3Genome architecture and tetrasomic inheritance of autotetraploid potato显示文摘Potato(Solanum tuberosum)is the most consumed non-cereal food crop.Most commercial potato cultivars are autotetraploids with highly heterozygous genomes,severely hampering genetic analyses and improvement.By leveraging the state-of-the-art sequencing technologies and polyploid graph binning,we achieved a chromosome-scale,haplotype-resolved genome assembly of a cultivated potato,Cooperation-88(C88).lntra-haplotype comparative analyses revealed extensive sequence and expression differences in this tetraploid genome.We identified haplotype-specific pericentromeres on chromosomes,suggesting a distinct evolutionary trajectory of potato homologous centromeres.Furthermore,we detected double reduction events that are unevenly distributed on haplotypes in 1021 of 1034 selfing progeny,a feature of autopolyploid inheritance.By distinguishing maternal and paternal haplotype sets in C88,we simulated the origin of heterosis in cultivated tetraploid with a survey of 3110 tetra-allelic loci with deleterious mutations,which were masked in the heterozygous condition bytwo parents.This study provides insights into the genomic architecture of autopolyploids and will guide their breeding.Zhigui Bao Canhui Li Guangcun Li Pei Wang Zhen Peng Lin Cheng Hongbo Li Zhiyang Zhang Yuying Li Wu Huang Mingwang Ye Daofeng Dong Zhukuan Cheng Peter VanderZaag Evert Jacobsen Christian W.B.Bachem Suomeng Dong Chunzhi Zhang Sanwen Huang Qian Zhou 2022Molecular Plant2022,15,7:4
4Optimization of dewatering schemes for a deep foundation pit near the Yangtze River, China显示文摘A deep foundation pit constructed for an underground transportation hub was excavated near the Yangtze River. Among the strata, there are two confined aquifers, between which lies an aquiclude that is partially missing. To guarantee the safety of pit excavation, the piezometric head of the upper confined aquifer, where the pit bottom is located, should be 1 m below the pit bottom, while that of the lower confined aquifer should be dewatered down to a safe water level to avoid uplift problem. The Yangtze River levee is notably close to the pit, and its deformation caused by dewatering should be controlled. A pumping test was performed to obtain the hydraulic conductivity of the upper confined aquifer. The average value of the hydraulic conductivity obtained from analytical calculation is 20.45 m/d, which is larger than the values from numerical simulation(horizontal hydraulic conductivity K_H = 16 m/d and vertical hydraulic conductivity K_V = S m/d). The difference between K_H and K_V indicates the anisotropy of the aquifer. Two dewatering schemes were designed for the construction and simulated by the numerical models for comparison purposes. The results show that though the first scheme could meet the dewatering requirements, the largest accumulated settlement and differential settlement would be94.64 mm and 3.3‰, respectively, greatly exceeding the limited values. 5Construction of homozygous diploid potato through maternal haploid induction显示文摘Reinventing the tetraploid potato into a seed-propagated,diploid,hybrid potato would significantly accelerate potato breeding.In this regard,the development of highly homozygous inbred lines is a prerequisite for breeding hybrid potatoes,but self-incompatibility and inbreeding depression present challenges for developing pure inbred lines.To resolve this impediment,we developed a doubled haploid(DH)technology,based on mutagenesis of the potato DOMAIN OF UNKNOWN FUNCTION 679 membrane protein(StDMP)gene.Here,we show that a deficiency in StDMP allows the generation of maternal haploids for generating diploid potato lines.An exercisable protocol,involving hybridization,fluorescent marker screening,molecular and flow cytometric identification,and doubling with colchicine generates nearly 100%homozygous diploid potato lines.This dmp-triggered haploid induction(HI)system greatly shortens the breeding process and offers a robust method for generating diploid potato inbred lines with high purity.Jinzhe Zhang Jian Yin Jiayi Luo Die Tang Xijian Zhu Jie Wang Zhihong Liu Pei Wang Yu Zhong Chenxu Liu Canhui Li Shaojiang Chen Sanwen Huang 2022aBIOTECH2022,3,3:2

17Application of thermal comfort theory in probabilistic safety assessment of a nuclear power plant显示文摘Human factor errors in probabilistic safety assessment(PSA) of a nuclear power plant(NPP) can be prevented using thermal comfort analysis.In this paper,the THERP+HCR model is modified by using PMV (Predicted Mean Vote) and PPD(Predicted Percentage Dissatisfied) index system,so as to obtain the operator cognitive reliability,and to reflect and analyze human perception,thermal comfort status,and cognitive ability in a specific NPP environment.The mechanism of human factors in the PSA is analyzed by operators of skill,rule and knowledge types.The THERP+HCR model modified by thermal comfort theory can reflect the conditions in actual environment,and optimize reliability analysis of human factors.Improving human thermal comfort for different types of operators reduces adverse factors due to human errors,and provides a safe and optimum decision-making for NPPs.ZHOU Tao SUN Canhui LI Zhenyang WANG Zenghui 2011Nuclear Science and Techniques2011,22,5:1
