1CRISPR/Cas9:A powerful tool for crop genome editing显示文摘The CRISPR/Cas9 technology is evolved from a type II bacterial immune system and represents a new generation of targeted genome editing technology that can be applied to nearly all organisms. Site-specific modification is achieved by a single guide RNA(usually about 20nucleotides) that is complementary to a target gene or locus and is anchored by a protospaceradjacent motif. Cas9 nuclease then cleaves the targeted DNA to generate double-strand breaks(DSBs), which are subsequently repaired by non-homologous end joining(NHEJ) or homology-directed repair(HDR) mechanisms. NHEJ may introduce indels that cause frame shift mutations and hence the disruption of gene functions. When combined with double or multiplex guide RNA design, NHEJ may also introduce targeted chromosome deletions,whereas HDR can be engineered for target gene correction, gene replacement, and gene knock-in. In this review, we briefly survey the history of the CRISPR/Cas9 system invention and its genome-editing mechanism. 2Effect of Salinity on the Survival, Ions and Urea Modulation in Red-eared Slider(Trachemys scripta elegans)显示文摘To understand the tolerance to salinity and osmoregulation of the introduced Trachemys scripta elegans, the salinity stress of four groups(salinity 5‰, 15‰, 25‰ and control group) were conducted. Inorganic ions, osmotic pressure, glucose and aldosterone of blood and urine in T. s. elegans(BW: 125.60 ± 19.84 g) were analyzed at 30 d, 60 d and 90 d stress. The results showed that: 1) inorganic ions concentration of blood and urine increased with ambient salinity, which indicated that high influx of ions was combined with higher outflow when exposed to saline water in T. s. elegans. However, blood aldosterone decreased with increasing salinity, which indicated that an increased sodium intake resulting in a diminished aldosterone production. However, with elapsed time, inorganic ions in urine decreased, which indicated that inorganic ions in blood would be accumulated, and Na+ and Cl– in the plasma inevitably build up to harmful levels, at last death was happening when T. s. elegans was exposed to salinity 25 during 90 d salinity stress; 2) blood osmotic pressure increased as ambient salinity increased, it would reach 400 mOsm/kg in the group of salinity 25, which was about 1.5 fold of the control group. Higher blood osmotic pressure was due to both higher blood ions and urea concentrations. There may be another mechanism to avoid an excess of NaCl together with an important loss of water using one of the end-products of nitrogen metabolism; 3) blood glucose in each group except the group of salinity 5 decreased with time elapsed and with salinity increased. Therefore, we can conclude that T. s. elegans is an osmoregulator that limits the entry of Na+ and Cl–, but can also tolerate certain degrees of increases in plasma Na+ and Cl–. When ambient salinity was lower than 15‰, T. s. elegans can increase blood osmotic pressure by balancing the entry of NaCl with the secretion of aldosterone decreased, and by accumulating blood urea for osmoregulation effectors, and survive for at least three months. 3Monthly change of community structure of zoobenthos in Xiangxi Bay after impoundment of the Three Gorges Reservoir,China显示文摘Xiangxi Bay,a typical bay of the Three Gorges Reservoir(TGR)in China,was formed in June 2003 after the impoundment of the TGR.We investigated the community structure of zoobenthos in Xiangxi Bay from August 2003 to August 2004.Benthic abundance and biomass were calculated per unit area of sediment.A total of 26 taxa were identified.They included two families of Lamellibranchia(7.7%of the total taxa number),two families of Oligochaeta(50%),Chironomidae(38.5%),and one other animal(3.8%).The average density and biomass were 276 ind./m2 and 0.301 g/m2,respectively.Oligochaetes and chironomids constituted the major groups,and Limnodrilus hoffmeisteri,Procladius sp.,Branchiura sowerbyi,Tubificidae sp1.and Polypedilum scalaenum group sp.were the common taxa in this reservoir system.Among them,L.hoffmeisteri and Procladius sp.were the dominant species,and contributed 37.0%and 28.3%of the total density,respectively.The standing crop of zoobenthos in Xiangxi Bay was relative low,compared with other similar reservoirs.Our studies demonstrate that the standing crop increased gradually with time.The total density and biomass in August 2004 were 8.8 and 14.3 times those of the initial impoundment,separately.Oligochaetes did not become the sole dominant group,indicating that the sediment loading in Xiangxi Bay may be not serious.SHAO Meiling XIE Zhicai YE Lin CAI Qinghua 2007Frontiers in Biology2007,2,2:1

12Connectivity of fish assemblages along the mangrove-seagrasscoral reef continuum in Wenchang, China显示文摘Understanding the connectivity of fish among different typical habitats is important for conducting ecosystembased management, particularly when designing marine protected areas(MPA) or setting MPA networks. To clarify of connectivity among mangrove, seagrass beds, and coral reef habitats in Wenchang, Hainan Province,China, the fish community structure was studied in wet and dry seasons of 2018. Gill nets were placed across the three habitat types, and the number of species, individuals, and body size of individual fish were recorded. In total, 3 815 individuals belonging to 154 species of 57 families were collected. The highest number of individuals and species was documented in mangroves(117 species, 2 623 individuals), followed by coral reefs(61 species,438 individuals) and seagrass beds(46 species, 754 individuals). The similarity tests revealed highly significant differences among the three habitats. Approximately 23.4% species used two habitats and 11.0% species used three habitats. A significant difference(p<0.05) in habitat use among eight species(Mugil cephalus, Gerres oblongus, Siganus fuscescens, Terapon jarbua, Sillago maculata, Upeneus tragula, Lutjanus russellii, and Monacanthus chinensis) was detected, with a clear ontogenetic shift in habitat use from mangrove or seagrass beds to coral reefs. The similarity indices suggested that fish assemblages can be divided into three large groups namely coral, seagrass, and mangrove habitat types. 15Distribution Characteristics and Health Risk Assessment of Antimony in Atmospheric Particulates in a Northern City of China显示文摘Over the last several decades,scientists have established a wealth of evidence to demonstrate the risks posed to human health by toxic elements in atmospheric particulate matter(PM).Antimony(Sb),as one of ever ignored PM-bound heavy metals,attracts more and more attentions and has been regarded as one emerging air pollutant with the change of pollution sources of particulate matter.To study the distribution of Sb in PM with different particle sizes is of great practical significance for understanding its source and health risks.In this study,the size distributions of Sb in PM(PM_(2.5),PM_(10)and TSP)in different seasons were studied from July 2018 to May 2019.The high concentration of PM-bound Sb was found and the health risk was evaluated.Sb was enriched in fine particles and showed higher values in winter,which was probably caused by coal combustion and meteorological conditions.It was also found that traffic-related non-exhaust emissions might become another main contribution to fine particle Sb.Health risk assessment demonstrated that the hazard quotient(HQ)of Sb in PM_(2.5)and PM_(10)for children(PM_(2.5):1.54,PM_(10):1.32)exceeded the acceptable threshold and ingestion contributed the most to the HQ for both children and adults.SHEN Yiwen ZHAO Hao ZHAO Changxian DONG Shuofei CAO Yuanming XIE Jiaojao LYU Meiling YUAN Chungang 2023Chemical Research in Chinese Universities2023,39,3:0
16A piezoelectric spectro- electrochemical study on the structural interconversion for poly(o-phenylenediamine) between its ladder structure with phenazine units and polyaniline-like chains显示文摘The combination of reflectance UV-Vis spec- troelectrochemistry with electrochemical quartz crystal microbalance (EQCM) and separate reflectance FTIR char- acterization were used to investigate the structural intercon- version for poly(o-phenylenediamine) (PoPD) between its ladder structure with phenazine units and polyaniline-like linear chains. The poly(o-phenylenediamine) films potentio- statically (0.8 V vs. SCE) grew on Au electrodes from 0.20 mol·L?1 H2SO4 (PoPD1) or 0.40 mol·L?1 NaOH (PoPD2) aque- ous solution containing 0.20 mol·L?1 Na2SO4 + 0.10 mol·L?1 o-phenylenediamine. By considering the mass of deposited PoPD2 film obtained from the EQCM data and the charge consumed under the current peak at ca. 0.6 V vs. SCE for oxidation of -NH2 groups in as-prepared PoPD2 during po- tential cycling in 0.10 mol·L-1 aqueous H2SO4, the molar per- centage of the polyaniline-like chains was estimated to be 19% (relative to total phenylenediamine units), being in agreement with the result obtained from a formaldehyde- combination experiment through the aminocarbonyl reaction. After 40-cycle potential sweeps between 0.2 and 0.8 V vs. SCE the polyaniline-like chains in PoPD2 could be com- pletely converted via intramolecular cyclization into the lad- der structure with phenazine units. However, PoPD1 was found to be perfectly composed of the ladder structure with phenazine units, and after 40-cycle potential sweeps between ?0.4 and 0.1 V vs. SCE only 2.5% in molar percentage of PoPD’s ladder structure could be converted into polyani line-like chains, suggesting that the ladder structure with phenazine units is thermodynamically more stable due to its possessing higher conjugation.TU Xinman XIE Qingji WANG Meiling ZHANG Youyu YAO Shouzhuo 2005Chinese Science Bulletin2005,50,15:0
