| 1 | Overexpression of a dominant-negative mutant of SIRT1 in mouse heart causes cardiomyocyte apoptosis and early-onset heart failure显示文摘SIRT1,a mammalian ortholog of yeast silent information regulator 2(Sir2),is an NAD+-dependent protein deacetylase that plays a critical role in the regulation of vascular function.The current study aims to investigate the functional significance of deacetylase activity of SIRT1 in heart.Here we show that the early postnatal hearts expressed the highest level of SIRT1deacetylase activity compared to adult and aged hearts.We generated transgenic mice with cardiac-specific expression of a dominant-negative form of the human SIRT1(SIRT1H363Y),which represses endogenous SIRT1 activity.The transgenic mice displayed dilated atrial and ventricular chambers,and died early in the postnatal period.Pathological,echocardiographic and molecular phenotype confirmed the presence of dilated cardiomyopathy.Terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling analysis revealed a greater abundance of apoptotic nuclei in the hearts of transgenic mice.Furthermore,we show that cardiomyocyte apoptosis caused by suppression of SIRT1 activity is,at least in part,due to increased p53acetylation and upregulated Bax expression.These results indicate that dominant negative form of SIRT1(SIRT1H363Y)overexpression in mouse hearts causes cardiomyocyte apoptosis and early-onset heart failure,suggesting a critical role of SIRT1 in preserving normal cardiac development during the early postnatal period. | MU WenLi ZHANG QingJun TANG XiaoQiang FU WenYan ZHENG Wei LU YunBiao LI HongLiang WEI YuSheng LI Li SHE ZhiGang CHEN HouZao LIU DePei | 2014 | Science China(Life Sciences)2014,57,9: | 12 |
| 2 | Abnormal subchondral bone remodeling and its association with articular cartilage degradation in knees of type 2 diabetes patients显示文摘Type 2 diabetes(T2 D) is associated with systemic abnormal bone remodeling and bone loss. Meanwhile,abnormal subchondral bone remodeling induces cartilage degradation, resulting in osteoarthritis(OA).Accordingly, we investigated alterations in subchondral bone remodeling, microstructure and strength in knees from T2 D patients and their association with cartilage degradation. Tibial plateaus were collected from knee OA patients undergoing total knee arthroplasty and divided into non-diabetic(n = 70) and diabetes(n = 51) groups. Tibial plateaus were also collected from cadaver donors(n = 20) and used as controls.Subchondral bone microstructure was assessed using micro-computed tomography. Bone strength was evaluated by micro-finite-element analysis. Cartilage degradation was estimated using histology. The expression of tartrate-resistant acidic phosphatase(TRAP), osterix, and osteocalcin were calculated using immunohistochemistry. Osteoarthritis Research Society International(OARSI) scores of lateral tibial plateau did not differ between non-diabetic and diabetes groups, while higher OARSI scores on medial side were detected in diabetes group. Lower bone volume fraction and trabecular number and higher structure model index were found on both sides in diabetes group. These microstructural alterations translated into lower elastic modulus in diabetes group. Moreover, diabetes group had a larger number of TRAP^+ osteoclasts and lower number of Osterix^+ osteoprogenitors and Osteocalcin^+ osteoblasts. T2 D knees are characterized by abnormal subchondral bone remodeling and microstructural and mechanical impairments, which were associated with exacerbated cartilage degradation. | 3 | The effect of interbeds on distribution of i ncremental oil displaced by a polymer flood显示文摘This paper discusses the effect of influ encing factors on the distribution of incremental oil displaced by a polymer flood(extra-displaced oil) using numerical reservoir simulation.These factors include the location,area and permeability of a thin low-permeability interbed,and the perforation location relative to the interbed.Simulation results show the locations from where the incremental oil was displaced by the polymer solution.The interbed position from the oil formation top affects the location of extra-displaced oil.The interbed area has a slight influence on the whole shape of extra-displaced oil.Larger interbed area leads to higher partition extent of extra-displaced oil.Higher vertical permeability of interbeds contributes to worse partition extent of extra-displaced oil and the partition effect disappears if the ratio of vertical to horizontal permeability is more than 0.05.The perforation location relative to the interbed affects polymer displacement efficiency,and also has a significant effect on the distribution of extra-displaced oil in polymer flooding. | Hou Jian Du Qingjun Lu Teng Zhou Kang Wang Rongrong | 2011 | Petroleum Science2011,8,2: | 3 |

| 8 | Structural, Optical and Electrical Properties of Ga Doped ZnO/Cu grid/Ga Doped ZnO Transparent Electrodes显示文摘Ga doped ZnO(GZO)/Cu grid/GZO transparent conductive electrode(TCE) structures were fabricated at room temperature(RT) by using electron beam evaporation(EBE) for the Cu grids and RF magnetron sputtering for the GZO layers. In this work, we investigated the electrical and optical characteristics of GZO/Cu grid/GZO multilayer electrode for thin film solar cells by using evaporated Cu grid and sputtered GZO thin films to enhance the optical transparency without significantly affecting their conductivity.The optical transmittance and sheet resistance of GZO/Cu grid/GZO multilayer are higher than those of GZO/Cu film/GZO multilayer independent of Cu grid separation distance and increase with increasing Cu grid separation distances. The calculation of both transmittance and sheet resistance of GZO/Cu grid/GZO multilayer was based on Cu filling factor correlated with the geometry of Cu grid. The calculated values for the transmittance and sheet resistance of the GZO/Cu grid/GZO multilayer were similar to the experimentally observed ones. The highest figure of merit ΦTC is 5.18 × 10-3Ω-1for the GZO/Cu grid/GZO multilayer with Cu grid separation distance of 1 mm was obtained, in this case, the transmittance and resistivity were 82.72% and 2.17 × 10-4Ω cm, respectively. | 13 | Peptide Functionalized Nanoplasmonic Sensor for Explosive Detection显示文摘In this study, a nanobiosensor for detecting explosives was developed, in which the peptide was synthesized with trinitrotoluene(TNT)-specific sequence and immobilized on nanodevice by Au–S covalent linkage, and the nanocup arrays were fabricated by nanoimprint and deposited with Au nanoparticles to generate localized surface plasmon resonance(LSPR). The device was used to monitor slight change from specific binding of 2,4,6-TNT to the peptide. With high refractive index sensing of ~10~4nm/RIU, the nanocup device can detect the binding of TNT at concentration as low as3.12×10^(-7)mg mL^(-1) by optical transmission spectrum modulated by LSPR. The nanosensor is also able to distinguish TNT from analogs of 2,4-dinitrotoluene and 3-nitrotoluene in the mixture with great selectivity. In this study, we proposed a simple light emitting diode(LED) based single-wavelength LSPR sensor modulated with bio-electron transfers for the detection of electroactive biomolecules. Indium tin oxide electrode loaded with nanocomposites of polyaniline coated gold nanorod was used as LSPR chip, and the applied electric potential was scanned at the LSPR chip for single-wavelength LSPR biosensing. Under the scanning of applied potentials, biological electron transfer of redox reaction was employed to demonstrate the bioelectronic modulation of single-wavelength LSPR for selective electroactive biomolecule detection. Without any additional recognition material, electroactive biomolecules uric acid and dopamine were detected directly with a sensitivity of 5.05 μmol/L and 7.11 μmol/L at their specific oxidation potentials, respectively.
