| 1 | USP19 suppresses inflammation and promotes M2-like macrophage polarization by manipulating NLRP3 function via autophagy显示文摘Macrophage polarization to proinflammatory M1-like or anti-inflammatory M2-like cells is critical to mount a host defense or repair tissue.The exact molecular mechanisms controlling this process are still elusive.Here,we report that ubiquitin-specific protease 19(USP19)acts as an anti-inflammatory switch that inhibits inflammatory responses and promotes M2-like macrophage polarization.USP19 inhibited NLRP3 inflammasome activation by increasing autophagy flux and decreasing the generation of mitochondrial reactive oxygen species.In addition,USP19 inhibited the proteasomal degradation of inflammasome-independent NLRP3 by cleaving its polyubiquitin chains.USP19-stabilized NLRP3 promoted M2-like macrophage polarization by direct association with interferon regulatory factor 4,thereby preventing its p62-mediated selective autophagic degradation.Consistent with these observations,compared to wild-type mice,Usp19−/−mice had decreased M2-like macrophage polarization and increased interleukin-1βsecretion,in response to alum and chitin injections.Thus,we have uncovered an unexpected mechanism by which USP19 switches the proinflammatory function of NLRP3 into an anti-inflammatory function,and suggest that USP19 is a potential therapeutic target for inflammatory interventions. | Tao Liu Liqiu Wang Puping Liang Xiaojuan Wang Yukun Liu Jing Cai Yuanchu She Dan Wang Zhi Wang Zhiyong Guo Samuel Bates Xiaojun Xia Junjiu Huang Jun Cui | 2021 | Cellular & Molecular Immunology2021,18,10: | 15 |
| 2 | The role of β18-β19 loop structure in insecticidal activity of Cry1Ac toxin from Bacillus thuringiensis显示文摘The β18-β19 loop in domain III of Cry1Ac toxin is unique among Bacillus thuringiensis Cry proteins. In this study, the role of the loop structure in insecticidal activity of Cry1Ac toxin was investigated. Alanine scanning mutations within the loop were initially generated and most mutants were over-expressed and reduced toxicity at different degrees, except mutant N546A that showed almost 2 times enhanced toxicity against Helicoverpa armigera larvae. Further mutagenic analysis of N546 revealed that a charged amino acid in this position would cause very unfavorable influence on insecticidal activity. In addition, the deletion of N546 led to protein instability because of destruction of the loop integrity. Besides, mutant W544F was much more toxic than W544Y, indicating that hydrophobic nature of the position was important for maintaining the stability and activity of Cry1Ac protein. | 4 | Metagenome analysis reveals potential microbial functions in topsoil of wheat-maize rotation system with five-year application of fertilizers显示文摘Fertilization mode affects soil quality and ecological health.The effects of four fertilization regimens on lignocellulose content,readily degradable carbohydrate decomposition,and potential microbial functions in the topsoil of a wheat-maize rotation system between 2012 and 2017 were investigated.The fertilization regimens of control(control NFNB),high chemical fertilizer(HCF),high biochar plus low chemical fertilizer(HBLCF),and biochar-based fertilizer(BBF)were compared on soil fundamental properties,microbial structure,and potential function in soil carbohydrate degradation based on metagenome analysis.The diversity of carbohydrate-active enzyme genes in the topsoil microbial consortia in the four trials was primarily distributed within the ten ecologically most dominant phyla.Application of BBF was associated with the lowest decline in total nitrogen and P2O5(2012-2017:6.5%and 28.1%,respectively)and the most effective carbohydrate decomposition(2015-2017:67.0%for cellulose and 59.9%for readily degradable carbohydrate).Carbohydrate transport and metabolism accounted for 6.0%of reads assigned functional classification under the BBF regimen.These findings reveal the ecologically functional diversity of topsoil microorganisms and suggest BBF application as a promising strategy for sustainable agriculture and beneficial to soil health. | Yujun Shen Lixin Zhao Haibo Meng Hongsheng Cheng Jingtao Ding Jiarui Wang Shanshan Dong Xi Zhang Liqiu Song Shengwei Zheng | 2019 | International Journal of Agricultural and Biological Engineering2019,12,6: | 3 |
| 5 | Synthesis and thermal conductivity of microfluidic copper nanofluids显示文摘A microfluidic chemical solution method is developed for the synthesizing Cu nanofluids. The method replaces batch-based macroreactors in the conventional chemical solution method by continuous-flow microfluidic microreactors, thereby enabling the synthesis of nanofluids with various microstructures. The Cu nanofluids synthesized by this technology show a better stability, remaining stable even after more than 100 h standing. | 8 | Prediction of Dynamic Wellbore Pressure in Gasified Fluid Drilling显示文摘The basis of designing gasified drilling is to understand the behavior of gas/liquid two-phase flow in the wellbore. The equations of mass and momentum conservation and equation of fluid flow in porous media were used to establish a dynamic model to predict wellbore pressure according to the study results of Ansari and Beggs-Brill on gas-liquid two-phase flow. The dynamic model was solved by the finite difference approach combined with the mechanistic steady state model. The mechanistic dynamic model was numerically implemented into a FORTRAN 90 computer program and could simulate the coupled flow of fluid in wellbore and reservoir. The dynamic model revealed the effects of wellhead back pressure and injection rate of gas/liquid on bottomhole pressure. Based on the single-phase-lagging heat conduction model, the condition for the occurrence of thermal vibration of heat conduction is established. In order to resolve the contradiction that the thermal vibration violates the second law of thermodynamics, the extended irreversible thermodynamics is improved and a generalized entropy definition is introduced.
