1Expression changes and bioinformatic analysis of Wallerian degeneration after sciatic nerve injury in rat显示文摘Wallerian degeneration (WD) remains an important research topic. Many genes are differentially expressed during the process of WD, but the precise mechanisms responsible for these differentiations are not completely understood. In this study, we used microarrays to analyze the expression changes of the distal nerve stump at 0, 1, 4, 7, 14, 21 and 28 days after sciatic nerve injury in rats. The data revealed 6 076 differentially-expressed genes, with 23 types of expression, specifically enriched in genes associated with nerve development and axonogenesis, cytokine biosynthesis, cell differentiation, cytokine/chemokine production, neuron differentiation, cytokinesis, phosphorylation and axon regeneration. Kyoto Encyclopedia of Genes and Genomes pathway analysis gave findings related mainly to the MAPK signaling pathway, the Jak-STAT signaling pathway, the cell cycle, cytokine-cytokine receptor interaction, the p53 signaling pathway and the Wnt signaling pathway. Some key factors were NGF, MAG, CNTF, CTNNA2, p53, JAK2, PLCB1, STAT3, BDNF, PRKC, collagen II, FGF, THBS4, TNC and c-Src, which were further validated by real-time quantitative PCR, Western blot, and immunohistochemistry. 2MicroRNA-23a promotes colorectal cancer cell migration and proliferation by targeting at MARK1显示文摘The functional role of microRNA-23a in tumorigenesis has been investigated;however, the exact mechanism of microRNA-23a (miR-23a) in colorectal cancer development has not been fully explored. In the present study, we aimed to investigate the molecular functional role of miR-23a in colorectal carcinogenesis. Quantitative real-time polymerase chain reaction was conducted to investigate the expression level of miR-23a in tissue samples and cell lines (HCT116 and SW480). CCK-8, colony formation and Transwell assay were used to explore the role of miR-23a in cell proliferation and migration. Dual luciferase reporter assay was used to identify the direct binding of miR-23a with its target, MARK1. Western blot analysis was used to analyze the expression level of MARK1, as well as a confirmed miR-23a target gene, MTSS1, in miR-23a-mimic and miR-23a-inhibit groups. Rescue experiments were conducted by overexpression of MARK1 in miR-23a-mimic-transfected cell lines. The results showed that miR-23a was highly expressed in colorectal cancer tissue and cell lines. MiR-23a could promote proliferation and migration of colorectal cancer cell lines. MARK1 was a direct target of miR-23a and the expression level of MARK1 was down-regulated in miR-23a-mimic-transfected cell lines but up-regulated in miR-23a-inhibit-transfected cells. Overexpression of MARK1 could partly reverse the cancer-promoting function of miR-23a. Our results suggested that miR-23a promotes colorectal cancer cell proliferation and migration by mediating the expression of MARK1. 3Gene expression profiling of the rat sciatic nerve in early Wallerian degeneration after injury显示文摘Wallerian degeneration is an important area of research in modern neuroscience.A large number of genes are differentially regulated in the various stages of Wallerian degeneration,especially during the early response.In this study,we analyzed gene expression in early Wallerian degeneration of the distal nerve stump at 0,0.5,1,6,12 and 24 hours after rat sciatic nerve injury using gene chip microarrays.We screened for differentially-expressed genes and gene expression patterns.We examined the data for Gene Ontology,and explored the Kyoto Encyclopedia of Genes and Genomes Pathway.This allowed us to identify key regulatory factors and recurrent network motifs.We identified 1 546 differentially-expressed genes and 21 distinct patterns of gene expression in early Wallerian degeneration,and an enrichment of genes associated with the immune response,acute inflammation,apoptosis,cell adhesion,ion transport and the extracellular matrix.Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed components involved in the Jak-STAT,ErbB,transforming growth factor-β,T cell receptor and calcium signaling pathways.Key factors included interleukin-6,interleukin-1,integrin,c-sarcoma,carcinoembryonic antigen-related cell adhesion molecules,chemokine(C-C motif) ligand,matrix metalloproteinase,BH3 interacting domain death agonist,baculoviral IAP repeat-containing 3 and Rac.The data were validated with real-time quantitative PCR.This study provides a global view of gene expression profiles in early Wallerian degeneration of the rat sciatic nerve.Our findings provide insight into the molecular mechanisms underlying early Wallerian degeneration,and the regulation of nerve degeneration and regeneration.Dengbing Yao Meiyuan Li Dingding Shen Fei Ding Shibi Lu Qin Zhao Xiaosong Gu 2012Neural Regeneration Research2012,7,17:5

5STAM2 knockdown inhibits proliferation,migration,and invasion by affecting the JAK2/STAT3 signaling pathway in gastric cancer显示文摘Signal transducing adaptor molecule 2(STAM2)is a phosphotyrosine protein,which regulates receptor signaling and trafficking of mammalian cells.However,its role in gastric cancer(GC)remains undiscovered.In this study,we aimed to investigate the functions of STAM2 in GC.The mRNA and protein expression levels of STAM2 were measured by quantitative real-time PCR,western blot analysis,and immunohistochemistry.STAM2 was stably silenced in AGS and HGC-27 cells using small interfering RNA.The function of STAM2 in GC cells was further investigated by CCK-8 assay,EdU incorporation assay,flow cytometry,and scratch wound healing and Boyden chamber assays.Additionally,we conducted biological pathway enrichment analysis and rescue assays to explore the effects of STAM2 on JAK/STAT signaling pathway.Our results showed that STAM2 is remarkably highly expressed in GC tissues and cells,and overexpressed STAM2 is correlated with tumor size,advanced tumor node metastasis stage,and poor prognosis.In addition,STAM2 knockdown could significantly inhibit proliferation,block cell cycle,and restrain migration and invasion capabilities of GC cells.Mechanistically,we found that STAM2 knockdown effectively decreased the expressions of MMP2 and MMP9 and the phosphorylation levels of JAK2 and STAT3.Taken together,this study revealed that STAM2 knockdown could suppress malignant process by targeting the JAK2/STAT3 signaling pathway in GC.Yang Yang Qi Zhang Jiakui Liang Meiyuan Yang Zheng Wang Dong Tang Daorong Wang 2021Acta Biochimica et Biophysica Sinica2021,53,6:2
6Experimental Study on the Anti-inflammatory Effect of Ficus pumila L.显示文摘[Objectives]To study the anti-inflammatory effects of extracts from different parts of Ficus pumila L.[Methods]Inflammatory models of xylene-induced ear swelling,glacial acetic acid-induced celiac capillary permeability and carrageenan-induced toe swelling were established to evaluate the effects of extracts from different parts of F.pumila L.on inflammation.The inflammatory model of ear swelling in mice with bilateral adrenalectomy induced by xylene was established to determine whether its anti-inflammatory effect depends on hypothalamus-pituitary-adrenal axis(HPAA)system.[Results]Compared with the blank control group,except that there was no statistical significance in the low dose group(P>0.05),the high and middle dose groups of extracts from different parts of F.pumila L.had significant inhibitory effects on xylene-induced ear swelling in mice.The high,middle and low dose groups of F.pumila L.extract could significantly increase the celiac capillary permeability of mice induced by glacial acetic acid,and significantly inhibit the toe swelling induced by carrageenan.Compared with the blank control group,the high and middle dose groups of extracts from different parts of F.pumila L.could significantly inhibit xylene-induced ear swelling and carrageenan-induced toe swelling in bilateral adrenalectomy mice.[Conclusions]The extracts from different parts of F.pumila L.had anti-inflammatory activity,and its anti-inflammatory effect was independent of hypothalamus-pituitary-adrenal axis(HPAA)system.Chao ZENG Meiyuan LU Tingting MO Bing WU Qingtong LAN 2020Medicinal Plant2020,11,4:2
