1Effects of inoculating fungi on agilawood formation in Aquilaria sinensis显示文摘Agilawood is a costly heartwood medicine obtained from Aquilaria sinensis with active ingredients mainly composed of volatile and semi-volatile substances. However, the formation time of agilawood is quite long and little is known about its formation mechanism. Two highly active fungi obtained from natural agilawood were inoculated on A. sinensis trees to understand their interaction processes and elucidate the transformation rules of induced chemical compositions within different test periods. The results demonstrated that the fungi could successfully colonize living tissues and cells and activate the host defense system, resulting in agilawood accumulation. With increasing time, the main components of A. sinensis converted into constituents or analogs of agilawood and the host exhibited 'self-injury' to prevent fungal intrusion and protect other tissues. 2Comprehensive Mineral Nutrition Analysis of Watermelon Grafted onto Two Different Rootstocks显示文摘Grafting is a widely used practice in fruit-bearing vegetables. However, why grafting affects plant growth, fruit yield, and quality, especially from the aspect of mineral nutrition, remains unclear. In this study, watermelon cultivar ‘Zaojia 8424' was grafted onto bottle gourd ‘Jingxinzhen1'(Lagenaria siceraria) and pumpkin ‘Qingyanzhen 1'(Cucurbita maxima × C. moschata). Non-grafted plants were used as the control. Results show that rootstock grafting significantly increases plant growth and single fruit weight of watermelon. Watermelon grafted onto rootstocks, especially pumpkin, exhibits significantly higher root volume, root surface area, and number of root tips and forks in comparison with non-grafted plants. Fruit flesh, rind firmness, and rind thickness were enhanced by grafting. However, fruit soluble solids and taste significantly decreased in plants grafted onto pumpkin. The total uptake(mg · plant^(-1)) and concentration(mg · g^(-1)DW) of N, K, Ca, Fe, Mg, and Mn in root, stem, leaf,fruit rind, and flesh were generally higher in grafted plants compared to non-grafted ones, especially for N of pumpkin rootstock-grafted plants.The total uptake of nutrients of plants grafted onto bottle gourd and pumpkin was increased by 30.41% and 49.14% at fruit development stage and by 21.33% and 47.46% at fruit maturation stage, respectively, compared with non-grafted plants. 3A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 4Comprehensive profiling of Lingzhihuang capsule by liquid chromatography coupled with mass spectrometry-based molecular networking and target prediction显示文摘Objective:Lingzhihuang capsule(LZHC)is a natural product that consists of 10 commonly used medicinal plants,and it is used in traditional Chinese medicine to promote people’s overall health.Previously,LZHC was successfully used as adjuvant therapy for treating patients with cancer.However,the chemical constituents of LZHC and their potential biological functions remain unclear.The aim of this study is to investigate the major bioactive compounds in LZHC and predict their pharmacological targets.Methods:The LZHC constituents were putatively identified by ultra-high performance liquid chromatography coupled with timeof-flight mass spectrometry combined with mass spectrometry-based molecular networking.The targets were predicted using SwissTargetPrediction software,and the associated gene ontology and Kyoto encyclopedia of genes and genomes pathways were analyzed using the Database for Annotation,Visualization,and Integrated Discovery.The mass spectrometry-based molecular network and compound-target-pathway network were constructed using Cytoscape 3.8.0 software.Results:We putatively identified 94 compounds of LZHC by mass spectrometry-based molecular networking,including triterpene(the main structural type)and other clusters(ie,flavonoids and organic acids).Our results suggested that multiple pivotal targets were regulated by LZHC,including tumor necrosis factor,nitric oxide synthase 2,glucocorticoid receptor,estrogen receptor,3-oxo-5-alpha-steroid 4-dehydrogenase 2,prostaglandin e2 receptor ep4 subtype,estrogen receptor beta,phosphatidylinositol 4,5-bisphosphate 3-kinase catalytic subunit alpha isoform,mitogen-activated protein kinase 3,and racalpha serine,which are related to signal transduction,positive regulation of transcription from RNA polymerase II promoters,oxidation-reduction processes,inflammatory responses,and other biological processes.Functional annotation of those targets suggested that several signaling pathways may be regulated by LZHC,such as cancer-related proteoglycans,the PI3K-Aktsignaling pathway,and the cAMP-signaling pathway.Conclusions:Our findings reveal the chemical constituents of LZHC and provided scientific support for the efficacy of LZHC in terms of immune regulation,anti-aging,and tumor suppression.Mengliang Huang Sijia Yu Qing Shao Hao Liu Yingchao Wang Hongzhang Chen Yansheng Huang Yi Wang 2022Acupuncture and Herbal Medicine2022,2,1:2

13Effect of Rare Earth Elements on Microstructure of Cu-6% Ag and Cu-24% Ag Alloys显示文摘The cast rods of the alloys with low Ag content, Cu-6% Ag, and high Ag content, Cu-24%,were prepared and homogenized. 15Accumulation and Distribution Dynamics of Soluble Carbohydrate in Helianthus tuberosus under Drought Stress显示文摘With Qingyu 1 and Qingyu 2 as test materials,variation of soluble carbohydrate content in different parts were studied under two types of artificially simulated drought stress( polyethylene glycol PEG-6000 stress and water-controlling stress). The results showed that under the stress of PEG,soluble carbohydrate content in leaves of Qingyu 1 and Qingyu 2 increased with the prolonging time of stress; soluble carbohydrate content in stems of Qingyu 1 did not show regular changes with the prolonging time of stress,while 30% PEG stress received the best effect on Qingyu 2 after 24h; soluble carbohydrate content in roots of Qingyu 1 and Qingyu 2 increased with the prolonging time of stress. 19Variability of fuel consumption and CO_(2) emissions of a gasoline passenger car under multiple in-laboratory and on-road testing conditions显示文摘An increasing divergence regarding fuel consumption(and/or CO_(2)emissions) between realworld and type-approval values for light-duty gasoline vehicles(LDGVs) has posed severe challenges to mitigating greenhouse gases(GHGs) and achieving carbon emissions peak and neutrality. To address this divergence issue, laboratory test cycles with more real-featured and transient traffic patterns have been developed recently, for example, the China Lightduty Vehicle Test Cycle for Passenger cars(CLTC-P). We collected fuel consumption and CO_(2)emissions data of a LDGV under various conditions based on laboratory chassis dynamometer and on-road tests. Laboratory results showed that both standard test cycles and setting methods of road load affected fuel consumption slightly, with variations of less than 4%. Compared to the type-approval value, laboratory and on-road fuel consumption of the tested LDGV over the CLTC-P increased by 9% and 34% under the reference condition(i.e., air conditioning off, automatic stop and start(STT) on and two passengers). On-road measurement results indicated that fuel consumption under the low-speed phase of the CLTC-P increased by 12% due to the STT off, although only a 4% increase on average over the entire cycle. More fuel consumption increases(52%) were attributed to air conditioning usage and full passenger capacity. Strong correlations(R2> 0.9) between relative fuel consumption and average speed were also identified. Under traffic congestion(average speed below 25 km/hr), fuel consumption was highly sensitive to changes in vehicle speed. 20Exploiting Complex-Type N-Glycan to Improve the in Vivo Stability of Bioactive Peptides显示文摘Peptides can be potentmolecules with high efficacy and selectivity in the development of biotherapeutics.However,the poor pharmacokinetic properties of peptides pose major challenges for their broader medicinal applications.Inspired by the proteinstabilizing role of natural N-glycosylation,we design and synthesize a series of parathyroid hormone(PTH)peptides(1-34),bearing either N-GlcNAc or biantennary complex-type N-glycan modification,and evaluate their serum stability and biological activities.The results indicate that an N-Asn-linked complex-type sialylundecasaccharide can increase the serum half-life and in vivo bioactivity of PTH peptides with a broad tolerance of modification sites.Further,hydrogen/deuterium exchange mass spectroscopy indicates that the larger-sized Nglycan can induce enhanced hydration dynamics in its surroundings,which may facilitate an improved resistance for the peptide against enzymatic proteolysis.This sialylundecasaccharide-based peptideengineering strategy has also been applied to glucagon-like peptide-1(7-37),leading to glycopeptides with enhanced hypoglycemic activity and acting time in vivo.Together,these results demonstrate the potential of using sialylated complextype N-glycan as a general engineering strategy for developing long-acting peptide therapeutics.Qijia Wei Jun Zhang Yuankun Dao Mengliang Ye Dangliang Liu Weidong Dong Ning Yuan Hongxing Li Chunli Song Mo Li Xiaomeng Shi Suwei Dong 2023CCS Chemistry2023,5,7:0
