| 2 | AFF3-DNA methylation interplay in maintaining the mono-allelic expression pattern of XIST in terminally differentiated cells显示文摘X chromosome inactivation and genomic imprinting are two classic epigenetic regulatory processes that cause mono-allelic gene expression.In female mammals,mono-allelic expression of the long non-coding RNA gene X-inactive specific transcript(XIST)is essential for initiation of X chromosome inactivation upon differentiation.We have previously demonstrated that the central factor of super elongation complex-like 3(SEC-L3),AFF3,is enriched at gamete differentially methylated regions(DMRs)of the imprinted loci and regulates the imprinted gene expression.Here,we found that AFF3 can also bind to the DMR downstream of the XIST promoter.Knockdown of AFF3 leads to de-repression of the inactive allele of X IST in terminally differentiated cells.In addition,the binding of AFF3 to the XIST DMR relies on DNA methylation and also regulates DNA methylation level at DMR region.However,the KAP1-H3K9 methylation machineries,which regulate the imprinted loci,might not play major roles in maintaining the mono-aUelic expression pattern of XIST in these cells.Thus,our results suggest that the differential mechanisms involved in the X IST DMR and gDMR regulation,which both require AFF3 and DNA methylation. | Yue Zhang Chao Wang Xiaoxu Liu Qian Yang Hongliang Ji Mengjun Yang Manman Xu Yunyan Zhou Wei Xie Zhuojuan Luo Chengqi Lin | 2019 | Journal of Molecular Cell Biology2019,11,9: | 3 |
| 3 | Heat Transfer Enhancement with Mixing Vane Spacers Using the Field Synergy Principle显示文摘The single-phase heat transfer characteristics in a PWR fuel assembly are important. Many investigations attempt to obtain the heat transfer characteristics by studying the flow features in a 5×5 rod bundle with a spacer grid. The field synergy principle is used to discuss the mechanism of heat transfer enhancement using mixing vanes according to computational fluid dynamics results, including a spacer grid without mixing vanes, one with a split mixing vane, and one with a separate mixing vane. The results show that the field synergy principle is feasible to explain the mechanism of heat transfer enhancement in a fuel assembly. The enhancement in subchannels is more effective than on the rod's surface. If the pressure loss is ignored, the performance of the split mixing vane is superior to the separate mixing vane based on the enhanced heat transfer. Increasing the blending angle of the split mixing vane improves heat transfer enhancement, the maximum of which is 7.1%. Increasing the blending angle of the separate mixing vane did not significantly enhance heat transfer in the rod bundle, and even prevented heat transfer at a blending angle of 50°. | 6 | Synthesis and biological evaluation of ^(18)F-FB-NGA as a hepatic asialoglycoprotein receptor PET imaging agent显示文摘Asialoglycoprotein receptor(ASGP-R)is a hepatic membrane receptor that uniquely exists on the surface of mammalian hepatocytes,and has been used as target of liver functional imaging agents for many years.We labeled the Galactosyl-neoglycoalbumin(NGA)with 18F to get a PET molecular probe 18F-FB-NGA and evaluated its ability as a liver functional PET imaging agent.The 18F-FB-NGA was prepared with NGA by conjugation with Nsuccinimidyl-4-18F-fluorobenzoate(18F-SFB)and purified with PD-10 desalting column.The radiolabeling yield and radiochemical purity of 18F-FB-NGA were determined by radio-HPLC.Starting with 18F-F–,the total time for 18F-FB-NGA was about 120±10 min.The decay-corrected radiochemical yield is about 25–30%.The radiochemical purity of purified 18F-FB-NGA was more than 98%.Labeled with 185–1850 MBq 18F-SFB,the specific activity of 18F-FBNGA was estimated to be 7.83–78.3 TBq/mmol.Biodistribution of 18F-FB-NGA in normal mice was investigated after injection through the tail vein.The results showed that the liver accumulated 39.47±3.42 and 12.12±6.11%ID/g at 10 and 30 min after injection,respectively.Dynamic MicroPET images in mice were acquired with and without block after injection of the radiotracer,respectively.High liver activity accumulation was observed at 5 min after injection in normal group.On the contrary,the liver accumulation was significantly lower after block,indicating the specific binding to ASGP-R.18F-FB-NGA is probably a potential PET liver imaging agent. | GU Xiaobo CAI Gangming JIANG Mengjun ZHOU Yaoyuan ZHANG Rongjun | 2013 | Nuclear Science and Techniques2013,24,6: | 2 |

| 9 | Image Generation of Tomato Leaf Disease Identification Based on Small-ACGAN显示文摘Plant diseases have become a challenging threat in the agricultural field.Various learning approaches for plant disease detection and classification have been adopted to detect and diagnose these diseases early.However,deep learning entails extensive data for training,and it may be challenging to collect plant datasets.Even though plant datasets can be collected,they may be uneven in quantity.As a result,the problem of classification model overfitting arises.This study targets this issue and proposes an auxiliary classifier GAN(small-ACGAN)model based on a small number of datasets to extend the available data.First,after comparing various attention mechanisms,this paper chose to add the lightweight Coordinate Attention(CA)to the generator module of Auxiliary Classifier GANs(ACGAN)to improve the image quality.Then,a gradient penalty mechanism was added to the loss function to improve the training stability of the model.Experiments show that the proposed method can best improve the recognition accuracy of the classifier with the doubled dataset.On AlexNet,the accuracy was increased by 11.2%.In addition,small-ACGAN outperformed the other three GANs used in the experiment.Moreover,the experimental accuracy,precision,recall,and F1 scores of the five convolutional neural network(CNN)classifiers on the enhanced dataset improved by an average of 3.74%,3.48%,3.74%,and 3.80%compared to the original dataset.Furthermore,the accuracy of MobileNetV3 reached 97.9%,which fully demonstrated the feasibility of this approach.The general experimental results indicate that the method proposed in this paper provides a new dataset expansion method for effectively improving the identification accuracy and can play an essential role in expanding the dataset of the sparse number of plant diseases. | Huaxin Zhou Ziying Fang Yilin Wang Mengjun Tong | 2023 | Computers, Materials & Continua2023,,7: | 1 |

External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model. | 18 | Theoretical and experimental study on the rheological properties of WIS grout and the dispersion and sealing mechanism显示文摘Recently a new grout material called water inflow sealing(WIS) was invented for sealing water inflow in tunneling and underground constructions. In this study, a special experimental method called intubated counter grouting(ICG) was proposed to investigate the influence of water dispersion on the rheological properties of the grout during the grouting process, and to testify the sealing performance of the grout,such as instant gelling ability(IGA) and anti-dispersion ability(ADA). In the experiment, dispersion was restricted in the downstream of the channel with a high turbulence intensity. The influences of ADA and IGA were therefore decoupled and evaluated separately. Experimental results revealed two distinctive sealing mechanisms of WIS. For a low initial velocity of water, WIS turned the shear flow of water into an overall movement of a plug by absorbing water into the particles. For a high initial velocity and the situation that the particles reached the outlet before sufficiently expanding, WIS modified the rheology of the water in the channel and reduced its velocity till the static state.
