离体模型(清除DPPH自由基和脂质体模型系统)中的粉末减少和抗氧化活性评价显示在各种提取物中使用甲醇作为萃取剂的提取物具有最高的抗氧化活性。甲醇提取物也具有最高的抗菌活性。最低抑制浓度(MIC)评价显示,在加入甲醇提取物对蜡状芽孢杆菌、凝结芽孢杆菌、枯草芽孢杆菌、单核细胞增生利斯特菌、小肠结肠炎耶尔森氏菌进行培养,MIC浓度分别为200,300,300,300,和350ppm。研究结果表明,沙棘种子应用于医药和食品储藏。 | P. | 2 | Over expression of resistin in adipose tissue of the obese induces insulin resistance显示文摘AIM: To compare resistin mRNA expression in subcutaneous adipose tissue (SAT) and its correlation with insulin resistance (IR) in postmenopausal obese women. METHODS: A total of 68 postmenopausal women (non obese = 34 and obese = 34) were enrolled for the study. The women of the two groups were age matched (49-70 years). Fasting blood samples were collected at admission and abdominal SAT was obtained during surgery for gall bladder stones or hysterectomy. Physical parameters [age, height, weight, body mass index (BMI)] were measured. Biochemical (plasma insulin and plasma glucose) parameters were estimated by enzymatic methods. RNA was isolated by the Trizol method.SAT resistin mRNA expression was done by real time- reverse transcription polymerase chain reaction (RT-PCR) by using Quanti Tect SYBR Green RT-PCR master mix. Data was analyzed using independent Student's t test, correlation and simple linear regression analysis. RESULTS: The mean weight (52.81 ± 8.04 kg vs 79.56 ± 9.91 kg; P < 0.001), BMI (20.23 ± 3.05 kg/m 2 vs 32.19 ± 4.86 kg/m 2 ; P < 0.001), insulin (8.47 ± 3.24 U/mL vs 14.67 ± 2.18 U/mL; P < 0.001), glucose (97.44 ± 11.31 mg/dL vs 109.67 ± 8.02 mg/dL; P < 0.001) and homeostasis model assessment index (2.01 ± 0.73 vs 3.96 ± 0.61; P < 0.001) were significantly higher in postmenopausal obese women compared to postmenopausal non obese women. The mean serum resistin level was also significantly higher in postmeno-pausal obese women compared to postmenopausal non obese women (9.05 ± 5.15 vs 13.92 ± 6.32, P < 0.001). Furthermore, the mean SAT resistin mRNA expression was also significantly (0.023 ± 0.008 vs 0.036 ± 0.009; P < 0.001) higher and over expressed 1.62 fold (upregulated) in postmenopausal obese women compared to postmenopausal non obese women. In postmeno-pausal obese women, the relative SAT resistin mRNA expression showed positive (direct) and significant correlation with BMI (r = 0.78, P < 0.001) and serum resistin (r = 0.76, P < 0.001). Furthermore, the SAT resistin mRNA expression in postmenopausal obese women also showed significant and direct association (r = 0.45, P < 0.01) with IR, while in postmenopausal non obese women it did not show any association (r = -0.04, P > 0.05). | 3 | Carbon stock estimation for tree species of Sem Mukhem sacred forest in Garhwal Himalaya, India显示文摘Carbon stock estimation was conducted in tree species of Sem Mukhem sacred forest in district Tehri of Garhwal Himalaya, Uttarakhand, India. This forest is dedicated to Nagraj Devta and is dominated by tree species, including Quercus floribunda, Quercus semecarpifolia and Rhododendron arboreum. The highest values of below ground biomass density, total biomass density and total carbon density were (34.81±1.68) Mg·ha-1, (168.26±9.04) Mg·ha-1 and (84.13±4.18) Mg·ha-1 for Pinus wallichiana. Overall values of total biomass density and total carbon density calculated were 1549.704 Mg·ha-1 and 774.77 Mg·ha-1 respectively. Total value of growing stock volume density for all species was 732.56 m3·ha-1 and ranged from (144.97±11.98) m3·ha-1 for Pinus wallichiana to (7.78±1.78) m3·ha-1 for Benthamidia capitata. | Nazir A. | 4 | Design and development of novel bioadhesive niosomal formulation for the transcorneal delivery of anti-infective agent:In-vitro and ex-vivo investigations显示文摘Gatifloxacin eye drops are frequently used in eye infections.However such formulations have a major drawback i.e.short duration of action and usually require 4e6 times installations daily.A chitosan coated niosomal formulation of gatifloxain was purposed to show a longer retention time on eyes and subsequent reduction in dosing frequency.Vesicles were prepared by solvent injection method using cholesterol and Span-60.An extensive optimization of formulation was done using different ratios of cholesterol,Span-60 and drug,revealed NS60-5(cholesterol:span-6050:50 and drug content of 20 mg)to be the optimized niosome formulation.NS60-5 had shown a highest entrapment efficiency of 64.9±0.66%with particle size 213.2±1.5 nm and zeta potential34.7±2.2 mV.Optimized niosomes were also coated with different concentrations of chitosan and evaluated.Permeation studies had revealed that optimized niosomes(86.77±1.31%)had increased the transcorneal permeation of Gatifloxacin more than two fold than simple drug solution(37.19±1.1%).Longer retention potential of the coated niosomes was further verified by fluorescence microscopy.Study revealed that simple dye solution got easily washed out with in 6 h.The uncoated niosomes(NS60-5)showed a longer retention(more than 6 h),which was further enhanced in case of coated niosomes i.e.CNS60-1(more than 12 h).Antimicrobial studies had shown the better efficacy of CNS60-1(zone of inhibition)when compared to marketed formulation.The final chitosan formulation was found to have shown better ocular tolerability as demonstrated by corneal hydration test histopathology investigations. | Yahaya Zubairu Lalit Mohan Negi Zeenat Iqbal Sushama Talegaonkar | 2015 | Asian Journal of Pharmaceutical Sciences2015,10,4: | 4 |

| 8 | CEACAM-1 Induced CSF3-receptor Downregulation in Bone Marrow Associated With Refractory Neutropenia in Advanced Cirrhosis显示文摘Background and Aims:Cirrhosis patients exhibit cyto-penia,and,at times refractory neutropenia to granulocyte colony-stimulating factor(G-CSF),which acts through the CSF3-receptor(CSF3R),and changes in CSF3R can affect the response.We conducted this study to assess the CSF3R status and its relevance in cirrhotic patients.Methods:Cirrhotic patients(n=127)and controls(n=26)with clini-cally indicated bone marrow(BM)examination were stud-ied.BM assessment was done by qRT-PCR and immunohis-tochemistry(IHC)for CSF3R.Circulating G-CSF,CSF3R,and carcinoembryonic antigen cell adhesion molecule-1(CEACAM1)were measured.BM hematopoietic precursor cells and their alterations were examined by flow cytom-etry.The findings were validated in liver cirrhosis patients who received G-CSF for severe neutropenia.Results:The mean age was 48.6±13.4 years,and 80.3%were men.Circulatory CSF3R reduction was noted with the advance-ment of cirrhosis,and confirmed by qRT-PCR and IHC in BM.CSF3R decline was related to decreased hematopoietic stem cells(HSCs)and downregulation of CSF3R in the re-maining HSCs.Cocultures confirmed that CEACAM1 led to CSF3R downregulation in BM cells by possible lysosomal degradation.Baseline low peripheral blood-(PB)-CSF3R also predisposed development of infections on follow-up.Decreased CSF3R was also associated with nonresponse to exogenous G-CSF treatment of neutropenia.Conclu-sions:Advanced liver cirrhosis was associated with low CSF3R and high CEACAM1 levels in the BM and circula-tion,making patients prone to infection and inadequate response to exogenous G-CSF. | Chhagan Bihari Sukriti Baweja Seggere Murlaikrishna Shasthry Deepika Lal Preeti Negi Swati Thangariyal Dinesh Mani Tripathi Shiv Kumar Sarin | 2022 | Journal of Clinical and Translational Hepatology2022,10,1: | 2 |
