| 2 | Anti-inflammatory and anti-thrombogenic effects of atorvastatin in acute ischemic stroke显示文摘Atorvastatin decreases inflammation and thrombogenesis in patients with carotid artery plaque. Atorvastatin is administered to lower lipid levels, but its anti-inflammatory and anti-thrombogenic effects remain unclear. Eighty-nine patients from northeastern China with acute ischemic stroke caused by large-artery atherosclerosis were randomly divided into the study and control groups. All patients received routine treatment, including antiplatelet therapy, circulatory support, and symp-tomatic treatment. The study group (n=43) also received daily atorvastatin 20 mg/d, and the control group (n=46) received daily placebo pills containing glucose. After 4 weeks, the levels of C-reactive protein, fibrinogen, and D-dimer were significantly lower in the study group than in the control group. Decreases in the levels of C-reactive protein, fibrinogen, and D-dimer were not associated with de-creases in the levels of triacylglycerol and low-density lipoprotein cholesterol. | 3 | Correlation between tongue manifestations and glucose, total cholesterol, and high-density lipoprotein cholesterol in patients with acute cerebral infarction显示文摘OBJECTIVE: To analyze the association between tongue manifestations and the levels of glucose (GLU), total cholesterol(TCH), and high-density lipoprotein cholesterol(HDL-C) in subjects with acute cerebral infarction.METHODS: Hospitalized patients with first unilateral cerebral infarction in the Neurological Department of Xuanwu Hospital were included and the correlation between tongue fur color, fur nature,and the levels of GLU,TCH, HDL-C were analyzed.RESULTS: HDL level in the thin fur group was higher than that in the thick fur group(P=0.02). The difference in the levels of GLU,TCH, and HDL-C among the groups was significant(P<0.05), classified in terms of slippery, moist, and dry fur. Further comparison between the groups by Student-Newman-Keuls test showed that GLU level in the dry fur group was the highest. | 4 | Protective effects of remote ischemic preconditioning in rat hindlimb on ischemia- reperfusion injury显示文摘Three cycles of remote ischemic pre-conditioning induced by temporarily occluding the bilateral femoral arteries (10 minutes) prior to 10 minutes of reperfusion were given once a day for 3 days before the animal received middle artery occlusion and reperfusion surgery. The results showed that brain infarct volume was significantly reduced after remote ischemic pre-conditioning. Scores in the forelimb placing test and the postural reflex test were significantly lower in rats having undergone remote ischemic pre-conditioning compared with those who did not receive remote ischemic pre-conditioning. Thus, neurological function was better in rats having undergone remote ischemic pre-conditioning compared with those who did not receive remote ischemic pre-conditioning. | 5 | Loss of Wip1 aggravates brain injury after ischaemia/reperfusion by overactivating microglia显示文摘Background and purpose The inflammatory response mediated by microglia/macrophages is closely related to cerebral ischaemia/reperfusion injury.Wild-type p53-induced protein phosphatase 1(Wip1),a serine/threonine phosphatase,is expressed in various tissues.A growing number of reports have suggested that Wip1 is a negative regulator of inflammation in peripheral tissue;however,its role in the central nervous system(CNS)remains unclear.This study aimed to clarify whether Wip1 can inhibit CNS inflammation by regulating microglia/macrophage functions after ischaemic injury.Methods A model of middle cerebral artery occlusion and reperfusion was established in mice.CNS inflammation was simulated by lipopolysaccharide treatment of primary microglia.Laser speckle imaging was used to monitor regional cerebral blood flow.Behavioural outcomes were assessed with a TreadScan gait analysis system.TTC staining was used to evaluate the infarct volume,and western blotting and immunofluorescence staining were applied to detect the phenotypical transformation of microglia.ELISA was performed to detect the levels of inflammatory factors.Results Wip1 expression was increased after ischaemia/reperfusion.Wip1-knockout(KO)mice displayed more severe brain injury than wild-type mice,as indicated by aggravated motor dysfunction,greater brain infarct volumes and higher expression of inflammatory cytokines(interleukin-6 and tumour necrosis factor alpha)in the brain.We also found that Wip1 depletion increased microglial/macrophage activation in both in vitro and in vivo models,which all showed activation of microglia/macrophages.Lentivirus-Ppm1d reversed the injury induced by Wip1-KO.Conclusions Our results suggest that Wip1 may inhibit neuroinflammation by inhibiting microglial/macrophage activation after brain ischaemia/reperfusion injury. | Feng Yan Xiang Cheng Ming Zhao Shenghui Gong Ying Han Liping Ding Di Wu Yumin Luo Wei Zuo Lingling Zhu Ming Fan Xunming Ji | 2021 | Stroke & Vascular Neurology2021,6,3: | 4 |
| 6 | A novel integrated non-targeted metabolomic analysis reveals significant metabolite variations between different lettuce (Lactuca sativa. L) varieties显示文摘Lettuce is an important leafy vegetable that represents a significant dietary source of antioxidants and bioactive compounds.However,the levels of metabolites in different lettuce cultivars are poorly characterized.In this study,we used combined GC×GC-TOF/MS and UPLC-IMS-QTOF/MS to detect and relatively quantify metabolites in 30 lettuce cultivars representing large genetic diversity.Comparison with online databases,the published literature,standards as well using collision cross-section values enabled putative identification of 171 metabolites.Sixteen of these 171 metabolites(including phenolic acid derivatives,glycosylated flavonoids,and one iridoid)were present at significantly different levels in leaf and head type lettuces,which suggested the significant metabolomic variations between the leaf and head types of lettuce are related to secondary metabolism.A combination of the results and metabolic network analysis techniques suggested that leaf and head type lettuces contain not only different levels of metabolites but also have significant variations in the corresponding associated metabolic networks.The novel lettuce metabolite library and novel non-targeted metabolomics strategy devised in this study could be used to further characterize metabolic variations between lettuce cultivars or other plants.Moreover,the findings of this study provide important insight into metabolic adaptations due to natural and human selection,which could stimulate further research to potentially improve lettuce quality,yield,and nutritional value. | Xiao Yang Shiwei Wei Bin Liu Doudou Guo Bangxiao Zheng Lei Feng Yumin Liu Francisco A.Tomás-Barberán Lijun Luo Danfeng Huang | 2018 | Horticulture Research2018,5,1: | 3 |
| 7 | lncRNA NEAT1 prompts autophagy and apoptosis in MPTP-induced Parkinson’s disease by impairing miR-374c-5p显示文摘Long non-coding RNAs(lncRNAs)play biological roles in brain disorder and neurodegenerative diseases.As the functions of lncRNA NEAT1 in Parkinson’s disease(PD)remain unknown,in the present study,we aimed to explore the roles and underlying molecular mechanisms of NEAT1 in PD.A PD mouse model induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)and a cell model of SH-SY5Y induced by N-methyl-4-phenylpyridinium(MPP+)were established.The ratio of tyrosine hydroxylase(TH+)cells was determined by immunofluorescence assay,and the behavioral changes in mice were observed using pole tests and rotarod tests.The cellular viability and apoptosis of SH-SY5Y were detected by MTT assay and flow cytometric analysis,respectively,and the number of autophagosomes was subsequently measured by transmission electron microscopy.High-performance liquid chromatography was performed to detect the content of dopamine,and a dual-luciferase reporter assay was used to clarify the target of NEAT1 simultaneously.The results demonstrated that the level of NEAT1 was upregulated in the MPTP-induced PD mice,dopamine neurons,and the SH-SY5Y cells treated with MPP+,whereas the level of miR-374c-5p was downregulated.NEAT1 level was positively correlated with MPP+in a concentration-dependent manner.NEAT1 inhibition efficiently facilitated cell proliferation but inhibited apoptosis and autophagy in the MPP+-treated SH-SY5Y cells.Additionally,silencing of NEAT1 increased the TH+rate of neurons and suppressed autophagy greatly in PD mice.As a possible target of NEAT1,miR-374c-5p could impact on the apoptosis and autophagy of the SH-SY5Y cells.NEAT1 inhibition upregulated the expression of miR-374c-5p,enhanced SH-SY5Y cell viability,and repressed autophagy and apoptosis in MPTP-induced PD mice.These findings indicated a potential therapeutic role of NEAT1 in treating PD. | Li Dong Yumin Zheng Lianbo Gao Xiaoguang Luo | 2021 | Acta Biochimica et Biophysica Sinica2021,53,7: | 3 |

| 16 | Delivery of an Rhs-family nuclease effector reveals direct penetration of the gram-positive cell envelope by a type VI secretion system in Acidovorax citrulli显示文摘The type VI secretion system(T6SS)is a double-tubular nanomachine widely found in gram-negative bacteria.Its spear-like Hcp tube is capable of penetrating a neighboring cell for cytosol-to-cytosol protein delivery.However,gram-positive bacteria have been considered impenetrable to such T6SS action.Here we report that the T6SS of a plant pathogen,Acidovorax citrulli(AC),could deliver an Rhsfamily nuclease effector RhsB to kill not only gram-negative but also gram-positive bacteria.Using bioinformatic,biochemical,and genetic assays,we systematically identified T6SS-secreted effectors and determined that RhsB is a crucial antibacterial effector.RhsB contains an N-terminal PAAR domain,a middle Rhs domain,and an unknown C-terminal domain.RhsB is subject to self-cleavage at both its N-and C-terminal domains and its secretion requires the upstream-encoded chaperone EagT2 and VgrG3.The toxic Cterminus of RhsB exhibits DNase activities and such toxicity is neutralized by either of the two downstream immunity proteins,RimB1 and RimB2.Deletion of rhsB significantly impairs the ability of killing Bacillus subtilis while ectopic expression of immunity proteins RimB1 or RimB2 confers protection.We demonstrate that the AC T6SS not only can effectively outcompete Escherichia coli and B.subtilis in planta but also is highly potent in killing other bacterial and fungal species.Collectively,these findings highlight the greatly expanded capabilities of T6SS in modulating microbiome compositions in complex environments. | Tong-Tong Pei Yumin Kan Zeng-Hang Wang Ming-Xuan Tang Hao Li Shuangquan Yan Yang Cui Hao-Yu Zheng Han Luo Xiaoye Liang Tao Dong | 2022 | mLife2022,1,1: | 1 |
