2Kallistatin,a new and reliable biomarker for the diagnosis of liver cirrhosis显示文摘Kallistatin,which protects organs and cells against inflammation,fibrosis and oxidative stress,is mainly synthesized and secreted in liver.However,its relationship to human liver disease remains unclear.The purpose of this study was to explore the relationship between serum kallistatin and clinical evidence of both cirrhosis and hepatocellular carcinoma(HCC),and to determine if serum kallistatin levels could be used as a diagnostic indicator of hepatic health status,especially human liver cirrhosis(LC).Our cohort consisted of 115 patients with clinically proven liver fibrosis(LF),LC,or HCC by liver biopsies,and 31 healthy controls(CON).Serum kallistatin levels were quantified by ELISA.Results of the present study demonstrated that irrespective of the underlying etiology,serum kallistatin levels were significantly lower in the LF/LC group when compared with the CON group.A decrease in serum kallistatin levels appeared to reflect the extent of cirrhosis,with the lowest levels associated with higher grades of cirrhosis.Patients with LC had a noticeable correlation between serum kallistatin levels and other serum biochemical indicators.The area under the curve(AUC) for LC,viral liver cirrhosis(VLC)and alcoholic liver cirrhosis(ALC) was 0.845,0.757 and 0.931,respectively.In conclusion,our findingsdemonstrated that kallistatin,a plasma protein produced by the liver,can be a useful and reliable diagnostic indicator of hepatic health status,especially for LC.Zhiyun Cheng Yinghui Lv Suqiu Pang Ruyu Bai Mingxi Wang Shuyu Lin Tianwen Xu Duncan Spalding Nagy Habib Ruian Xu 2015Acta Pharmaceutica Sinica B2015,5,3:7
3Environmental taxation and regional inequality in China显示文摘In order to combat environmental pollution, China enacted the Environmental Protection Tax Law in early 2018. Yet the impacts of the environmental tax on individual regions with different socioeconomic statuses, which are crucial for social justice and public acceptance, remain unclear. Based on a MultiRegional Input-Output(MRIO) table and a nationally regulated tax payment calculation method, this study analyzes the distributional impacts of an environmental tax based upon province’s consumption from both inter-provincial and rural-urban aspects. The national tax revenue based on the current levy mechanism is estimated to be only one seventh of the economic loss from premature mortality caused by ambient particulate matter(PM2.5). The taxation may slightly alleviate urban-rural inequality but may not be helpful with reducing inter-provincial inequality. We further analyze two alternative levy mechanisms. 4Capsid destabilization and epitope alterations of human papillomavirus 18 in the presence of thimerosal显示文摘Thimerosal has been widely used as a preservative in drug and vaccine products for decades.Due to the strong propensity to modify thiols in proteins,conformational changes could occur due to covalent bond formation between ethylmercury(a degradant of thimerosal)and thiols.Such a conformational change could lead to partial or even complete loss of desirable protein function.This study aims to investigate the effects of thimerosal on the capsid stability and antigenicity of recombinant human papillomavirus(HPV)18 virus-like particles(VLPs).Dramatic destabilization of the recombinant viral capsid upon thimerosal treatment was observed.Such a negative effect on the thermal stability of VLPs preserved with thimerosal was shown to be dependent on the thimerosal concentration.Two highly neutralizing antibodies,13H12 and 3C3,were found to be the most sensitive to thimerosal treatment.The kinetics of antigenicity loss,when monitored with 13H12 or 3C3 as probes,yielded two distinctly different sets of kinetic parameters,while the data from both monoclonal antibodies(mAbs)followed a biphasic exponential decay model.The potential effect of thimerosal on protein function,particularly for thiolcontaining proteinaceous active components,needs to be comprehensively characterized during formulation development when a preservative is necessary.Xiaofen Huang Yike Li Meifeng Nie Mingxi Yue Yufang Li Zhijie Lin Huirong Pan Mujin Fang Ting Wu Shaowei Li Jun Zhang Ningshao Xia Qinjian Zhao 2021Journal of Pharmaceutical Analysis2021,11,5:1

6A susceptibility locus rs7099208 is associated with non-obstructive azoospermia via reduction in the expression of FAM160B1显示文摘Non-obstructive azoospermia(NOA) is a severe defect in male reproductive health that occurs in 1%of adult men.In a previous study,we identified that rs7099208 is located within the last intron of FAM160B1 at10q25.3.In this study,we analysed expression Quantitative Trait Loci(eQTL) of FAM160B1,ABLIM1 and TRUB1,the three genes surrounding rs7099208.Only the expression level of FAM160B1 was reduced for the homozygous alternate genotype(GG) of rs7099208,but not for the homozygous reference or heterozygous genotypes.FAM160B1 is predominantly expressed in human testes,where it is found in spermatocytes and round spermatids.From 17 patients with NOA and five with obstructive azoospermia(OA),immunohistochemistry revealed that expression of FAM160B1 is reduced,or undetectable in NOA patients,but not in OA cases or normal men.We conclude that rs7099208 is associated with NOA via a reduction in the expression of FAM160B1.Yan Zhang Jing Qian Minghui Wu Mingxi Liu Kai Zhang Yuan Lin Xuejiang Guo Zuomin Zhou Zhibin Hu Jiahao Sha 2015The Journal of Biomedical Research2015,29,6:1

10Enhanced afterglow performance of CaAl_(2)O_(4):Eu^(2+),Nd^(3+) phosphors by co-doping Gd^(3+)显示文摘In order to effectively improve the afterglow properties of CaAl_(2)O_(4):Eu^(2+),Nd^(3+) phosphors,a series of Ca_(0.982-x)Al_(2)O_(4):0.012 Eu^(2+),0.006 Nd^(3+),xGd^(3+)(x=0,0.012,0.024,0.036,0.048,0.060 mol) phosphors were prepared by a high-temperature solid-phase approach.Crystalline composition and microstructure were characterized by XRD,TEM,HRTEM,and XPS,luminescence properties were systematically analyzed by fluorescence spectra,afterglow decay curves and TL glow curve.Results show that all of Ca_(0.982-x)Al_(2)O_(4):0.012 Eu^(2+),0.006 Nd^(3+),xGd^(3+)phosphors belong to monoclinic CaAl_(2)O_(4),without other cystalline phase.The blue emission at 442 nm is observed,which is assigned to the 4f^(6)5 d→4f^(7) transition of Eu^(2+) ions.Doping with appropriate amount of Gd^(3+) ions(x=0.036 mol) significantly improves the afterglow properties of phosphors,but the excessive doping of Gd^(3+) induces the fluorescent quenching.The doping of moderate Gd^(3+) changes the traps states,the trap depth varies from 0.598 to 0.644 eV and the trap concentration is also greatly improved,thus significantly improving afterglow performance.Mingxi Zhang Fengfeng Li Yongcheng Lin Yan Li Yi Shen 2021Journal of Rare Earths2021,39,8:0
11SARS-CoV-2-specific antibody response characteristics in COVID-19 patients of different ages显示文摘Age has been found to be one of the main risk factors for the severity and outcome of COVID-19.However,differences in SARS-CoV-2 specific antibody responses among COVID-19 patients of different age groups remain largely unknown.In this study,we analyzed the IgG/IgM responses to 21 SARS-CoV-2 proteins and 197 peptides that fully cover the spike protein against 731 sera collected from 731 COVID-19 patients aged from 1 to 92 years.We show that there is no overall difference in SARS-CoV-2 antibody responses in COVID-19 patients in the 4 age groups.By antibody response landscape maps,we find that the IgG response profiles of SARS-CoV-2 proteins are positively correlated with age.The S protein linear epitope map shows that the immunogenicity of the S-protein peptides is related to peptide sequence,disease severity and age of the COVID-19 patients.Furthermore,the enrichment analysis indicates that low S1 IgG responses are enriched in patients aged<50 and high S1 IgG responses are enriched in mild COVID-19 patients aged>60.In addition,high responses of non-structural/accessory proteins are enriched in severe COVID-19 patients aged>70.These results suggest the distinct immune response of IgG/IgM to each SARS-CoV-2 protein in patients of different age,which may facilitate a deeper understanding of the immune responses in COVID-19 patients.Linfang Lu Siqi Yu Min Liu Yang Li Qing Lei Mingxi Lin Danyun Lai Shujuan Guo Hewei Jiang Hongyan Hou Yunxiao Zheng Xuening Wang Mingliang Ma Bo Zhang Hong Chen Junbiao Xue Hainan Zhang Huan Qi Ziyong Sun Feng Wang Xionglin Fan Zhaowei Xu 2022Acta Biochimica et Biophysica Sinica2022,54,4:0
12RSBP15 interacts with and stabilizes dRSPH3 during sperm axoneme assembly in Drosophila显示文摘Flagellum in sperm is composed of over 200 different proteins and is essential for sperm motility. In particular, defects in the assembly of the radial spoke in the flagellum result in male infertility due to loss of sperm motility. However, mechanisms regulating radial spoke assembly remain unclear in metazoans.Here, we identified a novel Drosophila protein radial spoke binding protein 15(RSBP15) which plays an important role in regulating radial spoke assembly. Loss of RSBP15 results in complete lack of mature sperms in seminal vesicles(SVs), asynchronous individualization complex(IC) and defective '9 + 2'structure in flagella. RSBP15 is colocalized with dRSPH3 in sperm flagella, and interacts with dRSPH3 through its DD_R_PKA superfamily domain which is important for the stabilization of dRSPH3. Moreover,loss of dRSPH3, as well as dRSPH1, dRSPH4 a and dRSPH9, showed similar phenotypes to rsbp15 KO mutant.
