1Natural variations in grain length 10(GL10)regulate rice grain size显示文摘Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly characterized.Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 plants.The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages.Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape.Penglin Zhan Shuaipeng Ma Zhili Xiao Fangping Li Xin Wei Shaojun Lin Xiaoling Wang Zhe Ji Yu Fu Jiahao Pan Mi Zhou Yue Liu Zengyuan Chang Lu Li Suhong Bu Zupei Liu Haitao Zhu Guifu Liu Guiquan Zhang Shaokui Wang 2022Journal of Genetics and Genomics2022,49,5:3

4Characteristics of TP53 germline variants and their correlations with Li-Fraumeni syndrome or Li-Fraumeni-like syndrome in Chinese tumor patients显示文摘Li-Fraumeni syndrome(LFS),a rare autosomal-dominant inheritance condition,is associated with a family cancer history as well as pathogenic/likely-pathogenic TP53 germline variants(P/LP TP53 GV).The current clinical methods for detecting LFS are limited.Here,we retrospectively investigate P/LP TP53 GV among Chinese cancer patients by next-generation sequencing and evaluate its relationship with a family cancer history.A total of 270 out of 19,226 cancer patients have TP53 GV,including 53 patients with P/LP TP53 GV.Patients with P/LP TP53 GV are mainly found in male with glioma,lung cancer or sarcoma.The median age of diagnosis for P/LP TP53 GV patients is significantly lower than that of non-P/LP TP53 GV patients(31-years vs.53-years;P<0.01).One LFS patient and 3 Li-Fraumeni-like syndrome(LFL)patients are among the 26 followed-up P/LP TP53 GV patients.Among 25 types of P/LP TP53 GV,the highest variant frequencies occurred at codon 175 and 248.p.M237 I,p.R158 H,p.C238 Y and p.C275 R,are firstly identified among the Chinese LFS/LFL patients.This study reports the(P/LP)TP53 GV characteristics of Chinese pan-cancer patients.These findings suggest analyzing the P/LP TP53 GV in cancer patients is an effective strategy for identifying cancer predisposition syndrome.Panwen Tian Xiaoyan Zhang Sheng Yang Yu Fang Hongling Yuan Wei Li Honglin Zhu Fangping Zhao Jinlei Ding Yunshu Zhu Sizhen Wang Guochen Sun Hongbin Ni Tonghui Ma Ting Lei 2022Journal of Genetics and Genomics2022,49,7:0
5Genome-wide investigation of defensin genes in peanut(Arachis hypogaea L.)reveals AhDef2.2 conferring resistance to bacterial wilt显示文摘Peanut(Arachis hypogea L.)bacterial wilt(BW)is a devastating disease caused by Ralstonia solanacearum that results in severe yield and quality losses.Plant defensins are short cysteine-rich peptides with antimicrobial activity.The role of defensin genes(Ah Def)in peanut is unclear.A genome-wide investigation of Ah Def genes was undertaken,and 12 identified Ah Def genes were classified into two groups containing the gamma-thionin domain formed by four disulfide pairs:Cys1-Cys8,Cys2-Cys5,Cys3-Cys6,and Cys4-Cys7.In silico analysis revealed that Ah Def genes showed highly conserved architectural features and contained cis-elements associated with phytohormone signaling and defense responses.A highly resistant cultivar,H108(R)and a susceptible accession,H107(S)were tested by R.solanacearum inoculation.H108(R)showed fewer symptoms than H107(S)owing to inhibition of bacterial reproduction and spread in the vascular bundles of roots and stems.In a transcriptomic expression profile,Ah Def genes,particularly Ah Def1.6 and Ah Def2.2,were up-regulated in H108(R)compared with H107(S)under R.solanacearum infection and phytohormone treatment.Subcellular localization showed that the Ah Def1.6 and Ah Def2.2 proteins were both expressed specifically on the plasma membrane.Overexpression of protein fusion Ah Def2.2-YFP in Nicotiana benthamiana and peanut leaves increased resistance to R.solanacearum,suggesting its role in response to BW infection.Ah Def2.2 may be valuable for peanut resistance breeding.Kai Zhao Rui Ren Xingli Ma Kunkun Zhao Chengxin Qu Di Cao Qian Ma Yingying Ma Fangping Gong Zhongfeng Li Xingguo Zhang Dongmei Yin 2022The Crop Journal2022,10,3:0
