3Transplantation of placenta-derived mesenchymal stem cells reduces hypoxic-ischemic brain damage in rats by ameliorating the inflammatory response显示文摘Hypoxic-ischemic brain damage(HIBD)is a common cause of infant death.The purpose of our research was to explore the immunoregulatory mechanism of placenta-derived mesenchymal stem cells(PD-MSCs)in HIBD treatment.Seven-day-old rat pups were randomly divided into HIBD,PD-MSC,fibroblast,and control groups.Forty-eight hours after HIBD induction,cells at a density of 5×104 cells/10µl were injected into the cerebral tissue in the PD-MSC and fibroblast groups.The TNF-α,interleukin-17(IL-17),interferon-γ(IFN-γ),and IL-10 levels were detected through quantitative real-time polymerase chain reaction(RT-PCR)and enzyme-linked immunosorbent assay(ELISA).Regulatory T cell(Tregs)populations were detected through flow cytometry,and forkhead box P3(Foxp3)was measured through western blot analysis.Behavioral tests and gross and pathological examinations showed that PD-MSC treatment exerted significantly stronger neuroprotective effects than the other treatments.The expression levels of pro-inflammatory cytokines were substantially upregulated after HI injury.Compared with fibroblast treatment,PD-MSC treatment inhibited the production of pro-inflammatory cytokines and increased the production of IL-10 in the ischemic hemispheres and peripheral blood serum(all P<0.01).Flow cytometry results showed a notable increase in the number of Tregs within the spleen of the HIBD group.Moreover,the number of Tregs and the Foxp3 expression levels were higher in the PD-MSC treatment group than in the HIBD and fibroblast groups(all P<0.01).Our research suggests that the mechanism of PD-MSC treatment for HIBD partially involves inflammatory response suppression.Hongfang Ding Hui Zhang Huifang Ding Dong Li Xinhao Yi Xiaoxu Ma Ruijuan Li Mei Huang Xiuli Ju 2017Cellular & Molecular Immunology2017,14,8:10
4Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 5Dynamic changes of phosphatidylinositol and phosphatidylinositol 4-phosphate levels modulate H^(+)-ATPase and Na^(+)/H^(+) antiporter activities to maintain ion homeostasis in Arabidopsis under salt stress显示文摘Plant metabolites are dynamically modified and distributed in response to environmental changes.How-ever,it is poorly understood how metabolic change functions in plant stress responses.Maintaining ion ho-meostasis under salt stress requires coordinated activation of two types of central regulators:plasma membrane(PM)H^(+)-ATPase and Na^(+)/H^(+) antiporter.In this study,we used a bioassay-guided isolation approach to identify endogenous small molecules that affect PM H^(+)-ATPase and Na^(+)/H^(+) antiporter activities and identified phosphatidylinositol(PI),which inhibits PM H^(+)-ATPase activity under non-stress conditions in Arabidopsis by directly binding to the C terminus of the PM H^(+)-ATPase AHA2.Under salt stress,the phosphatidylinositol 4-phosphate-to-phosphatidylinositol(PI4P-to-PI)ratio increased,and PI4P bound and activated the PM Na^(+)/H^(+) antiporter.PI prefers binding to the inactive form of PM H^(+)-ATPase,while PI4P tends to bind to the active form of the Na^(+)/H^(+) antiporter.Consistent with this,pis1 mutants,with reduced levels of PI,displayed increased PM H^(+)-ATPase activity and salt stress toler-ance,while the pi4kβ1 mutant,with reduced levels of PI4P,displayed reduced PM Na^(+)/H^(+) antiporter activity and salt stress tolerance.Collectively,our results reveal that the dynamic change between PI and PI4P in response to salt stress in Arabidopsis is crucial for maintaining ion homeostasis to protect plants from un-favorable environmental conditions.Yongqing Yang Xiuli Han Liang Ma Yujiao Wu Xiao Liu Haiqi Fu Guoyong Liu Xiaoguang Lei Yan Guo 2021Molecular Plant2021,14,12:4
6Significant Inhibition of Porcine Epidemic Diarrhea Virus In Vitro by Remdesivir,Its Parent Nucleoside and β-D-N^(4)-hydroxycytidine显示文摘Porcine epidemic diarrhea(PED)caused by porcine epidemic diarrhea virus(PEDV)is widespread in the world.In recent years,the increased virulence of the virus due to viral variations,has caused great economic losses to the pig industry in many countries.It is always worthy to find effective therapeutic methods for PED.As an important class of antivirals,nucleoside drugs which target viral polymerases have been applied in treating human viral infections for half a century.Herein,we evaluated the anti-PEDV potential of three broad-spectrum antiviral nucleoside analogs,remdesivir(RDV),its parent nucleoside(RDV-N)andβ-D-N^(4)-hydroxycytidine(NHC).Among them,RDV-N was the most active agent in Vero E6 cells with EC_(50) of 0.31μmol/L,and more potent than RDV(EC_(50)=0.74μmol/L)and NHC(EC_(50)=1.17μmol/L).The activity of RDV-N was further confirmed using an indirect immuno-fluorescence assay.Moreover,RDV-N exhibited a good safety profile in cells and in mice.The high sequence similarity of the polymerase functional domains of PEDV with other five porcine coronaviruses indicated a broader antiviral spectrum for the three compounds.Generally,RDV-N is a promising broad-spectrum antiviral nucleoside,and it would be worthy to make some structural modifications to increse its oral bioavailability.Yuanchao Xie Xiaozhen Guo Tianwen Hu Daibao Wei Xiuli Ma Jiaqiang Wu Bing Huang Jingshan Shen 2021Virologica Sinica2021,36,5:3
7Nodal Promotes mir206 Expression to Control Convergence and Extension Movements During Zebrafish Gastrulation显示文摘Nodal,a member of the transforming growth factor b(TGF-b)superfamily,has been shown to play a role in mesendoderm induction and gastrulation movements.The activity of Nodal signaling can be modulated by microRNAs(miRNAs)as previously reported,but little is known about which miRNAs are regulated by Nodal during gastrulation.In the present study,we found that the expression of mir206,one of the most abundant miRNAs during zebrafish early embryo development,is regulated by Nodal signaling.Abrogation of Nodal signal activity results in defective convergence and extension(CE)movements,and these cell migration defects can be rescued by supplying an excess of mir206,suggesting that mir206 acts downstream of Nodal signaling to regulate CE movements.Furthermore,in mir206 morphants,the expression of cell adhesion molecule E-cadherin is significantly increased,while the key transcriptional repressor of E-cadherin,snail1a,is depressed.Our study uncovers a novel mechanism by which Nodal-regulated mir206 modulates gastrulation movements in connection with the Snail/E-cadherin pathway.Xiuli Liu Yuanqing Ma Congwei Zhang Shi Wei Yu Cao Qiang Wang 2013Journal of Genetics and Genomics2013,40,10:2
8Cyclic Shearing Deformation Behavior of Saturated Clays显示文摘The apparatus for static and dynamic universal triaxial and torsional shear soil testing is employed to perform stress-controlled cyclic single-direction torsional shear tests and two-direction coupled shear tests under unconsolidated-undrained conditions. Through a series of tests on saturated clay, the effects of initial shear stress and stress reversal on the clay’s strain-stress behavior are examined, and the behavior of pore water pressure is studied. The experimental results indicate that the patterns of stress-strain relations are distinctly influenced by the initial shear stress in the cyclic single-direction shear tests. When the initial shear stress is large and no stress reversal occurs, the predominant deformation behavior is characterized by an accumulative effect. When the initial shear stress is zero and symmetrical cyclic stress occurs, the predominant deformation behavior is characterized by a cyclic effect. The pore water pressure fluctuates around the confining pressure with the increase of cycle number. It seems that the fluctuating amplitude increases with the increase of the cyclic stress. But a buildup of pore water pressure does not occur. The de- formations of clay samples under the complex initial and the cyclic coupled stress conditions include the normal deviatoric deforma- tion and horizontal shear deformation, the average deformation and cyclic deformation. 13Study on Synthesis of Mesoporous M-MCM-48(M=Zr,Mg) and Its Activity for Isomerization of n-Heptane显示文摘The mesoporous materials supported zirconium(Zr-MCM-48) and magnesium(Mg-MCM-48) with three-dimensional pore structures and different Zr or Mg contents were prepared via the hydrothermal method, respectively. The prepared samples were characterized by means of X-ray diffraction(XRD), scanning electron microscope(SEM), transmission electron microscope(TEM), Fourier transform infrared spectroscopy(FT-IR), NH_3 temperature programmed desorption(NH_3-TPD) and N_2 adsorption-desorption. The XRD, TEM and N_2 adsorption-desorption results suggested that M-MCM-48(where M = Zr, Mg) samples still maintained typical cubic mesoporous framework of MCM-48, with slight decrease of specific surface areas and mesopore orders. The isomerization of n-heptane was carried out as a probe reaction at various factors including the zirconium or magnesium content, reaction time, reaction temperature and weight hourly space velocity(WHSV). Preliminary results demonstrated that the Mg-MCM-48 catalyst exhibited a higher catalytic activity with a maximum heptane conversion of 91.5% and a corresponding isomerization selectivity of 72.3% as compared to that of ZrMCM-48. However, the catalytic stability of Mg-MCM-48 for n-heptane isomerization was poorer than Zr-MCM-48, because the Mg-MCM-48 catalyst could maintain a relatively high catalytic activity for only 130 min, while the Zr-MCM-48 catalyst could maintain a relatively high catalytic activity for more than 190 min without any obvious decrease in performance.Wang Yingjun Dong Xiuli Ma Shoutao Wang Jian Li Xiaoqing Suo Yanhua Zhang Wei 2018China Petroleum Processing & Petrochemical Technology2018,20,3:2

16The Study of Visible Implant Elastomer(VIE)Tag on Small-sized Chinese Shrimp(Fenneropenaeus chinensis)显示文摘[Objective] The paper was to prove the feasibility of using visible implant elastomer( VIE) tags to mark Chinese shrimp of about 1 cm in length.[Method]In June 2014,2 856 Chinese shrimps with an average length of 1. 14 cm were tagged with red VIE. After temporary rearing for one day,2 321 tagged shrimps were cultured in the same pond with 60 000 untagged shrimps in this experiment. In predation test,spottedtail gobies( Synechogobius ommaturus) were selected as the predator,and the predation rate of spottedtail gobies on small-sized Chinese shrimps was measured. [Result]On September 21,2014,206 kg of Chinese shrimp was harvested in the pond. There were 389 tagged shrimps,with an average body weight of 16. 84 g,and an average length of 11. 15 cm. The untagged shrimps had an average body weight of 17. 08 g,and average length of 11. 23 cm. The results indicated that there was no significant difference in growth between the tagged and untagged shrimps. The survival rate of the tagged shrimps was 2. 7% lower than the untagged ones. Although the fluorescent tag dispersed and the position changed,the tag was still distinct both under ultraviolet light and to the naked eye. The predation test revealed that the fluorescent tag of shrimps did not increase the predation rate by spottedtail gobies. [Conclusion]Red VIE tagging is feasible in Chinese shrimps.
