1Early Serum HBsAg Kinetics as Predictor of HBsAg Loss in Patients with HBeAg-Negative Chronic Hepatitis B after Treatment with Pegylated Interferonα-2a显示文摘Hepatitis B surface antigen(HBsAg)loss is an ideal treatment endpoint for patients with chronic hepatitis B(CHB).We investigated the predictive value of on-treatment HBsAg levels for HBsAg loss in hepatitis B e antigen(HBe Ag)-negative CHB patients who received 120-week PEG-IFNα-2a treatment.Serum HBV DNA,HBsAg,and anti-HBs levels were assayed at baseline and every 3 months during the treatment.Of 81 patients,12 achieved HBsAg loss,20 achieved HBsAg\100 IU/mL,and 49 maintained HBs Ag C 100 IU/mL.HBsAg loss rate was only 3.7%at 48 weeks,while it reached to 11.1%and 14.8%after treatment of 96 weeks and 120 weeks.The cutoff HBs Ag levels at 12 weeks predicting HBsAg loss at 96 weeks and 120 weeks of treatment were 400 IU/mL and 750 IU/mL,with AUC 0.725 and 0.722,positive predictive value(PPV)29.41%and 30.56%,and negative predictive value(NPV)93.75%and 97.78%,respectively.The cutoff HBsAg levels at 24 weeks predicting HBsAg loss at 96 weeks and 120 weeks of treatment were 174 IU/m L and 236 IU/mL respectively,with AUC 0.925 and 0.922,PPV 40.0%and 46.15%,and both NPV 100%.The predictive ability of the cutoff HBsAg levels at 24 weeks was better than that at 12 weeks for HBs Ag loss at either 96 or 120 weeks(χ~2=3.880,P=0.049 andχ~2=4.412,P=0.036).These results indicate that extended therapy is critical to HBsAg loss in HBe Ag-negative CHB patients during PEG-IFN treatment,and the HBsAg level at 24 weeks can be used to predict HBsAg loss during tailoring PEG-IFN therapy.Minghui Li Lu Zhang Yao Lu Qiqi Chen Huihui Lu Fangfang Sun Zhan Zeng Gang Wan Linqing Zhao Yao Xie 2021Virologica Sinica2021,36,2:15
2An A-D-A′-D-A type unfused nonfullerene acceptor for organic solar cells with approaching 14%efficiency显示文摘In recent years,power conversion efficiency(PCE)of organic solar cells(OSCs)has made significant improvement.A large number of studies were reported to achieve high PCEs through exploring new active layer materials,especially the high efficiency fused ring acceptors(FRAs).Compared with FRAs,another type of so-called unfused-ring acceptors(UFAs),possessing some advantages such as simple synthesis and low cost,have attracted a lot of attention.Herein,a new UFA BTzO-4F,incorporating with a benzotriazole moiety and S···O intramolecular noncovalent interactions,has been successfully synthesized.The photovoltaic device based on PBDB-T:BTzO-4F achieved a record PCE of 13.8%for UFAs,which indicates that introducing the benzotriazole moiety is an effective strategy for high quality acceptors.Thus,these findings of this work demonstrate the great potential of UFAs for high performance OSCs.Xingzheng Liu Yanan Wei Xin Zhang Linqing Qin Zhixiang Wei Hui Huang 2021Science China Chemistry2021,64,2:7
3Application of Human Adenovirus Genotyping by Phylogenetic Analysis in an Outbreak to Identify Nosocomial Infection显示文摘Nosocomial infections are common in pediatric patients and can be fatal in infants and immunocompromised patients. In September 2018, a high positive rate of human adenovirus HAdV was occurred among hospitalized children in the Children's Hospital Affiliated to the Capital Institute of Paediatrics in Beijing. To investigate whether this outbreak of HAdV was related to nosocomial infections or the result of community infections, we collected respiratory specimens from patients with acute respiratory infections in a respiratory ward during June to December 2018, and screened for respiratory viruses. Among 1,840 cases included, 95(5.2%, 95/1840) were positive for HAdV and 81 were genotyped based on phylogenetic analysis, including seven as HAdV-1(8.6%), 30 HAdV-3(37.0%), two HAdV-6(2.5%), and 42 HAdV-7(51.9%). More HAdV-positive samples were collected in August(4.7%, 12/255), September(15.0%, 41/274) and October(6.9%, 17/247), with a peak in September 2018. By combining the results of HAdV phylogenetic analysis with clinical data of patients, there were 77 cases(4.2%, 77/1840;81.1%, 77/95) excluded from nosocomial infections, eight cases representing possible infections transmitted by visitors or attending parents, three cases without sequences that might have been due to infection transmitted by roommates positive for HAdV, one case of a roommate without an HAdV sequence, and six cases that shared highly homologous sequences with those of their roommates, for which nosocomial infections might be considered. 7Sulfur vs. tellurium: the heteroatom effects on the nonfullerene acceptors显示文摘The effect of chalcogen heteroatom variation on donor materials has been systematically investigated. However, this effect on acceptors has rarely been explored. Herein, nonfullerene acceptors BFPSP and BFPTP were reported by simply changing the chalcogen atoms from S to Te. The differences between BFPSP and BFPTP in light absorption, energy levels, excited-state lifetimes, energy loss, charge mobilities, morphology, and photovoltaic properties were systematically investigated to understand the heteroatom effects. 8Distance-controllable and direction-steerable opto-conveyor for targeting delivery显示文摘Opto-conveyors have attracted widespread interest in various fields because of their non-invasive and non-contact delivery of micro/nanoparticles.However,the flexible control of the delivery distance and the dynamic steering of the delivery direction,although very desirable in all-optical manipulation,have not yet been achieved by optoconveyors.Here,using a simple and cost-effective scheme of an elliptically focused laser beam obliquely irradiated on a substrate,a direction-steerable and distance-controllable opto-conveyor for the targeting delivery of microparticles is implemented.Theoretically,in the proposed scheme of the opto-conveyor,the transverse and longitudinal resultant forces of the optical gradient force and the optical scattering force result in the transverse confinement and the longitudinal transportation of microparticles,respectively.In this study,it is experimentally shown that the proposed opto-conveyor is capable of realizing the targeting delivery for microparticles.Additionally,the delivery distance of microparticles can be flexibly and precisely controlled by simply adjusting the irradiation time.By simply rotating the cylindrical lens,the proposed opto-conveyor is capable of steering the delivery direction flexibly within a large range of azimuthal angles,from-75°to 75°.This study also successfully demonstrated the real-time dynamic steering of the delivery direction from-45°to 45°with the dynamical rotation of the cylindrical lens.Owing to its simplicity,flexibility,and controllability,the proposed method is capable of creating new opportunities in bioassays as well as in drug delivery.Zhen Che Wenguo Zhu Yaoming Huang Yu Zhang Linqing Zhuo Pengpeng Fan Zhibin Li Huadan Zheng Wenjin Long Wentao Qiu Yunhan Luo Jun Zhang Jinghua Ge Jianhui Yu Zhe Chen 2020Photonics Research2020,8,7:1
9Consolidation treatment needed for sustained HBsAg-negative response induced by interferon-alpha in HBeAg positive chronic hepatitis B patients显示文摘Hepatitis B surface antigen(HBsAg)clearance is considered as functional cure in patients with chronic hepatitis B(CHB).This study aimed to assess the durability of HBsAg clearance achieved by interferon-based therapies in patients with CHB who were originally positive for hepatitis B envelope antigen(HBeAg).In this prospective study,HBeAg-positive CHB patients with confirmed HBsAg loss under interferon-based therapies were enrolled within 12 weeks from end of treatment and followed up for 48 weeks.Virological markers,biochemical indicators,and liver imaging examinations were observed every 3–6 months.Sustained functional cure was analysed as primary outcome.Factor associated with sustained HBsAg loss or reversion was also investigated.The rate of HBsAg loss sustainability was 91.8%(212/231).Patients receiving consolidation treatment for 12–24weeks or≥24 weeks had higher rates of sustained HBsAg negativity than those receiving consolidation treatment for<12 weeks(98.3%and 91.2%vs.86.7%,P=0.068),and the former groups had significantly higher anti-HBs levels than the later(P<0.05).The cumulative incidence of HBsAg reversion and HBV DNA reversion was 8.2%and 3.9%,respectively.Consolidation treatment of≥12 weeks[odd ratio(OR)3.318,95%confidence interval(CI)1.077–10.224,P=0.037)was a predictor of sustained functional cure,and HBeAg-positivity at cessation of treatment(OR 12.271,95%CI 1.076–139.919,P=0.043)was a predictor of HBsAg reversion.Interferon-alpha induced functional cure was durable and a consolidation treatment of≥12–24 weeks was needed after HBsAg loss in HBeAg-positive CHB patients.Minghui Li Fangfang Sun Xiaoyue Bi Yanjie Lin Liu Yang Yao Lu Lu Zhang Gang Wan Wei Yi Linqing Zhao Yao Xie 2022Virologica Sinica2022,37,3:1
10Relationship Between F_1,F_2 Hybrid Yield, Heterosis and Genetic Distance Measured by Molecular Markers and Parent Performance in Cotton显示文摘Genetic distance among 36 cotton cultivars measured by molecular markers of RAPDs, ISSRs,and SSRs was from 0.0701 to 0.4255 with the mean of 0.2844, and from 2.18 to 12.60 with the mean of 7.04 based on the genotype performance in two-year field experiments, which has a significant positive correlation (r = 0.3350). The correlative coefficients for boll number per plant, boll weight, yield per plant, lint percent and lint yield per plant were 0. 8035,0. 8877,0. 7135,0. 9640 and 0. 8956 between F1 and F2 hybrid performance assessed by three-environment field experiments, respectively. The mean of F1 and F2 hybrid heterosis of yield per plant and lint yield per plant were 13.62%, 16.31%, 7.90% and 9.02%, and the correlative coefficients between them were 0.3689 and 0.3787, respectively. 11Gravitational wave astronomy: the current status显示文摘In the centenary year of Einstein's General Theory of Relativity, this paper reviews the current status of gravitational wave astronomy across a spectrum which stretches from attohertz to kilohertz frequencies. Sect. 1 of this paper reviews the historical development of gravitational wave astronomy from Einstein's first prediction to our current understanding the spectrum. It is shown that detection of signals in the audio frequency spectrum can be expected very soon, and that a north-south pair of next generation detectors would provide large scientific benefits. Sect. 2 reviews the theory of gravitational waves and the principles of detection using laser interferometry. The state of the art Advanced LIGO detectors are then described. These detectors have a high chance of detecting the first events in the near future. Sect. 3 reviews the KAGRA detector currently under development in Japan,which will be the first laser interferometer detector to use cryogenic test masses. Sect. 4 of this paper reviews gravitational wave detection in the nanohertz frequency band using the technique of pulsar timing. Sect. 5 reviews the status of gravitational wave detection in the attohertz frequency band, detectable in the polarisation of the cosmic microwave background, and discusses the prospects for detection of primordial waves from the big bang. The techniques described in sects. 1–5 have already placed significant limits on the strength of gravitational wave sources. Sects. 6 and 7 review ambitious plans for future space based gravitational wave detectors in the millihertz frequency band. 12Variant rs8400 enhances ALKBH5 expression through disrupting miR-186 binding and promotes neuroblastoma progression显示文摘Objective:AlkB homolog 5(ALKBH5)has been proven to be closely related to tumors.However,the role and molecular mechanism of ALKBH5 in neuroblastomas have rarely been reported.Methods:The potential functional single-nucleotide polymorphisms(SNPs)in ALKBH5 were identified by National Center for Biotechnology Information(NCBI)dbSNP screening and SNPinfo software.TaqMan probes were used for genotyping.A multiple logistic regression model was used to evaluate the effects of different SNP loci on the risk of neuroblastoma.The expression of ALKBH5 in neuroblastoma was evaluated by Western blotting and immunohistochemistry(IHC).Cell counting kit-8(CCK-8),plate colony formation and 5-ethynyl-2'-deoxyuridine(EdU)incorporation assays were used to evaluate cell proliferation.Wound healing and Transwell assays were used to compare cell migration and invasion.Thermodynamic modelling was performed to predict the ability of miRNAs to bind to ALKBH5 with the rs8400 G/A polymorphism.RNA sequencing,N6-methyladenosine(mA)sequencing,mA methylated RNA immunoprecipitation(MeRIP)and a luciferase assay were used to identify the targeting effect of ALKBH5 on SPP1.Results:ALKBH5 was highly expressed in neuroblastoma.Knocking down ALKBH5 inhibited the proliferation,migration and invasion of cancer cells.miR-186-3p negatively regulates the expression of ALKBH5,and this ability is affected by the rs8400 polymorphism.When the G nucleotide was mutated to A,the ability of miR-186-3p to bind to the 3'-UTR of ALKBH5 decreased,resulting in upregulation of ALKBH5.SPPI is the downstream target gene of the ALKBH5 oncogene.Knocking down SPP1 partially restored the inhibitory effect of ALKBH5 downregulation on neuroblastoma.Downregulation of ALKBH5 can improve the therapeutic efficacy of carboplatin and etoposide in neuroblastoma.Conclusions:We first found that the rs8400 G>A polymorphism in the m6A demethylase-encoding gene ALKBH5 increases neuroblastoma susceptibility and determines the related mechanisms.The aberrant regulation of ALKBH5 by miR-186-3p caused by this genetic variation in ALKBH5 promotes the occurrence and development of neuroblastoma through the ALKBH5-SPP1 axis.Qian Guan Huiran Lin Wenfeng Hua Lei Lin Jiabin Liu Linqing Deng Jiao Zhang Jiwen Cheng Zhonghua Yang Yong Li Jun Bian Haixia Zhou Suhong Li Li Li Lei Miao Huimin Xia Jing He Zhenjian Zhuo 2023Chinese Journal of Cancer Research2023,35,2:0

14Epidemiological Characteristics of Human Parainfluenza Viruses Infections-China,2019-2023显示文摘Introduction:A retrospective study based on sentinel surveillance was conducted in 10 provinciallevel administrative divisions(PLADs)in China to enhance the understanding of the epidemiological characteristics of human parainfluenza viruses(HPIVs).Methods:From January 2019 to June 2023,respiratory specimens were collected from individuals with acute respiratory infections(ARIs)and screened for four HPIVs serotypes and other common respiratory viruses using multiplex real-time polymerase chain reaction(PCR).This study analyzed the association of HPIVs infections with seasonal patterns,geographical distribution,demographic profiles,clinical features,and co-infection status.Results:During the study period,a total of 12,866 ARIs were included.The overall detection rate of HPIVs was 6.15%,varying from 5.04%in 2022 to 9.70%in 2020.The median age of HPIVs-infected patients was 3 years.HPIV2 was more prevalent among individuals aged 5–17 years(42.57%),while HPIV4 was more common in those over 65 years(12.24%).HPIV3(54.16%)and HPIV1(27.18%)were the predominant serotypes,and their prevalence exhibited significant seasonal fluctuations postcoronavirus disease 2019(COVID-19)pandemic.The peak of HPIV3 shifted three months later in 2020 compared to 2019 and returned to a summer peak thereafter.Two peaks of HPIV1 were observed in 2021 following the peak of HPIV3.Additionally,coinfections were frequent in HPIVs cases(overall rate:22.12%),with human rhinovirus being the most common co-infecting virus.Conclusions:The prevalence of HPIVs in China was predominantly due to HPIV3 and HPIV1,and their seasonal patterns were altered by pandemic restrictions.Hence,continuous surveillance of HPIVs is essential.Yixuan Gao Yingwei Ma Daxing Feng Feng Zhang Biao Wang Xiaoqing Liu Bing Zhu Hui Xie Linqing Zhao Xiaoru Long Ying Chen Bing Wang Jie Jiang Zhen Zhu Yan Zhang Aili Cui Baicheng Xia Naiying Mao 2024China CDC weekly2024,6,12:0
15Interfacial Characteristics and Formation Mechanisms of Copper–steel Multimaterial Structures Fabricated via Laser Powder Bed Fusion Using Different Building Strategies显示文摘Laser powder bed fusion(LPBF)is an innovative method for manufacturing multimaterial components with high geometrical resolution.The LPBF-printing sequences of materials may be diverse in the actual design and application of multimaterial components.In this study,multimaterial copper(CuSn10)–steel(316 L)structures are printed using different building strategies(printing 316 L on CuSn10 and printing CuSn10 on 316 L)via LPBF,and the characteristics of two interfaces(the 316 L/CuSn10 or“L/C”and CuSn10/316 L or“C/L”interfaces)are investigated.Subsequently,the interfacial melting mode and formation mechanisms are discussed.At the L/C interface,the keyhole melting mode induced by the high volumetric energy density(EL/C=319.4 J/mm3)results in a large penetration depth in the pre-solidified layer and enhances laser energy absorption,thus promoting the extensive migration of materials and intense intermixing of elements to form a wide diffusion zone(∼400μm).At the C/L interface,the conduction mode induced by the low volumetric energy density(EC/L=74.1 J/mm3)results in a narrow diffusion zone(∼160μm).The interfacial defects observed are primarily cracks and pores.More cracks appeared at the C/L interface,which is attributable to the weak bonding strength of the narrow diffusion zone.This study provides guidance and reference for the design and manufacturing of multimaterial components via LPBF using different building strategies.Linqing Liu Di Wang Guowei Deng Yongqiang Yang Jie Chen Jinrong Tang Yonggang Wang Yang Liu Xusheng Yang Yicha Zhang 2022Chinese Journal of Mechanical Engineering(Additive Manufacturing Frontiers)2022,1,3:0
16simple and effective aerosol pathogen disinfection test for a flowing air disinfector显示文摘Aerosol transmission is an important disease transmission route and has been especially pertinent to hospital and biosafety laboratories during the SARS-CoV-2 pandemic.The thermal resistance of airborne SARS-CoV-2 is lower than that of Bacillus subtilis spores,which are often used to test the effectiveness of SARS-CoV-2 and other pathogen disinfection methods.Herein,we propose a new method to test the disinfection ability of a flowing air disinfector(a digital electromagnetic induction air heater)using B.subtilis spores.The study provides an alternative air disinfection test method.The new test system combined an aerosol generator and a respiratory filter designed in-house and could effectively recover spores on the filter membrane at the air outlet after passing through the flowing air disinfector.The total number of bacterial spores used in the test was within the range of 5×10^(5)–5×10^(6)colony-forming units(CFUs)specified in the technical standard for disinfection.The calculation was based on the calculation method in Air Disinfection Effect Appraisal Test in Technical Standard for Disinfection(2002 Edition).At an air speed of 3.5 m/s,we used a digital electromagnetic induction air heater to disinfect flowing air containing 4.100×10^(6)CFUs of B.subtilis spores and determined that the minimum disinfection temperature was 350℃for a killing rate of 99.99%.At 400℃,additional experiments using higher spore concentrations(4.700×10^(6)±1.871×10^(5)CFU)and a higher airspeed(4 m/s)showed that the killing rate remained>99.99%.B.subtilis spores,as a biological indicator for testing the efficiency of dry-heat sterilization,were killed by the high temperatures used in this system.The proposed method used to test the flowing air disinfector is simple,stable,and effective.This study provides a reference for the development of test systems that can assess the disinfection ability of flowing air disinfectors.Xuling Liu Zhiran Qin Linqing Wang Xiaoting Xie Yifang Fu Jianhai Yu Zuxin Liang Xiaoen He Jingshu Li Hong Dai Jinxiu Yao Qinghua Wu Weiwei Xiao Li Zhu Chengsong Wan Bao Zhang Wei Zhao 2023Journal of Biosafety and Biosecurity2023,5,1:0
17More common RNAemia in the early stage of severe SARS-CoV-2 BF.7.14 infections in pediatric patients显示文摘The risk factors of severe infections in children during the severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)outbreak in Beijing remain elusive.SARS‐CoV‐2‐positive children admitted to the intensive care unit(ICU)with collected plasma specimens were enrolled and screened for common pathogens using capillary electrophoresis‐based multiplex PCR from December 12,2022,to January 24,2023.The SARS‐CoV‐2 subvariants were identified using next‐generation sequencing.Plasma was positive for two(positive;P),one(suspicious;S),or no(negative;N)SARS‐CoV‐2 genes were classified as plasmatic RNA‐positive(RNAemia;P+S)or without RNAemia(N).Clinical and laboratory data of the enrolled cases were then collected and analyzed.The 34 enrolled children included 26 males and 24 younger than three years.All were negative for other respiratory pathogens.BF.7.14(18/29)was the predominant subvariant.Viral loads in respiratory specimens,hours from symptom onset to the first respiratory specimen collection(time‐variable),with comorbidities and BF.7.14 and BA.5.2 distributions were significantly different in P vs.N and RNAemia vs.without RNAemia group.Among most cases,the T lymphocyte ratios decreased,while the cytokine level and the B lymphocyte ratio increased.The time variables were 2.22±2.05 and 4.00±2.49 days in BF.7.14 and BA.5.2 infections,respectively.In conclusion,SARS‐CoV‐2 was more likely to cause severe infections among males aged≤3 years old with comorbidities during the SARS‐CoV‐2 outbreak in Beijing,while RNAemia is more common in children at the early stage of severe BF.7.14 infections,and most had high cytokine levels and B‐cell activation.Yu Sun Runan Zhu Yang Pan Ri De Shuang Liu Liping Jia Bing Lv Xiaoyun Li Dongmei Chen Yao Yao Dong Qu Daitao Zhang Linqing Zhao 2024Biosafety and Health2024,6,1:0
18The wrinkling and buckling of graphene induced by nanotwinned copper matrix:A molecular dynamics study显示文摘The three-dimensional(3D)graphene-based materials have raised significant interest due to excellent catalytic performance and unique electronic properties,while the preparation of uniform and stable 3D graphene structures remains a challenge.In this paper,using molecular dynamics simulations,we found that the nanotwinned copper(nt-Cu)matrix with small twin spacing can induce the wave-shaped wrinkling and sawtooth-shaped buckling graphene structures under uniaxial compression.The nt-Cu matrix possesses a symmetrical lattice structure for the lattice rotation with the dislocation annihilation,resulting in the transition of sandwiched graphene from 2D to 3D structures with good uniformity.The newly formed twin boundaries(TBs)in the nt-Cu matrix improve the resistance of graphene against the out-of-plane deformation so that graphene can maintain a stable wrinkling or buckling morphology in a wide strain range.These 3D texturing structures show great flexibility and their micro parameters can be controlled by applying different compressive strains.Furthermore,we propose a simple sliding method for decoupling graphene from the nt-Cu matrix without any damage.This work provides a novel strategy to induce and transfer the uniform wrinkling and buckling of graphene,which may expand the application of graphene in energy storage and catalysts.Che Zhang Cheng Lu Linqing Pei Jiaqing Li Rui Wang 2021Nano Materials Science2021,3,1:0
19A novel peptide-based probe^(99m)Tc-PEG_(6)-RD-PDP2 for the molecular imaging of tumor PD-L2 expression显示文摘Tumor-related PD-L2 expression is associated with the clinical efficacy of PD-1/PD-L1 blockade therapy.PD-L2-specific imaging can help selecting patients for appropriate immunotherapy.In this study,a PD-L2-targeting peptide(PDP2)was screened by the one-bead one-compound combinatorial library approach.Using the retro-inverso D-peptide of PDP2(RD-PDP2)and PEGylation strategies,we developed a novel Tc-99m-labeled PD-L2-targeting peptide as a SPECT tracer(^(99m)Tc-PEG_(6)-RD-PDP2)for imaging of tumor PD-L2 expression.The radiolabeling yield of ^(99m)Tc-PEG_(6)-RD-PDP2 was greater than 95%by the standard HYNIC/tricine/TPPTS labeling procedure.^(99m)Tc-PEG_(6)-RD-PDP2 displayed high PD-L2-binding specificity both in vitro and in vivo.SPECT/CT imaging with^(99m)Tc-PEG_(6)-RD-PDP2 showed that the A549-PD-L2tumors were clearly visualized,whereas the signals in PD-L2-negative A549 tumors were much lower.In vivo blocking study suggested that the tumor uptake of^(99m)Tc-PEG_(6)-RD-PDP2 was PD-L2 specifically mediated.^(99m)Tc-PEG_(6)-RD-PDP2 is a promising SPECT probe for the non-invasive imaging of tumor PD-L2expression and has a great potential in guiding the anti-PD-1 or anti-PD-L1 immunotherapy of cancer.Qi Luo Yunwei Zhang Zihua Wang Yining Sun Linqing Shi Yue Yu Jiyun Shi Zhiyuan Hu Fan Wang 2022Chinese Chemical Letters2022,33,7:0
