Characterization and assessment of volatile organic compounds (VOCs) emissions from typical industries显示文摘VOCs play an increasingly important role in affecting air quality and threatening human health in China in recent years, where industry activities show a significant contribution to VOCs emission. In this article we report our long term study of industrial VOCs emissions of six major industries (vehicle manufacturing, printing, equipment coating, electronic manufacturing, furniture manufacturing and bio-pharming) on the aspects of emission characteristics, environmental impact and health risk assessment, and control challenge analysis with the purpose to obtain in-depth understanding of industry VOCs emissions and offer some original basements for national control and management of industry VOCs emissions. This study shows that all these industries give middle or low emission with total VOCs concentration less than 1000 mg/m3 at each exhaust pipe.Benzenes, esters, alcohols, ketones, alkanes, chloroalkanes and alkenes were detected as the major emission components and the most frequently monitored VOCs were benzenes, which varied obviously with different processes and industries. The environmental impact assessments indicate that vehicle manufacturing and benzenes should be prior controlled with the purpose to reduce air pollution. While, health risk assessments suggest that furniture manufacturing and chloroalkanes should be firstly controlled.Control analysis indicates that developing technologies with low cost and high efficiency and establishing and completing specific industry emission standards/regulations are the two key issues in VOCs emission management at present stage.WANG HaiLin NIE Lie LI Jing WANG YuFei WANG Gang WANG JunHui HAO ZhengPing 2013Science China(Technological Sciences)2013,56,3:60
Polymorphisms in PPARD,PPARG and APM1 associated with four types of Traditional Chinese Medicine constitutions显示文摘Based on the theory of constitution of Traditional Chinese Medicine (TCM), the human population is divided into nine constitutions including one balanced constitution (Normality) and eight unbalanced constitutions (Yang-deficiency, Yin-deficiency, Phlegm-wetness, Qi-deficiency, Wetness-heat, Blood stasis, Depressed constitution, and Inherited special constitution). Different constitutions have specific metabolic features and different susceptibility to certain diseases. However, whether a genetic basis accounts for such constitution classifica-tion is yet to be determined. Here we performed a genetic study to assess the association between genetic variations of metabolic genes in-cluding PPARD, PPARG and APM1 and the constitutions. A total of 233 individuals of the Han population in China were classified into four groups, Normality, Yang-deficiency, Yin-deficiency and Phlegm-wetness with whom 23 single nucleotide polymorphisms (SNPs) in the three genes were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Biased distri-bution of PPARD rs2267669 and rs2076167, APM1 rs7627128 and rs1063539 in Yang-deficiency, PPARG Pro12Ala in Yin-deficiency and PPARD rs2076167, APM1 rs266729 and rs7627128 in Phlegm-wetness were observed. The frequencies of Haplotype13 (Hap13) of PPARG in Yin-deficiency, Hap25 of APM1 in Yang-deficiency and Hap2 of PPARD and Hap14 of PPARG in Phlegm-wetness, were significantly different from those in Normality, suggesting those might be group-associated haplotypes. These results suggested that single SNP and hap-lotypes of PPARD, PPARG and APM1 may underlie the genetic basis of the constitutions classified in TCM.Yanrui Wu Yina Cun Jing Dong Jingru Shao Shengjun Luo Shengjie Nie Haijing Yu Bingrong Zheng Qi Wang Chunjie Xiao 2010Journal of Genetics and Genomics2010,37,6:36

CPSF30-L-mediated recognition of mRNA m^(6)A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis显示文摘N6-methyladenosine(m^(6)A),a ubiquitous internal modification of eukaryotic mRNAs,plays a vital role in almost every aspect of mRNA metabolism. However, there is little evidence documenting the role of m^(6)A in regulating alternative polyadenylation(APA)in plants.APA is controlled by a large protein-RNA complex with many components, including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30(CPSF30).In Arabidopsis,CPSF30 has two isoforms and the longer isoform(CPSF30-L)contains a YT512-B Homology(YTH)domain, which is unique to plants. In this study,we showed that CPSF30-L YTH domain binds to m^(6)A in v itro.In the cpsf30-2 mutant, the transcripts of many genes including several important nitrate signaling-related genes had shifts in polyadenylation sites that were correlated with m^(6)A peaks, indicating that these gene transcripts carrying m^(6)A tend to be regulated by APA.Wild-type CPSF30-L could rescue the defects in APA and nitrate metabolism in cpsf30-2,but m^(6)A-binding-defective mutants of CPSF30-L could not.Taken together, our results demonstrated that m^(6)A modification regulates APA in Arabidops is and revealed that the m^(6)A reader CPSF30-L affects nitrate signaling by controlling APA,shedding new light on the roles of the m^(6)A modification during RNA 3-end processing in nitrate metabolism.Yiteng Hou Jing Sun Baixing Wu Yangyang Gao Hongbo Nie Zhentian Nie Shuxuan Quan Yong Wang Xiaofeng Cao Sisi Li 2021Molecular Plant2021,14,4:12
Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. Therefore the observation of Insight-HXMT constitutes an important chapter in the full context of multi-wavelength and multi-messenger observation of this historical GW event.TiPei Li ShaoLin Xiong ShuangNan Zhang FangJun Lu LiMing Song XueLei Cao Zhi Chang Gang Chen Li Chen TianXiang Chen Yong Chen YiBao Chen YuPeng Chen Wei Cui WeiWei Cui JingKang Deng YongWei Dong YuanYuan Du MinXue Fu GuanHua Gao He Gao Min Gao MingYu Ge YuDong Gu Ju Guan ChengCheng Guo DaWei Han Wei Hu Yue Huang Jia Huo ShuMei Jia LuHua Jiang WeiChun Jiang Jing Jin YongJie Jin Bing Li ChengKui Li Gang Li MaoShun Li Wei Li Xian Li XiaoBo Li XuFang Li YanGuo Li ZiJian Li ZhengWei Li XiaoHua Liang JinYuan Liao CongZhan Liu GuoQing Liu HongWei Liu ShaoZhen Liu XiaoJing Liu Yuan Liu YiNong Liu Bo Lu XueFeng Lu Tao Luo Xiang Ma Bin Meng Yi Nang JianYin Nie Ge OU JinLu Qu Na Sai Liang Sun Yin Tan Lian Tao WenHui Tao YouLi Tuo GuoFeng Wang HuanYu Wang Juan Wang WenShuai Wang YuSa Wang XiangYang Wen BoBing WU Mei Wu GuangCheng Xiao He Xu YuPeng Xu LinLi Yan JiaWei Yang Sheng Yang YanJi Yang AiMei Zhang ChunLei Zhang ChengMo Zhang Fan Zhang HongMei Zhang Juan Zhang Qiang Zhang Shu Zhang Tong Zhang Wei Zhang WanChang Zhang WenZhao Zhang Yi Zhang Yue Zhang YiFei Zhang YongJie Zhang Zhao Zhang ZiLiang Zhang HaiSheng Zhao JianLing Zhao XiaoFan Zhao ShiJie Zheng Yue Zhu YuXuan Zhu ChangLin Zou 2018Science China(Physics,Mechanics & Astronomy)2018,61,3:12
Emergency material scheduling optimization model and algorithms:A review显示文摘In the emergency management of disruptions, efficient emergency material scheduling(EMS) is a key factor to save people’s lives and reduce loss.Based on the literature of EMS and related areas in recent years, the research was summarized from two aspects of EMS optimization model and algorithms. It is concluded that the EMS optimization models mainly aim at the shortest time, shortest distance, minimum cost, maximum satisfaction and fairness, etc. The constraints usually include the quantity of supply depots, relief supply and vehicles, the types of commodities, the road network conditions, the budgets and the demand forecast of emergency materials.Multi-objective model is more complex and it usually considers more than one objective. To find the optimized solution, the multiobjective model with complex constraints needs more efficient algorithms. The existing algorithms, including mathematic algorithm and heuristic algorithm, have been categorized. For NP-hard(non-deterministic polynomial hard) problems, heuristic algorithms should be designed, which mainly include genetic algorithm(GA),ant colony optimization(ACO),particle swarm optimization(PSO),etc.Based on the characteristics of the optimization model and various algorithms, appropriate algorithm or tools should be chosen and designed to obtain the optimized solution of EMS model. Finally, the development trends of EMS optimization model and algorithm in the future are proposed.Hui Hu Jing He Xiongfei He Wanli Yang Jing Nie Bin Ran 2019Journal of Traffic and Transportation Engineering(English Edition)2019,6,5:11

Effects of permissive hypocaloric vs standard enteral feeding on gastrointestinal function and outcomes in sepsis显示文摘BACKGROUND Intestinal mucosal barrier injury and gastrointestinal dysfunction are important causes of sepsis. However, few studies have investigated the effects of enteral underfeeding on gastrointestinal function in sepsis. Moreover,no consensus on goal enteral caloric intake has been reached in sepsis.AIM To investigate the effects of different goal caloric requirements of enteral nutrition on the gastrointestinal function and outcomes in the acute phase of sepsis.METHODS Patients were randomly assigned to receive 30%(defined as group A),60%(group B),or 100%(group C)of goal caloric requirements of enteral nutrition in this prospective pilot clinical trial. The acute gastrointestinal injury(AGI)grades, incidence of feeding intolerance(FI),daily caloric intake, nutritional and inflammatory markers, and biomarkers of mucosal barrier function were collected during the first 7 d of enteral feeding. The clinical severity and outcome variables were also recorded.RESULTS A total of 54 septic patients were enrolled. The days to goal calorie of group C(2.55±0.82)were significantly longer than those of group A(3.50±1.51;P=0.046)or B(4.85±1.68;P<0.001).The FI incidence of group C(16.5%)was higher than that of group A(5.0%)or B(8.7%)(P=0.009).No difference in the incidence of FI symptoms was found between groups A and B.The serum levels of barrier function biomarkers of group B were significantly lower than those of group A(P<0.05)on the 7th day of feeding. The prealbumin and IL-6 levels of group A were lower than those of group B(P<0.05)on the 7th day of feeding. No significant differences in the clinical outcome variables or 28-d mortality were found among the three groups.CONCLUSION Early moderate enteral underfeeding(60%of goal requirements)could improve the intestinal barrier function and nutritional and inflammatory status without increasing the incidence of FI symptoms in sepsis. However, further large-scale prospective clinical trials and animal studies are required to test our findings. Moreover, the effects of different protein intake on gastrointestinal function and outcomes should also be investigated in future work.Jia-Kui Sun Shuai Nie Yong-Ming Chen Jing Zhou Xiang Wang Su-Ming Zhou Xin-Wei Mu 2021World Journal of Gastroenterology2021,27,29:9
DNA methylation-mediated repression of miR-181a/135a/302c expression promotes the microsatellite-unstable colorectal cancer development and 5-FU resistance via targeting PLAG1显示文摘Micro sate Hite instability(MSI) defines a subtype of colorectal cancer(CRC) with typical clinicopathologic characteristics. CRCs with MSI(MSI CRCs) frequently acquire accelerated carcinogenesis and 5-FU resistance, and the exact underlying mechanism remains incompletely understood. Our previous study has identified the microRNA(miRNA) expression profile in MSI CRCs. In this study, three miRNAs(miR-181 a, miR-135 a and miR-302 c) were validated by qRT-PCR to be dramatically decreased in 67 CRC samples. Proliferation and apoptosis assays demonstrated that miR-181 a/135 a/302 c function as tumor suppressors via repressing PLAG1/IGF2 signaling. Moreover, we presented compelling evidence that restoration of miR-181 a/135 a/302 c expression promoted sensitivity of MSI CRC cells to 5-FU treatment. miR-181 a/135 a/302 c exerted their effect on chemoresistance through attenuating PLAG1 expression. Notably, the hypermethylation status of MSI CRC accounts for the decrements of miR-181 a/135 a/302 c. Our results contribute to a better understanding of the mechanism of chemo?resistance in MSI CRCs, and provide a clue for digging the bio markers and therapeutic targets for CRC patients.Lu Shi Xiang Li Zhiqiang Wu Xiaolei Li Jing Nie Mingzhou Guo Qian Mei Weidong Han 2018Journal of Genetics and Genomics2018,45,4:9
Simultaneous Determination of Plant Growth Regulators in Fruits Using a Modified Qu ECh ERS Procedure and UPLC–MS/MS显示文摘Six plant growth regulators frequently used in fruit were detected using UPLC–MS/MS including chlormequat chloride, GA3, forchlorfenuron, paclobutrazol, thidiazuron and 2,4-D. A modified Qu ECh ERS method was used in pretreatment: each compound was extracted with acetonitrile containing 1% acetic acid and cleaned with a mixture of ostade-cylsilane(ODS) and Mg SO_4. The detection was performed using electrospray ionization(ESI) and multiple reaction monitoring mode(MRM) and quantified using the external standard method. Results indicated that the calibration curves of six plant growth regulators were linear within the range of 1–100 μg·kg^(-1) with correlation coefficients over 0.999. The recovery at four spiked concentrations, 1, 20, 50, and 100 μg·kg^(-1), ranged from 60.77% to 119.43% and relative standard deviation(RSD) were between 0.18% and 32.94%. The limits of detection(LOD) and limits of quantitation(LOQ) were 0.01–0.14 μg·kg^(-1) and 0.02–0.46 μg·kg^(-1), respectively, for apples, 0.01–0.18 μg·kg^(-1) and 0.03–0.59 μg·kg^(-1) for grapes, 0.01–0.08 μg·kg^(-1) and 0.04–0.28 μg·kg^(-1) for kiwis, 0.003–0.110 μg·kg^(-1) and 0.01–0.38 μg·kg^(-1) for peaches, and 0.01–0.08 μg·kg^(-1) and 0.03–0.25 μg·kg^(-1) for oranges.YAN Zhen NIE Jiyun XU Guofeng LI Haifei LI Jing LI Zhixia WU Yonglong KUANG Lixue 2016Horticultural Plant Journal2016,2,4:8

CIVB flashback analysis of hydrogen flame based on azimuthal vorticity at mixing zone exit显示文摘Swirl-premixed combustion systems exhibit potential to meet future regulations on pollution emissions. However, combustion induced vortex breakdown(CIVB) flashback is frequently observed in these systems, especially for high hydrogen content fuel. In this study, a swirl-premixed burner with diverging centerbody was used to investigate CIVB flashback based on azimuthal vorticity at mixing zone exit. Through 2D axisymmetric model, it was found that there was a maximal azimuthal vorticity at mixing zone exit for each equivalence ratio. The physical meaning of these maximal azimuthal vorticity values was the minimally required azimuthal vorticity to trigger CIVB flashback. At the same time, the required azimuthal vorticity declined with the increase of equivalence ratio since turbulent burning velocity started to control flashback. Nevertheless, azimuthal vorticity offered by heat release increased with the increase of equivalence ratio, which promoted flame propagating upstream continually.TIAN Xiao Jing XING Shuang Xi CUI Yu Feng FANG Ai Bing NIE Chao Qun 2014Science China(Technological Sciences)2014,57,12:7
Phase I study of CAR-T cells with PD-1 and TCR disruption in mesothelin-positive solid tumors显示文摘Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid tumors. We generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior lymphodepletion. No dose-limiting toxicity or unexpected adverse events were observed in any of the 15 patients. The best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 month.TCR-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after infusion. We further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal models. Our results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors.Zhenguang Wang Na Li Kaichao Feng Meixia Chen Yan Zhang Yang Liu Qingming Yang Jing Nie Na Tang Xingying Zhang Chen Cheng Lianjun Shen Jiaping He Xun Ye Wei Cao Haoyi Wang Weidong Han 2021Cellular & Molecular Immunology2021,18,9:7
Nusap1 is essential for neural crest cell migration in zebrafish显示文摘Microtubules play important roles in mitotic spindle assembly and chromosome segregation to maintain normal cell cycle progression.A number of microtubule-associated proteins have been identified in epithelial and neural cell cultures;however, their physiological significance is not well characterized due to the lack of appropriate in vivo animal models.Nucleolar spindleassociated protein(NuSAP)is a microtubule-binding protein and is reported to be involved in mitosis by cell culture studies. In this report,we identified the zebrafish homologue of human NuSAP and investigated its expression profile and functions.Using in situ hybridization,we demonstrated that transcripts of zebrafish nusap1 are specifically expressed in the retina, forebrain, hindbrain and neural crest. When the in vivo expression of nusap1 was knocked down through antisense oligonucleotide morpholino technology, the morphants of nusap1 showed impaired morphogenesis in the trunk and yolk extension, implying the involvement of Nusap1 in cell migration.Mechanistic studies revealed that nusap1 morphants have an altered expression pattern of neural crest markers crestin and sox9b,but normal expression of blood vessel and notochord markers gata1 and shh.In addition,nusap1 mRNA injection caused serious apoptosis in retina and hindbrain tissue, and these phenotypes can be rescued by co-injection of morpholino against nusap1.These observations not only suggest a role for Nusap1 in connecting apoptosis with cell migration, but also provide strong evidences that Nusap1 is potentially involved in morphogenesis in vertebrates.Jing Nie Hua Wang Fuchu He Huizhe Huang 2010Protein & Cell2010,1,3:7
Health risk assessment of dietary exposure to polycyclic aromatic hydrocarbons in Taiyuan, China显示文摘Sixteen polycyclic aromatic hydrocarbons(PAHs) were determined in 24 duplicate-diet samples from people in Taiyuan during summer and winter in 2009. Dietary intake of PAHs for 2862 participants was subsequently estimated by a survey in Taiyuan. Results from these 24 samples were compared with a raw food study in Taiyuan in 2008. Three main sources of dietary PAHs are vegetables, wheat flour and fruits, the sum of which contributes 75.95% of PAHs in dietary food. Compared to the estimated value in raw food, much more B[a]Peq(benzo[a]pyrene equivalents) were detected in food samples collected in the duplicate-diet study in Taiyuan(60.75 ng/day). The cooking process may introduce more B[a]Peq into food, and the relative contribution of 16 PAHs in the diet would be changed during the cooking procedure.Jing Nie Jing Shi Xiaoli Duan Beibei Wang Nan Huang Xiuge Zhao 2014Journal of Environmental Sciences2014,26,2:6
Risk Factors for Strangulated Ovarian Hernia in Female Infants:the Role of Ovarian Volume显示文摘The risk factors associated with strangulated ovarian hernia (SOH)in female patients (<1 year old)were identified.A retrospective analysis was conducted regarding the data from 2006 to 2017.The patients were divided into 2groups:SOH group (n=9)and non-SOH group (n=23). Patient demographics, clinical signs, preoperative examinations and intraoperative findings were compared between the two groups, and risk factors for SOH were tested using a binary logistic regression model. To explore whether greater ovary was more likely to be twisted, leading to SOH, all the patients were divided into ovary volume <5cm^3 and ≥5cm^3 groups and the association between ovarian volume and ovary torsion was assessed.Among a total of 32 female patients (<1year old)with incarcerated ovarian herniation,9 patients developed SOH.The single variate analysis revealed that times of manual reduction, ovarian volume, ovary with or without multiple cysts, ovary torsion or not and angle of ovary torsion were found to be significant factors associated with SOH.The multivariate analysis showed ovarian volume was evidenced as an independent risk factor for SOH.Furthermore, the incidence of ovary torsion was significantly higher in ovarian volume ≥5cm^3 group than in ovarian volume <5cm^3 group, indicating that larger ovary was more likely to result in ovary torsion, leading to SOH.Our study demonstrated that the odds of SOH increased with increasing ovarian volume in female patients (<1 year old)because the relatively greater ovary at this age was more likely to be incarcerated and twisted, leading to SOH.Yu CHEN Xiang-zhi PENG Wei LU Kai ZHENG Jian GUO Hua NIE Xiao-jie SONG Yan ZHANG Jing YANG 2018Current Medical Science2018,38,6:6
Micro RNAs: Novel immunotherapeutic targets in colorectal carcinoma显示文摘Colorectal carcinoma(CRC) is one of the most common types of cancer worldwide and the prognosis for CRC patients with recurrence or metastasis is extremely poor. Although chemotherapy and radiation therapy can improve survival, there are still numerous efforts to be performed. Immunotherapy is frequently used, either alone or in combination with other therapies, for the treatment of CRC and is a safe and feasible way to improve CRC treatment. Furthermore, the significance of the immune system in the biology of CRC has been demonstrated by retrospective assessments of immune infiltrates in resected CRC tumors. Micro RNAs(mi RNAs) are short, non-coding RNAs that can regulate multiple target genes at the post-transcriptional level and play critical roles in cell proliferation, differentiation and apoptosis. Mi RNAs are required for normal immune system development and function. Nevertheless, aberrant expression of mi RNAs is often observed in various tumor types and leads to immune disorders or immune evasion. The immunomodulatory function of mi RNAs indicates that mi RNAs may ultimately be part of the portfolio of anti-cancer targets. Herein, we will review the potential roles of mi RNAs in the regulation of the immune response in CRC and then move on to discuss how to utilize different mi RNA targets to treat CRC. We also provide an overview of the major limitations and challenges of using mi RNAs as immunotherapeutic targets.Xiang Li Jing Nie Qian Mei Wei-Dong Han 2016World Journal of Gastroenterology2016,22,23:6
Expression and effect of TLR4 in rats with diabetic nephropathy显示文摘Objective: To observe the expression of TLR4 in kidney tissue of rats with diabetic nephropathy and discuss the role of TLR4 in the occurrence and development of the diabetic nephropathy. Methods: A total of 60 clean male SD rats were selected and randomly divided into the modeling group and control group after 1 week of breeding, including 30 rats in each group. Biochemical indices as well as the protein expression of TLR4 were observed and compared between two groups at 2 w, 4 w, 6 w, 8 w and 12 w after the modeling, and the correlation between TLR4 and each biochemical indexes was analyzed. Results: Rats in the modeling group had higher levels of blood glucose, 24-hour urine protein and blood urea nitrogen after the modeling, and showed the increase in the serum creatinine, kidney/body weight ratio, CRP and serum TNF-毩 at 4w after the modeling, with the significant difference compared to results of the control group ( P <0.05). The cross-section area and mean volume of glomerulus in the modeling group at 4 w, 6 w, 8 w and 12 w were significantly higher than those in the control group, with the statistically significant difference ( P <0.05). The expression of TLR4 at each time point in the control group was relatively low. Rats in the modeling group had the high expression of TLR4 in kidney's glomerular basement membrane, proximal convoluted tubule and renal interstitial area since 2 w, with the significant difference compared to the control group ( P <0.05). The expression in rats of the modeling group was higher than the one of the control group since the 2nd week. As the time flied, its expression increased, with the statistically significant difference between two groups ( P <0.05). There was certain correlation between the protein expression of TLR4 and the increased serum titer of 24hour urine protein excretion, serum creatinine, CRP and TNF-毩. Conclusions: TLR4 may activate the immuno-inflammatory reactions to play a role in the occurrence and development of the diabetic nephropathy.Si-Yang Liu Xiang-Zhi Nie Wen-Yang Zhou Jing Chen 2013Asian Pacific Journal of Tropical Medicine2013,6,8:6
In vitro and in vivo activity of D-serine in combination with β-lactam antibiotics against methicillin-resistant Staphylococcus aureus显示文摘As D-amino acids play important roles in the physiological metabolism of bacteria, combination of D-amino acids with antibiotics may provide synergistic antibacterial activity. The aim of the study was to evaluate in vitro and in vivo activity of D-serine alone and in combination withβ-lactams against methicillin-resistant Staphylococcus aureus(MRSA) strains, and to explore the possible sensitization mechanisms. The activity of D-serine, β-lactams alone and in combinations was evaluated both in vitro by standard MICs, time–kill curves and checkerboard assays, and in vivo by murine systemic infection model as well as neutropenic thigh infection model. An in vitro synergistic effect was demonstrated with the combination of D-serine and β-lactams against MRSA standard and clinical strains.Importantly, the combinations enhanced the therapeutic efficacy in the animal models as compared toβ-lactam alone groups. Initial mechanism study suggested possible revision of D-alanine-D-alanine residue to D-alanine-D-serine in peptidoglycan by adding of D-alanine in the medium, which may cause decreased affinity to PBPs during transpeptidation. In conclusion, D-serine had synergistic activity in combination with β-lactams against MRSA strains both in vitro and in vivo. Considering the relatively good safety of D-serine alone or in combination with β-lactams, D-serine is worth following up as new anti-MRSA infection strategies.Qing Wang Yuemeng Lv Jing Pang Xue Li Xi Lu Xiukun Wang Xinxin Hu Tongying Nie Xinyi Yang Yan Q.Xiong Jiandong Jiang Congran Li Xuefu You 2019Acta Pharmaceutica Sinica B2019,9,3:6
