1SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. 3Longitudinal study of a community hospital integrated model for diabetes management in the Beijing Jingsong community显示文摘Objective:To determine the effects of a community hospital integrated model on the longitu-dinal management of diabetic patients.Methods:Four hundred forty-one patients with type 2 diabetes residing in Jingsong Commu-nity were randomly assigned to intensive and standard groups.Metabolic parameters were meas-ured in the two groups at baseline and after 36 months of management to compare the rate of goal achievement.Results:After comprehensive management,the overall control rate in the intensive group was increased from 2.7%to 9.6%compared with 2.3%-4.5%in the standard group.Specifically,the control rates for fasting plasma glucose,glycosylated hemoglobin,and blood pressure in the intensive group were significantly increased from 54.1%,40.0%,and 68.2%to 85.5%,74.6%,and 89.1%,respectively.The control rate for glycosylated hemoglobin in the intensive group was significantly higher than the standard group after 36 months of treatment.Conclusion:The community hospital integrated model for longitudinal management effec-tively improved the control rate of glycosylated hemoglobin and the overall control rate in patients with type 2 diabetes.Jiandong Zhang Ning Zhuang Mingxia Yuan Lijuan Gao Chuan Bian Huaining Chen Qing Chi Suqing Hui Guiying Li Yanli Su Hongsong Bao Gang Wan Guangran Yang Shenyuan Yuan 2014Family Medicine and Community Health2014,2,1:7

7Allosteric inhibition reveals SHP2-mediated tumor immunosuppression in colon cancer by single-cell transcriptomics显示文摘Colorectal cancer(CRC), a malignant tumor worldwide consists of microsatellite instability(MSI) and stable(MSS) phenotypes. Although SHP2 is a hopeful target for cancer therapy, its relationship with innate immunosuppression remains elusive. To address that, single-cell RNA sequencing wasperformed to explore the role of SHP2 in all cell types of tumor microenvironment(TME) from murine MC38 xenografts. Intratumoral cells were found to be functionally heterogeneous and responded significantly to SHP099, a SHP2 allosteric inhibitor. The malignant evolution of tumor cells was remarkably arrested by SHP099. Mechanistically, STING-TBK1-IRF3-mediated type I interferon signaling was highly activated by SHP099 in infiltrated myeloid cells. Notably, CRC patients with MSS phenotype exhibited greater macrophage infiltration and more potent SHP2 phosphorylation in CD68;macrophages than MSI-high phenotypes, suggesting the potential role of macrophagic SHP2 in TME. 8Effect of Shot Peening and Plasma Electrolytic Oxidation on the Intergranular Corrosion Behavior of 7A85 Aluminum Alloy显示文摘The effects of shot peening(SP) and plasma electrolytic oxidation(PEO) on the intergranular corrosion behavior of the novel high strength aluminum alloy 7A85(AA 7A85) were investigated by electrochemical polarization and electrochemical impedance tests.The intergranular corrosion mechanism of SP,PEO and PEO combined with sealingtreated AA 7A85 was studied by the metallographic analysis,residual stress testing,X-ray diffractometer analysis and scanning electron microscopy.The results show that AA 7A85-T7452 is very sensitive to intergranular corrosion.SP would significantly improve its intergranular corrosion resistance.This is attributed to the combination action of residual compressive stress and grain refinement.PEO would reduce the largest corrosion depth by 41.6%.Moreover,PEO without sealing did not eliminate the intergranular corrosion due to the existence of the micropores and microcracks in the oxide coating.However,PEO combined with the SiO2sol–gel sealing treatment could effectively protect the AA 7A85-T7452 from intergranular corrosion because of the good corrosion resistance and barrier function of the sealed coating.Zuoyan Ye Daoxin Liu Chongyang Li Xiaoming Zhang Zhi Yang Mingxia Lei 2014Acta Metallurgica Sinica(English Letters)2014,27,4:4

By analyzing the DPSIR model and its correlation and relation with the evaluation index system, the indexes for geographic national conditions were selected from the aspects of resources, ecology, environment, economy, society, etc to build an evaluation index system for geographic national conditions that consists of three hierarchies: index, criterion, and target levels. 10Thermodynamic properties of metamizol monohydrate in pure and binary solvents at temperatures from(283.15 to 313.15) K显示文摘The thermodynamic properties of metamizol monohydrate in pure solvents(methanol,ethanol,n-propanol and isopropanol) and two binary mixed solvent systems including(methanol+ethanol) and(methanol+isopropanol) were measured from 283.15 K to 313.15 K by gravimetric method under atmospheric pressure thought as 0.1 MPa.The modi fied Apelblat equation,the CNIBS/R-K equation,the Hybrid model and the NRTL model were used to correlate the solubility of metamizol monohydrate,respectively.The results show that the solubility of metamizol monohydrate in all the tested solvents increases with the rising temperature which means that it has temperature dependence.What's more,the effects of solvent components of the binary solvent mixtures on solubility were discussed,it illustrates that the increasing of the molar fraction of methanol gives the system a greater dissolving power.Furthermore,according to the NRTL model,the enthalpy,the Gibbs energy and the entropy of the mixing process were also obtained and discussed.Mingxia Guo Qiuxiang Yin Chang Wang Yaohui Huang Yang Li Zaixiang Zhang Xia Zhang Zhao Wang Meijing Zhang Ling Zhou 2017Chinese Journal of Chemical Engineering2017,25,10:3

12Loss of hnRNP A1 in murine skeletal muscle exacerbates high-fat diet-induced onset of insulin resistance and hepatic steatosis显示文摘Impairment of glucose(Glu)uptake and storage by skeletal muscle is a prime risk factor for the development of metabolic diseases.Heterogeneous nuclear ribonucleoprotein A1(hnRNP Al)is a highly abundant RNA-binding protein that has been implicated in diverse cellular functions.The aim of this study was to investigate the function of hnRNP A1 on muscle tissue insulin sensitivity and systemic Glu homeostasis.Our results showed that conditional deletion of hnRNP Al in the muscle gave rise to a severe insulin resistance phenotype in mice fed a high-fat diet(HFD).Conditional knockout mice fed a HFD showed exacerbated obesity,insulin resistance,and hepatic steatosis.In vitro interference of hnRNP Al in C2C12 myotubes impaired insulin signal transduction and inhibited Glu uptake,whereas hnRNP Al overexpression in C2C12 myotubes protected against insulin resistance induced by supraphysiological concentrations of insulin.The expression and stability of glycogen synthase(gysl)mRNA were also decreased in the absence of hnRNP A l.Mechanistically,hnRNP Al interacted with gys l and stabilized its mRNA,thereby promoting glycogen synthesis and maintaining the insulin sensitivity in muscle tissue.Taken together,our findings are the first to show that reduced expression of hnRNP Al in skeletal muscle affects the metabolic properties and systemic insulin sensitivity by inhibiting glycogen synthesis.Mingxia Zhao Lihong Shen Zijun Ouyang Manru Li Guoliang Deng Chenxi Yang Wei Zheng Lingdong Kong Xuefeng Wu Xudong Wu Wenjie Guo Ye Yin Qiang Xu Yang Sun 2020Journal of Molecular Cell Biology2020,12,4:2
13Baduanjin exercise improved premenstrual syndrome symptoms in Macao women显示文摘OBJECTIVE: To investigate the effect of Baduanjin,a traditional Chinese medical exercise, on improving premenstrual syndrome(PMS) symptoms in women.METHODS: Forty reproductive age women with PMS in Macao practiced standardized Baduanjin exercise for three menstrual cycles. A questionnaire,the daily record of severity of problems(DRSP), was used to measure symptom severity. DRSP was filled out every days tarting from the last cycle before exercise(i.e., the first menstrual cycle) to the third cycle after exercise initiation(i.e., the fourth menstrual cycle). The total scores and the scores of each item during the 5 premenstrual days and follicular phase(5-9 postmenstrual days)were calculated.RESULTS: After exercise, the total DRSP scores dur-ing the 5 premenstrual days and differences in the total DRSP scores between the 5 premenstrual days and the follicular phase were both significantly reduced. Of note, the physical symptom total scores in the 5 premenstrual days and the differences between its total score in the 5 premenstrual days and in the follicular phase were both significantly reduced. 15Multiple forms of vitamin B_(6) regulate salt tolerance by balancing ROS and abscisic acid levels in maize root显示文摘Salt stress causes osmotic stress,ion toxicity and oxidative stress,inducing the accumulation of abscisic acid(ABA)and excessive reactive oxygen species(ROS)production,which further damage cell structure and inhibit the development of roots in plants.Previous study showed that vitamin B_(6)(VB_(6))plays a role in plant responses to salt stress,however,the regulatory relationship between ROS,VB_(6) and ABA under salt stress remains unclear yet in plants.In our study,we found that salt stress-induced ABA accumulation requires ROS production,in addition,salt stress also promoted VB_(6)(including pyridoxamine(PM),pyridoxal(PL),pyridoxine(PN),and pyridoxal 5′-phosphate(PLP))accumulation,which involved in ROS scavenging and ABA biosynthesis.Furthermore,VB_(6)-deficient maize mutant small kernel2(smk2)heterozygous is more susceptible to salt stress,and which failed to scavenge excessive ROS effectively or induce ABA accumulation in maize root under salt stress,interestingly,which can be restored by exogenous PN and PLP,respec-tively.According to these results,we proposed that PN and PLP play an essential role in balancing ROS and ABA levels under salt stress,respectively,it laid a foundation for VB_(6) to be better applied in crop salt resistance than ABA.Chongchong Lu Yuan Tian Xuanxuan Hou Xin Hou Zichang Jia Min Li Mingxia Hao Yanke Jiang Qingbin Wang Qiong Pu Ziyi Yin Yang Li Baoyou Liu Xiaojing Kang Guangyi Zhang Xinhua Ding Yinggao Liu 2022Stress Biology2022,2,1:1

19Combination of extraction tip and MALDI-TOF-MS for efficient separation and analysis of cysteine-containing peptides显示文摘In our work,a new extraction tip with gold-modified polymer is developed.The simple,self-made and extremely economical tips were successfully applied to capture cysteine-containing peptides.The loading capacity of a tip(column bed:0.3 mm diameter,5 mm length)is 2–4μg peptides.We can make one tip in 30 s and each costs less than 0.1 cent.The use of these tips can achieve a stable analysis with less background interference,even for 10 ng target peptides.Compared with other separation techniques,our method can save much time and energy while providing a means to selectively capture cysteine-containing peptides from complex analyte due to the strong interaction.All results showed that our new extraction tips have minimal cost and perfect selectivity;thus they have great potential in sample pretreatment systems for proteomics.WAN Dan CHEN Qi GAO MingXia ZHANG XiangMin YANG PengYuan 2014Science China Chemistry2014,57,5:1
